$\alpha$-Glucosidase Inhibition and Glucose-uptake Stimulation by Ethanol Extracts from Edible Mushroom Hinmogi (Tremella fuciformis)

  • Ethanol extracts from the edible mushroom hinmogi (Tremella fuciformis, TF) were used in the investigation of effects on $\alpha$-glucosidase in vitro and on glucose-uptake in 3T3-L1 mature adipocytes. Addition of the extract significantly inhibited $\alpha$-glucosidase from small intestine of porcine and of rat (about 42 and 35% of control, respectively), and stimulated glucose uptake (about 100% of control), of which activity was higher than that of maitake (Grifola frondosa) X-fraction, a well known anti-diabetic substance. When the ethanol extracts were further partitioned successively by organic solvents and purified by silica gel chromatography, the non-polar layer (F-7) from hexane layer showed highest stimulatory activity of glucose-uptake among layers tested. The major components of the F-7 were 1-monooleoylglycerol and 1-monopalmitoylglycerol. Our report is the first description of TF with stimulatory activity of glucose-uptake. These results suggest that TF extracts may constitute a new source of glucose transport activator and could be employed as a potential anti-diabetic material for treatment and preventing diabetes.

Cytotoxicity of Dichloromethane Extracts of Asian Dust

  • The appearance of Asian Dust (AD) originating from China and Mongolia during spring each year is a meteorological phenomenon periodically observed in extensive regions of East Asia. According to a previous epidemiological study, AD has adverse effects on both human beings and ecosystems. In this study, we collected total suspension particles (TSP) in the AD period and Non-AD (NAD) period. We extracted organic components from TSP using dichloromethane (DCM), and the polyaromatic hydrocarbons (PAHs) were analyzed. The DCM extracts contained PAHs such as benzo(b)fluoranthene, benzo[g,h,i]perylene, benzo(k)fluoranthene, benzo(a)pyrene, and pyrene. No significant difference was observed in cytotoxicity of the DCM extracts from AD versus NAD when tested on the human bronchial epithelial cells, BEAS-2B. e also examined the toxic mechanisms of AD extracts in cultured BEAS-2B cells and RAW264.7 cells, and in BEAS-2B cells observed increased levels of reactive oxygen species (ROS), decreased glutathione (GSH), and induced caspase-3 activity. Increased expression of oxidative stress-related and inflammation- related genes were also observed in BEAS-2B cells, while nitric oxide (NO) levels were increased in RAW264.7 cells. Taken together, the results suggest that in these cultured cells, AD may induce cytotoxicity through oxidative stress and pro-inflammatory signals.

여러 가지 천연 염재를 이용한 면, 견, 모직물의 염색 및 소취 특성 (Dyeing and Deodorizing Properties of Cotton, Silk, and Wool Fabrics Dyed with Various Natural Colorants)

  • Four kinds of natural dying solutions (natural colorant extracts)were obtained by extraction from sappan wood, black tea, peony, and clove using water as extracting solvent at $90^{\circ}C$ for 90 min with liquor ratio (solid natural colorant material/solvent water, weight ratio) of 1/10. The dyeing, colour fastness and deodorizing properties of fabrics (cotton, silk, and wool fabrics) dyed with natural colorant extracts were compared. It was found that these properties were significantly dependent on the concentration of extracts, the structure of colorant, and the kind of fabrics. The K/S value of dyed cotton fabric increased in the order of peony < sappan wood < clove < black tea, however, the values of dyed silk and wool fabrics were in the order of peony < sappan wood < black tea < clove. Colour fastness (light, water, and perspiration fastness) was in the range of 3 - 5 grade except for sappan wood. The deodorizing performance of fabrics dyed with various natural colorants extracts was in the range of 56 - 99%. The deodorizing performance increased in the order of peony < black tea < sappan wood < clove. Especially, the deodorizing performance of all fabrics dyed with clove was found to be the highest at 98-99%.

  • 몇 가지 유기용매를 사용하여 인삼 추출액을 분별 분리하여 몇 개의 분칠들을 얻어서 이들이 RNA중합효소의 활동성에 미치는 영향을 조사하였다. 백삼과 홍삼에서 모두 RNA 중합효소의 활동성에 양성적인 효과를 가진 분획들을 얻었다. 백삼의 경우 RNA 중합효소에 양성적인 효과를 미치는 성분들은 전체 메탄을 추출액 분획, 초산에틸 추 출모액 및 이 모액의 수용성 분획들에서 확인되었고 흥삼의 경우에는 전체 메탄을 추출액 분획과 에테르 분획들에서 확인되었다. 이 사실은 중합효소에 양성적인 효과를 나타내는 성분들이 무극성 및 극성 부분들로 이루어져 있으며, 그들이 홍삼의 가공과 정에서 두 부분으로 분해되었을 것이라는 것을 암시의 준다.

프로폴리스 농도가 항균활성에 미치는 영향 (The effect of propolis concentration on the antibacterial activity)

  • The objetives of this study are to set up optimum extraction temperature, time and organic solvent for propolis extraction, to investigate chemical properties, and to develop health foods from propolis preparation. In this study, ethanol and ultrasonic extracts method performed to optimum extraction temperature was at 60, $20^{\circ}C$, optimum extraction time was at 12, 4 hours and optimum extraction amount of solvent was at 20, 15 times of propolis weight. When various ethanol solutions were used, whereas flavonoid content was highest in 70, 80% aqueous ethanol, respectively. So the ultrasonic extracts method used gave better results than the ethanol extracts method in this work. Extraction of propolis with etanol and ultrasonic extracts method was performed by using the water and various concentrations of aqueous ethanol as solvent. Sensitivity of propolis samples to Staphylococcus aureus was investigated and the results were shown. Samples of water extract did not inhibit microbial growth, where as 50% aqueous ethanol extract the largest inhibitory zone for Staphylococcus aureus, then decreased inhibition with increasing ethanol concentrations.

짚신나물, 삼백초의 항산화와 항암활성 효과 (Antioxidant and Anticancer Effects of Agrimony (Agrimonia pilosa L.) and Chinese Lizardtail (Saururus chinensis Baill))

  • The antioxidant activities of Agrimony (Agrimonia pilosa L.) and Chinese lizardtail (Saururus chinensis Baill) according to extraction methods were measured. SOD-like activity showed greater antioxidant effects with ethanol solvent than those with water. Ethanol extracts of Agrimony leaves showed the highest SOD-like activity of 94.4%. SOD-like activity differed according to the extraction solvents. The contents of polyphenolic compounds were higher in water extracts than those in ethanol extracts. The contents were 161.4 mg for Agrimony roots, 100.2 mg for Agrimony leaves, and 79.1 mg for Agrimony stalks in order. EDA in Agrimony leaves that were highest among medicinal plants were 83.4% in the water extract and 81.7% in the ethanol extract. The anticancer effects of the extracts by water and ethanol from Agrimony and Chinese lizardtail were experimented. The growth of stomach cancer cells, SNU-719 was inhibited 94.5% by the hexane fractions of Agrimony and also the growth of liver cancer cells, Hep3B was inhibited 83.2% by the hexane fractions of Agrimony, while the growth of normal cell, DC2.4 was not affected.

Physiological and Whitening Effects of Morus alba Extracts

  • Mulberry extracts can be incorporated into skin-whitening products. The compound attributed to lighten the skin is arbutin, a form of hydroquinone that inhibits melanin release by suppressing the tyrosinase enzyme. For the cosmetic applications, the physiological effects of mulberry (Morus alba) extracts were investigated. The water soluble fraction of mulberry contains higher amount of protein (16.28~4.47%) in contrast to fat (1.55~1.41%). In addition, the fraction abundantly contains succinic acid (972.4-275.8 mg/g) and phosphoric acid (1,628.4-121.9 mg/g) in different parts of mulberry. The free radical scavenging ability in water soluble fraction was found to display remarkable effects in comparison with methanol and ethyl acetate fraction. The ethyl acetate-soluble of root and leaf showed remarkable tyrosinase inhibition activity by IC 50 (${\mu}g/ml$). The anticancer activity of methanol fraction obtained from mulberry using human cancer cell lines showed growth inhibition effect (270.14 mg/ml in Calu-6 cells, 295.29 mg/ml in HCT-116, and 332.29 mg/ml in MCF-7 cells, respectively). Based on the results, Morus alba extracts include cosmetic ingredients with antioxidizing and whitening properties.

PILS-TOC 및 GC/MS-TD를 이용한 서울시 대기 중 초미세먼지(PM2.5) 유기탄소의 특성 분석 (Characteristics of PM2.5 Carbonaceous Aerosol using PILS-TOC and GC/MS-TD in Seoul)

  • Continuous Water-Soluble Organic Carbons (WSOC) by the Particle Into Liquid Sampler - Total Organic Carbon (PILS-TOC) analyzer were measured at the Seoul intensive monitoring site from June 17 through July 5 in 2014. In addition, the 24 hour integrated PM2.5 collected by Teflon and Quartz filters were analyzed for water soluble ions by Ion chromatography (IC), WSOC by TOC from water extracts, organic carbon (OC), elemental carbon (EC) by carbon analyzer using the thermal optical transmittance (TOT) method, and mass fragment ions (m/z) related to alkanes and PAHs (Poly Aromatic Hydrocarbons) by Gas Chromatography-Mass Spectrometer-Thermal Desorption (GC/MS-TD). Based on the statistical analysis, four different Carbonaceous Thermal Distributions (CTDs) from OCEC thermal-gram were identified. This study discusses the primary and secondary sources of WSOC based on the Classified CTD, organic mass fragments, and diurnal patterns of WSOC. The results provide knowledge regarding the origins of WSOC and their behaviors.

유기 게르마늄 농축수로 재배한 농산물의 항염 및 함암효과 (Anti-inflammatory and Anti-cancer Effects of Agricultural Produce Grown with Organic Germanium-enriched Water)

  • The study was conducted to identify the anti-inflammatory and anti-cancer effects in sprouts of mouse-eyed bean (Rhynchosia nulubilis), ginseng (Panax ginseng), perilla (Perilla frutescens), broccoli (Brassica oleracea var. italica), and lettuce (Lactuca sativa) grown with organic germanium concentrate. Western blot analysis was performed to assess the anti-inflammatory activity of the extract. All extracts exhibited noticeable anti-oxidant activity, indicating a significant correlation between the germanium content and anti-oxidant activity (p<0.05). In particular, rat-eyed bean sprouts with the highest germanium content showed significant anti-inflammatory activity (p<0.05) by significantly inhibiting the expression of the inflammatory complexes, NLRP3, cytokines IL-1β and caspase-1. Ginseng and broccoli sprouts showed strong anti-cancer properties and had high anti-oxidant effects (p<0.001). Germanium-concentrated water allows the mass production of agricultural products containing high concentrations of organic germanium. Agricultural produce grown with germanium concentrate add organic germanium to various physiological active ingredients, increasing the anti-oxidant and anti-cancer effects. These results strongly suggest that agricultural products containing high germanium concentrations can be used as novel health supplements to improve health.

잎담배의 팽화방법에 따른 팽화율 및 내용성분 특성 (Physico-chemical Properties of leaf tobacco by expansion processes.)

  • Flue-cured tobaccos(cv. NC82, grade AB3O-1) expanded by super steam, volatile organic solvents(tricuorofluoromethane, propane), carbon dioxide, nitrogen, and by freeze-drying were examined for their physico-chemical properties. The expanding rate by using the organic solvents or carbon dioxide at the impregnated pressure was 70% or more, whereas the rate by super steam and freeze-drying was 49.4 and 42.9%, respectively. The contents of total sugar, alkaloid and ether extracts of the expanded tobacco by organic solvents or carbondioxide were decreased to 27.6, 20% or more, and 13%, respectively, when compared to non-expanded one. The contents of citric acid of the tobacco expanded by using trichlorofluoromethane and carbon dioxide were increased to 19.1, 14.0%, respectively. Linoleic acid in the tobacco expanded by super steam or trichlorofluoromethane was also increased to 17.3%, but malonic acid in the tobacco expanded by super steam was 18.5% less than that in the non-expanded one. Nineteen essential oil among 22 assayed in this experiment were decreased in the expanded tobaccos. Key words : expanding method, tobacco, physico-chemical properties.
