Mulberry Paper Bag μ-Solid Phase Extraction for the Analysis of Five Spice Flavors by Gas Chromatography-Mass Spectrometry

  • Headspace micro solid phase extraction using mulberry paper bag (HS-MPB-$\mu$-SPE) has been developed and validated for the analysis of volatile flavor compounds from five spice by gas chromatography-mass spectrometry (GC/MS). HS-MPB-$\mu$-SPE was performed with adsorbent particles enclosed inside a mulberry paper bag. Four different kinds of adsorbents such as Tenax TA, Porapack Q, dimethylpolysiloxane and polyethylene glycol were tested. The extraction solvents compared were petroleum ether, methylene chloride, and chloroform. Better results were obtained when Tenax TA and petroleum ether were used. The limit of detection (LOD) and the limit of quantitation (LOQ) were in the range of 1.3 ng/mL and 4.3 ng/mL, respectively, for o-cymene as a model compound of monoterpene. Proposed method showed good reproducibility (3.3%, RSD) and good recoveries (94.0%). The HS-MPB- μ-SPE is very simple to use, inexpensive, rapid, requires small sample amounts and solvent consumption. Because the solvent for extraction is reduced to only a very small volume (0.6 mL), there is minimal waste or exposure to toxic organic solvent and no further concentration step. This method allows successful characterization of the headspace in contact with the five spice sample. Strong trans-anethole from star anise or fennel is a characteristic flavor of five spice powders. HS-MPB-$\mu$-SPE combined with GC/MS can be a promising technique for the broad spectrum measurement of volatile aroma compounds from solid spices.

  • The crystal structure of the title compounds with both coumarin and sulfonamide moieties were examined. These two groups have very special for their pharmaceutical and medicinal properties have been determined from single crystal X-ray diffraction data. In the title compound crystallizes in the monoclinic space group C2/c with unit cell dimension a = 28.633(3) ${\AA}$, b= 9.3215(7) ${\AA}$ and c= 24.590(2) ${\AA}$ [alpha & gamma=$90^{\circ}$ beta= $115.976(3)^{\circ}$]. In the structure The S1 atom shows a distorted tetrahedral geometry, with O1-S1-O2 [119.74 $(2)^{\circ}$] and N1-S1-C5 [$105.57(1)^{\circ}$] angles deviating from ideal tetrahedral values are attributed to the Thrope-Ingold effect. The sum of bond angles around N1 ($316.2(1)^{\circ}$) indicates that N1 is in sp2 hybridization. The Pyridine ring adopts boat conformation and pyran rings adopt a sofa conformation. The carboxylate group of atoms were disordered over two positions with site occupancy factors 0.598 (9):0.402 (9). Crystal structure and packing is stabilized by $C-H{\ldots}O$ intra and inter molecular hydrogen bond interactions.

Screening of Functional Materials from Solvent Fractions of Apple Flower Leaf Extract (사과꽃잎 추출물의 용매 분획으로부터 기능성 소재의 탐색)

  • Fractional solvent extraction by organic solvents such as hexane, chloroform, ethylacetate, and butanol was carried out using 70% ethanol extract of apple flower leaves. Biological activities including antioxidant, whitening, antimicrobial and anti-wrinkle activities were investigated and bio-active materials of the extracts were identified using GC/MSD. Among the tested solvent fractions, ethylacetate fraction showed the highest total polyphenol content (1218.94 ${\mu}g/mL$), and flavonoid (140 ${\mu}g/mL$). The DPPH radical scavenging activities was over 80% at a dry matterbased concentration of 200 ${\mu}g/{\mu}L$ and SOD-like activity was over 90% at 50 ${\mu}g/mL$ concentration in ethylacetate fraction that was slightly lower than of ascorbic aicd. Tyrosinase inhibition activity related to skin-whitening was over 60% by ethylacetate fraction of 100 ${\mu}g/mL$. As an anti-aging effect, elastase inhibitory activity was about 45% in ethylacetate fraction. Also, it showed a significantly antimicrobial activity against P. acenes. From GC/MSD analysis, a characteristic peak of high content in ethylacetate fraction was identified as kaempferol, which has been reported as a bioactive compound.

Plant Growth Substances Produced by Methylobacterium spp. and Their Effect on Tomato (Lycopersicon esculentum L.) and Red Pepper (Capsicum annuum L.) Growth

  • Bacteria from the Methylobacterium genus, called pink-pigmented facultative methylotrophic bacteria (PPFMs), are common inhabitants of plants, potentially dominating the phyllosphere population, and are also encountered in the rhizosphere, seeds, and other parts of plants, being versatile in nature. The consistent success of the Methylobacterium plant association relies on methylotrophy, the ability to utilize the one-carbon compound methanol emitted by plants. However, the efficiency of Methylobacterium in plant growth promotion could be better exploited and thus has attracted increasing interest in recent years. Accordingly, the present study investigated the inoculation effects of Methylobacterium sp. strains CBMB20 and CBMB 110 on seed imbibition to tomato and red pepper on the growth and accumulation of phytohormone levels under gnotobiotic conditions. Seeds treated with the Methylobacterium strains showed a significant increase in root length when compared with either the uninoculated control or Methylobacterium extorquens $miaA^-$ knockout mutanttreated seeds. Extracts of the plant samples were used for indole-3-acetic acid (IAA), trans-zeatin riboside (t-ZR), and dihydrozeatin riboside (DHZR) assays by immunoanalysis. The treatment with Methylobacterium sp. CBMB20 or CBMB 110 produced significant increases in the accumulation of IAA and the cytokinins t-ZR and DHZR in the red pepper extracts, whereas no IAA was detected in the tomato extracts, although the cytokinin concentrations were significantly increased. Therefore, this study proved that the versatility of Methylobacterium as a plant-growth promoting bacteria could be better exploited.

Application of Non-Thermal Plasma for the Simultaneous Removal of Odor and Sludge (무기악취와 슬러지 동시처리를 위한 저온플라즈마의 적용)

  • In this study, odorous compounds emitted from various wastewater treatment were treated with using the non-thermal plasma reaction, and the effluent gas from the plasma reactor was introduced to a waste sludge reactor to achieve simultaneous sludge reduction. Hydrogen sulfide, the model odorous compound, was removed at 70% using the plasma reaction, and greater than 99% removal efficiency was observed when treated by the sludge reactor. In addition, the sludge reactor showed a high efficiency of ozone removal. As ozone reacted with sludge, oxidation with organic matters took place, and total COD decreased by 50~60% and soluble COD increased gradually. As a result, the integrated process consisting of the non-thermal plasma and the sludge reactor can be successfully applied for the simultaneous treatment of malodorous gas and waste sludge.

Effects of Horseradish, Spent Mushroom Compost and Almond Hull on Odorous Compound Concentration of Pig Slurry for Recycling in Grassland (슬러리 환경개선물질이 초지환원용 양돈 슬러리의 악취물질 농도에 미치는 효과)

  • This study investigated the effect of slurry additives on reducing the concentration of odorous compounds. Slurry treatments included control, horseradish, spent mushroom, and almond hull (n = 4 each group). Odorous compounds including volatile organic compounds (VOCs phenols and indoles) and volatile fatty acids (VFAs short chain FA, SCFA, and branched chain FA, BCFA) were analyzed. Pigs of 85~120 kg body weight were fed diets formulated to meet the Korean Feeding Standard (2012) and their excretion was sampled from slurry pits. VOCs decreased (p<0.05) in horseradish, spent mushroom, and almond hull, compared to the control. SCFA was lower (p<0.05) in the horseradish group and BCFA was lower (p<0.05) in all three treatment groups compared to the control. Taken together, the tested additives are effective in reducing odorous compounds in pig slurry, indicating that the use of spent mushroom and almond hull can not only recycle the agri-byproduct but also reduce the impact of odors on surrounding communities.

Compositional Characterization and Colorant Identification of Omija (Schizandra chinensis) Fruit Extract

  • A major polyphenolic compound extracted from omija (Schisandra chinensis) fruit was structurally identified, and its composition of major nutrients was investigated as well in this study. A dominating high performance liquid chromatography (HPLC) peak of water-extracted anthocyanin represented 94.1% of total absorbable compounds at 520 nm, which was further identified with HPLC-mass spectrometry (MS). As a result, mass-to-charge ratio (m/z) of the predominant anthocyanin was determined to be 727, and it was identical to molecular mass of cyanidin-3-xylosylrutinoside (Cya-3-O-xylrut). This is the first report that colorant of omija is predominantly composed of Cya-3-O-xylrut. Omija fruit contained exclusively 3 types of monosaccharide such as glucosc (0.68 g), galactose (0.01 g), and fructose (0.52 g) per 100 g of fruits. Several organic acids, citric (3.29 g), malic (1.4 g), acetic (0.4 g), and succinic acids (0.36 g) per 100 g of fruits, were detected by high performance anion exchange chromatography (HPAEC) analysis. During the compositional analysis of tree amino acid by HPLC, it was noticed that omija fruit contained substantial amount (0.01 g/100 g of fruits) of $\gamma$-amino butyric acid (GABA).

  • An investigation on the synthesis of aluminum tartrate, one of the aluminum organic compounds, has been performed using aluminum chloride solution as a raw material. For this aim, the effect of the ratio of ethanol/Al solution and pH on the synthesis of aluminum tartrate has been examined and aluminum tartrate synthesized has also been characterized in terms of the chemical composition, X-ray diffraction pattern, particle size distribution, and SEM analysis. As a result, the synthesis more than 97% could be obtained under the conditions of pH more than 3.0 at the ratio of ethanol/Al solution of 3.0. From the chemical analysis of aluminum tartrate synthesized in this work, the content of $NH_4$, Al and C was found to be 9.10, 4.83 and 25.8%, respectively. In addition, aluminum tartrate synthesized from the aluminum chloride solution was found to be $(NH_4)_3Al(C_4H_4O_6)_3$.

Effect of Multiple Reflows on the Mechanical Reliability of Solder Joint in LED Package (LED 패키지 솔더 접합부의 기계적 신뢰성에 미치는 리플로우 횟수의 영향)

  • The research efforts on GaN-based light-emitting diodes (LEDs) keep increasing due to their significant impact on the illumination industry. Surface mount technology (SMT) is widely used to mount the LED packages for practical application. In surface mount soldering both the device body and leads are intentionally heated by a reflow process. We studied on the effects of multiple reflows on microstructural variation and joint strength of the solder joints between the LED package and the substrate. In this study, Pb-free Sn-3.0Ag-0.5Cu solder and a finished pad with organic solderability preservatives (OSP) were employed. A $Cu_6Sn_5$ intermetallic compound (IMC) layer was formed during the multiple reflows, and the thickness of the IMC layerincreased with an increasing number of reflows. The shear force decreased after three reflows. From the observation of the fracture surface after a shear test, partially brittle fractures were observed after five reflows.

  • Acetylsalicylic acid (ASA) has been issued recently in contaminated water environments because of potential impacts on ecosystem and public health. This study was aimed at investigating the possibility of ASA degradation using gamma ray irradiation. In addition, the use of sodium persulfate, hydrogen peroxide, ferrous sulfate were tested in order to examine a synergistic effect with gamma ray. The absorbed dose was ranged from 0.2 to 10 kGy and the concentration of oxidants were from 0.1 to 10 mM in this study. The concentration of ASA was gradually decreased corresponding to the increase of the absorbed dose. When soudium persulfate was simultaneously applied, most of the parent compound was completely degraded even at a low dose of 0.8 kGy. The removal efficiency of total organic carbon was 90% even at the highest dose of 10 kGy without sodium persulfate. However, the efficiency was dramatically enhanced up to 98% at the same dose by adding 10 mM of oxidants. It was suggested that hydroxyl radical ($OH{\cdot}$) and sulfate radical ($SO{_4}^-{\cdot}$) were formed in the system and made roles in degrading ASA at the same time.
