The present study identified the favorable conditions for isolation, enrichment and in vitro culture of highly purified, undifferentiated pig spermatogonial stem cell (SSC) lines that proliferate for long periods of time in culture. The colonies displayed morphology similar to miceSSC and were positive for markers of SSC (PGP9.5), proliferating germ cell (PigVASA), pre-meiotic germ cell (DAZL) and pluripotency (OCT4, SSEA-1, NANOG, and SOX2) based on immuno-cytochemistry and RT-PCR. The purity of these colonies was confirmed by negative expression of markers for sertoli cell (GATA4 and SOX9), peritubular myoid cell (${\alpha}$-SMA), differentiating spermatogonial and germ cells (c-KIT). The colonies could be maintained with undifferentiated morphology for more than two months and passaged more than 8 times with doubling time between 6-7 days. Taken together, we conclude that pigSSC could be successfully isolated and cultured in vitro and they possess characteristics similar to miceSSC.
This study was to assess the developmental capacity of oocytes matured in vitro after 20, 15, 10, 5 and 0 days of organ culture when ovaries were isolated from juvenile mice at 0-, 5-, 10-,15- and 20-day old, respectively, and to develop in vitro culture system that observed a view to morphology of ovaries and nucleus maturation of oocytes. The size of ovaries decreased 35.9%, 8.7%, 1.2% and 14.4% after 20, 15, 10, 5 days of organ culture when the ovaries were isolated from 0-, 5-, 10 and 15-day old mice, respectively. After organ culture, the recovery rates, diameters of oocytes and the number of oocytes progressed from GV to MII were increased as increasing age of mice.

Morphology and characteristics of floral organ in highbush blueberry cultivars were studied to select suitable cultivars of highbush blueberry for domestic cultivation. The stamen consists an anther and a tape-like hairy filament with well-developed trichomes. When the anther was opened, the wall of anther was not dehiscent, and pollen grains were discharged into two tubes. Pollen was mature tetrad type without being separated after meiosis (Late March). The number of pollen granules per anther was 400~1,300, the germination rate was higher in the cultivars having many pollen grains. Pistil was composed of five carpels and a shipper without separate part. The number of ovules per ovary was 39~67, therefore, the coefficient of ranged from 11.6 to 31.0%. The seed pod formation by combination of ‘Bluejay’ and ‘Sharpblue’ was higher in the cross-pollination than in the self-pollination.
The organ culture model of the whole mammary gland has many advantages for the study of branching morphogenesis and biological characteristics, including tumorigenesis. Prior to whole gland organ culture, rats were treated with 7,12-dimethylbenz[a]anthracene (DMBA) or N-methyl-N-nitrosourea (MNU) for one week. The tramdorming effect and the morphological changes were assessed by the whole mount preparations and histopathological examination in terminal end buds (TEB), terminal ducts (TD), alveolar buds (AB), alveolar lobules (AL) and hyperplastic alveolar nodules (HAN) of the mammary gland. Grossfindings of the mammary glands at dissection were higher branching morphogenesis and larger volume in carcinogen-treated groups than in carcinogen-non-treated groups. Results of the whole mount method were coincided with those of the histopathological observations. Circular TEB, normally maintained AB, AL, and high cellular density were more frequently observed in carcinogen-treated groups than in carcinogen-nan-treated groups. Histopathologically, as a preneoplastic marker, HAN was maintained only in mammary organ culture of the carcinogen-treated groups. These findings suggest that in vivo trans-formation effects by carcinogens persisted during the mammary organ culture. These results were more characteristic in DMBA than in MNU-treated group. Ducts and terminal ducts appeared to have lost morphology during their growths in case of without diethylstilbestrol (DES). The fact that in vitro organ culture without DES was resulted in abnormal ductular morphogenesis confirms that DES is a physiological regulator of ductular epithelial cell growth.
The olfactory organ of Carassius auratus and C. cuvieri was compared morphologically and morphometrically using stereomicroscopy and statistical program (SPSS version 18.0). The external morphology of the olfactory organ consists of the open semicircular anterior and posterior nostril, and nasal flap. The internal structure showed the oval rosette consisting of several lamellae arranged radiately. In statistical analysis of standard length (SL), olfactory lamellar number (LN), and SL/LN ratio between two species using independent two sample t-test and Pearson's correlation and coefficient, C. cuvieri is the longer SL than that of C. auratus and C. auratus LN (14~20) is lager than that of C. cuvieri (14~16) (P<0.001) and C. auratus LN/SL ratio (12.7±0.7%) is larger than that of C. cuvieri (8.2±0.6%). These results suggest that i) open semicircular nostrils is functional morphology to offset the boundary layer and ii) the lager LN of smaller C. auratus may be a morphological adaptation to reflect its higher olfactory dependence than C. cuvieri and iii) such interspecific difference in LN and LN/SL ratio could be applied as a new taxonomic trait for identification.
The olfactory organ of a Korean endemic species, Pseudobagrus koreanus, was investigated to describe its morphology and histology and analyze correlation between habitat environment and its ecological habits and characters, using a stereo microscope, a light microscope, and a scanning electron microscope. Its external morphology showed tubular anterior nostril at the tip of the snout and slit posterior nostril at the base of the nasal barbel. The olfactory chamber showed the rosette structure consisting of numerous lamellae radial and parallel to the medium raphe. The olfactory lamella has the sensory epithelium with olfactory receptor nuerons, supporting cells, basal cells, lymphatic cells, plasma cells and the non-sensory epithelium with stratified epithelial cells, mucous cells, lymphatic cells. In particular, 1) 41~43 (n=20) lamellae, 2) lymphatic cells and plasma cells, 3) mucous cells including neutral polysaccharid may be olfactory characteristics relevant to a stagnant or a standing habitat environment, nocturnal habit dependent on olfaction, and taxonomic characters compared at least to other catfish species.
Since 1993, sudden and massive death has occurred among penaeid shrimp, Penaeus orientalis cultured in Korea. We investigated the pathological characterizations on the spontaneously and experimentally infected penaeid shrimp. The major gross findings of the spontaneous cases were 2-6mm sized-white spots on the inside of the carapace and reddish discolorization. Histopathologically, massive necrosis of lymphoid organ, degeneration and necrosis of epithelia of epidermis and foregut were observed in both spontaneous and experimental cases. Amphophilic to basophilic intranuclear inclusion bodies were readly observed epithelia of epidermis, foregut and gills. Electron microscopy revealed enveloped, non-occuluded ellipsoid to rod shaped virus particles, within the nucleus, in the lymphoid organs and interstitial cells of hepatopancreas of both spontaneously and experimentally infected shrimps. The size of the virion was $375{\times}167nm$, and the nucleocapsid was $290{\times}75nm$. The causative agent causing massive death in penaeid shrimps in Korea resembles baculovirus associated with white spot syndrome (WSBV) occurred in Taiwan in virus morphology and gross and histological changes of the shrimps.
To study the immunotoxicity of mouse injected with fungal mycotoxin, T-2 toxin (Fusarium mycotoxin) was treated to 6 week-old female C3H/He mouse and the body, organ weight and morphological change were investigated. The weights of body, liver and kidney of mouse injected the 2 mg/kg of toxin was decreased to 17, 20 and 3%, respectively, compared to control animal and the comsumption of feed was also decreased with lapsing the time. The fat dropleting phenomenon and destruction of Golgi apparatus in liver and histopathological changes of tissue and mitochondria in small intestine were found by scanning electron microscopic observation.
The effect of ccocaine on reproductive system were studied in male ICR mice. daily injection of cocaine HCL (10, and 40 mg/kg s.c for 15 days) disrupted epididymal sperm levels and testicular morphology. These disruptions were manifested in a decreased number of sperum, an increased number of dead sperm as compared with control vehicle, and wre evidenced by vacuolations, multinucleated giant cell formations and coagulation necrosis in the testicular seminiferous tubules. Responses in the current study were trended in dose dependent pattem. These results suggest that thronic cocaine intoxication can perturb on male reproductive organ functioning.
The morphometry and morphology of the olfactory organ of the southern king spine loach Iksookimia hugowolfeldi were investigated by stereo microscopy and statistical analysis. Its external morphology consists of two holes, the anterior and posterior nostrils. The anterior nostril (0.35~0.53 mm in diameter) forms tubular process protruding vertically from each side of the snout. The posterior nostril (0.32~0.68 mm in major diameter) is a eye-like hole parallel to the skin. Internally, the rosette structure is set with 14 to 21 lamellae and medium raphe on the inner floor. In morphometry, its lamellar number (male vs. female; 18±1.8 vs. 17±1.6, P<0.05) and lamellar number to standard length ratio (24.2±1.3% vs. 21.7±2.5%, P<0.05) was larger in male than female. The lamellar number and standard length have high correlation in male (P<0.001) but not in female (P=0.170). It seems that such characteristics of the olfactory organ of I. hugowolfeldi are related to its bottom-dwelling life which digs and takes a rest under sand and sexual dimorphism by a distinct degree in olfactory dependence for reproduction of each sex.
