Analytical Quality by Design Methodology Approach for Simultaneous Quantitation of Paeoniflorin and Decursin in Herbal Medicine by RP-HPLC Analysis

  • Simultaneous quantification of multiple marker compounds in herbal medicine by high performance liquid chromatography (HPLC) analysis is still a challenge due to the complexity in various parameters to be considered and co-existing multi-components. As a case study, a reliable HPLC method for simultaneous quantification of paeoniflorin from Paeoniae Radix and decursin from Angelicae Gigantis Radix in various commercial herbal medicine was developed based on analytical quality by design (AQbD) strategy. As a first step, risk assessment was performed to select the critical method parameters (CMPs) which were decided as organic mobile phase ratio and column oven temperature. In order to evaluate the effect of the CMPs on critical method attributes (CMAs) of peak resolution and tailing, central composite design (CCD) was employed. The final chromatographic conditions were optimized as follows: column- C18, 4.6 × 250 mm, 5 ㎛ particle size; mobile phase- A: acetonitrile, B: 0.1% acetic acid water; detection wavelength- 235 nm for paeoniflorin, 325 nm for decursin; column oven temperature- 25℃; flow rate- 1.0 mL/min; gradient mobile phase system as Time (min) : % A, 0:14, 25:14, 30:50, 60:50, 61:100, 65:100, 66:14, 75:14. The method was successfully validated according to the International Conference on Harmonization (ICH) guidelines and piloted for ten commercial herbal medicines.

D-$\Pi$-A designed dye chromophores and nanoparticles: optical properties, chemosensor effects and PE/Aramid fiber colorations

  • Studies on attractive color changing property of dye chromophore and fluorophore have been greatly enjoyed in the related industrial and research fields such as optoelectronics, chemosensor, biosensor and so on. The optical property based on D-$\Pi$-A intramolecular charge transfer (ICT) system of chromophore molecules can be utilized as suitable sensing probes for checking media polarity and determining colorimetric chemosensing effect, especially heavy metal detection. These finding are obtained by absorption and emission properties. In this work, donor-acceptor D-$\Pi$-A type fluorescent dyes were designed and synthesized with the corresponding donor and acceptor groups. The selected donor moieties might be provided prominent amorphous properties which are very useful in designing and synthesizing functional polymers and in fabricating devices. Another reasons to choose are commercial availabilities in high purity and low price. Donor-bridge-acceptor (D-A) type dyes can produce impressive optical-physical properties, yielding them potentially suitable for applications in the synthesis of small functional organic molecules. Small organic functional molecules have unique advantages, such as better solubility, amorphous character, and represent an area of research which needs to be explored and developed. Currently, their applications in metalorganic compounds is rapidly expanding and becoming widespread in self-assembly processes, photoluminescence applications, chiral organocatalysts, and ingrafts with nanomaterials. Colloidal nanoparticles have received great attentions in recent years. The photophysical properties of nanoparticles, particularly in terms of brightness, photostability, emission color purity and broad adsorption range, are very attractive functions in many applications. To our knowledge background, colloidal nanoparticles have been enjoyed their applications in bio-probe research fields. This research interest can be raised by the advantages of the materials such as high photoluminescence quantum yields, sharp emission band, long-term photostability and broad excitation spectra. In recent, the uses of nanoparticles being embedded in a polymer matrix and binded on polymer surface have been explored and their properties such as photo-activation and strong photoluminescence have been proposed. The prepared chromophores and nanoparticles were investigated with absorption and emission properties, solvatochromic behaviors, pH induced color switching effects, chemosensing effects and HOMO/LUMO energy potentials with computer simulation. In addition, synthesized fluorophore dyes and particles were applied onto PE/Aramid fiber fluorescing colorations. And the related details were then discussed.

Cobalt Phthalocyanine 탄소유기 전극을 이용한 농약 잔류량 측정 센서개발 (Development of Electro-Biosensor for the Residual Pesticides using Organic Carbon and Cobalt Phthalocyanine)

  • 본 연구에서는 탄소 유기물과 CoPh를 혼합하여 농약 잔류량을 측정 할 수 있는 바이오센서를 구현 하였다. 작동 전극은 탄소유기물과 CoPh를 섞어 사용하였고 비율은 CoPh를 7%로 제작하였다. CoPh가 7%인 경우 저농도 thiocholine 농도에서도 민감하게 반응하였다. 대표적인 농약인 카보후란에 대하여 센서의 감도 한계는 약$0.5{\mu}g/L$ 수준이며, 농약 농도에 따른 전극의 출력이 선형적인 결과를 얻었다. 또한 EPN 및 클로로피리포스 농약에 대하여 실험 한 결과 카보후란과 같이 농도에 따라 효과적으로 반응하는 것을 확인하였다. 이러한 센서는 현재 농약 잔류량 측정에 사용되는 비색법과 비교하여도 감도 및 선형성 면에서 뒤떨어지지 않으며, 간이 농약 잔류량 측정기 센서로 사용가능성이 있다.

휘발성 유기화합물의 배출량 산정 및 관리 소프트웨어 개발 (A Study on Process Integrated Innovation System for a LNG Industry)

  • 본 연구에서는 국내 휘발성 유기화합물질의 주요 취급시설에 대한 조사를 통하여 배출원별 배출메카니즘을 정립하고, 새로운 배출량산정모델을 제안하였다. 또한 각 배출원에 적용가능한 배출저감기술과 각 기술의 경제성 평가기법을 제안하였다. 여기에 배출원 DB, 화학물질물성치 DB, 기상정보 DB, 배출계수 DB 등의 정보를 연계하여 휘발성유기화합물질 배출량산정 및 관리 소프트웨어 VEER(VOCs Emission Estimation and Reduction)를 개발하였다. 결국 본 연구를 통해 개발된 VEER를 이용하여, 휘발성 유기화합물질 취급업체, 관리기관, 연구기관 등에서 쉽고 정확하게 배출원 인벤토리를 구축하고, 배출량을 산정하며, 계산된 결과를 바탕으로 각 배출원의 배출유량과 농도에 적합한 휘발성 유기화합물질 배출 저감기술을 선택하고, 여러 저감기술들에 대한 경제성을 평가함으로써, 저비용의 배출량 저감 및 배출원 관리기술을 선택하고, 설계할 수 있을 것으로 기대된다.

Wine quality grading by near infrared spectroscopy.

  • The ability to accurately assess wine quality is important during the wine making process, particularly when allocating batches of wines to styles determined by consumer requirements. Grape payments are often determined by the quality category of the wine that is produced from them. Wine quality, in terms of sensory characteristics, is normally a subjective measure, performed by experienced winemakers, wine competition judges or winetasting panellists. By nature, such assessments can be biased by individual preferences and may be subject to day-to-day variation. Taste and aroma compounds are often present in concentrations below the detection limit of near infrared (NIR) spectroscopy but the more abundant organic compounds offer potential for objective quality grading by this technique. Samples were drawn from one of Australia's major wine shows and from BRL Hardy's post-vintage wine quality allocation tastings. The samples were scanned in transmission mode with a FOSS NIR Systems 6500, over the wavelength range 400-2500 ㎚. Data analysis was performed with the Vision chemometrics package. With samples from the allocation tastings, the best correlations between NIR spectra and tasting data were obtained with dry red wines. These calibrations used loadings in the wavelengths related to anthocyanins, ethanol and possibly tannins. Anthocyanins are a group of compounds responsible for colour in red wines - restricting the wavelengths to those relating to anthocyanins produced calibrations of similar accuracy to those using the full wavelength range. This was particularly marked with Merlot, a variety that tends to have relatively lower anthocyanin levels than Cabernet Sauvignon and Shiraz. For dry white wines, calibrations appeared to be more dependent on ethanol characteristics of the spectrum, implying that quality correlated with fruit maturity. The correlations between NIR spectra and sensory data obtained using the wine show samples were less significant in general. This may be related to the fact that within most classes in the show, the samples may span vintages, glowing areas and winemaking styles, even though they may be made from only one grape variety. For dry red wines, the best calibrations were obtained with a class of Pinot Noir - a variety that tends to be produced in limited areas in Australia and would represent the least matrix variation. Good correlations were obtained with a tawny port class - these wines are sweet, fortified wines, that are aged for long periods in wooden barrels. During the ageing process Maillard browning compounds are formed and the water is lost through the barrels in preference to ethanol, producing “concentrated” darkly coloured wines with high alcohol content. These calibrations indicated heaviest loadings in the water regions of the spectrum, suggesting that “concentration” of the wines was important, whilst the visible and alcohol regions of the spectrum also featured as important factors. NIR calibrations based on sensory scores will always be difficult to obtain due to variation between individual winetasters. Nevertheless, these results warrant further investigation and may provide valuable Insight into the main parameters affecting wine quality.

캠프 캐럴 인근 주민의 혈중 및 요중 휘발성 유기화합물 평가 (Assessment of Volatile Organic Compounds in Blood and Urine among Residents around Camp Carroll)

  • Objectives: Exposure to volatile organic compounds such as trichloroethylene(TCE) and perchloroethylene(PCE), along with Agent Orange, that were issued around Camp Carroll US Army Base situated in Waegwan, Chilgok-gun, Gyeongsangbuk-do Province, Korea. The main objective of this study was to assess the exposure to TCE and PCE of residents of the area surrounding Camp Carroll. Methods: The TCE, PCE and trichloroethanol(TCEOH) concentrations in blood and trichlroroacetic acid(TCA) and TCEOH concentrations in urine were measured and analyzed in a total of 1,033 residents around Camp Carroll. TCA and TCEOH are metabolites of TCE and PCE, respectively. The information on demographic characteristics and exposure variables in relation to underground water were obtained through a questionnaire completed by the subjects. Results: TCE, PCE and TCEOH concentrations were not detected in blood. Detection rates of TCA and TECOH concentrations in urine were 98.5% and 36.6%, respectively. Creatinine-corrected average TCA and TCEOH concentrations were $12.23{\pm}23.81{\mu}g/g$ and $0.66{\pm}4.31{\mu}g/g$, respectively. A significant difference was not shown between the drinking group and no drinking group for underground water, which was assumed as a potential route of exposure to TCE and PCE through the consumption of ground water. However, females drinking ground water showed a significantly higher mean level of TCA in urine than did males. There was no significant difference according to drinking ground water as a potential source of exposure to TCE and PCE in residents around Camp Carroll. Conclusions: Considering the statistical analysis of factors affecting exposure to TCE and PCE in ground water along with previous reports, TCA in urine as exposure to TCE and PCE might not be appropriate because it is found in chlorinated drinking water. Therefore, TCA concentration in urine may be the result of drinking of chlorinated water.

낙동강 수계의 섬유 및 화학 산업폐수로부터 발생하는 미량유해화학물질의 모니터링 (Monitoring of Micro Noxious Chemicals Caused by Fiber and Chemistry Industrial Wastewater on the Nakdong River Water System)

  • 다양한 산업의 발달은 우리 인간이 사용해야 하는 수자원으로 여러 가지 형태의 위해성을 함유하는 유해화학물질들의 수적 양적 증가를 초래하였다. 이로 인해 효율적인 수자원의 이용을 위해서는 수자원으로 유입되는 오염원의 사전 처리가 필수적인 것으로 인지되어 국내에서도 수자원으로 유입되는 오염물질에 대해 총량으로 방류업체의 수질을 규제하기 시작했다. 그러나 아직까지도 규명되지 않은 화학물질들이 대다수를 차지하므로 이에 대한 모니터링이 매우 중요한 역할로 자리하고 있다. 본 연구에서는 낙동강 수계로 유입되는 섬유 및 화학 산업폐수를 중심으로 미량유해화학물질을 계측하여 이들이 낙동강 수계에 미치는 유해성을 검토하였다. 대상으로 한 7개 업체로부터의 유입수와 방류수에서 검출되는 VOCs(volatile organic compounds) 중에서는 클로로포름(chloroform)이 가장 높은 빈도로 검출되었으며, 동일한 업체에서도 노출수준이 일정하지 않아 이의 관리에 어려움이 예상된다. 그러나 이들 업체의 방류수가 유입되는 하수처리장에서는 콜로로포름의 처리효율이 높아(88% 이상) 낙동강 수계의 영향은 그리 크지 않을 것으로 추정된다. 한편, 섬유/화학관련 산업체의 유입수와 방류수로부터 노출되는 EU지정우선물질에 대해 조사한 결과, 검출현황에 대한 일정한 경향을 찾을 수 없을 뿐만 아니라 국내자료의 축적도 거의 없는 상태임을 감안하여 지속적인 모니터링을 통해 국가정책에 활용 할 수 있는 자료구축을 해 나가야 할 것이다.

휘발성 질소화합물 감응형 색변환 센서를 활용한 연어 신선도 모니터링 (Freshness Monitoring of Raw Salmon Filet Using a Colorimetric Sensor that is Sensitive to Volatile Nitrogen Compounds)

  • A colorimetric paper sensor was used to detect volatile nitrogen-containing compounds emitted from spoiled salmon filets to determine their freshness. The sensing mechanism was based on acid-base reactions between acidic pH-indicating dyes and basic volatile ammonia and amines. A sensing layer was simply fabricated by drop-casting a dye solution of bromocresol green (BCG) on a polyvinylidene fluoride substrate, and its color-change response was enhanced by optimizing the amounts of additive chemicals, such as polyethylene glycol, p-toluene sulfonic acid, and graphene oxide in the dye solution. To avoid the adverse effects of water vapor, both faces of the sensing layer were enclosed by using a polyethylene terephthalate film and a gas-permeable microporous polytetrafluoroethylene sheet, respectively. When exposed to basic gas analytes, the paper-like sensor distinctly exhibited a color change from initially yellow, then to green, and finally to blue due to the deprotonation of BCG via the Brønsted acid-base reaction. The use of ammonia analyte as a test gas confirmed that the sensing performance of the optimized sensor was reversible and excellent (detection time of < 15 min, sensitive naked-eye detection at 0.25 ppm, good selectivity to common volatile organic gases, and good stability against thermal stress). Finally, the coloration intensity of the sensor was quantified as a function of the storage time of the salmon filet at 28℃ to evaluate its usefulness in monitoring of the food freshness with the measurement of the total viable count (TVC) of microorganisms in the food. The TVC value increased from 3.2 × 105 to 3.1 × 109 cfu/g in 28 h and then became stable, whereas the sensor response abruptly changed in the first 8 h and slightly increased thereafter. This result suggests that the colorimetric response could be used as an indicator for evaluating the degree of decay of salmon induced by microorganisms.

석산개발에 따른 주변 환경오염 및 석분토 처리를 위한 연구 (Environmental Contamination and Best Management of Stone-dust from Quarry Mine)

  • 이번 연구에서는 석산 내부(광산) 및 외부(주변 환경)에서 채취한 모든 물 시료와 응집제를 함유한 석분토에 대해서 중금속, 시안, 독성 유기화합물 및 유기인의 총함량을 분석하였다. 그리고 EDTA 용출실험법 및 산농도 변화에 따른 용출실험법 등을 이용하여 석분토를 장기간 석산 내부에 보관할 경우 발생할 수 있는 중금속 원소의 용출특성을 평가하였다. 모든 물 시료와 석분토에서 검출된 $Cr^{6+}$, Hg, CN, TCE/PCE 및 총 인의 농도는 모두 검출한계 이하로, 오염되지 않았음을 확인하였다. 석산 내부 및 외부에서 채취한 물 시료에서는 Pb과 CD이 검출되지 않았고 구리와 아연은 일부 시료에서만, 비소는 일반적으로 검출되었으나 검출된 비소 함량은 모두 먹는 물 수질기준보다 훨씬 낮은 값을 보여주고 있다. EDTA 및 산도변화를 이용한 용출실험 등의 결과에서 Pb, Cr, Cd, Cu 및 Zn의 용출함량은 검출한계 이하 혹은 매우 낮은 함량만이 검출되었으며, 비소는 pH 3의 용출실험에서 검출되었다. 따라서 석산 개발에 따른 수계 오염은 없는 것으로 판단된다.

국내에 사용되어 금지된 유기염소계 농약류의 다중분석법과 모니터링 (Development of Multi-residue Analysis and Monitoring of Persistent Organic Pollutants (POPs) - Used Organochlorine Pesticides in Korea)

  • 본 연구에서는 참외, 풋고추 및 상추 시설재배지 토양 및 작물 중 스톡홀름협약에서 대부분 잔류성유기오염물질(POPs)로 지정된 잔류성유기염소계 농약의 잔류량을 조사하였다. 토양 및 작물 중 잔류성유기염소계 농약 분석을 위한 추출 및 정제는 QuEChERS 방법을 다소 개선한 방법으로 수행되었다. 시설재배지 토양, 참외, 풋고추 및 상추 중 잔류성유기염소계 농약의 회수율은 각각 78.2-110.6, 85.2-94.2, 78.3-92.4 및 73.3-92.6% 이었으며, 토양과 3개 작물에서의 검출한계는 각각 0.01-0.08과 $0.11-0.17{\mu}g/kg$ 수준이었고, 상대표준편차는 토양 및 3개 작물에서 각각 1.0-4.1, 1.9-3.9, 2.8-4.8 및 2.5-5.8%로 20% 미만으로 적합한 범위이었다. 참외, 풋고추 및 상추 시설재배지 토양에서 dieldirn, ${\beta}-endosulfan$ 및 endosulfan sulfate 3 성분이 각각 1.4-72.5, 0.1-78.7 및 $0.0-214.1{\mu}g/kg$ 수준으로 검출되었으나 참외, 풋고추 및 참외의 모든 시료에서는 검출되지 않았다. 이러한 결과는 참외, 풋고추 및 상추 시설재배지 토양이 농산물 안전성을 위협할 정도로 잔류성유기염소계 농약으로 오염되지 않았음을 나타냈다.
