홍모기(Culex pipiens pallens) 지방체와 난소에서의 난황단백질합성에 관한 연구 (Ovarian and Fat Body Yolk Protein Synthesis in Culex piplens pallens)

  • 홍모기 난소에서 일어나는 난황단백질의 합성을 조사하였다. 지방체의 난황단백질합성양과 난소에 축적되는 난황단백질의 양을 rocket immunoelectorphoresis 방법과 in vitro organ culture 방법으로 조사한 결과 지방체에서의 난황단백질 합성을 흡혈 후 6시간째 부터 시작되어 24시간에 최대의 합성양을 나타낸 후 48시간 이내에 완료되었으며, 난소내로의 축적은 6시간부터 시작되어 60시간까지 계속되었다. 흡혈 후 0, 24, 48, 72시간된 난소추출물을 전기영동 및 Western blotting한 결과 24시간된 난소에서는 한종류의 난황단백질이 존재한 반면, 48, 72시간된 난소에서는 두종류의 난황단백질이 나타났다. 흡혈 후 48시간된 난소와 지방체를 $^3$H-leucine이 함유된 배지에서 배양하였을 때 지방체에서는 단백질의 합성이 거의 일어나지 않았으나 난소에서는 난황단백질을 포함한 다량의 단백질합성이 일어났다. Crossed immunoelectrophoreses의 결가 YP1과 YP2는 분자량에서는 차이를 보이나 면역성은 동일한 것으로 나타났다. 이상의 결과 홍모기에서는 난소에서도 난황단백질의 합성이 일어나는 것으로 나타났다.

A successful management after preterm delivery in a patient with severe sepsis during third-trimester pregnancy

  • A 33-year-old woman visited the emergency department presenting with fever and dyspnea. She was pregnant with gestational age of 31 weeks and 6 days. She had dysuria for 7 days, and fever and dyspnea for 1 day. The vital signs were as follows: blood pressure 110/70 mmHg, heart rate 118 beats/minute, respiratory rate 28/minute, body temperature $38.7^{\circ}C$, and oxygen saturation by pulse oximetry 84% during inhalation of 5 liters of oxygen by nasal prongs. Crackles were heard over both lung fields. There were no signs of uterine contractions. Chest X-ray and chest computed tomography scan showed multiple consolidations and air bronchograms in both lungs. According to urinalysis, there was pyuria and microscopic hematuria. She was diagnosed with community-acquired pneumonia and urinary tract infection (UTI) that progressed to severe sepsis and acute respiratory failure. We found extended-spectrum beta-lactamase producing Escherichia coli in the blood culture and methicillin-resistant Staphylococcus aureus in the sputum culture. The patient was transferred to the intensive care unit with administration of antibiotics and supplementation of high-flow oxygen. On hospital day 2, hypoxemia was aggravated. She underwent endotracheal intubation and mechanical ventilation. After 3 hours, fetal distress was suspected. Under 100% fraction of inspired oxygen, her oxygen partial pressure was 87 mmHg in the arterial blood. She developed acute kidney injury and thrombocytopenia. We diagnosed her with multi-organ failure due to severe sepsis. After an emergent cesarean section, pneumonia, UTI, and other organ failures gradually recovered. The patient and baby were discharged soon thereafter.

사생관 척도의 개발 (A Study on Development of a View of Life and Death Scale)

  • 본 연구에서는 일반 대학생을 대상으로 청년기 사생관의 전체적 구조를 밝히기 위한 도구를 개발하고자 하였다. 즉, 자살, 임신중절, 장기기증 등의 청년기와 관련이 깊은 생명윤리문제에 대한 태도는 죽음에 대한 태도와 그 배경에 있는 내세관의 영향을 받아 형성될 것이라는 가정 하에 이들 사이의 구조적 관계를 밝혀 현상을 이해하기 위한 하나의 종합적인 모형을 구축하기 위해 우선 선행 연구들에 대한 검토 결과를 토대로 하여 종합적인 새로운 사생관 척도 제작을 시도하고 신뢰도와 타당도를 확인하였다. 본 연구를 통해, 내세관 특성(내세지향성, 현세회귀성), 죽음의 의미(해방, 자연, 집대성, 좌절, 충격, 허무), 죽음불안, 죽음관여도(죽음수용, 죽음관심), 생명존중의지(자살억제의지, 중절억제의지, 장기기증의도) 등의 개념적 정의 및 조작적 정의를 재정립하였다. 이로써 개인이 가지고 있는 사생관을 전체적으로 이해할 수 있게 되었으며 사생관의 내부구조를 밝힐 수 있는 도구가 마련된 것이다. 또한 그 동안 일부 요소(예컨대 죽음불안, 죽음관여도 등)와의 관계만이 연구가 가능했던 정신건강이나 심리적 적응 및 심리 사회적 발달과 죽음에 대한 태도의 관계를 사생관 전반에 걸쳐서 보다 체계적으로 검토할 수 있게 되었다.

Vitamin C promotes the early reprogramming of fetal canine fibroblasts into induced pluripotent stem cells

  • Background: Canine induced pluripotent stem cells (iPSCs) are an attractive source for veterinary regenerative medicine, disease modeling, and drug development. Here we used vitamin C (Vc) to improve the reprogramming efficiency of canine iPSCs, and its functions in the reprogramming process were elucidated. Methods: Retroviral transduction of Oct4, Sox2, Klf4, c-Myc (OSKM), and GFP was employed to induce reprogramming in canine fetal fibroblasts. Following transduction, the culture medium was subsequently replaced with ESC medium containing Vc to determine the effect on reprogramming activity. Results: The number of AP-positive iPSC colonies dramatically increased in culture conditions supplemented with Vc. Vc enhanced the efficacy of retrovirus transduction, which appears to be correlated with enhanced cell proliferation capacity. To confirm the characteristics of the Vc-treated iPSCs, the cells were cultured to passage 5, and pluripotency markers including Oct4, Sox2, Nanog, and Tra-1-60 were observed by immunocytochemistry. The expression of endogenous pluripotent genes (Oct4, Nanog, Rex1, and telomerase) were also verified by PCR. The complete silencing of exogenously transduced human OSKM factors was observed exclusively in canine iPSCs treated with Vc. Canine iPSCs treated with Vc are capable of forming embryoid bodies in vitro and have spontaneously differentiated into three germ layers. Conclusions: Our findings emphasize a straightforward method for enhancing the efficiency of canine iPSC generation and provide insight into the Vc effect on the reprogramming process.

꽃창포 화기조직 절편체 배양으로부터 식물체 분화에 미치는 광.온도.당의 영향 (Effects of Light, Temperature, and Sucrose on Plant Regeneration from the Flower Organ Explant in Iris ensata)

  • Iris 속 식물인 꽃창포의 번식 특성에서 종자의 잡종성과 적은 분얼로 인한 분주 번식의 어려움을 해결하고 고유한 품종의 특성을 유지하기 위해서는 조직배양을 통한 대량 번식의 구명이 필요하므로 Iris 속 식물의 화기부위 별 절편체를 이용하여 기내 번식에 의한 대량 증식방법을 정립하고자 본 실험을 수행하였다. 자생꽃창포의 화피기부조직, 자방, 소화경, 화경을 치상하여 기내배양환경에 적절한 환경을 구명하고자 조도 2000 Lux에서 일장 (0~24시간), 온도 (10~3$0^{\circ}C$), sucrose (l~9%)의 조건에서 배양하였다. 자생꽃창포의 화기 절편체로부터 유식물체 재분화에 적합한 일장은 16시간의 장일조건이, 온도 조건은 $25^{\circ}C$에서, sucrose 농도는 3%에서 가장 적합한 것으로 나타났으며 특히 화기 부위 중 화피기부조직과 자방 배양에서 높은 신초 형성을 보였다. 화기조직 치상절편체로부터 부정근의 분화는 암상태에서 촉진되어지고 sucrose는 6%에서 뿌리를 생장시키며 1$0^{\circ}C$, 15$^{\circ}C$의 저온에서는 신초,뿌리의 분화가 저조하였다.

Cytomegalovirus Infection in Infantile Hepatitis

  • Purpose: The aims of this study was to compare and evaluate the clinical characteristics, laboratory data, and prognosis for infants under age 1 year with CMV hepatitis and those with viral hepatitis of unknown etiology. Methods: A retrospective study was conducted of infants under age 1 year who were admitted with acute hepatitis. The exclusion criteria consisted of: autoimmune, genetic, metabolic, toxic, HAV, HBV, HCV, toxoplasma, rubella, herpes simplex, and Epstein-Barr virus. The 30 patients included were divided into two groups based on markers for CMV (IgM anti-CMV, CMV PCR in urine, CMV culture in urine). Results: The median age of patients (n=15) was 2.8 months. No other organ involvement was detected in any patient. Peak serum total bilirubin levels (n=4) ranged from 2.6 to 6.7 mg/dL. Peak serum ALT levels ranged from 51 to 1,581 IU/L. The duration of ALT elevation ranged from 1.5 weeks to 26 weeks (median 9 weeks). All had recovered in full without ganciclovir; there were no cases of hearing loss. The median age of controls (n=15) was 2.5 months. Peak serum total bilirubin levels (n=4) ranged from 1.6 to 9.1 mg/dL. Peak serum ALT levels ranged from 26 to 1,794 IU/L. No significant differences were observed between both groups regarding the peak serum ALT levels, peak serum total bilirubin levels, duration of hyperbilirubinemia and ALT elevation. Conclusion: Although it was not possible to differentiate congenital infection with perinatal infection in this study, the prognosis of patients with CMV hepatitis without other organ involvement was good without ganciclovir treatment.

STUDIES ON THE TISSUE CULTURE OF PANAX GINSENG

  • Unlike the tissue culture in animals and human being, in higher plants various parts of the plant are cultured for varied purposes, and they are named variously depending on which parts are used as explants or what purposes they are cultured for. Followings are some of the names of culture used frequently: organ culture, tissue culture, callus culture, single cell culture, meristem culture, mericlone culture, ovary culture, ovule culture, embryo culture, endosperm culture, anther culture, pollen culture, protoplast culture, etc.. As the names of the culture indicate, in some kinds of culture the explants used for culture are actually not tissues, but organs, single cells, or protoplasts. It seems, however, convenient to call all of the above-mentioned cultures grossly as tissue culture. Several kinds of tissue culture were attempted using Panax ginseng as material and some of the results were summarized below. 1. Callus culture After dormancy of the sed was broken, whole embryo or parts (hypocotyl, cotyledon and epicotyl) of partly grown embryo were cultured in the media supplemented with growth regulators. Rapid swelling occurred in a few weeks, but most of the swelling was observed only in the basal part of epicotyl, changes in the other parts of embryo appearing in much later stages. The swelling or increase in size, however, was resulted not from the divisions of cells, but from the mere expansion of cell. Real calli were formed about two months after inoculation of explants. Callus tissues developed from cortex, pith, and vascular bundle in the cases of hypo- and epicotyl, from mesophyl tissue in the case of cotyledon. Shoots developed more easily from cotyledons regardless of whether they are detached from or attached to the embryo proper. 2. Culture in the Knudson C medium When cotyledons, detached from or attached to the embryo proper, were cultured in the growth regulator-free Knudson C medium comprision only several kinds of mineral compounds and sucrose, shoot primordium or callus developed profusely and finally plantlets were produced directly from shoot primordium or indirectly through callus. In this medium epidermal cells as well as mesophyl cells of the cotyledon became meristematic and divided, changing into multinucleate cells or multicellular bodies, developing eventually into either shoot primordia or calli. 3. Anther culture Anthers were cultured in the media supplemented with various growth regulators applied singly or in combinations. Callus was formed mostly in the connective tissue of anther. Cells of anther wall layers changed in appearance, but no division occurred. Microspores of all stages in development were not changed, ruling out the possibility that microspore-originated callus might be formed. 4. Isolation of protoplast Protoplasts were isolated from young root, leaf, and epicotyl, using 0.7M D-mannitols as osmoticum and using macerozyme and cellulase respectively for maceration and digestion of the cell wall. Production in large number of naked intact protoplast was rather difficult as compared with other plant species. Fusion of protoplasts occurred infrequently mainly due to the fewer number of naked protoplasts in the solution.

하악두연골의 성장에 관한 시험관적 연구 (GROWTH OF MANDIBULAR CONDYLAR CARTILAGE OF THE RAT IN VITRO)

  • The purpose of this study was to evaluate the effect of intrinsic factor and extrinsic factor for growth of the mandibular condylar cartilage of 4 day-old rats in a serum-free medium for 1, 4, 7, 14 days. They were compared with normal growth in vivo and with growth of spheno-occipital synchondrosis in serum-free medium. The cellular kinetics of cartilages were evaluated by auto-radiography of tritiated thymidine. 1. Condylar cartilage was enlarged with rounded head on day 14 of experiment while in vivo the rounded-headed shape changed into functionally flattened appearance. 2. On day 14 of experiment, a severe reduction of the proliferative zone and a considerable increase of the hypertrophic zone were observed while in normal control group endochondrol bone formation and bone marrow were observed. 3. The proliferative activity in the proliferative zone of condylar cartilage detected by $^3H-thymidine$ incorporation was lower than that of normal control group and decreased more than that of spheno-occipital synchondrosis, but it continued during the 14 days of culture. 4. The continued maintenance of condylar cartilage and morphologic change were disturbed in this culture system, but cell division within the proliferative zone was continued and probably linked to intrinsic factor.

Intestinal organoids as advanced modeling platforms to study the role of host-microbiome interaction in homeostasis and disease

  • After birth, animals are colonized by a diverse community of microorganisms. The digestive tract is known to contain the largest number of microbiome in the body. With emergence of the gut-brain axis, the importance of gut microbiome and its metabolites in host health has been extensively studied in recent years. The establishment of organoid culture systems has contributed to studying intestinal pathophysiology by replacing current limited models. Owing to their architectural and functional complexity similar to a real organ, co-culture of intestinal organoids with gut microbiome can provide mechanistic insights into the detrimental role of pathobiont and the homeostatic function of commensal symbiont. Here organoid-based bacterial co-culture techniques for modeling host-microbe interactions are reviewed. This review also summarizes representative studies that explore impact of enteric microorganisms on intestinal organoids to provide a better understanding of host-microbe interaction in the context of homeostasis and disease.

조직배양으로 Kalanchoe pinnata의 직접기관분화를 유도하기 위한 호르몬 연구 (Hormonal Study to Induce Direct Organ Differentiation of Kalanchoe pinnata by Tissue Culture)

  • 식물 세포는 각 세포가 발생을 통해 새로운 완전한 개체를 생산하는 전형성능이 있다. 이것을 응용하여 식물의 증식 방법으로 기내에서 호르몬을 처리하여 체세포 배 발생의 광범위한 적용으로 여러 기술이 발전하고 있다. 이 기술을 이용하기 위해서 보다 규칙적인 세포 분열을 하는 무성생식이 가능한 식물인 Kalanchoe pinnata 에 cytokinin에 속하는 kinetin과 auxin에 속하는 호르몬들 중 picloram을 서로 조합하여 첨가한 뒤 8주 동안 처리한 후 전형성능을 실험하였다. 우리의 실험 결과로 잎 절편에서 발근 효과로는 picloram 농도가 0.1 mg/L에서 70%의 발생율을 보였다. kinetin과 picloram의 비율이 1:5-1:10의 농도차이가 효과적이라는 것이 입증 되었다. auxin의 효과가 Kalanchoe 뿌리 발생에 필수적이라는 실험 결과이다. 경엽부 발생 효과로는 picloram 농도가 0.5 mg/L에서 60%의 발생율을 보였다. kinetin 농도는 0.5 - 1.0 mg/L이며 발생에 중요한 영향을 준다. kinetin과 picloram의 비율이 1:1-1:2의 농도 차이가 효과적이라는 것이 입증 되었다. cytokinin과 auxin의 조합이 결정적으로 경엽부 발생에 중요하다는 것을 보여 주는 결과이다. 호르몬의 조합으로 직접기관형성을 유도하여 기내에서 대량 증식을 유도하는 기술의 기초가 될 수 있다고 사료된다.
