A Study on Environment Characteristics of the Angelica gigas Nakai Population (참당귀(Angelica gigas Nakai) 개체군의 환경 특성 분석)

  • This study was carried out to investigate vegetation structure and soil properties of Angelica gigas population distributed in Jeombongsan, Bangtaesan, Odaesan, Gyebangsan and Jirisan. From August 2007 until September 2008, 5m${\times}$5m quadrat was established in native area of Angelica gigas in order to record a dominants and coverage, and soil factors at 20 sites. It was found that the altitude in the distributed areas for Angelica gigas population was 710m or more. Angelica gigas population was classified into Parasencio auriculata var. kamtschatical dominant population, Pseudostellaria palibiniana dominant population, Isodon excisus dominant population and typical dominant population. In the site of study, soil pH, electrical conductivity, soil organic matter, available phosphorous, and exchangeable potassium, exchangeable calcium, exchangeable magnesium, exchangeable natrium concentration and total nitrogen were ranged from 5.1~6.2, 0.1~0.79dS/m, 2.21~22.11%, 9.56~37.97mg/kg, 1.85~23.88cmol+/kg, 0.21~1.18cmol+/kg, 1.07~5.09cmol+/kg, 0.04~0.14cmol+/kg and 0.09~1.04% respectively. The Parasencio auriculata var. kamtschatical dominant population was found in area of high altitude and had high slope degree. But Pseudostellaria palibiniana dominant population was found in area of low altitude and low slope degree. Isodon excisus dominant population was found to have higher soil pH and exchangeable potassium concentration than the other dominant population.

M-dephanox Process with Rotating Biological Contactor (RBC) in Nitirification Reactor (회전원판형 질화조를 이용한 M-dephanox 공정)

  • This study was focused on improving nitrification efficiencies of M-dephanox (Modified-Dephanox) process. Rotating biological contactor (RBC) was used instead of floating sponge type media in nitrification reactor. High ammonia removal efficiencies were observed in nitrification reactor, regardless of organic loading from contactor of M-dephanox process. Denitrification efficiencies were also increased to maintain low $NO_3-N$ concentration in effluent. This enhanced phosphate release in anaerobic contactor and resulted in high removal efficiencies of phophorus. Average removal efficiencies of $TCOD_{Cr}$ and $SCOD_{Cr}$ were 93.8% and 81.6%, respectively, while those of TKN and ${NH_4}^+-N$ were 80.9% and 74.4%, respectively. As for phosphorous treatment, the average removal efficiencies of TP and OP were 94.7% and 94.3%, respectively. Also, effect of operating temperature on nitrogen removal was examined. Average removal efficiency of TN was 65.8 % at $15^{\circ}C$ or below (at average temperature of $13.3^{\circ}C$), while that was 82.8% at $15^{\circ}C$ or above (at average temperature of $21.9^{\circ}C$).

Effect of Soil Mixture on the Growth of Chrysanthemum zawadskii var. latilobum KITAMURA (분화용 배양토가 구절초의 생육에 미치는 영향)

  • This study was conducted to investigate the combination of proper culture medium for differentiation culture of Chrysanthemum zawadskii var. latilobum KITAMURA. The combinations of culture medium were consisted with an inorganic materials such as vermiculite, perlite, saprolite, sand and upland top soil, and with an organic materials such as peat-moss, leaf mold and compost. For the plant growth characteristics, the plant height, number of tiller, root length and root weight in the combination of leaf mold was relatively great as compared to the treatment with peat-moss and compost. It was considered that humus might be contributed to improve the physical properties of soil as well as the sequential nutrient supply. For the combination of peat-moss, the plant growth was not good because of not sufficient nutrient supply, Also, the plant growth in the treatment of compost was so bad because of enhancement of culture medium pH 8.9 and increasement of phosphorous content.

Performance Evaluation of Advanced Municipal Wastewater Treatment by Advanced Phase Isolation Ditch (APID) Process (Advanced Phase Isolation Ditch 공정에 의한 하수 고도처리 성능평가)

  • Advanced Phase Isolation Ditch (APID) process was studied to develop economic retrofitting technology, for the plants where retrofitting of common activated sludge process is required. In this study, to evaluate the effluent BOD, SS, T-N, and T-P concentrations as process capable and stable parameters for treating municipal wastewater, a demonstration plant was installed and operated in the existing sewage treatment plant of P city. During this study, the average effluent BOD, SS, T-N, and T-P concentrations were 4.56, 5.20, 9.30, and 1.75 mg/L at the conventional mode and 3.95, 3.17, 7.65, and 1.18 mg/L at the modified mode. The modified mode (BOD: 3.69, SS: 3.19, T-N: 1.27, and T-P: 0.69) increased the process capability more than the conventional mode (BOD: 1.80, SS: 1.05, T-N: 2.17, and T-P: 0.15) in this study. If process capability over 1.0, this process is capable and stable to treat wastewater. Therefore, newly developed APID process with modified intermittent aeration mode can be one of the useful processes for stable organic matter and nutrients removal.

Evaluation of Operation Parameters for the Removal of Algae by Electro-Coagulation (수계 내 조류 제거를 위한 전기응집 운전 특성 평가)

  • Electro-coagulation experiments were conducted with aluminum (Al) or iron (Fe) electrode in order to determine the optimal electrode material and operation conditions for algae removal. Al electrode showed higher removal rate of algae than Fe electrode because Al flocs have positive surface charges which electrostatically attract algae species having negative surface charges. Removal rate of algae and total phosphorous (T-P) was increased as current density and electrode area increases. It was also found that initial pH with neutral range was optimum for T-P removal by electro-coagulation. Bench-scale continuous flow experiments consisted of electro-coagulation reactor, agitation tank and settling tank were conducted. In electro-coagulation reactor, a large fraction of Al flocs were distributed to scum layer, due to the gas bubbles generated by electrolysis reaction. In agitation tank, most of Al flocs were settled and the optimal mixing intensity was found to be 50 rpm to achieve good settleability. The removal rate of algae was about 90-95%. Additionally, the removal rate of the T-P and COD was observed to be $73.8{\pm}8.0%$ and $75.0{\pm}3.8%$, respectively. Meanwhile, the removal rate of total nitrogen (T-N) was relatively low at only 24%.

Synergistic effect of co-inoculation with phosphate-solubilizing bacteria

  • The synergistic effect on phosphate solubilization of single- and co-inoculation of two phosphate solubilizing bacteria, Burkholderia anthina PSB-15 and Enterobacter aerogenes PSB-16, was assessed in liquid medium and green gram plants. Co-inoculation of two strains was found to release the highest content of soluble phosphorus ($519{\mu}g\;mL^{-1}$) into the medium, followed by single inoculation of Burkholderia strain ($492{\mu}g\;mL^{-1}$) and Enterobacter strain ($483{\mu}g\;mL^{-1}$). However, there was no significant difference between single inoculation of bacterial strain and co-inoculation of two bacterial strains in terms of phosphorous release. The highest pH reduction, organic acid production, and glucose consumption were observed in the culture medium co-inoculated with PSB-15 and PSB-16 strains rather than that of single inoculation. Based on the plant growth promotion bioassay, co-inoculated mung bean seedlings recorded 9% and 8% higher shoot and root growth, respectively, compared to the control. Therefore, in conclusion, co-inoculation of the strains B. anthina and E. aerogenes displayed better performance in stimulating plant growth than inoculation of each strain alone. However, considering the short assessment period of the present study, we recommend engaging in further work under field conditions in order to test the suitability of these strains as bio-inoculants.

Biological Improvement of Reclaimed Tidal Land (I) Desalination Effects of Saline Soil by the Growth of certain Halophytes (해안간척지 토양의 생물학적 토성개량에 관한 연구 (제1보) 수종 염생식물에 의한 간 탁지토양의 제염효과에 대하여)

  • Korea has a lots of margin for security of farm land from her coastal region. The area of saline soil may be reached about 10% of present farm land if the reclamation works are finished. This paper was conducted as a part of studying the possibilities of desalination of saline soil through the experiment of some halophytes. The halophytes in this works were Salicornia herbacea L., Suaeda glauca Bunge, chenopodium acuminatum Willd, and Scirpus triquerter L. Of the above halophytes, Salicornia was proved the most effective plant for desalination of saline soil referring to the following results; 1) The seasonal uptake of chloride by Salicornia was the highest of all. However, the general tendencies of all plants showed a decrease on August. 2) Salinity of soil showed the lowest value on the site where Salicornia was grwon densely. Comparing the other sites grouped by age of saline soil with the above site, the salinity of rice-paddy (10 years after reclamation) is similar to those of the site wehre Salicornia were as well as the 50 cm below the surface soil. 3) The maximum water holding capacity of surface soil appeared in the site of Salicornia, but in 50 cm below the surface, the maximum water holding capacity are almost on equat terms having no connection with the age of saline soil. Soil pH, other chemical compositions such as organic matter, magnesium, potassium, phosphorous, and nitrate were determined to elucidate the relationship between the changes of soil properties and chemical uptakes by certain halophytes. It is assumed that the above chemical compositions are frequently affected by the factors such as coastal circulation of salts, exchangeable base, microbial growth, climatic conditions, and irrigation of water.

Synergistic Phosphate Solubilization by Burkholderia anthina and Aspergillus awamori

  • Single or co-inoculation of phosphate solubilizing bacterial and fungal strains (Burkholderia anthina and Aspergillus awamori respectively) was performed separately to assess their synergistic and antagonistic interactions and the potential to be used as bio-inoculants. Co-inoculation was found to release the highest content of soluble phosphorus (1253 ${\mu}g\;ml^{-1}$) into the medium, followed by single inoculation of fungal strain (1214 ${\mu}g\;ml^{-1}$) and bacterial strain (997 ${\mu}g\;ml^{-1}$). However, there was no significant difference between single inoculation of fungal strain and co-inoculation of fungal and bacterial strain in terms of the phosphorous release. The highest pH reduction, organic acid production and glucose consumption were observed in the sole A. awamori inoculated culture medium. According to the plant growth promotion bioassays, co-inoculation of the microbial strains resulted in 21% and 43% higher shoot and root growth of the mung bean seedlings respectively as compared to the respective controls. Therefore, co-inoculation of B. anthina and A. awamori showed better performance in stimulating plant growth than that in inoculation of each strain alone. However, assessment period of the present study being short, we recommend in engaging further experimentation under field conditions in order to test the suitability of the strains to be used as bio-inoculants.

Optimized Culture Condition and Enzyme Activity of the Mycelia of Clavicorona pyxidata (좀나무싸리버섯(Clavicorona pyxidata)의 균사체 최적 배양조건 및 세포효소활성 특성)

  • The culture conditions for the enhanced mycelial of Clavicorona pyxidata DGUM 29005 were investigated. The optimal temperature and pH for the mycelial growth were $24^{\circ}C$ and 5.0, respectively. It was shown that trehalose was the best supplement of carbon sources in Czapek-Dox medium as a minimal medium for enhanced mycelial growth. In general, inorganic nitrogen sources were better than organic ones for mycelial growth. Calcium nitrate was the best out of the inorganic nitrogen test. The appropriate phosphorous and vitamin were $Na_2HPO_4$ and p-aminobenzoic acid, respectively. After the mycelial of C. pyxidata DGUM 29005 was cultivated at $24^{\circ}C$ for 20 days in MEM broth(pH 5.0), the specific activities of both exomycelial and endomycelial enzymes were determined. Among the exomycelial enzyme assayed, the specific activity of laccase was much higher than those of other enzymes. However, little or no enzyme activities of ${\alpha}$-amylase, chitinase, lipase and pretense were found.

Comparison to Soil Environment of Tricholoma matsutake and Sarcodon aspratus at Uljin Sokwang-ri Pinus densiflora for. erecta Uyeki Forest (울진 소광리 금강소나무림의 송이발생지와 능이발생지의 토양환경 비교)

  • This study was carried out in order to produce useful material for the forest multiple use and forest protection by physico-chemical soil analysis of studied area in Sokwang-ri Forest Genetic Resource Protection Forest which was divided into in standard plots include Tricholoma matsutake and Sarcodon aspratus production forest. The result of physico-chemical soil analysis represented as following. The soil type of T. matsutake production forest was Dry brown forest soil(B1), while on the other hand the soil type of S. aspratus production forest was Moderately moist brown forest soil(B3). Between T. matsutake and S. aspratus production forest did not result in significant changes in soil pH(5.22-5.60) and soil depth(47cm), but available phosphorus, carbon, and nitrogen contents were different results. CN ratio of the fairy ring of T. matsutake was quite lower than that in S. aspratus production forests, which indicated that T. matsutake production forest was built up in the relatively immature soils which contain little organic matter. Generally, it was predicted that Pinus densiflora for. erecta forest succeeded to deciduous tree forest in stable soil environments. To conserve these T. matsutake and S. aspratus production forest, the contents of available phosphorous and exchangeable cation should be increased by continuous soil environment management and it should be established the secondary growth forests of old aged Pinus densiflora for. erecta trees as soon as possible.
