Vitronectin regulates osteoclastogenesis and bone remodeling in a mouse model of osteoporosis

  • Vitronectin (VN) is an extracellular matrix protein with a crucial role in regulating bone remodeling. In this study, we aimed to investigate the effect of VN deficiency in a mouse model of osteoporosis induced by ovariectomy (OVX). The findings revealed that the absence of VN led to an increase in the activity of tartrate-resistant acid phosphatase (TRAP), a marker for osteoclasts, in the plasma of OVX-operated mice. TRAP staining further demonstrated that VN deficiency resulted in a higher number of osteoclasts within the femurs of OVX-operated mice. X-ray micro-computed tomography analysis of the femurs in OVX-operated mice indicated that VN deficiency significantly suppressed the OVX-induced increase of marrow area and total volume of bone. Additionally, we assessed structural model index (SMI) and degree of anisotropy (DA) as indices of osteoporosis. The results showed that VN deficiency effectively attenuated the OVX-induced increase in SMI and DA among OVX-operated mice. In summary, our study demonstrates the vital role of VN in regulating osteoclastogenesis and bone remodeling in the mouse model of osteoporosis.

  • Objective & Methods : The purpose of this study is to observe the effects of Cyperi Rhizoma Herbal-acupuncture infusion solution at Umgok($KI_{10}$) on Osteoporotic Rats Induced by Ovariectomy. The author performed several experimental items to analyze cytotoxic, osteoporosis evaluation, change of ALP, osteocalcin, Ca and histological change of tibia. Result : 1. BMD were increased non-meaningly in CR-HA than control group. 2. ALP was not inhibited by CR-HA, osteocalcin was decreased meaningly in CR-HA than control group. 3. Osteoclast like cell in tibia was increased meaningly in control group, decreased meaningly in CR-HA. 4. In the histological study in tibia, TBV was significantly increased, GPL was significantly decreased in the CR-HA than control group. Conclusions : From the result above, the results suggest that CR-HA at $KI_{10}$ has significant effect on osteoporosis, and to be put to practical use in the future osteoporosis clinic.

A Efficacy of Ginseng radix on the Preservation of Spinal Bone Mineral Density and Bone Inorganic Substance of Oophorectomized Rats

  • This study was undertaken to investigate the effects of the Ginseng radix in osteoporosis of the oophorectomized rats. In this experiment, the rats were oophorectomized and administered the water extracts of the Ginseng radix. The spinal bone mineral density, calcium, phosphorus and ash weight of the bones were measured. The spinal bone mineral density was significantly increased in the ovariectomized (OVX)-Ginseng radix group at 8 weeks as compared to the OVX-saline group. Furthermore, the calcium and phosphorus contents of the femoral and fibula-tibia were significantly increased in the OVX-Ginseng radix group as compared to the OVX-saline group. The ash weights of the femoral and fibula-tibial bones were increases in the OVX-Ginseng Radix group, although it was not statistically significant. On reviewing these experiments, it appears that the Ginseng radix possess efficacy for the prevention of osteoporosis. Further study would be of value to confirm the efficacy of the Ginseng radix for the treatment and/or prevention of osteoporosis in humans.

The Effects of Proprioceptive Neuromuscular Facilitation Exercise on the BMD, Balance and Lower Muscular Strength in Patients with Osteoporosis (고유수용성신경근촉진법 운동이 골다공증 환자의 골밀도, 균형 및 하지 근력에 미치는 효과)

  • Purpose: This study examined the effects of an exercise program involving the Proprioceptive Neuromuscular Facilitation (PNF) lower-extremity patterns on the Bone Mineral Density (BMD), balance and lower muscular strength in patients with osteoporosis. Methods: The participants were allocated randomly to 2 groups: experimental group and control group. To evaluate the effects of exercise, the subjects were evaluated using the BMD and Time Up & Go Test and One Leg Stance Test for balance, and the Sit To Stand for lower muscular strength. Results: After the 8 week study, the experimental group showed a significant difference in all items compared to the control groups. Conclusion: These results suggest that a PNF exercise program is effective in patients with osteoporosis. Furthermore, an exercise program involving these patterns can be used in a follow-up program for patients with osteoporosis. Overall, more study will be needed to develop and apply more efficient exercise programs for such patients.

  • It is reported that the mechanical properties of vertebral trabecular bone depend on the density and the mass of bones. Osteoporosis is a systemic skeletal disease caused by low bone mass and microstructure deterioration of trabecular bone. Silva and Gibson (1997) studied the treatment of age-related bone loss using drug therapy. Vertebroplasty is a minimally invasive surgery for the treatment of osteoporosis vertebrae. This procedure includes puncturing vertebrae and filling with Polymethylmethacrylate (PMMA). However, the relative effect of drug therapy and bone cement for osteoporosis treatment is not reported yet. In this study, several 2D models of human vertebral trabecular bone are analyzed by finite element method. The mechanical behaviors of the vertebral trabecular bone treated by the drug therapy and the bone cement are compared. This study shows that bone cement treatment is more effective strategy than drug therapy to prevent the degradation of bone strength.

The Correlation between Dietary Intakes of Calcium and Vitamin D and Osteoporosis, Hypertension and Diabetes Mellitus (식이 칼슘, 비타민 D의 섭취와 골다공증, 고혈압 및 당뇨병의 상관관계)

  • In the light of the increasing concern over the prevalence of osteoporosis, hypertension, diabetes mellitus, we performed this study to review the correlation between the dietary calcium and vitamin D intake and these diseases. To this end, we investigated the effects of dietary calcium and vitamin D on these diseases. We observed that the intake of dietary calcium and vitamin D had a negative correlation with the incidences of osteoporosis, hypertension, and diabetes mellitus. Further, the intake of these two nutrients is expected to improve related mechanisms such as the renin-angiotensin system. Therefore, we suggest that dietary calcium and vitamin D have a beneficial effect on these diseases.

Relationship Between Osteoporosis and Body Mass Index among the Elderly: A Theoretical Review (노인의 골다공증과 체질량지수의 관계에 관한 이론적 고찰)

  • Purpose: The purpose of this study was to review the literature to explore the relationship between body mass index (BMI) and bone mineral density (BMD) in elderly using an integrative review. Methods: The keywords 'osteoporosis,' 'body mass index,' and 'bone mineral density' were used to search peer-reviewed publications through four databases. Among 1,389 searched articles, eight articles were selected after excluding those that did not meet the inclusion criteria. Results: Seven articles stated that BMI was positively associated with BMD among elderly. In the elderly, the prevalence of osteoporosis was 25-55.7% in women, and 11-13% in men. Conclusion: This study found that high BMI may be helpful to increase BMD among elderly although the mechanism was not clear. It is necessary to identify BMD and fractures in elderly according to body composition in future research.

AN EXPERIMENTAL STUDY ON THE CHANGE OF BONE MINERAL METABOLISM AFTER IRRADIATION (방사선조사가 골무기질함량에 미치는 영양에 관한 실험적 연구)

  • Irradiation is widely used for the treatment of malignant diseases, and possibly cause the osteoporosis. The bone densitometry and bone scintigraphy are valuable when used to monitor the patients longitudinally to access the progression of osteoporosis and risk of osteoradionecrosis. To evaluate the osteoporosis after irradiation of cobalt-60 gamma ray on the lumbar spines of New Zealand white rabbits, bone densitometry by dual photon absortiometry and bone scintigraphy were performed weekly. The decrease of bone density began at the first week after irradiation, and were in the nadir at 4-6th week. The osteoblastic activity measured by bone scintigraphy decreased in the first week, and was in the nadir at 4-6th week. The severity of these changes were related to the radiation dose. In conclusion, the osteoporosis before presentation of the osteoradionecrosis can be developed at low dose irradiation and confirmed by bone densitometry, bone scanning, and histopathology.

Effects of Dioscorea batatas on Estrogen-deficient Osteoporosis (Estrogen 결핍성(缺乏性) 골다공증(骨多孔症)에 미치는 산약(山藥) 추출물(抽出物)의 영향(影響))

  • Osteoporosis is characterized by bone loss and mobidity with osteoporotic fracture. This study was performed to evaluate the effect of on the bone mass and its related factors in estrogen-deficient animal model. The model rats of osteoporsis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 14th day after ovariectomy-surgery, rats were administered with DBE, extract of Dioscorea batatas, per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of DBE. Osteoporetic rats showed lower serum estrogen level, higher body weight than normal rats, and showed atrophy of uterine horns. DBE showed inhibitory effect on bone loss in osteoporetic condition, and reduced the increase of ALP activity and osteocalcin level in serum, and reduced the increase of OH-proline level in urine. But, DBE had no effect on cell proliferation and ALP activity in rat calvarial cell culture.
