Improvement of the macro- and micro-elements density of rice (Oryza sativa L.) is gradually becoming a new breeding objective. In this study, the genomic regions associated with potassium, calcium, magnesium and iron content in rice grain were identified and characterized by using a doubled haploid (DH) population. Fifty-six simple sequence repeat (SSR) and one hundred and twelve sequence tagged site (STS) markers were selected to construct the genetic linkage map of the DH population with a full length of 1808.3cM scanning 12 rice chromosomes. Quantitative trait loci (QTLs) were detected, and QTL effects and QTL interactions were calculated for five traits related to macro- and micro-elements in the DH population from a cross between 'Samgang' (Tongil) and 'Nagdong' (Japonica). Twelve QTLs were located on five chromosomes, consisting of two QTLs for potassium, three QTLs for calcium, two QTLs for magnesium, one QTL for iron content and four QTLs for the ratio of magnesium to potassium (Mg/K). Among them, qca1.1 was detected on chromosome 1 with an LOD value of 8.58 for calcium content. It explained 27% of phenotype variations with increasing effects from 'Samgang' allele. Furthermore, fifteen epistatic combinations with significant interactions were observed on ten chromosomes for five traits, which totally accounted for 4.19% to 12.72% of phenotype variations. The screening of relatively accurate QTLs will contribute to increase the efficiency of marker-assisted selection (MAS), and to accelerate the establishment of near-isogenic lines (NILs) and QTL pyramiding.
The concerns on the crop damage by ultraviolet (UV) radiations is increasing owing to the decrease of their absorbing stratospheric ozone in the tropospheric. Cultivar differences on early growth of UV radiation among five Korean rice cultivars, four japonica types and one Tongil type (indica-japonica cross hybrid), were studied. Pot-seeded rice plants were grown under four different radiation conditions, i.e., visible radiation only, visible radiation with supplemented with high or low dose of UV-B (280~320 nm in wavelength) and UV-C (less than 280 nm in wavelength). The inhibitory degree on plant height, shoot and root weight and length of leaf blade and leaf sheath were determined at 40 days after seeding. UV-C showed the most severe inhibitory effect on the degree of biomass gain and leaf growth in most cultivars examined, followed by high UV-B and low UV-B. Among the cultivars used, the Kuemobyeo was the most sensitive cultivar and had not repair or showed resistance ability to continued irradiation of UV radiation. However, Janganbyeo and Jaekeon showed different responses that the elongation of leaf blades was promoted on 2nd and 3rd leaves and inhibited on 4th and 5th leaves but this inhibitory degree was reduced on 6 th and 7th leaves. Such tendency on leaf growth means that both cultivars had low sensitivity and most resistant ability to continued irradiation of UV radiation. While Tongil showed different response to enhanced UV radiation, ie., low UV-B promoted leaf growth but the inhibitory was severely increased by continued irradiation of high UV-B and UV-C, which means that Tongil had high threshold of UV radiation for response as an inhibitory light of plant growth. The results of this study indicate that the differences on sensitivity or resistant to the effects of UV radiation were existed among Korean rice cultivars.
Varietal differences of grain filling duration and rate are generally recognized in rice (Oryza sativa L.). But the differences of grain filling characteristics between the parts within a panicle might be present since it takes 6-8 days to finish anthesis in a panicle. To elucidate this, 6 Japonica and 6 Japonica/Indica varieties were grown under three nitrogen levels at the Experimental Farm of Chungbuk National University in 1986, and grain filling characteristics of top, middle and bottom parts of a panicle were analyzed. Rice grain filling duration could be divided into 3 phases, i.e., Lag phase(LP) of 5 days after heading. Linear increasing period (LIP) of 5-20 days after heading and Late filling period (LFP) thereafter. Grain weight difference between the panicle parts was greater during LIP in all the varieties and was decreased during LFP. The difference was greater in Japonica/Indica varieties. Samgangbyeo and Seomjinbyeo had the shortest and the longest grain filling durations with 22.6 and 37.1 days, respectively. In average, grain filling duration of the bottom part of a panicle was 1-2 days shorter than that of top or middle part. The differences were negligible in Daecheongbyeo, Taebaegbyeo and Milyang 23, while it was greater in Tongil with 4.9 days. Grain filling rates were ranged from 0.612 to 1.097 mgㆍgrain$^{-1}$ㆍday$^{-1}$ㆍMilyang 23 showed the lowest rate with 0.612-0.682 mgㆍgrain$^{-1}$ㆍday$^{-1}$, while the rates of Baegyangbyeo, Yongmoonbyeo, Samgangbyeo, Nongbaeg and Daeseongbyeo were about 1 mgㆍgrain$^{-1}$ㆍday$^{-1}$ㆍGrain filling rate of bottom part of a panicle was lower than that of top or middle part. The difference was samll in Nongbaeg and Daeseongbyeo, while it was the greatest in Tongil with 0.222 mgㆍgrain$^{-1}$ㆍday$^{-1}$. Grain filling duration and rate were not afftected by nitrogen level, but varietal and spatial differences of grain filling characteristics were highly significant. However, the spatial difference of grain filling rates within a variety was not significant. Grain filling characteristics of each part of a panicle were significantly correlated (r=0.7207-0.9879) with those of a whole panicle.
This study was carried out to get infirmation for the breeding varieties and developing the cultural methods with representative 32 cultivars which were diffused from 1900 to 1982 and two cultural methods which were old one i.e., low dressing and late planting and modern one i.e., medium dressing and early planting. Duration of transplanting between cultural practices were 15 days but the duration of heading date were 5days. Panicle projecting duration was 5-10 days in all cultivars, especially, it is 5 days in early maturing varieties, 6-7 days in medium maturing varieties and 7-10 days in late maturing varieties. Periods from transplanting date to effective ripening date were 99.8 days in low dressing and late planting method, 106.2 days in modern one but the periods from heading date to effective ripening date were 31.4 days in low dressing and late planting method, 30.2 days in modern one. Ripening speed was very active from 5 to 10 days after heading and the daily increase of 1000 grain weight was from 847.8mg to 1130mg in that time and it slightly increased from 5 to 35 days after heading in Japonica varieties but increased after heading and suddenly decreased at 30 days after heading in Indica/Japonica varieties. Meteorological factors (temperature and amount of solar radiation) were positively correlated with the ripening speed but negatively correlated with the period of physiological and effentive ripening.
This study was conducted to investigate the chromosomal locations and effects of quantitative trait loci (QTL) associated with chemical properties of rice (Oryza sativa L.). One hundred sixty four recombinant inbred lines (MGRILs) of $F_{11}$ were derived from the cross between Milyang 23, Tongil type, and Gihobyeo, japonica type. They were evaluated for 7 traits of chemical property in rice. Transgressive segregation was observed for all traits examined. Eight significant QTLs were detected (LOD$\geq$2.0) for five traits, including two QTLs for amylose content, two QTLs for potassium content, one QTL for ratio of magnesium to potassium, one QTL for fat content and two QTLs for ash content. Phenotypic variation explained by each QTL ranged from 7.2% to 14.4%. However, no significant QTL was detected for magnesium and protein contents. In amylose content and ash content M alleles originated from Milyang 23 were responsible for increasing these traits and J alleles originated from Gihobyeo also responsible for increasing these traits. Pleiotropic effects of single QTLs on different traits are observed.
The objective of this study is to identify main-effect QTLs, epistatic QTLs, and the interactions between QTL and the environment associated with grain appearances of brown rice. A genetic linkage map was created with 172 DNA markers spanning 12 rice chromosomes based on 120 DH lines, which derived from a cross between 'Samgang'(Tongil) and 'Nagdong'(Japonica). One thousandgrain weight, length, width, length-to-width ratio, and thickness were evaluated regarding the DH population. Twenty independent QTLs and fourteen epistatic QTLs were identified in using CIM by two programs, known as WinQTLcart2.5 and QTLMAPPER. The QTLs of qgw9.1 in an interval of RM434-RM242 on chromosome 9 and qgw11.1 at a peak marker of RM287 on chromosome 11 for one thousand-grain weight, qgwi2.2 for grain width at a peak marker of RM450, qlw2.1 for length-to-width ratio flanked by RM492 and RM324, and qgt2.1 for thickness flanked by 2009 and RM492 on chromosome 2 were detected over two years, which can be considered as stable QTLs. The epistatic effect might be an important component for genetic basis of one thousand-grain weight and width. The main-effect QTLs of grain width and length to width ratio were easily influenced by environments.
The amino acid composition of rice is a major concern of rice breeders because amino acids are among the most important nutrient components in rice. In this study, a genetic map was constructed with a population of 134 recombinant inbred lines (RILs) from a cross between Dasanbyeo (Tongil-type indica) and TR22183 (temperate japonica), as a means to detect the main and epistatic effect quantitative trait loci (QTLs) for the amino acid content (AAC). Using a linkage map which covered a total of 1458 cM based on 239 molecular marker loci, a total of six main-effect QTLs (M-QTLs) was identified for the content of six amino acids that were mapped onto chromosome 3. For all the M-QTLs, the TR22183 allele increased the trait values. The QTL cluster (flanked by id3015453 and id3016090) on chromosome 3 was associated with the content of five amino acids. The phenotypic variation, explained by the individual QTLs located in this cluster, ranged from 10.2 to 12.4%. In addition, 26 epistatic QTLs (Ep-QTLs) were detected and the 25 loci involved in this interaction were distributed on all nine chromosomes. Both the M-QTLs and Ep-QTLs detected in this study will be useful in breeding programs which target the development of rice with improved amino acid composition.

The objective of this study was to map the gene for reduced embryo size in rice using DNA markers. The reduced embryo size mutant was induced from N-methyl-N-nitrosourea (MNU) treated Taichung 65. Genetic analysis revealed that the phenotype of the reduced embryo was controlled by a single recessive gene, designated as re-1. For mapping the gene controlling embryo size, an $F_2$ population was developed from a cross between the Korean Tongil-type, Milyang 23 (Oryza sativa ssp. indica) and the mutant. The ratio of $F_2$ seeds nearly fitted to 3:1 ratio, indicating that this phenotype was controlled by a single recessive gene. Bulked sergeant analysis was performed with SSR markers. The gene for the reduced embryo size was detected on chromosome 1. The gene was further mapped between two SSR markers, RM315 and RM265 on chromosome 1 (approximately 1.5 Mb interval). The linked markers will facilitate selection of this grain character in a breeding program and provide the foundation for positional cloning of this gene.
Virus diseases often cause serious damage to rice production in Asia. The lack of information on virus resistance genes has been a major obstacle for the breeding of resistant varieties. In order to identify DNA marker associated with resistance against rice stripe virus (RSV), the quantitative trait locus (QTL) was carried out using advanced backcross population developed from a cross between RSV-resistant tongil type cultivar Shinkwang and susceptible japonica cultivar Ilpum. A RSV resistance QTL $qSTV11^{SG}$ explaining 44.2% of the phenotypic variation was identified on chromosome 11 of Tongil type rice cultivar 'Shingwang'. $qSTV11^{SG}$ was tightly linked to DNA marker RM6897. The RM6897 divided as resistance type allele and susceptible type alleles. Twenty seven resistant varieties showed the resistant-type allele and 23 susceptible varieties were susceptible-type allele to the marker of RM6897. This results and the molecular markers presented here may be useful in rice breeding for improving RSV resistance in japonica rice.
In order to find out the basic information for salt tolerance in vice (Oryza sativa L.), effects of NaCl treatment on absorption of inorganic components and growth were investigated in 6 japonica and 5 tongil varieties and compared to the salt tolerance variety, Annapurna. The absorption of N and $\textrm{P}_2\textrm{O}_5$ was less repressed than that of $\textrm{K}_2\textrm{O}$, MgO, CaO, and $\textrm{Si}\textrm{O}_2$ which were a little affected by NaCl treatment. $\textrm{K}_2\textrm{O}$ was the most highly repressed component in the absorption of inorganic components, followed by MgO, CaO, total-N, $\textrm{P}_2\textrm{O}_5$ and $\textrm{Si}\textrm{O}_2$. $\textrm{Na}_2\textrm{O}$ content was increased about twenty times to the control at 30days after NaCl treatment, and tonsil varieties more than absorbed japonica ones. Dry weight of japonica varieties by NaCl treatment was less reduced than that of tonsil varieties, followed by Seomjinbyeo, Sinseonchalbyeo, Nakdongbyeo, Daechoungbyeo, Dongjinbyeo and Chuchoungbyeo in japonica group, and Milyang30, Gayeabyeo, Jangseongbyeo, Chilseongbyeo and Taebackbyeo in tonsil group. The relationship between dry weight reduction and Na/K ratio showed positively significant correlation in rice.
