In order to develop a method of economical production and to reduce energy-consumption in fuel alcohol production, we investigated the fermentation characters of two newly selected thermotolerant yeasts. The RA-74-2 showed stable and superior fermentability between 30 and $40^{\circ}C$ in 20% glucose media in comparison to the industrial strains. The optimum concentration of glucose for economical fermentation at $40^{\circ}C$ was 15-18%, and organic nitrogen was necessary for a satisfactory fermentation. The optimum pH was 4.0 and aeration was adversed for high temperature fermentation. Agitation was an important factor at $40^{\circ}C$ and the addition of magnesium ion 0.2% was required in this experiment. When the inoculum was increased, ethanol productivity as well as the speed of fermentation increased. On the other hand RA-912, which can grow at $48^{\circ}C$, showed similar fermentability between 30-$45^{\circ}C$ in 20% glucose media As the concentration of substrate decreased, fermentation ratio increased at $45^{\circ}C$ (45%, 65%, 95% fermentation ratio in 20%, 15%, 10% glucose media, respectively). Also, requirement of organic nitrogen and magnesium ion in RA-912 was similar in RA-74-2. The optimum pH for fermentation was 5.0, and the effects of agitation were enhanced at $37^{\circ}C$ than at $45^{\circ}C$. As the inoculum was increased, fermentation speed became more enhanced but the ethanol productivity was less affected. RA-912 showed fermentability with various substrates. Among the substrates used, inulin was the most promising substrate for the high-temperature fermentation. When 14.5% inulin was used as the substrate, 93% and 55% fermentation ratios were shown at $37^{\circ}C$ and $45^{\circ}C$, respectively.
${\alpha}-Tocopherol$ was found to be a fluorescent probe in determination of the phasetransition temperature of liposome. Since this was a discovery of a new aspect of ${\alpha}-tocopherol$ as an important biochemical molecule, its molecular spectroscopic characterization was carried out in order to obtain some informations on its spectral and, structural properties in various media, anticipating that the compound may entertain a wide applications in biochemical systems as a spectroscopic probe. Two species of α${\alpha}-tocopherol$, monomer and dimer, were found to exist in organic media, especially in solvents of nonhydrogen bonding ability. Monomer with maximum UV-absorption around $(291{\sim}294nm)$ is highly fluorescent, while dimer which is formed by intermolecular hydrogen bonds and absorbes with spectral peak at 298nm is nonfluorescent. ${\alpha}-tocopherol$ incorporated to liposome exhibits emission property quite different from that in various organic media showing broad and red-shifted fluorescence excitation and emission spectra. This spectral abnormality is to be interpreted to arise from chromanolate-type ion, H-dissociated ${\alpha}-tocopherol$.
This study was carried out to determine the effects of media compositions on cell growth and casein hydrolysis for cell production in order to add Micrococcus sp. LL3 as a potential agent for industrial application for the shortening of ripening period. Monosaccharides like glucose, mannose and fructose were mare excellent as carbon source, but arabinose and xylose markedly inhibited cell growth and caseinolysis. Among the organic nutrients, yeast extract was more effective for cell growth and for caseolysis. However, inorganic nitrogen sources were less effective than organic sources. Urea inhibited cell growth severely. Cell growth and caseinolysis were rather increased a little in the broth containing 1% NaCl, and the organism tolerated and grew in relatively high concentrations of NaCl up to 9%. Addition of vitamin did not affect cell growth and caseolysis in level of $0.1\;{\mu}g/ml$ concentration. Cell growth and caseinolysis were stimulated by addition of glutamic acid and $MgSO_4$ with concentration of 0.2% and 0.05% respectively.
Two step rapid filter system as a pre-treatment for the injected water into aquifer storage and recovery (ASR) in Korea was developed to reduce physical blockage and secure the volume of the injected water. First, single rapid sand filters with three different media sizes (0.4~0.7, 0.7~1.0 and 1.0~1.4 mm) were tested. Only two sizes (0.4~0.7 and 0.7~1.0 mm) satisfied target turbidity, below 1.0 NTU. However, they showed the fast head loss. To prevent the fast head loss and secure the volume of the injected water, a rapid anthracite filter with roughing media size (2.0~3.4 mm) were installed before a single rapid sand filter. As results, both the target turbidity and reduction of head loss were achieved. It was determined that the media size for a rapid sand filter in two step rapid filter system (i.e. a rapid anthracite filter before a rapid sand filter) was 0.7~1.0 mm. In addition, the effects of coagulant doses on the removal of natural organic matter (NOM), which might cause a biological clogging, were preliminarily evaluated, and the values of $UV_{254}$, dissolved organic carbon (DOC) and SUVA were interpreted.
This paper was carried out to isolate and identify Aeromonas caviae which can degrade Sodium Dodecyl Benzene Sulfonate(SDBS) effectively. And the affecting factors for the ability of bacterial degradation were also studied. Frm October 1991 to February 1992, two hundred samples from sweage in Taegu area and Nakdong river waters in Talsung Gun area were tested. Minimal salt medium which contain SDBS only as a carbon source was used as a culture medium. The isolated new strain was identified as Aeromonas caviae Kim & Kweon. The optimal pH for SDBS degradation were 7.0 and temperature, $32^{\circ}C.$ It was taken 24 hours to degrade SDBS of 20mg/l completely under the optimal pH and temperature. And in the case of 30 mg/l of SDBS, it was taken 36 hours. The nitrogen sources were added to the minimal salt media containing 20mg/l of SDBS, and they were incubated at $32^{\circ}C$ for 14 hours. 86.9% SDBS were degraded after addition of 0.03% peptone as a organic nitrogen source. And 70.5% SDBS after addition of 0.05% ammonium sulfate as a inorganic nitrogen source. In the case of metal compounds(0.015%), the degradation rate for SDBS were 3.5 fold increased in the media containing magnesium chloride and calcium chloride than in the media that were not containing these metal compounds. And where the media containing magnesium chloride was 0.05%, the degradation rate was 65.8%. And above 0.3% NaCI, the degradation rate was decreased slowly.
This study was undertaken to determine the effect of reservoirs on water quality and the distribution of pathogenic and indicator bacteria in a drinking water distribution system (total length 14km). Raw water, disinfected water, and water samples from the distribution system were subjected to physicochemical and microbiological analyses. Most factors encountered at each season included residual chloride, nitrate, turbidity, and phosphorus for heterotrophic bacterial distribution, and hardness, heterotrophic bacteria, sampling site, and DOC (dissolved organic carbon) for bacteria on selective media. No Salmonella or Shigella spp. were detected, but many colonies of opportunistic pathogens were found. Comparing tap water samples taken at similar distances from the water treatment plant, samples that had passed through a reservoir had a higher concentration of heterotrophic bacteria, and a higher rate of colony formation with 10 times as many bacteria on selective media. Based on the results with m-Endo agar, the water in reservoirs appeared safe; however, coliforms and opportunistic pathogenic bacteria such as Pseudomonas aeruginosa were identified on other selective media. This study illustrates that storage reservoirs in the drinking water distribution system have low microbiological water quality by opportunistic pathogens, and therefore, water quality must be controlled.
Treatability tests were conducted using EMC process to study the feasibility of applying this process as recycling-water treatment system in high density seawater aquaculture farm. To study the effect of organic and ammonia nitrogen loading on system performance, hydraulic retention time of reactor was reduced gradually from 12hr to 10min. The conclusions are can be summarized as follows. When the system HRT was reduced from 12hr to 10 min gradually, there was little noticeable change(reduction) in ammonia nitrogen removal efficiencies until 2hr of HRT, however, removal efficiencies were decreased dramatically when the system was operated under the HRT of less than 2hr. In case of organics(COD), there was no dramatic deterioration in removal efficiencies depending on HRT reduction. More than 90% of removal efficiencies were maintained successfully when the system was operated at the HRT of 10 min. In case of system performance depending on media packing ratio in reactor, there was little difference in each reactor performance depending on media packing ratio in reactor when the reactors were operated under the HRT of longer than 1hr, however, differences in reactor performances were considerably evident when the reactors were operated under the HRT of shorter than 1hr. That is, the more reactor was packed, the better reactor performed. When comparing reactor performance among 25%, 50%, 75% packed reactor, it can be judged that media packing ratio more than 50% plays no significant role in increasing reactor performance. For this reason, packing the media less than 50% is more reasonable way in view of economic. Such a tendency well agreed with the variation of ammonia-nitrogen removal efficiencies according to the media packing ratio in reactors at each HRT. Difference in effluent ammonia-nitrogen concentration between 50% media packing reactor and 75% media packing reactor was negligible. When comparing with the results of 25% packing reactor, difference was not so great.
The cheaper and high-quality media are necessary to produce the year-round mushroom. For low input and high productivity, this experiment was carried out to screen the proper supplementary materials on media among agricultural by-products; bean-curd residues, dried orange peel, chinese drug residues and celery cabbage leaf. Judging by chemico-physicals, the used agricultural by-products was possible for an additives on mushroom media. Bean-curd residues added to media raised the amount of organic matter, nitrogen and carbon. In Pleurotus ostreatus, the conventional media yielded 20 pilus cap, 72g per a bottle, but adding 10 percent bean-curd residues produced 12 pilus cap, 77g and an increase yielded 7%, and 10 percent orange peel took 12 pilus cap and an increase yieled 11%. In Agrocybe aegerita, added to media, except chinese drug residues, increased performance compared with conventional media, produced 98g per a bottle. Especially, the bean-curd residue increase 15 percent in productivity.

This experiment was designed to assess the physical and chemical properties of growing media substituted with a range of increasing concentrations of pretreated wood and to relate these properties to plant growth responses. For preparing the growing media, each material was combined with rural soil, peat, perlite and pretreated wood. Physicochemical properties studied were similar to ideal substrate ranges for plant growth on growing media, including pretreated wood. Physical properties were also well maintained over time. In comparison to plants growing in 100% rural soil, tall fescue(Festuca arundinacea) in the prepared growing media achieved better growth, especially when using the 50% rural soil + 50% PPW(peat + perlite + pretreated wood, 3:1:6(w/w/w)) and 30% rural soil + 70% PPW (peat + perlite + pretreated wood, 3:1:6(w/w/w)), and showed improved germination percentage. We confirmed the potential use of growing media, including pretreated wood. Furthermore, our results show a correlation among the physicochemical properties of tall fescue(Festuca arundinacea); physical properties were significantly influenced by germination and aerial parts. The root length of physicochemical properties was correlated with bulk density and organic compound (p<0.01).
The identification of explosive residues on specimens obtained from an explosion event is a crucial factor for assessing the cause of the explosion. In order to detect the components of explosives, the explosive residues deposited on surfaces are commonly extracted using swabbing materials pre-wetted with an organic solvent. The residues are then analyzed with analytical instruments such as LC/MS and CE/MS. Most conventionally used swabbing media such as cotton swabs or cotton tip swabs seem unsuitable for extracting explosive residues from the surface of a large area of clothes because the swabbing materials tend to be damaged easily, and because only a relatively small amount of explosives is collected. To overcome these problems, we have introduced a novel wiper ($215{\times}210mm$, single layer, Yuhan-Kimberly, Republic of Korea) as a swabbing material to recover representative organic explosives, namely, TNT, RDX, tetryl, HMX, PETN, and NG, from a large area of clothes. Different sides of the wiper, which was folded in half five times, was used to swab the surface of a clothing. We compared this novel wiper with a cotton swab and a cotton tip swab in terms of the recovery efficiency for the aforementioned organic explosives by pre-wetting with methanol, acetone, and acetonitrile, respectively. We identified that this novel wiper collected a significantly higher amount of organic explosive residues than a cotton swab or a cotton tip swab when using methanol as an extracting solvent.
