Reproductive Toxicity of DA-125, A New Anthracycline Anticancer Agent: Teratogenicity Study in Rabbits (새로운 안트라사이클린계 항암제 DA-125의 생식독성연구: 토끼 최기형시험)

  • DA-125, a new anthracycline antitumor antibiotic, was administered at dose levels of 0, 0.2, 0.6 and 1.8 mg/kg/day intravenously to pregnant New Zealand White rabbits from day 6 through 18 of gestation. The does were subjected to the caesarean section on day 28 of gestation. Effects of test agent on general toxicity of does and embryonic development of F1 fetuses were examined. At 1.8 mg/kg, the organ weight for ovary of does was significantly decreased. The decrease in the number of corpus lutea, implantations and litter size, and the increase in the rate of resorptions were also observed. In addition, various types of external, visceral and skeletal malformations occurred in fetuses at an incidence of 7.7, 7.7 and 20.6%, respectively. The results show that the no effect dose levels (NOELs) of DA-125 are 0.6 mg/kg/day for does and F1 fetuses.

The protective effect of Red Giseng on the organotin compounds(TBTO) poison : Focusing on the Immunity and sex hormone (유기주석화합물의 독성에 대한 홍삼의 방어효과 - 면역 및 성 호르몬물질 중심으로 -)

  • This study was carried out effect of red ginseng extract(1.0g/kg) on organotin compounds (10, 20 and 40 mg/kg) which poisons against some organs like thyroid gland, liver, kindey, testis, ovary, serum immuinty and sex hormone activity of rats were examinned by gastric tubing for 3 weeks. The weight of each organ in TBTO treated group were significantly increased other organs which excepted kinedy in males and only liver in females.(p<0.05, p<0.01). In case of Immunity activity of each sex, IgM level was small change comparsion with that of control group in all sex. but IgG level was significantly decreased females rather than males comparsion with that of control group.(p<0.05, p<0.01) In case of sex hormone activity, the testosterone activity of males and the estradiol activity of females were significantly decreased rather than the control group. on the other hand, red singsong treated group was only significantly increased estradiol activity.( p<0.05, p<0.01)

  • The sequence of hydroxypyruvate isomerase gene was obtained in NCBI. In this study, the hydroxypyruvate isomerase gene of Bombyx.mori was identified and annotated with bioinformatics tools. The result was confirmed by RT-PCR, prokaryotic expression, mass spectrographic analysis and sub-cellular localization. The hydroxypyruvate isomerase cDNA comtains a 783bp ORF, and has 4 exons. The deduced protein has 260 amino acid residues with the predicted molecular weight of 29169.30 Da, isoelectric point of 6.10, and contains conserved PRK09997 and Hfi domains. The hydroxypyruvate isomerases of Nasonia vitripennis and Bombyx mori have a high homology. Through RTPCR analysis, we found that this transcript was present in testis, ovary, blood-lymph, fat body, midgut, silk gland and tuba Malpighii. This protein was located in cytoplasm through immunohistochemistry. We submitted the cloned gene under the accession number EU344910. The enzyme has been classified under accession number EC 5.3.1.22.

Effect on Sex hormone activity of TBTO against injected red Ginseng extract (홍삼 투여가 TBTO의 성호르몬 활성에 미치는 영향)

  • The purpose of this study finds out the effect of red ginseng extract (1.0g/kg) on TBTO (10, 20, and 40mg/kg) which poisons against some organs like thyroids gland, liver, kidney, testis, ovary, sex hormone activity of rats are examined by gastric tubing for 3 weeks. The weight of each organ in treated group were increased, especially liver in female and those of testis in males were significantly increased at 10, 20 and 40mg/kg (P<0.05, P<0.01). In case of sex hormone activity of each sex, the estradiol activity of female and testosterone activity of males were significantly decreased rather than the control group (P<0.05, P<0.01) According to between the TBTO treated group and 10+ rGe group of the testosterone activity each sex were significantly increased (P<0.01).

  • Human chorionic gonadotropin hormone(H.C.G.) is secreted from the villus tissues of the placenta and excreted in lange amount into the urine. Its isolation is chiefly made from the urine of a pregnant woman. Recently, Matsushima attempted isolation of H.C.G. directly from the placenta itself. In order to prepare H.C.G. from human placenta, general method of extractiag and purifying proteins was applied. Its way was as follow: Crude H.C.G. was extracted from placenta with pH 9.0 and pH 5.0 aqua ammonia, and purified with pH 8.0 ammonia and 50% ethanol at pH 4.8. The purified H.C.G. showed two moving bands on the anode by paper electrophoresis. On the other hand, the H.C.G. from pregnancy urine (Standard. Pharm. Co.) showed same two bands but their moving ratio were different. The purified H.C.G. showed gonadotropin effect when it was injected young fomale rats 40r/cc per day for 5 days and weighted the increased ovary weight.

  • Directional (sinistral) asymmetry (DA) occurs when the traits of one side of a supposedly bilaterally symmetrical organism differ in a random way from those of the other side. We examined asymmetries in gonadal growth traits within both sexes of hatchery reared ayu, Plecoglossus altivelis. The pattern of the gonadal growth from hatching to 320 days after hatching was a type of DA except for the growth in gonad weight from 140 to 180 days after hatching, although the ovary of the right side tended to exhibit more pronounced DA phenomenon.

  • Pediatric obesity has become a serious public health issue. The prevalence of obesity in children and adolescents has increased worldwide and in Korea over several decades. Obese children are more likely to be obese adults with an increased cardiovascular risk. Therefore, maintaining a healthy weight and preventing obesity during childhood are of critical importance. Moreover, obese children and adolescents often have endocrine comorbidities such as prediabetes, type 2 diabetes, dyslipidemia, metabolic syndrome, polycystic ovary syndrome, and central precocious puberty. Hence, the early implementation of obesity management using a multidisciplinary team approach and screening for these comorbidities in obese children and adolescents are required with the appropriate management of each comorbidity and/or specialist referral.

  • Over the past few decades, there has been a notable increase in the incidence of pediatric obesity, which is a significant public health concern. Children who are obese have a greater risk of type 2 diabetes, hypertension, dyslipidemia, polycystic ovary syndrome, obstructive sleep apnea, and adult obesity. Lifestyle modification therapy is typically the initial approach to treat pediatric obesity. For patients who do not achieve success with lifestyle modification therapy alone, pharmacotherapy is the next logical treatment option. When selecting an anti-obesity medication (AOM), it is essential to first ascertain the medical background of the patient, including current medications and obesity-associated comorbidities. Evaluation of obesity phenotypes in patients may also be beneficial. AOMs for pediatric obesity include metformin, orlistat, glucagon-like peptide 1 agonists, phentermine, and the phentermine/topiramate combination. Sufficient lifestyle modification therapy should be administered before considering pharmacotherapy and continued after the initiation of AOM. To ensure healthy development, monitoring growth and puberty development during anti-obesity treatments is essential.

Localization and Accumulated Concentration Changes of Mercury Compound in Reproductive Organs of Female Mice with Time (암컷 마우스 생식기관 내 수은 화합물의 위치와 시간에 따른 축적된 수은 농도 변화)

  • This study was performed to investigate the localization and concentration changes of mercury compound in female reproductive organs with time. Methylmercuric chloride was subcutaneously injected weekly into pubescent female mice for 3 weeks. For the concentration changes of mercury with time, the mice were sacrificed at 10, 150, and 300 days post treatment (DPT). Body and organ weights were not significantly different between the control and mercury-treated groups, except for 10 DPT in body weight. Localization of accumulated mercury was identified by the autometallography method. Localization of mercury compounds in the uterus, ovary, and ovum was analyzed with a light microscope. In the uterus, mercury was densely located in the stroma cells and surface epithelium of the perimetrium at 10 DPT. Mercury concentration was decreased at 150 DPT and did not appear at 300 DPT. In the ovary, mercury particles were distributed in the stroma cells of the cortex region, cells of the theca around the follicle, and the corpus luteum at 10 DPT. Mercury was concentrated in the medulla region at 150 DPT and was not distributed at 300 DPT. In the ovum, mercury particles were mainly located in the marginal region at 10 and 150 DPT. Mercury concentration was decreased and evenly distributed at 300 DPT. These results suggest that hormone synthesis, implantation, and developing embryos will be affected by mercury compound in the female mouse.

Superovulation Treatment with PMSG and $\textrm{PG600}^{\textregistered}$ in Prepubertal Gilts (돼지에서 PMSG와 $\textrm{PG600}^{\textregistered}$의 과배란 처치 효과)

  • The objective of this study was to compare different superovulation treatments using PMSG or PG600$^{ }$ and to determine the optimal time of oocyte recovery after hCG administration. A total of 90 prepubertal Yorkshire x Landrace gilts crossed with Duroc, 6~7 months old and 100~120 kg of body weight, were used. PMSG (1,500 IU/head) or 5~7.5 ml of PG600$^{ }$(400 IU of PMSG and 200 IU of hCG) were administrated subcutaneously, and then 1,000 IU of hCG were administered intramuscularly at 72 hours after PMSG or PG600$^{ }$ injection. At carious time of 44, 46, 48 and 50 hours after hCG injection, superovulated gilts were slaughtered in a local abattoir. Ovaries together with oviducts were excised from the body immediately after slaughtered and transported to laboratory in 39$^{\circ}C$ saline. Ovaries were examined fur the number of corpus hemorrhagicum and unovulated follicles present in the surface of ovary. The unovulated follicles were categorized into small (1~3 mm in diameter) and large (4~8 mm) groups according to their diameter. Oocytes were recovered by flushing both oviducts with micropipette tip (1~100 $\mu$l) attached to a 10-ml disposable syringe. The number of CH on ovary and recovered oocytes at 46, 48 and 50 hr after hCG injection in PG600$^{ }$ treated groups were significantly higher than the other group. Group of phCG 50 hr among PMSG treated groups had a greater number of CH and recovered oocytes(P<0.05). The number of CH on ovary and recovered oocytes at 50 hr after hCG injection in 1$\frac{1}{2}$ vial(7.5 ml) of PG600$^{ }$ treated groups was significantly higher than 1 vial(5 ml) of PG600$^{ }$ treated group(P<0.05). In conclusions, considering a number of corpus hemorrhagicum and recovered oocytes after superovulation in gilts, effective time of oocyte recovery by treatment with PMSG and hCG was post-hCG 50 hr and with PG600$^{ }$ plus hCG was post-hCG 46, 48 and 50 hr. Also, admini-stration of 1$\frac{1}{2}$ vial(7.5 ml) of PG600$^{ }$ treated group had a great number of CH and recovered oocytes.covered oocytes.
