광양만의 해조류에 관한 연구 2. 해조류의 유기염소계 농약잔류량에 대하여 (A Study on the Marine Algae in the Kwang Yang Bay)

  • The appearence of organochlorine pesticide residues in some marinealgae, e.g. Sargassum thunbergii, Ulva pertusa, Codium fragile, and Enteromorpha linza, collected from various sites of Kwang Yang Bay, southern coast of Korea, was surveyed through the year from May 1974 to March 1975. The residues were found to appear in the order to July>September>May>November≒March≒January; $\alpha$-BHC>heptachlor epoxide>dieldrin>aldrin>endrin; and in the range of 0-93 $\mu\textrm{g}$/kg dry matter.

  • 농산물에 잔류되어 있는 16종의 유기염소제 농약들을 추출하여 cleanup한 것을 기체 액체 크로마토그래피법으로 분리, 정량하였다. 5% QF-1 정지상의 분리관을 주로 사용하였으며 3% OV-17관과 DC-200/2.5% QF-1 혼합관을 보조적으로 사용하였다. $^{63}Ni$ electron capture detector를 사용하여 각 농약 성분을 검출하였다. 대부분의 농약에 대한 감응곡선은 직선성을 나타냈고 일부는 약간 휘었으나 정량에는 별 지장을 주지 않았다. 16종의 농약을 13종의 농산물에 가하여 분석한 결과 대부분은 85% 이상의 평균회수율을 보였으며 각 5회 분석결과에 대한 상대평균편차는 12.1% 이하로서 AOAC에서 정한 기준을 만족하였다.

  • 농산물에 잔류되어 있는 16종의 유기염소제 농약들을 기체-액체 크로마토그래피(GLC)로 동시에 정량하기 위하여 필요한 전처리 과정인 용매추출과 불순물 제거(Cleanup) 처리에 대하여 조사 검토 하였다. 농산물로 부터 농약들을 $H_3PO_4$와 물을 가한 강산성(pH 1.5이하)의 actone용액으로 녹여낸 다음, petroleum ether를 분배 용매로 사용하여 농약을 추출시켰다. 추출물에서 용매를 증발시켜 날려보내고 ethylether-petroleum ether (6 : 94) 용리액으로 녹여서 650$^{\circ}$C에서 활성화 시킨 Florisil관을 통과시켜 불순물을 제거 하였다. Petroleum ether에 의한 추출효과는 94%이상으로 AOAC법 보다는 우수 하였고, Florisil관 크로마토그래피법에 의해 불순물이 제거되었다.

  • 충남지역(忠南地域)의 포장(圃場)에서 채취(採取)한 마늘에 대한 유기염소제(有機鹽素劑) 10종(種)의 잔류수준(殘留水準)을 $1982{\sim}1983$년(年)에 걸쳐 조사하였다. ${\alpha}-BHC$는 흔적${\sim}0.004ppm$(평균(平均) 0.002ppm), ${\gamma}-BHC$$0.015{\sim}0.176 ppm$(평균(平均) 0.083 ppm), heptachlor는 $0.001{\sim}0.008 ppm$(평균(平均) 0.006 ppm), aldrin은 부검출(不檢出)${\sim}0.006 ppm$(평균(平均) 0.003 ppm), ${\alpha}-endosulfan$은 흔적${\sim}0.011ppm$(평균(平均) 0.003ppm), heptachlor epoxide는 부검출(不檢出)${\sim}0.004 ppm$(평균(平均) 0.001 ppm), dieldrin은 흔적${\sim}0.002ppm$(평균(平均) 0.001ppm), ${\beta}-endosulfan$은 흔적${\sim}0.008ppm$(평균(平均) 0.001ppm), p,p'-DDD 및 p,p'-DDT는 흔적으로 나타났다.

대전시 유통 과실류의 농약 잔류 실태 및 위해성 평가 (Pesticide Residue Survey and Risk Assessment of Fruits in Daejeon)

  • 대전시 노은 도매 시장에서 채취한 과실류와 6종의 농약잔류량을 조사한 후 이에 대한 위해성 평가를 실시하였다. 전체 120건의 분석시료 중 70.8%가 농약 성분이 검출되었고 그중 1.67%가 잔류허용기준치을 초과하여 검출되었다. 검출 비율이 높은 시료는 사과 복숭아, 포도 등이었다. 검출된 성분에서 살충제는 유기인계, 합성 pyrethroid계, 유기염소계가 비슷한 비율로 검출되었고, 살균제에서는 특히 procymidone(dicarboximide계)과 chlorothalonil(유기염소계)의 검출비율이 높았다. 검출빈도가 높은 성분은 chlorothalonil, chlorpyrifos, fenvalerate, pocymidone, deltamethrin EPN의 순이었다. 위해성을 평가하기 위해서 각 농작물에 대한 농약의 추정섭취량과 ADI를 비교한 결과 추정섭취량의 비율이 1.221% 이하로 나타나 그 영향이 매우 낮은 것으로 판단되었다. 이것은 농약 잔류 허용기준치를 초과한 농작물인 복숭아에서 chlorothalonil, EPN에 대해서도 동일한 결과를 나타내었다.

Interaction Between Persistent Organic Pollutants and C-reactive Protein in Estimating Insulin Resistance Among Non-diabetic Adults

  • Objectives: Chronic inflammation is now thought to play a key pathogenetic role in the associations of obesity with insulin resistance and diabetes. Based on our recent findings on persistent organic pollutants (POPs) including the lack of an association between obesity and either insulin resistance or diabetes prevalence among subjects with very low concentrations of POPs, we hypothesized that POP concentrations may be associated with inflammation and modify the associations between inflammation and insulin resistance in non-diabetic subjects. Methods: Cross-sectional associations among serum POPs, C-reactive protein (CRP), and homeostasis model assessment of insulin resistance (HOMA-IR) were investigated in 748 non-diabetic participants aged ${\geq}20$ years. Nineteen types of POPs in 5 subclasses were selected because the POPs were detectable in ${\geq}60%$ of the participants. Results: Among the five subclasses of POPs, only organochlorine (OC) pesticides showed positive associations with CRP concentrations, while polychlorinated biphenyls (PCBs) showed inverse associations with CRP concentrations. There were statistically significant interactions between CRP and OC pesticides and between CRP and PCBs, in estimating HOMA-IR (P for interaction <0.01 and <0.01, respectively). CRP was not associated with HOMA-IR among subjects with low concentrations of OC pesticides or PCBs, while CRP was strongly associated with HOMA-IR among subjects with high concentrations of these POPs. Conclusions: In the current study, OC pesticides were associated with increased levels of CRP, a marker of inflammation, and both OC pesticides and PCBs may also modify the associations between CRP and insulin resistance.

A Single Cell Multimedia Fate Model for Endocrine Disrupting Chemicals

  • To understand environmental paths of the transport and accumulation of endocrine disrupting chemicals (EDCs), a single cell multimedia fate model has been constructed and evaluated. The EDCs of concern were PAHs, Organochlorine Pesticides (OCPs), PCBs, Alkyl phenols, and phthalates. An evaluation model was designed for the multimedia distribution, including air, water, soil, sediment and vegetation. This model was verified using reported values and via monitoring data. Based on collected data, the distribution trends of EDCs with respect to environmental media were analyzed. Those results have applied to the model for the prediction of the spatial and temporal distribution of EDCs in Seoul. Especially, phenol compound, phthalates, PAHs, PCBs and organochlorine pesticides were estimated and the model was verified. This model was successfully conducted to environmental media, such as air (vapor and suspended particles), soils (forest soil, bare soil, and cement-concrete covered soil), water (dissolved and suspended solids), sediment, trees (deciduous and coniferous). The discrepancies between the model prediction and the measured data are approximately within or near a factor of 10 for the PAHs of three rings through that of six rings, implying that multimedia distribution of the PAHs could be predicted with a factor of 10. Concerning about the air equilibrium may be assumed, a fugacity at steady state is similar in all environmental media. Considering the uncertainties of this model, the use of equilibrium models may be sufficient for assessing chemical fates. In this study, a suggestion was made that modeling and estimation of chemicals in environmental multimedia be rigorously evaluated using the measured flux data. In addition, these data should be obtained, for example, from the precise and standardized inventory of the target chemicals. The model (EDC Seoul) will be refined in an on-going research effort and will be used to support decision-making concerning the management of EDCs.

Ecological and human health risk from polychlorinated biphenyls and organochlorine pesticides in bivalves of Cheonsu Bay, Korea

  • Cheonsu Bay, one of the most important in Korea as a coastal fishery is a semi-enclosed bay that is surrounded by large farmlands and industrial areas. This coastal environment has been affected by anthropogenic pollutants, such as polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs). The objectives of this study were to investigate the distribution of PCBs and OCPs in sediment, Manila clams, and mussels from Cheonsu Bay; the accumulation pattern of these chemicals in these bivalves in relation to seasonal changes; and the ecological risk from sediments and the risk to the Korean population from the consumption of these bivalves. The levels of ${\Sigma}PCBs$, ${\Sigma}DDT$, and ${\Sigma}HCHs$ were 69.3-109, 40.3-49.3 and 6.25-17.8 ng/g lipid in Manila clams, and 70.6-159, 38.6-102 and 9.00-13.5 ng/g lipid in Mussels. Significant seasonal variations in PCBs and OCPs concentrations were observed in the two bivalves, suggesting that the accumulation of PCBs and OCPs in these species is related to their spawning times. The dietary intake of these two bivalves and the resulting lifetime cancer risk (LCR) and non-cancer risk were calculated for the human population. The consumption of these bivalves seemed to be safe in relation to human health with negligible LCR and non-cancer risk.

한국인의 지방조직에서 검출되는 유기염소제 농약의 에스트로겐 활성 (Estrogenic Potentials of Organochlorine Pesticides which Detected in Korean Adipose Tissue)

  • 유기염소계 농약은 화학적으로 안정하고, 지용성이 크며 체내 대사에 대해 저항성을 가지고 있어서 인체 및 생태계의 생물체 중 축적성이 매우 크다. 또한 대부분 내분비계 장애물질로 분류되어 있어 관심의 대상이 되고 있다. 유기염소계 농약은 잔류성으로 인하여 사용이 금지되었음에도 불구하고 인체에서 검출되고 있으며 한국인의 지방조직을 분석한 결과 9종의 유기염소계 농약이 주로 축적되어 있음이 보고된 바 있다. 본 연구에서는 이들 유기염소계 농약의 에스트로겐 활성을 MCF-7 BUS cell을 이용한 E-screen assay competitive binding assay 및 pS$_2$ gene experession assay에 의해 조사하였다. o,p'-DDT,p, p'-DDT,p,p'-DDD,p,p'-DDE등 4종의 유기염소계 농약은 에스트로겐 수용체에 대한 ligand의존적인 작용기전에 의해 에스트로겐 활성을 가지며, $\alpha$-, $\beta$-, ${\gamma}$-, $\delta$-BHC, dieldrin등 5종의 유기염소계 농약은 lingand-비의존적 작용기전에 의해 에스트로겐 활성을 보였다. 또한 이들 유기염소계 농약을 혼합 투여하여 에스트로겐 활성을 관찰한 결과 DDT류의 경우에는 단독투여시 보다 그 대사체와 혼합 투여할 때 에스트로겐 활성에 상승적 효과가 나타났으며 o,p'-와 p,p'-DDT의 두 이성질체를 혼합 투여할 경우가 단독 투여시 보다 상승적 효과가 나타났다. 따라서 지방조직에서 검출되는 유기염소계농약은 상호작용에 의해 개별 물질이 나타내는 내분비계장애작용 보다 실제로는 강한 효과를 나타낼 것으로 추정되었다.

Enhanced Biodegradation of Lindane Using Oil-in-Water Bio-Microemulsion Stabilized by Biosurfactant Produced by a New Yeast Strain, Pseudozyma VITJzN01

  • Organochlorine pesticide residues continue to remain as a major environmental threat worldwide. Lindane is an organochlorine pesticide widely used as an acaricide in medicine and agriculture. In the present study, a new lindane-degrading yeast strain, Pseudozyma VITJzN01, was identified as a copious producer of glycolipid biosurfactant. The glycolipid structure and type were elucidated by FTIR, NMR spectroscopy, and GC-MS analysis. The surface activity and stability of the glycolipid was analyzed. The glycolipids, characterized as mannosylerythritol lipids (MELs), exhibited excellent surface active properties and the surface tension of water was reduced to 29 mN/m. The glycolipid was stable over a wide range of pH, temperature, and salinity, showing a very low CMC of 25 mg/l. Bio-microemulsion of olive oil-in-water (O/W) was prepared using the purified biosurfactant without addition of any synthetic cosurfactants, for lindane solubilization and enhanced degradation assay in liquid and soil slurry. The O/W bio-microemulsions enhanced the solubility of lindane up to 40-folds. Degradation of lindane (700 mg/l) by VITJzN01 in liquid medium amended with bio-microemulsions was found to be enhanced by 36% in 2 days, compared with degradation in 12 days in the absence of bio-microemulsions. Lindane-spiked soil slurry incubated with bio-microemulsions also showed 20-40% enhanced degradation compared with the treatment with glycolipids or yeast alone. This is the first report on lindane degradation by Pseudozyma sp., and application of bio-microemulsions for enhanced lindane degradation. MEL-stabilized bio-microemulsions can serve as a potential tool for enhanced remediation of diverse lindane-contaminated environments.
