Present Status and Problems of Chemical Seed Treatment of Seedborne Diseases (종자소독의 현황과 문제점)

  • A wide variety of pathogens are known io be seedborne, carried either as infectious mycelium internally or as contaminants on the seed coat. When seed is infected with a pathogen, the seed nay be rendered nonviable or it may remain viable but produce weak seedling. In some cases, the Infected seedling nay not be severely weakened, but nay serve as a source of primary inoculum within a community of plants. A recent problem nay be the dissemination of seedborne pathogens occurring as a result of the massive movements of seed, as a part of the 'Green revolution' Disease of great danger to agriculture may be introduced with seed from other parts of world. Seed treatment with organic mercury compounds in liquid form had become popular since about 1955. Organic mercury compounds contributed considerably to the increase in production of many crops and vegetables. In 1975, however, the use of organic mercury compound was forbidden because of doubts regarding their residual mammalian toxicity in agricultural products. Benomyl-thiram mixture, thiophanate methyl-;hiram mixture and TCMB have now been registered as seed disinfectants for the use of rice blast, brown spot and Bakanae disease. Oxathiinsthiram mixture has been registered as seed disinfectant for barley and wheat loose smut and leaf stripe of barley. Agricultural techniques have made such rapid progress that the nursery methods changed from the use of paddy nursery to box nursery designed for machine-transplanting. The spread of rice transplanting machines has caused increase of seedborne diseases. Among seedborne diseases, Bakanae disease has remarkably increased and causes much damage recently. In order to counter this trend, seed disinfectants must also be diversified. First, effective non-selective disinfectants need to be developed, and second, appropriate control methods always need to be prepared in parallel with the development of new techniques for cultivation.

A Study on the Separation of Mercury from Spent Mercury Batteries (단추형 폐수은 전지로부터 수은 분리에 관한 연구)

  • Mercury in spent button type batteries can be separated and recovered with vacuum distillation method. It was found that mercury in the battery began to distill at $150^{\circ}C$ and organic substanced like a packing material was decomposed at$ 300^{\circ}C$. More than 99.9% of mercury contained in the battery was distiled and separated at about $250^{\circ}C$ and 20 torr with 8 hours' reaction time. The dissolution tests of the residue after distillation showed that mercury concentration in the solution were lower than 5 ppb and this values satisfied the environ-mental condition. Also as the furnace heating rate was above $15^{\circ}C$/min, it was found that the spent battery was destroyed because of increased pressure in the battery inside.

  • Methylmercury, an organic derivative, is the principal form of mercury that biomagnifies and causes neurodegenerative symptoms in animals. In recent years, living modified organism (LMO) resulting from biotechnology has played a highly visible and controversial role. Despite the potential benefits of this technology, public concerns have been raised about the environmental risk of LMO. The concern on the risk from LMO release has urged efforts to evaluate and manage the risks of the LMO. To build up the capacity building of risk assessment method for LMO used environmental remediation, we engineered Solanum nigrum L, expressing the modified bacterial gene, merB, encoding organomercurial lyase. Two independently isolated transgenic lines produced merB RNA. Transgenic Solanum nigrum leaf discs expressing merB gene showed organic mercury resistance, forming shoots well on growth medium containing $0.5{\mu}M$ methylmercury (II) chloride and $1{\mu}M$ phenylmercuric acetate while control plants breached. Transgenic merB seeds germinated and grew on growth medium containing $2{\mu}M$ methylmercury (II) chloride and phenylmercuric acetate. The merB transgenic plants will be used for risk assessment of natural environment.

Simultaneous Determination of Mercury and Arsenic by Reductive Vapor Generation-ICP-AES (환원 증기 발생법-유도결합 플라즈마 원자방출 분광계를 이용한 수은과 비소의 동시 분석법)

  • Simultaneous determination of mercury and arsenic has been studied by reductive vapor generation-ICP-AES. Samples were digested with a microwave digestion system and extracted with acids. Reductive vapor generation was carried out with 6N HCI and 2% $NaBH_4$. Detection limit of Hg and As are found to be 0.06 ppb and 0.08 ppb, respectively. Measured values of Hg and As in inorganic samples agree well with reference value of SRMs, but the recovery of As from organic samples is obtained approximately 80% of the reference values.

Effect of Allopurinol on Methylmercuric Chloride-Induced Cytotoxicity in $C_6$ Cultured Glioma Cells

  • It is demonstrated that inorganic mercury has cytotoxic effect on glial cells. Recently, oxygen radicals is involved in methylmercuric chloride (MMC)-induced cytotoxicity. But, the toxic mechanism of MMC is left unknown. The purpose of this study was to examine the cytotoxicity of MMC on $C_6$ glioma cells. The cytotoxicy was measured by cell viability using XTT assay in $C_6$ glioma cells. Colorimetric assay is regarded as a very sensitive screening method for the determination of the cell viability on various agents. In this study, MMC decreased cell viability according to the dose- and time dependent manners after $C_6$ glioma cells were grown with various concentrations of MMC for 48 hours. In the protective effect of allopurinol on MMC-induced cytotoxicity, allopurinol was effective in the prevention of MMC-induced cytotoxicity in these cultures. These results suggest that MMC has highly cytotoxic effect on $C_6$ glioma cells by the decrease of cell viavility, and free radical scavenger such as allopurinol was effective on organic mercury-induced cytotoxicity in these cultures.

  • The energy levels are very important to investigate properties of organic dye materials. These values of energy levels can be calculated and compared with absorption spectra, cyclic voltammetric measurement and computer simulative calculation. In this study, absorption and emission changes were observed by complexation between rhodamine 6G based dye and mercury. This is related to spirolactam ring system of rhodamine 6G based dye. According to structural change of this dye, HOMO and LUMO energy levels were investigated and determined by their values with different approaches.

  • The solvent extraction of an inorganic trace mercury in sea water samples was studied using zinc diethyldithiocabamate $Zn(DDC)_2$ as chelating agent. The $Zn(DDC)_2$ which maintained the stability of DDC- in the acidic aqueous solution in the course of extraction was synthesized from NaDDC and $ZnSO_4$ in this laboratory. The trace of mercury(Ⅱ) was extracted at pH 3.0 from 100 ml of sea water into 10ml of chloroform containing 0.05 M $Zn(DDC)_2$ by shaking for 5 minutes. And from the organic phase, the $Hg(DDC)_2$ was back-extracted into 10ml of 1 to 1 mixed acid of each 3% (v/v) nitric acid and hydrochloric acid by shaking for 25 minutes. The mercury back-extracted was determined by a cold vapor atomic absorption spectrophotometry. The trace mercury(Ⅱ) was so successfully extracted that this procedure could be applied to its determination in the sea water. That is, the recoveries of mercury in two kinds of samples into which as given amount of Hg(Ⅱ) was spiked were 90.0% and 93.3%, respectively.

  • This is a study to determine the content of the mercury compound and the distribution of microorganisms in the waste waters flowing from the chemical plants in the Ulsan area (at 4 stations). The summary of the result of this study is as follows: 1. The content of the mercury compound has ranged from non-detection to 0.075 ppm with an average of 0.03 ppm. The highest content has been detected from the water at station A. 2. As for the distribution of the microorganisms, one species each of bacteria, Mucor, Aspergillus, Cladosporium, and Trichothecium (T. roseum); 4 species of Penicillium; and 3 species of Sterile hyphae; a total of 12 species have been isolated. 3. The following results have been found with regard to the growth of these microorganisms in terms of the content of the mercury compound. In the case of an inorganic mercury compound, most of the microbes can grow in water with a content of up to 10 ppm. Pe. sp No. 1 particularly can grow even in 50 ppm. In the case of an organic mercury compound, the growth of the microbes seems to be somewhat restrained even in 2 ppm.

Quality Characteristics and Antioxidant Potential of Seeds of Native Korean Persimmon Genotypes

  • Persimmon seeds contain considerable amounts of minerals, amino and organic acids, natural antioxidants and phenolic compounds. The objective of this study was to investigate quality characteristics and antioxidant potential of Korean persimmon seeds. The pH (4.88-4.94), color values, contents of minerals, free amino acids, organic acids, and phenolic compounds and DPPH free radical scavenging potentials of persimmon seed extracts significantly (p < 0.05) varied with the genotypes. This study showed that the seeds could be used as a source of different mineral elements (47.14-85.07 mg/kg) without any measureable amount of heavy metals such as arsenic, cadmium, lead and mercury. Similarly, considerable amounts of organic (1550.13-2413.08 mg/kg) and essential amino (50.85-54.03 mg/kg) acids and total phenolic compounds ($1227.91-1307.78{\mu}g$ gallic acid equivalent/g) were also found in the seed extracts, indicating their potential food value as a natural antioxidant. Results of the present study imply that prethanol-A, a food preservative, can be used as an effective extraction to obtain the minerals, organic and free amino acids, and phenolic compounds from the persimmon seeds, which possess a big potential to be commercially used in food, cosmetic and pharmaceutical industries.
