Ethanolic extract of Condurango (Marsdenia condurango) used in traditional systems of medicine including homeopathy against cancer can induce DNA damage and apoptosis in non small lung cancer cells, A549 and H522, in vitro

  • In traditional systems of medicine including homeopathy, the Condurango extract (Con) is often used to cure stomach cancer mainly, without having any scientific validation of its anti-cancer ability. Con has therefore been tested against non-small-cell lung cancer cells (NSCLC) A549 and NCI-H522 (H522) known to contain the KRAS mutation, making them resistant to most chemotherapeutic agents. As cancer cells generally defy cytotoxicity developed by chemopreventive agents and escape cell death, any drug showing the capability of preferentially killing cancer cells through apoptosis is worth consideration for judicious application. A549 and H522 cells were exposed to $0.35{\mu}g/{\mu}l$ and $0.25{\mu}g/{\mu}l$ of Con, respectively, for 48 h and analysed based on various protocols associated with apoptosis and DNA damage, such as MTT assay to determine cell viability, LDH assay, DNA fragmentation assay, comet assay, and microscopical examinations of DNA binding fluorescence stains like DAPI, Hoechst 33258 and acridine orange/ethidium bromide to determine the extent of DNA damage made in drug-treated and untreated cells and the results compared. Changes in mitochondrial membrane potential and the generation of reactive oxygen species were also documented through standard techniques. Con killed almost 50% of the cancer cells but spared normal cells significantly. Fluorescence studies revealed increased DNA nick formation and depolarized membrane potentials after drug treatment in both cell types. Caspase-3 expression levels confirmed the apoptosis-inducing potential of Con in both the NSCLC lines. Thus, overall results suggest considerable anticancer potential of Con against NSCLC in vitro, validating its use against lung cancer by practitioners of traditional medicine including homeopathy.

Symptomatology of Citrus mosaic sadwavirus (CiMV) in Some Citrus Cultivars and Effect of CiMV Infection on Citrus Fruit Quality

  • Citrus mosaic sadwavirus (CiMV) is a closely related virus with the Satsuma dwarf virus (SDV) along with Navel orange infectious mottling virus (NIMV), Natsudaidai dwarf virus (NDV), and Hyugagatsu virus (HV). The present study found that the typical symptoms of CiMV-infected citrus fruits include the appearance of dark blue speckles or ringspots on fruit rinds and the browning of oil glands in the spots as rind coloring began. As rind coloring progressed, the spots gradually faded, whereas the browning of the oil glands worsened to the point that the tissues surrounding the oil glands became necrotic. In very early satsuma mandarins (Citrus unshiu 'Miyamoto Wase') and 'Setoka' cultivar (C. hybrid 'Setoka') of late-maturity citrus, the symptomatic fruits were eventually dropped. And in early satsuma mandarin (C. unshiu 'Miyakawa Wase'), the peel hardness of the virus-infected fruit (1,618.3 ± 305.5, g-force) was more than twice as hard as that of the healthy fruit (636.5 ± 39.1, g-force). The ratio of flesh weight to total fruit weight was higher for the healthy fruit (77.3 ± 1.7%) than for the infected fruit (70.7 ± 0.6) and peel puffing was more severe in the infected fruit (2.9 ± 0.4 mm) than in the healthy fruit (0.9 ± 0.2 mm). The soluble solids content in infected citrus fruits was less values than the healthy fruit by 0.5-1.5 °Brix. These findings reveal that CiMV infection on citrus trees reduces the fruit quality of citrus.

Moraxella catarrhalis의 광역학적 비활성화 (Photodynamic Inactivation of Moraxella catarrhalis)

  • The aim of this study was to evaluate the bacterial effects of Moraxella catarrhalis in otitis media with effusion (OME) by photodynamic therapy (PDT). Bacterial suspensions (10000 CFU/mL) were prepared. The colony forming units (CFU) of Moraxella catarrhalis have been measured after an application of photogem plus 632 nm diode laser irradiation. One ml of the bacterial suspensions have been incubated in the dark for 3h with various concentrations of photogem ($0.625{\sim}5.0_{\mu}g/mL$) and then irradiated with 632 nm diode laser ($15J/cm^2$). After, the PDT Moraxella catarrhalis suspensions ($50{\mu}L$) were inoculated on chocolate agar plate and cultured in the dark at $37^{\circ}C$, 5% $CO_2$ condition for 18h. The colony forming units off the bacteria were measured. Also transmission electron microscopy (TEM) was employed to evaluate the effect of otitis media pathogens by PDT. The nucleus of Moraxella catarrhalis was stained using green fluorescent nucleic acid dye thiazole orange and the fluorescence intensity of the nucleus was measured by flow cytometry. The PDT was effective in killing Moraxella catarrhalis at the photogem dose of $5.0_{\mu}g/mL$, respectively, As assessed by flow cytometry analysis the fluorescence intensity of the nucleus got lower after PDT. TEM result appeared to able to cause damage to the bacterial membranes. On the basis of these findings, bacterial photodynamic therapy with photogem can be considered to be a promising new therapeutic approach for OME.

광촉매를 병합한 플라즈마 공정을 이용한 폐수에 함유된 살충제 분해 (Degradation of Pesticides in Wastewater Using Plasma Process Coupled with Photocatalyst)

  • 광촉매 혼성 저온 플라즈마는 폐수에 함유된 유기물을 분해시키는 효과적인 기술이다. 본 연구에서는 광촉매가 결합된 특별히 설계된 유전체 방전 시스템을 골프장이나 감귤농가에서 흔히 살포되는 디크로보스, 카보퓨란 및 메치다치온 살충제의 분해에 적용하였다. 단독 및 병합 시스템에서 살충제의 분해를 평가하였다. 단독 시스템은 UV의 차폐 유무 및 산소기체와 공기에 의한 오존(각종 반응 활성종들 포함) 플라즈마를 이용하였다. 혼성 시스템은 UV로 활성화된 산화아연, 이산화티타늄과 그래파이트 옥사이드와 결합하여 공기에 의한 플라즈마 반응에 적용하였다. 그래파이트 옥사이드는 모사 허머스 법으로 제조하여 FT-IR 분광기로 성능을 측정하였다. 반응시간 60 min에서 UV를 차폐하고 공기를 이용한 플라즈마 반응에 의한 분해성능과 비교하였으며, UV로 활성화된 그래파이트 옥사이드(0.01 g/L)와 결합된 플라즈마 반응은 디크로보스와 카보퓨란의 각각 100% 분해도를 보였다. UV를 활용한 광촉매 혼성 플라즈마는 살충제를 분해시키는 효과적인 대안으로 입증되었다.

  • 퇴비로부터 분리한 Niabella ginsenosidivorans $BS26^T$ 균주의 유전체서열을 분석하였다. 균주 $BS26^T$의 유전체는 G + C 비율이 44.48%이며, 4,800개의 유전자와 4,704개의 단백질 코딩 유전자, 85개의 위유전자 그리고49개의 RNA유전자를 포함한 단일 원형 염색체로 구성되었으면 그 크기는 5,627,734 bp였다. 균주 $BS26^T$는 인삼사포닌의 당 분해에 관여하는 여러 타입의 글라이코시다제 유전자를 가지고 있었다. 이러한 유전체 분석은 주요 진세노사이드 전환에 관여하는 유전자 특징을 이해하는데 큰 기여가 되었다.

조생종 추석용 단감 '초시' 육성 ('Chosi', Early Maturing Sweet Persimmon (Diospyros kaki Thunb.) for 'Chuseok')

  • 조생종 추석용 품종을 육성하기 위해서 2000년 '조홍시'에 '서촌조생'을 교배하여 얻은 실생묘를 전라남도 영암군 소재 배연구소 교배육성 포장에 식재하였고 그 중에서 과실 특성이 우수한 '00-10-135' 계통을 1차 선발하여 검토한 결과, 연차간 변이가 적고 특성이 우수하여 최종 선발하여 '초시'로 명명하였다. 과실의 숙기는 9월 19일경으로 조생종이며, 평균 과중은 161 g이고, 당도는 15.3 °Bx이었으며, 과즙이 많고, 과육이 아삭하여 식미가 우수하다. 과형은 편원형이고 과피색은 오렌지색으로 외관이 수려하고, 과정부 열과, 과피미세균열 등 생리장해가 발생하지 않아 재배관리가 쉽다(등록번호: 제7075호).

Adsorption process efficiency of activated carbon from date pits in removing pollutants from dye wastewater

  • The presence of high amounts of organic and inorganic contaminants in textile wastewater is a major environmental concern. Therefore, the treatment of textile wastewater is an urgent issue to save the aquatic environment. The disposal of large quantities of untreated textile wastewater into inland water bodies can cause serious water pollution. In this study, synthetic dye wastewater samples were prepared using orange dye in the laboratory. The synthetic samples were then treated by a batch adsorption process using the prepared activated carbon (AC) from date pits. The wastewater parameters studied were the pH, total dissolved solids (TDS), total suspended solids (TSS), electrical conductivity (EC) and salinity. The activated adsorption process showed that the maximum removal efficiencies of electric conductivity (EC), salinity, TDS and TSS were 65%, 92%, 89% and 90%, respectively. The removal efficiencies were proportional to the increase in contact time (30-120 min) and AC adsorbent dose (1, 3 and 5 g/L). The adsorption profile indicates that 5 g/L of adsorbent delivers better results for TDS, EC, TSS and salinity at contact time of 120 min. The adsorption characteristics are better suited to the pseudo-second-order kinetic model than to the pseudo-first-order kinetic model. The Langmuir and Freundlich isotherms were well suited for describing the adsorption or contact behavior of EC and TSS within the studied system.

연하보조식 점도 조절 가이드를 위한 국과 음료류의 점도증진제 첨가 조건 설정 (Study on Setting the Amount of Thickening Agent in Soup and Beverages as a Guide for Modifying the Viscosity of Dysphagia Diets)

  • This study aimed to establish the optimal amount of thickening agent for the appropriate viscosity in soups and beverages, which are part of the dysphagia diet. The soups were bean sprout soup and soybean paste soup; the beverages were orange juice, regular milk, and low-fat milk; the thickening agent was one type of xanthan gum product. After adding the thickening agents (from 1 g to 5 g per 200 mL of the test food), syringe tests were conducted over time (5, 10 and 15 minutes) to verify the effects of the amount of thickening agent added per sample and the time between addition and achieving the resulting viscosity, and to establish the optimal addition conditions to reach IDDSI levels 1, 2, and 3 of the dysphagia diet. Water (based on 200 mL) was used as the standard control. These results provide a useful basis for customized diets based on the patient's dysphagia severity. On the other hand, this study is limited by including only liquid foods in the dysphagia diet and one type of xanthan gum-based thickening agent. Therefore, it is necessary to conduct continuous research, based on the study results, to modify the viscosity of the dysphagia diet using various thickening agents and foods and prevent nutritional deficiencies by managing the diet according to the patient's swallowing ability.

한국인에서 Aldehyde Dehydrogenase 2 유전자 변이가 알코올의 신경인지 기능, 정신운동성 수행 및 주관적 반응에 미치는 영향 (Effects of Alcohol on Neurocognitive Function, Psychomotor Performance and Subjective Response in Koreans with Different ALDH2 Genotypes)

  • Objective : The purpose of this study was to evaluate the effects of alcohol on neurocognitive function, psychomotor performance and subjective response in healthy Korean adults with different ALDH2 genotypes. Method : A total of 24 males, half with active $ALDH2^*1/2^*1$ and the other with inactive $ALDH2^*1/2^*2$, was selected through genotyping using restriction fragment length polymorphism. In a double-blind, placebo-controlled cross-over design, each subject consumed 0.5g/kg dose of alcohol, given as a mixture of 40% vodka and orange juice, and placebo(orange juice) on two separate occasions on an average of weekly intervals. The blood alcohol concentrations(BACs) were measured using a breath analyzer at baseline and at 30, 60 minutes after drinking. P300s were measured at baseline and at 30 minutes after alcohol and placebo intake. Vital signs and psychomotor performance[Critical Flicker Fusion Threshold(CFFT), Choice Reaction Time (CRT), Digit Symbol Substitution(DSS)] were measured at baseline and at 60 minutes after alcohol and placebo intake. Subjective responses were measured at the end of the study. The statistical analysis focused on whether there were any differences between groups with different ALDH2 genotypes. Results : The major results are as follows. 1) BACs in the inactive group were overall equivalent to those in the active group. Only in terms of time, BACs were significantly higher overall at 30 minutes than at 60 minutes after alcohol intake. 2) Pulse rates were significantly increased after alcohol intake compared with placebo, and the increase was greater in the inactive than in the active group. 3) P300 latencies in leads Fz(frontal), Cz(cental) and Pz(parietal) were significantly increased after alcohol intake compared to placebo, and the increase was greater in the inactive than in the active group. P300 amplitudes in leads Cz and Pz were significantly decreased overall after alcohol intake compared to placebo. 4) Compared with placebo, alcohol produced significant effect on the psychomotor performance : impairment in the inactive group, improvement in the active group. 5) Compared with placebo, alcohol significantly induced a negative or an intense effect on the subjective responses in the inactive group, but little negative and even a somewhat positive effect in the active group. Conclusions : These results suggest that ALDH isozyme variance might be an important factor to determine the effects of acute dose of alcohol on the various psychobehavioural functions and also to determine the alcohol use pattern and to predict the future development of alcohol overuse and/or abuse.

산란계에 녹차 첨가 사료가 계란의 이화학적 성질에 미치는 영향 (Effects of a Diet Containing Green Tea Powder on the Physicochemical Properties of Eggs)

  • 녹차가 계란의 품질 특성에 미치는 영향을 조사하기 위해 녹차 분말을 기존 산란계 사료(대조군)에 4%(A), 8%(B) 첨가하여 10주간 식이하게 한 후 생산된 계란의 이화학적 특성을 분석하여 나타낸 결과는 다음과 같다. Ca, K, P, Fe, Mg 등의 주요 무기질 함량은 계란의 부위(흰자, 노른자)에 관계없이 녹차 함량에 비려하여 증가하는 경향을 보였으나 무기질의 절대 양은 Mg 함량을 제외하면 흰자에서 보다는 노른자에 많았다. 무기질 원소의 증가 비가 가장 큰 원소는 흰자에서는 Ca이온으로 41.0 mg%에서 119.8 mg%로 2.9배 증가하였고, 노른자에서는 Fe이온으로 3.7 mg%에서 12.6 mg%로 3.4배 증가하였다. 계란의 평균 중량은 녹차 식이 양에 비례하여 감소(68.8 g에서 64.4 g)하는 경향이었고, 난황의 총 콜레스테롤 함량은 1899.1 mg%∼2011.3 mg% 으로서 녹차 식이와는 유의 차이가 없었다. 흰자는 녹차 식이와 관계없이 떫은맛을 내고 옅은 회색을 나타내나, 노른자는 녹차 식이를 증가시킴에 따라서 약간 검붉은 색으로, 그리고 비린내가 적은 경향이었다. 계란의 수소이온농도 값은 녹차 식이 양의 증가와 더불어 감소하는 경향이었는데 B 처리군의 흰자, 노른자 및 전체의 값은 각각 8.45, 5.96 및 7.66이었다. 계란 노른자의 주요 지방산은 palmitic acid, stearic acid, oleic acid, linoleic acid 으로서 전체 지방산의 90% 이상을 차지하였고, 녹차 식이 양을 증가시킴에 따라서 포화지방산은 32.0%에서 27.4% 으로 감소하고, 불포화지방산은 68.0%에서 72.6% 으로 증가하는 경향으로 나타났다.
