Oxygen Transfer Rate Coefficient of Membrane Aeration Bioreactor for Vero Cell Culture

  • Oxygen is a key substrate in animal cell metabolism and its consumption is thus a parameter of great interest for monitoring and control in animal cell culture bioreactor. The use of a gas-permeable membrane offered the possibility to provide the required quantity of oxygen into the culture. while avoiding problems of foaming or shear damage generally linked to sparging. For determining the optimum DO control strategy of this gas-permeable membrane aeration bioreactor, the oxygen transfer rate coefficient was measured with varying $N_2$ ratio in inlet air. The results showed that an increasing mass flow rate of nitrogen reduced the $K_La$ value. and 5% nitrogen in air did not result in any oxygen limitation.

Oxygen Permeability Characteristics of the Multi-Cathode Type Dissolved Oxygen Sensor Using the Low Noise Measuring Circuit (저잡음화 계측회로에 의한 다음극형 용존산소센서의 산소투과특성)

  • An evaluation method for oxygen permeable characteristics of the membrane covering to each cathode of multiple cathode - single anode type dissolved oxygen sensor, which has high reproducibility and is capable of measuring multiple components in solutions. For this purpose, a measuring circuit for the multiple cathode type DO sensor was designed to lower the noise signal by adapting a digital LPF to readout the sensor output accurately. Digital LPF is designed by setting up the transfer function to set the cutoff frequency to 10Hz, and the transfer function is programmed by C language, and then the filtering characteristics are evaluated with the simulation and experiments. Using this LPF added measuring circuit for the multiple cathode type DO sensor, we have obtained the calibration factor for each cathode to calibrate the variation of the output signals. The calibration factor was obtained by measuring the sensor output signal followed by oxygen partial pressure, using the same oxygen permeable membrane at each cathode of the multiple cathode type DO sensor.

  • The existing permeability measurements based on pressure differential between the polymer membrane that is permeable to measure the amount of oxygen used, but these methods must be kept in a vacuum, and the measurement of the membrane with low permeability in the membrane is too time consuming. In recent years by using electrochemical method polymer membrane currents caused by the amount of oxygen is a measure of how much is used. In this study, apparatus consisting of one anode and six cathodes for multi-oxygen permeability tester used the same number of membranes produced by electrochemical oxygen permeation characteristics. In this study, one silver/silver chloride anode electrochemical method with a hexagonal sensor to put various kinds of polymer membranes with the six oxygen permeability for simultaneous measurement in real-time systems. Six cathodes (Pt), and one of the coil-shaped anode (Ag/AgCl) to form a hexagonal one of the polarographic oxygen sensor in a single measurement system by six sensors. Each sensor for making hexagonal specificity of the sensor to compensate for the conditions obtained in a pure nitrogen gas and pure oxygen gas conditions. With this study, self-developed hexagonal sensor capable of measuring sensors and oxygen permeability tester, for a multi-six different oxygen permeability characteristics of the membrane measured at the same time.

  • The detection range of this galvanic oxygen sensor for oxygen concentration was 0.0% to 20.0%. By using gold or silver as cathode, reproducible response time and sensitivity to change of oxygen concentration were observed. The anode was Pb-Sn-Ca alloy. Oxygen selective permeable membrane was hydrophobic and porous Teflon film. The effect of the membranes varying in thickness have been studied on the temperature($10{\sim}50^{\circ}C$) and relative humidity(R. H 0~99%). Lead acetate buffer solution as the electrolyte has shown a high output voltage and longer life.

  • Polyimide membrane system was designed for manufacturing nitrogen-enriched gas, and basic technical data was suggested for appling this system to controlled atmosphere storage. The permeability characteristics of pure oxygen and nitrogen could be explained by dual-mode sorption model. There was substantial decrease in the permeation rates of oxygen, which is the more permeable gas, through the polyimide membrane due to the presence of nitrogen in comparison with pure oxygen. However, the permeation rates of nitrogen was increased by the presence of oxygen. The ideal separation factor was in the range of 5 to 6 in the range of temperature and pressure difference studied, and the separation factor of air was lower than the ideal separation factor. The increase of ideal separation factor with increasing temperature is due to the fact that the activation energy for oxygen is larger than that for nitrogen. Nitrogen concentration decreased rapidly with increasing product recovery, and it was found that this is a major operating factor to obtain nitrogen concentration required for controlled atmosphere storage. A relation equation, by which nitrogen concentration in storehouse can be predicted, was suggested under the establishment of a hypothetical model for controlled atmosphere storage process using polyimide membrane system.

  • The BOD (biochemical oxygen demand) sensor was fabricated by covering a dissolved oxygen (DO) probe with a microbe-impregnated membrane and a dialysis membrane. Various microorganisms isolated from the soils, water and activated sludge have been evaluated for measuring biochemical oxygen demand (BOD); Bacillus species HN24 and HN93 were selected as they exhibited rapid oxygen consumption and fast recovery. Improved BOD sensor could be prepared by using mixed microbes (Bacillus subtilis, Bacillus sp. HN24 and Bacillus sp. NH93) and silicon rubber gas-permeable membrane for DO probe, and by bubbling 50% $O_2$ ($N_2$ valence) through background buffer solution. This system exhibited excellent analytical performance resulting in good linearity ($r^2=0.9986$) from 0 to 100 mg/L level of BOD.

  • This paper presents a microfabricated Clark-type sensor which exactly can measure dissolved oxygen in the cell containing solution. We designed, fabricated, and characterized a microfabircated Clark-type oxygen sensor for measuring dissolved oxygen. The microfabricated oxygen sensor consists of 3-electrodes on a glass substrate, a FEP (Fluorinated ethylene propylene) oxygen-permeable membrane, and PDMS (Polydimethylsiloxane) reservoir for storing sample solution. Thin-film Ag/AgCl was employed as a reference electrode and its durability was verified by obtaining a stable open circuit potential for 2 hours against a commercial Ag/AgCl electrode and a stable cyclic voltammetry curve. Selectivity, response time, and linearity of the fabricated oxygen sensor were also verified well by cyclic voltammetry and amperometry depending. The fabricated oxygen sensor showed a 90% response time of 40sec and an excellent linearity with a correlation coefficient of 0.994.

Effect of Depletion and Oxidation of Cellular GSH on Cytotoxicity of Mitomycin Small Cell Lung Cancer Cells

  • Effect of the depletion or oxidation of GSH on mitomycin c (MMC)-induced mitochondrial damage and cell death was assessed in small cell lung cancer (SCLC) cells. MMC induced cell death and the decrease in the GSH contents in SCLC cells, which were inhibited by z-LEHD.fmk (a cell permeable inhibitor of caspase-9), z-DQMD.fmk (a cell permeable inhibitor of caspase-3) and thiol compound, N-acetylcysteine. MMC caused nuclear damage, release of cytochrome c and activation of caspase-3, which were reduced by N-acetylcysteine. The depletion of GSH due to L-butionine-sulfoximine enhanced the MMC-induced cell death and formation of reactive oxygen species in SCLC cells, whereas the oxidation of GSH due to diamide or $NH_2Cl$ did not affect cytotoxicity of MMC. The results show that MMC may cause cell death in SCLC cells by inducing mitochondrial dysfunction, leading to activation of caspase-9 and -3. The MMC-induced change in the mitochondrial membrane permeability, followed by cell death, in SCLC cells may be significantly enhanced by the depletion of GSH. In contrast, the oxidation of GSH may not affect cytotoxicity of MMC.
