Rapid molecular authentication of three medicinal plant species, Cynanchum wilfordii, Cynanchum auriculatum, and Polygonum multiflorum (Fallopia multiflorum), by the development of RAPD-derived SCAR markers and multiplex-PCR

  • Definitive identification of original plant species is important for standardizing herbal medicine. The herbal medicines Cynanchi Wilfordii Radix (Baekshuoh in Korean and Beishuwu in Chinese) and Polygoni Multiflori Radix (Hashuoh in Korean and Heshuwu in Chinese) are often misidentified in the Korean herbal market due to morphological similarities and similar names. Therefore, we developed a reliable molecular marker for the identification of Cynanchi Wilfordii Radix and Polygoni Multiflori Radix. We used random amplified polymorphic DNA (RAPD) analysis of three plant species, Polygoni multiflorum, Cynanchum wilfordii, and Cynanchum auriculatum, to obtain several species-specific RAPD amplicons. From nucleotide sequences of these RAPD amplicons, we developed six sequence characterized amplification region (SCAR) markers for distinguishing Polygoni Multiflori Radix and Cynanchi Wilfordii Radix. Furthermore, we established SCAR markers for the simultaneous discrimination of the three species within a single reaction by using multiplex-PCR. These SCAR markers can be used for efficient and rapid authentication of these closely related species, and will be useful for preventing the distribution of adulterants.

Simultaneous Determination of Seven Compounds in Houttuynia cordata using UPLC-PDA

  • A UPLC method for the simultaneous determination of seven compounds was established for the quality control in H. cordata. The UPLC was performed on a $C_{18}$ HSS T3 $2.1{\times}100$ mm, 1.8 ${\mu}m$ column during a 13 minute gradient elution of 0.2% aqueous acetic acid and acetonitrile with the flow rate of 0.2 mL/min at $30^{\circ}C$. The UPLC method was validated according to the ICH guideline of analytical procedures with respect to precision, accuracy, and linearity. The limit of determination and quantitation for the seven compounds were 0.01-0.09 and 0.03-0.28 ${\mu}g/mL$, respectively. The calibration curves of all seven compounds showed good linearity ($r^2$ > 0.999). The intra-day and inter-day the RSD values used to evaluate the precision of analysis were less than 0.9%. The recoveries of quantified compounds ranged from 98.63 to 103.85%. The developed UPLC method was found to be effective, convenient and sensitivity for quantitative analysis of seven compounds in H. cordata. This work could be provided a baseline source for quality control of H. cordata.

현곡(玄谷) 보비탕(補脾湯)의 구성한약과 그 기미배오(氣味配伍) 분석 (Hyeongok's Bobee-tang was Combined according to the Theory for Properties and Tastes of Herbal Medicines)

  • The objectives of this study lie in theoretical establishment of Bobee-tang for curing the asthenic syndrome of spleen through analyzing the component medicines and combination principles of Hyeongok's Bobee-tang, and furthermore, maximizing the clinical use of Bobee-tang. This study analyzed the component medicines and combination principles of Hyeongok's Bobee-tang based on the theory for properties and tastes of herbal medicines from the ${\ulcorner}$Yellow Emperor's Canon of Internal Medicine${\lrcorner}$, the theory for principal herbal medicine, assistant herbal medicine, adjuvant herbal medicine, dispatcher herbal medicine, and the five elements doctrine. Hyeongok's Bobee-tang is an odd prescription, composed of 7 kinds of ingredients : No.1 Radix Ginseng (2don;7.5 g), No.2 Rhizoma Atractylodis Macrocephalae (1don;3.75 g), No.3 Carapax Et Plastrumtestudinis (1don), No.4 Concha Ostreae (1don), No.5 Cortex Cinnamomi (1don), No.6 Rhizoma Zingiberis Praeparatae (1don), and No.7 Radix Glycyrrhizae (1don). First, to cure the asthenic syndrome of the spleen, the methods of invigorating the spleen and the heart, and purging the liver should be used according to the five elements doctrine. Secondly, herbal medicines appropriate for those treatment methods should be chosen according to the theory for properties and tastes of herbal medicine and thirdly, the combination of those herbal medicines should be carried out according to the theory for principal herbal medicine, assistant herbal medicine, adjuvant herbal medicine, dispatcher herbal medicine. As a good example, Hyeongok's Bobee-tang is combined according to the above theories. In conclusion, this formula was created by applying to the theory for properties and tastes of herbal medicines.

반하후박탕으로 호전된 뇌경색 환자의 연하곤란 치험 1례 (A Case Report of Treatment of Dysphagia in a Stroke Patient Treated with Banhahubak-tang)

  • Objectives: The aim of this report is to report the effects of Banhahubak-tang on dysphagia in a stroke patient. Methods: A case of a 67-year-old female stroke patient with dysphagia is presented. The patient was treated with the herbal medicine Banhahubak-tang. Results : After administration of the herbal medicine Banhahubak-tang, dysphagia was improved, which reduced time per meal. Conclusion : Banhahubak-tang was an effective treatment for dysphasia in a stroke patient.

제천 한약재의 역사에 기반한 한방산업 전략 (Herbal medicine bio industry strategy based on the historical topography of Jecheon city)

  • Revising 11 historical topography of Jecheon city we could notice that this territory was traditionally producing high quality medicinal plants. This view is supported as it was tributed to the King. Production of medicinal plants continue these days, Jecheon being one of the largest production of medicinal plants in Korea. Abundant production of diverse medicinal plants of high quality seems to be linked with the soil, climate and environmental characteristics unique of Jecheon. Therefore we propose to develop new type of herbal medicin drugs like Gobang(膏方) which could distinguish from other places. Gobang matchs well to the requirement of current days being convenient to carry and of good taste. Also is appropriate to treat chronic diseases. Its primordial abundant herbal medicines of high quality for the development of Gobang where Jecheon can provide.

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata THUNB extracts on A549 (lung cancer), MDA-MB231 (breast cancer), SNU-C4 (colon cancer) and B16 (mouse melanoma) cell lines. We have determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The 150 $\mu$g/$m\ell$ concentration of methanol extract (63.81 %) of Houttuynia cordata THUNB was shown significantly antitoxic activity on A549 cell lines. (omitted)

  • Traditional herbal medicine is widely used among the Korean people, and other eastern Asian countries employ similar therapies as well. In recent years, due to increasing interest in herbal medicines, many researches have been made on the toxicity and side effects of herbal medications. Through private and public media, there have been many opinions suggesting taking herbal medicines is very harmful, especially on the liver and kidney functions. This assertion has been mainly presented by the doctors that practice western medicine, But this assertion is never based on adequate knowledge of herbal medicine. This study aims to provide the evidences that taking herbal medicines is safe on the renal functions. Four frequently used herbal medications(Sipjeondaebotang, Bojungigitang, Ojeoksan, and Yukmijihwangtang) were used to test the toxicity of herbal medicine oh the lab animal model(SD-Rat). There is no significant difference in body weight and kidney weight after herbal medication for 1 month. In all experimental groups, no abnormal findings were observed in histological study, and lab renal function index(BUN, creatinine, uric acid). These results say that four herbal multi-used-medicines, when medicated, is safe from the renal toxicity in lab animal model.

다낭신의 중의치료에 대한 임상연구 동향 - CNKI검색을 중심으로 (A Review of Clinical Studies of Chinese Medicine Treatment of Polycystic Kidney Disease Using the CNKI Database)

  • Objective: This research reviews and investigates the trends in recent clinical studies of polycystic kidney disease (PKD) in China. Method: We searched for clinical studies discussing Oriental medicine-based treatments for PKD in the China National Knowledge Infrastructure (CNKI) database. Thirteen clinical articles published from 2001 to 2019 were analyzed. The search focused on the authors, publication year, type of study, purposes of study, method and duration of treatment, evaluation criteria, and results of the selected articles. Results: Of the articles from the database, 9 case series and 4 randomized controlled trials (RCTs) were analyzed. Ten articles used herbal medicine; 4 used herbal medicine for external use. Gamigyejibokryeong-hwan was the most common herbal prescription. The most frequently used herb was Polia Sclerotium (茯苓), and Cnidii Rhizoma (川芎) was employed in all the external uses. All 13 studies confirmed the efficacy of Oriental medicine treatments. Conclusion: 1. Scientifically designed and more varied clinical studies are required to develop treatments for PKD. 2. The current study could be used as basic data in future clinical studies on treatment and further studies of PKD.

한방병원 입원 허혈성 뇌경색 환자에서 한방 의료보험용 엑스산제와 항혈소판제 및 항응고제의 병용투여 현황 (The current status of the combination therapy of frequently used herbal extracts and anti-platelet drug, anti-coagulant drug in ischemic stroke patients hospitalized in oriental medical hospital)

  • Objective: Nowadays the combined use of herbal extracts and western medicines has been prevalent, but concern about its risk is also increasing. Even though the importance of clinical trials is well recognized, there have been only a few studies on the combined use of herbal extracts and western medicines. This study was aimed to examine which herbal extracts and antiplatelets or anticoagulants are most commonly prescribed together to inpatient ischemicstroke patients and investigate the combined prescription rate. Methods: We investigated the most frequently prescribed herbal extracts from two different sources. First, we chose herbal medicine extracts from 2008 Traditional Korean Medicine Utilization Status. Then, among patients who were admitted to Kyung-Hee Oriental Medical Center diagnosed with ischemic stroke, we found patients who were administered with these herbal medicine extracts and among these patients, we investigated how many were concomitantly administered with antiplatelets or anticoagulants. Second, we chose other herbal medicine extracts that were most often prescribed within Kyung-Hee Oriental Medical Center and found patients who were administered with these herbal medicine extracts, then investigated how many of them were concomitantly administered with specific antiplatelets or anticoagulants. Results: The most commonly prescribed herbal medicine extracts among ischemic stroke patients were Ojeok-san, Bojungikgi-tang, Sochungryong-tang and Samsoeum. About 46 to 69 percent were concomitantly administered with either specific antiplatelets or anticoagulants. Conclusions: The most often prescribed herbal medicine extracts in inpatient ischemic stroke patients are Ojeok-san, Bojungikgi-tang, Sochungryong-tang, and Samsoeum. Among patients who were prescribed with Ojeok-san, Bojungikgi-tang, Sochungryong-tang, Samsoeum, patients who were concomitantly administered with either antiplatelets or anticoagulants were about 46% to 69%.

사향약침액(麝香藥鍼液)이 Triton WR-1339로 유발된 흰쥐의 고지혈증(高脂血症)에 대한 실험적(實驗的) 연구(硏究) (Effects of Moschus Herbal-acupuncture on Hyperlipidemia Rats induced by Triton WR-1339)

  • In order to study the effects of Moschus Herbal-acupuncture on hyperlipidemia, we divided the rats into three groups(Normal group, Control group & Sample group) and performed the experimental research. Hyperlipidemia rats were induced by Triton WR-1339(150mg/kg) for 3days. Sample group was injected Moschus Herbal-acupuncture 0.1 ml/200g for 3 days and control group was injected equal dose of saline. And then we measured the amount of serum total cholesterol, triglyceride, phospholipid and HDL-cholesterol. The results were as follows : 1.Moschus Herbal-acupuncture showed decreasing effects on total cholesterol, triglyceride and phospholipid levels in serum significantly(p<0.05). 2.Moschus Herbal-acupuncture showed increasing effects on HDL-cholesterol level in serum significantly(p<0.05). According to the above results, Moschus Herbal-acupuncture showed significant decreasing effects on hyperlipidemia and it is considered that it is appropriate to apply for hyperlipidemia.
