Growth and Cultural Characteristics of Cordyceps cardinalis Collected from Korea

  • Cordyceps cardinalis was reported in Japan and the USA in 2004, and its fruiting bodies have recently been cultured in Korea. Herbarium specimens preserved at the Cordyceps Research Institute, Mushtech, Korea were revised and identified as C. cardinalis, based on morphological characters and conidial structures. Most of the C. cardinalis specimens were collected from Mt. Halla in Jeju-do. The effects of various nutritional sources and environmental conditions such as temperature and pH on mycelial growth of C. cardinalis were studied. Oatmeal agar, Martin's peptone dextrose agar, and Schizophyllum (mushroom) genetics complete medium plus yeast extract resulted in the best mycelial growth. Among carbon sources, cereals, and nitrogen sources, maltose, oatmeal, and peptone resulted in the best mycelial growth respectively. Mineral salts helped to increase growth rate but only resulted in thin mycelial density, similar to water agar. A temperature of $25^{\circ}C$ and a pH of 7 resulted in the highest mycelial growth. Based on these results, a Cordyceps cardinalis composite medium (CCM) was formulated with 1% maltose, 2% oatmeal, 1% peptone, and 2% agar. Use of the CCM resulted in slightly better mycelial growth than that of other commonly used agar media. Only organic nitrogen sources imparted a reddish pigmentation to the agar media, but this character diminished after several subcultures. A 7 day culture duration resulted in the best mycelial growth.

Enhanced pectinase and β-glucosidase enzyme production by a Bacillus subtilis strain under blue light-emitting diodes

  • Bacillus subtilis B22, a chemotrophic and aerobic bacterial strain was isolated from homemade kimchi, identified by 16S rRNA gene sequencing. B22 was primarily screened by biochemical, carbon source utilization tests. B22 was used to produce pectinase and ${\beta}$-glucosidase by submerged fermentation under different light sources. B22 was incubated in pectin media and basal media (pH 7.0) under blue, green, red and white light-emitting diodes (LEDs), fluorescent white light, and in darkness at $37^{\circ}C$, orbital shaker 150 rpm for 24 hours. Fermentation under blue LEDs maximized pectinase production ($71.59{\pm}1.6U/mL$ at 24 h) and ${\beta}$-glucosidase production ($56.31{\pm}1.6U/mL$ at 24 h). Further, the production of enzyme increased to pectinase ($156{\pm}1.28U/mL$) and ${\beta}$-glucosidase ($172{\pm}1.28U/mL$) with 3% glucose as a carbon source. Activity and stability of the partially purified enzymes were higher at pH 6.0 to 8.0 and $25-55^{\circ}C$. The effect on the metal ions $Na^+$ and $K^+$ and (moderateactivity) $Mn^{2+}$ and $Ni^{2+}$ increased activity, while $Hg^{2+}$, $Cu^{2+}$, $Fe^{2+}$, and $Fe^{2+}$ inhibited activity. EDTA, phenylmethylsulfonyl fluoride and 5,5-dithiobis (2-nitrobenzoicacid) reduced activity, while tetrafluoroethylene and 1,10-phenanthroline inhibited activity. The amylase was highly tolerant of the surfactants TritonX-100, Tween-20, Tween-80 and compatible with organic solvents methanol, ethanol, isoamylalcohol, isopropanol, t-butylalcohol and the oxidizing agents hydrogen peroxide, sodium perborate and sodium hypochlorite, although potassium iodide and ammonium persulfate reduced activity. These properties suggest utility of pectinase and ${\beta}$-glucosidase produced by B. subtilis B22 under blue LED-mediated fermentation for industrial applications.

Effects of Hydrocarbon Additions on Gas-liquid Mass Transfer Coefficients in Biphasic Bioreactors

  • The effects of aliphatic hydrocarbons (n-hexadecane and n-dodecane) on the volumetric oxygen mass transfer coefficient $(k_L\;a)$ were studied in flat alveolar airlift reactor and continuous stirred tank reactors (CSTRs). In the flat alveolar airlift reactor, high aeration rates (>2vvm) were required in order to obtain efficient organic-aqueous phase dispersion and reliable $k_La$ measurements. Addition of 1% (v/v) n-hexadecane or n-dodecane increased the $k_La$ 1.55- and 1.33-fold, respectively, compared to the control (superficial velocity: $25.8{\times}10^{-3}m/s$, sparger orifice diameter: 0.5 mm). Analysis of the gas-liquid interfacial area a and the liquid film mass transfer coefficient $k_L$ suggests that the observed $k_La$ increase was a function of the media's liquid film mass transfer. Addition of 1% (v/v) n-hexadecane or n-dodecane to analogous setups using CSTRs led to a $k_La$ increase by a factor of 1.68 and 1.36, respectively (superficial velocity: $2.1{\times}10^{-3}m/s$, stirring rate: 250 rpm). These results propose that low-concentration addition of oxygen-vectors to aerobic microbial cultures has additional benefit relative to incubation in purely aqueous media.

전기영동 디스플레이 패널용 OTFT-하판 제작 연구 (Study on OTFT-Backplane for Electrophoretic Display Panel)

  • 본 논문에서는 플라스틱 기판에 OTFT를 스위칭 소자로 사용하여 유연한 EPD 패널을 제작하였다. OTFT의 채널 폭과 길이의 비(W/L)는 EPD의 응답속도를 고려하여 15이상으로 설계를 하였다. 게이트전극은 Al, 절연층은 cross-linked PVP, 반도체층은 펜타센, 중간층은 PVA/Acryl를 사용하였다. 플라스틱 기판은 보호층 처리를 통하여 열처리 공정 시 발생하는 입자를 제거하였고, 거친 표면을 평탄화하였다. 반도체층의 크기는 게이트 전극 보다 작도록 제한하여 누설전류를 줄일 수 있었다. EPD-상판과 OTFT-하판 사이에 픽셀전극을 삽입하고 또한 OTFT-하판을 보호하기 위하여 PVA/Acryl로 구성된 중간층을 상빙하였다. 완성된 OTFT-하판에서 OTFT의 이동도는 $0.21cm^2/V.s$, 전류점멸비(Ion/Ioff)는 $10^5$ 이상의 성능을 보였다.

분산지역의 생활하수 처리를 위한 고친수성 Biofilter 개발 (Development of high-hydrophilic Biofilter for Decentralized Regions and Rural Communities)

  • The feasibility of the high-hydrophilic biofilter was examined for application in rural wastewater treatment in Korea. The intermittent trickling biofiter was developed for wastewater treatment of media and examined instantaneous wetting water and immersional wetting water. Melamin foam absorbed 120 times it's weight in water and maintained wetting status than other materials. These characteristics are improvement for application in rural areas showing large variance of amount of influent. The biofilter process was effective in treating organic pollutants; mean removal efficiencies of $BOD_5$ and TSS were above 80%. The average SS concentrations of effluent was showed below $10mg/L^{-1}$ and meet guidelines in special regions, however, the average concentration of $BOD_5$ was about $20mg/L^{-1}$. The removals of T-N and T-P were relatively less effective and removal efficiencies were below 40%. It might meet the guidelines for T-P because of low levels of influent T-P concentration. However, the T-N concentration were too high and further treatment is required. The effluent concentration of $NH_4-N$ showed a significant reduction rate about 43.8%, but part of $NH_4-N$ was transformed to $NO_2-N$ and $NO_3-N$ inside the biofilter through nitrification process. The effluent concentration of org-P was removed about 78.8% of influent concentration by filtration. Considering stable performance and effective removal of pollutant in wastewater, low maintenance, and cost-effectiveness, the hydrophilic biofilter system was thought to be an effective and feasible alternative for decentralized rural areas.

제주도 삼양 수원지 RO 시설 도입 연구 (A Study on the Introduction of RO Facility for Jeju Samyang Water Source)

  • Pollutants removal efficiency in pretreatment(GAC filter, multi-media filter, disk filter) and RO facilities was investigated for the Jeju Samyang spring water source where raw water intake has been stopped due to sea water intrusion. In addition, preliminary feasibility analysis was conducted between RO and groundwater intake systems. Turbidity removal in 4 different pretreatment processes was less than 25% due to low concentration of turbidity(i.e., less than 0.21 NTU), while multi-media filter is recommended for the pretreatment facility based on the low organic content in raw water as well as cheaper operation and maintenance cost. The average concentration of $Cl^-$ in raw water was 691.4 mg/L, while that of RO permeate was 9.1 mg/L(i.e., removal efficiency was 98.4%). In addition, TDS removal efficiency was 98.1%, which was quite high. The production cost for RO system($Q=4,000m^3/d$) was $362.1won/m^3$ considering installation, operation and maintenance cost for 30 years. While that of groundwater was $262.6won/m^3$ which was low compared to the RO system. However, it is recommended to introduce RO system for Samyang water source rather than new groundwater development because Samyang water source has been discharged to the sea without any usage, while groundwater can be used for other purpose as a sustainable water source.

능이 균사체의 액체배양을 위한 배양 인자 (Liquid Culturing Factors of Sarcodon aspratus Mycelia)

  • 능이 균사체의 액체배양 조건을 구명하고자 배지별 및 배양조건을 규명하고자 하였다. 능이 균사체 배양에 적합한 기존의 배지로는 GYS배지였고, 적정한 온도는 $25^{\circ}C$인 것으로 나타났다. 능이 균사체 배양에 적합한 탄소원은 전분, 말토오스 및 포도당인 것으로, 질소원으로는 유기복합 질소원으로 소이톤(soytone)이, 무기 질소원으로는 암모니아태 질소가 균사체 생장에 유리한 것으로 나타났다. 무기인산염은 제일인산암모늄이 적합한 것으로 나타났다. 비타민 종류로는 니코틴산 처리에서 생장이 우수한 것으로 나타났다. 이상의 적합한 조건으로 20일간 능이 균사체를 배양하여 5.7 g dw/L을 생산할 수 있었다.

생체계면활성제 소포로리피드의 생합성과 계면 특성 (Biosynthesis and Interfacial Properties of Sophorolipids As a Biosurfactant)

  • Sophorolipids were biosynthesized using a strain of yeast, Torulopsis bombicola ATCC22214. It has been reported that this yeast gives the highest yields for the production of biosurfactant sophorolipids. Hence, this yeast was used in this study. One of the objectives of this study is to increase the yield of the sophorolipid synthesis. To meet this end, basic culture medium was formulated on the basis of literature research to-date. When this medium was used, the increase in yield from 15% to 150% was observed compared to using the media in the literature. To examine how the interfacial characteristics of sophorolipids change with substrate, glucose (the first carbon source) was maintained in the media and after being cultured for three days, the second carbon sources such as alkanes, vegetable oils, alcohols or organic acids were added. The whole broth was extracted twice with ethyl acetate and the extract was analyzed by thin layer chromatograhy(TLC). After qualitative analyses by TLC, surface tensions of sophorolipids were measured by the Wilhelmy plate method and critical micelle concentration(CMC) was determined using these surface tension data. Also, interfacial tensions were measured by the spinning drop method and emulsions of the three-component water/decane/sophorolipid system were tested. Sophorolipids were effective and efficient in terms of surface tension reduction and CMC, but they were ineffective as emulsifiers because emulsions were separated within 30 minutes.

합성 Pyrethroid 계 살충제인 Bifenthrin의 토양환경중 동태 제1보. Bifenthrin 및 Cyhalothrin의 토양 및 수용액중에서의 분해양상 (Behavior of Synthetic Pyrethroid Insecticide Bifenthrin in Soil Environment I) Degradation Pattern of Bifenthrin and Cyhalothrin in Soils and Aqueous Media)

  • 합성 pyrethroid계 살충제의 화학구조중 alcohol부분이 2-methyl biphenyl 그룹이 있는 bifenthrin과 ${\alpha}$-cyanobenzyl ester 그룹이 있는 cyhalothrin의 분해양상을 비교하기 위하여 토양의 환경조건을 호기적, 혐기적 및 살균조건으로 하였을 때와 pH를 변화시킨 수용액중에서 분해양상을 조사한 결과는 다음과 같다. Bifenthrin의 호기적 토양조건에서의 분해반감기는 칠곡 및 복현 토양에서 85.1 및 61.2일이었으며 cyhalothrin은 각각 54.6 및 32.2일로서 유기물함량이 많은 복현토양에서 두 약제 모두 분해가 빨랐다. 두 약제 모두 혐기적 조건 및 살균 조건의 토양에서는 매우 느린 분해양상을 보였다 Bifenthrin과 cyhalothrin의 분해는 주로 호기성 미생물이관여하는 것으로 나타났다. 두 약제간에는 cyhalothrin이 bifenthrin보다 분해반감기가 30일 정도 빨랐다. 수용액의 pH가 10인 알카리성 조건에서 cyhalothrin이 bifenthrin보다 상당히 빨리 분해되 었으며 pH 2 및 6의 산성조건에서도 두 약제 모두 분해가 지연되었다.

Biological characteristics of Paenibacillus polymyxa GBR-1 involved in root rot of stored Korean ginseng

  • Background: This study aims to describe the characterization of Paenibacillus polymyxa GBR-1 (GBR-1) with respect to its positive and negative effects on plants. Methods: The morphological characteristics of GBR-1 were identified with microscopy, and subjected to Biolog analysis for identification. Bacterial population and media optimization were determined by a growth curve. The potential for GBR-1 as a growth promoting agent, to have antagonistic activity, and to have hydrolytic activity at different temperatures was assessed. The coinoculation of GBR-1 with other microorganisms and its pathogenicity on various stored plants, including ginseng, were assessed. Results: Colony morphology, endospore-bearing cells, and cell division of GBR-1 were identified by microscopy; identification was performed by utilizing the Biolog system, gas chromatography of fatty acid methyl esters (GC-FAME). GBR-1 showed the strongest antagonistic activity against fungal and bacterial pathogens. GBR-1 cell numbers were relatively higher when the cells were cultured in brain heart infusion (BHI) medium when compared with other media. Furthermore, the starch-hydrolytic activity was influenced by GBR-1 at higher temperature compared to low temperatures. GBR-1 was pathogenic to some of the storage plants. Coinoculation of GBR-1 with other pathogens causes differences in rotting on ginseng roots. A significant growth promotion was observed in tobacco seedlings treated with GBR-1 suspensions under in vitro conditions, suggesting that its volatile organic compounds (VOCs) might play a role in growth promotion. Conclusion: The results of this study indicate that GBR-1 has both positive and negative effects on ginseng root and other stored plants as a potential biocontrol agent and eliciting in vitro growth promotion.
