In order to evaluate the genotoxicity of airborne particulate matter extracted with dichloromethane (APE), the rat microsome mediated (S-9) or DNA repair enzyme treated Comet assays were performed using the single cell gel electrophoresis in A549 human lung carcinoma cells. It was found that the cells interacting with APE showed more DNA single-strand breaks relative to untreated cells. The genotoxicity of APE was increased with the treatment of S-9 mixture. Microsome mediated DNA damage was inhibited by CYP1Al inhibitor, quercetin. The APE also showed oxidative DNA damage evaluated by endonuclease III treatment. Oxidative DNA damage of APE was inhibited by antioxidants such as vita- min C and Trolox. We also found that the vegetables or fruits extract may reduce APE-induced genotoxicity by their anti- oxidant activity and CYP1A1 inhibition.
The present study attempts to evaluate antioxidant activities of extracts from Abeliophyllum distichum. Nakai flower. The samples were collected in Jangyyeon-myeon, Goesan-gun, Korea and extracted with either hot-water or ethyl acetate (EtOAC). In DPPH, hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity of EtOAC extracts were 93.41%, 98.43%, and 7.38%, while those of hot-water extracts were 86.93%, 41.33% and 47.68% at 200 ${\mu}g/ml$, respectively. In ${\varphi}X$-174 RF I plasmid DNA cleavage assay, the protective effects of EtOAC and hot-water extracts against oxidative DNA damage were 82% and 17% at 200 ${\mu}g/ml$, respectively. Both extracts showed the protective effect of DNA migration by oxidative stress in intracellular DNA migration assay. Both extracts had no cytotoxity in NIH3T3 cells. Several polyphenolic compounds were identified such as 2-methoxy-benzoic acid, vanillic acid, phytol and pulegone by GC/MS. These results indicated that extracts of Abeliophyllum distichum Nakai flower showed antioxidant activities and protective activities against oxidative DNA damage and showed the possibility to be used as an effective natural antioxidants.
Abeliophyllum distichum is Korea Endemic Plants and its genetic resources found from Korea only. Bioactivities of A. distichum such as antioxidant, anti-cancer, and anti-inflammatory studies have been proved through many researches. Whereas, there are no studies on the biological activity of its callus extracts. In this study, we investigated the antioxidant activities of callus extracts derived from A. distichum and its inhibitory effect on oxidative DNA damage. The antioxidant activities were assessed using radical scavenging assays with DPPH, ABTS, and reducing power assay and the inhibitory effects on oxidative DNA damage were measured using ${\varphi}-174$ RF I plasmid DNA cleavage assay. In addition, callus extracts derived from A. distichum showed high antioxidant acitivties and no cytotoxicity in NIH/3T3. Also, it has significantly suppressed expression of ${\gamma}$-H2AX and p53 protein and mRNA levels in NIH/3T3 cells exposed to oxidative stress. Therefore, the callus extracts derived from A. distichum has potential antioxidant activity that can provide protective effects against the oxidative DNA damage caused by free radicals. This study suggest that it is valuable as cosmetics and medicine for antioxidant and cancer preventive materials.
오가피 에탄올 추출물이 제초제로 인한 DNA와 적혈구 손상을 억제할 수 있는지 알아보기 위해 코멧 어세이와 용혈반응을 사용하여 오가피 추출물 존재하에서 DNA 산화손상 억제와 적혈구 손상 억제 정도를 측정하였다. 페녹시계 제초제인 2,4-D (2,4-dichlorophenoxyacetic acid), 2,4,5-T (2,4,5-trichlorophenoxyacetic acid) 그리고 바이피리딜계 제초제인 paraquat은 임파구 DNA에 산화적 손상을 유발하였다. 그러나 2,4-D, 2,4,5-T, 혹은 paraquat으로 인한 DNA 산화손상은 오가피 추출물 처리에 의해 시험관에서 억제되었다. 또한 적혈구 손상도 오가피 추출물 처리에 의해 시험관에서 억제되었다.
Glutathione S-transferase (GST) is a multigene family of phase II detoxifying enzymes that metabolize a wide range of exogenous and endogenous electrophilic compounds. GSTM1 and GSTT1 gene polymorphisms may account for inter-individual variability in coping with oxidative stress. We investigated the relationships between the level of lymphocyte DNA and antioxidative parameters and the effect on GST genotypes. GSTM1 and GSTT1 were characterized in 301 young healthy Korean adults and compared with oxidative stress parameters such as the level of lymphocyte DNA, plasma antioxidant vitamins, and erythrocyte antioxidant enzymes in smokers and non smokers. GST genotype, degree of DNA damage in lymphocytes, erythrocyte activities of superoxide dismutase, catalase, and glutathione peroxidase (GSH-Px), and plasma concentrations of total radical-trapping antioxidant potential (TRAP), vitamin C, ${\alpha}$- and ${\gamma}$-tocopherol, ${\alpha}$- and ${\beta}$-carotene, and cryptoxanthin were analyzed. Lymphocyte DNA damage assessed by the comet assay was higher in smokers than that in non-smokers, but the levels of plasma vitamin C, ${\beta}$-carotene, TRAP, erythrocyte catalase, and GSH-Px were lower than those of non-smokers (p < 0.05). Lymphocyte DNA damage was higher in subjects with the GSTM1- or GSTT1-present genotype than those with the GSTM1-present or GSTT1- genotype. No difference in erythrocyte antioxidant enzyme activities, plasma TRAP, or vitamin levels was observed in subjects with the GSTM1 or GSTT1 genotypes, except ${\beta}$-carotene. Significant negative correlations were observed between lymphocyte DNA damage and plasma levels of TRAP and erythrocyte activities of catalase and GSH-Px after adjusting for smoking pack-years. Negative correlations were observed between plasma vitamin C and lymphocyte DNA damage only in individuals with the GSTM1-present or GSTT1- genotype. The interesting finding was the significant positive correlations between lymphocyte DNA damage and plasma levels of ${\alpha}$-carotene, ${\beta}$-carotene, and cryptoxanthin. In conclusion, the GSTM1- and GSTT1-present genotypes as well as smoking aggravated antioxidant status through lymphocyte DNA damage. This finding confirms that GST polymorphisms could be important determinants of antioxidant status in young smoking and non-smoking adults. Consequently, the protective effect of supplemental antioxidants on DNA damage in individuals carrying the GSTM1- or GSTT1-present genotypes might show significantly higher values than expected.
We examined the effects of the NADPH oxidase p22phox C242T polymorphism on endurance exercise performance and oxidative DNA damage in response to acute and chronic exercises. One hundred three subjects were recruited, among which 26 healthy subjects (CC: 12, TC: 12, and TT: 2) were studied during rest, exercise at 85% $VO_2max$, and recovery before and after 8 weeks of treadmill running. Lymphocyte DNA damage increased significantly in response to exercise (p < 0.05). There were no significant differences in plasma MDA, SOD concentrations and lymphocyte DNA damage between CC genotype and T allele group, but significant endurance training differences were observed. Endurance training increased exercise time to exhaustion in both the CC genotype and T allele groups (p < 0.05) but no significant difference was found between groups. The results of the current study with young, healthy, Korean men are interpreted to mean that 1) the majority had the CC genotype of the NADPH oxidase p22phox C242T polymorphism (82.5%: CC, 15.5%: TC, 1.9%: TT), 2) acute exercise increased lymphocyte DNA damage, 3) endurance training significantly increased exercise time to exhaustion, and alleviated lymphocyte DNA damage, and 4) The NADPH oxidase p22phox C242T polymorphism, however, did not alter lymphocyte DNA damage or exercise performance at rest, immediately after exercise, or during recovery.
Previous studies have shown that one of the primary causes of increased iron content in the brain may be the release of excess iron from intracellular iron storage molecules such as ferritin. Free iron generates ROS that cause oxidative cell damage. Carnosine and related compounds such as endogenous histidine dipetides have antioxidant activities. We have investigated the protective effects of carnosine and homocarnosine against oxidative damage of DNA induced by reaction of ferritin with $H_2O_2$. The results show that carnosine and homocarnosine prevented ferritin/$H_2O_2$-mediated DNA strand breakage. These compounds effectively inhibited ferritin/$H_2O_2$-mediated hydroxyl radical generation and decreased the mutagenicity of DNA induced by the ferritin/$H_2O_2$ reaction. Our results suggest that carnosine and related compounds might have antioxidant effects on DNA under pathophysiological conditions leading to degenerative damage such as neurodegenerative disorders.
Among aging diseases, the most basic problem is a decrease in bone mineral density. Patients with osteoporosis are steadily increasing in the world. This study was investigated to show the relationship between osteoporotic effects and oxidative damage. Water extracts of 15 medicinal plants and ethanol extracts of 14 medicinal plants with known anti-osteoporotic effects, were tested for their radical scavenging activity using DPPH, ABTS, SRSA and FRAP assay. Water extract of Cornus officinalis, Rubus coreanus and ethanol extract of Rubus coreanus, Viscium album var. coloratum, Cimicifuga heracleifolia showed about 15-20 mg/g of total phenolic contents. Water extract of Cornus officinalis, Rubus coreanus and Epimedium koreanum showed high radical scavenging activity. Ethanol extract of Drynaria fortunei, Cornus officinalis, Rubus coreanus, Gentiana scabra and Astragalus membranaceus showed high radical scavenging activity. Water extract of Drynaria fortunei, Cornus officinalis, Nelumbo nucifera, Epimedium koreanum, and Gentiana scabra showed very strong protective effect against oxidative damage to DNA. These results suggest the correlation between anti-osteoporotic effects and antioxidative effects.

Welding fume (WF) induces pulmonary disease including pneumoconiosis. To investigate whether reactive oxygen species-induced oxidative DNA damage occurs during welding fume exposure and the upregulation of DNA repair mechanisms is accompanied, SPF SD rats were exposed to welding fumes with the concentrations of 65.6${\pm}$2.9 mg/㎥(low dose) and 116.8${\pm}$3.9 mg/㎥ (high dose) of total suspended particulate for 2 hrs per day in an inhalation chamber for a total of 2hrs, 15 or 30 days.(omitted)
Oxidative DNA damage negatively affects humans and the research is currently ongoing to find ways to reduce oxidative stress. Oxidative stress has been identified as a key factor in triggering various diseases. Thus, its alleviation is important for human health. Broussonetia kazinoki (B. kazinoki) has been used in traditional Korean medicine as a dermatological therapy to treat burns, pruritus, and acne. B. kazinoki is generally segregated into peeled root (PR), root bark (RB), peeled stem (PS), and stem bark (SB). To assess these components for their antioxidant activity and protection against DNA damage, their ethyl acetate fractions were examined by 1,1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging assay. As a result of confirming the expression of factors involved in attenuating DNA damage, the protective effect of SB on oxidative stress suppressed the expression of p-p53 and γ-H2AX. Additionally, the levels of p53 and H2AX mRNA were significantly downregulated. In conclusion, these results indicated that the SB component of B. kazinoki had the potential to be used as an effective natural antioxidant compared to the other parts of the plant.
