Sialadenoma papilliferum (SP) is a rare benign neoplasm that normally arises from the minor salivary glands, particularly in the palate. SP is normally encountered in older men with an exophytic papillary surface growth. In the present study, an SP of the hard palate of a 69-year-old woman was examined immunohistochemically. Myoepithelial cell markers, such as S-100, smooth muscle actin and vimentin, were observed in the basal or luminal layer of tumor cells, indicating that myoepithelial cells participate in the pathogenesis of SP. In addition, cytokeratin 7 was also strongly detected in the tumor cells, suggesting that excretory ductal epithelial cells have a role in its histogenesis. A review of the literature of immunohistochemical studies on SP showed that the expression and co-expression of cytokeratins and myoepithelial cell markers have been reported in tumor cells. These results suggested that excretory duct cells and myoepithelial cells participate in the pathogenesis of SP.
The chitosan-coated liposomes (chitosomes) were designed to improve the stability in the gastrointestinal (GI) tract and to enhance the efficacy for oral drug delivery of liposomes. The phosphatic acid (PA)-incorporated anionic liposomes were surface-coated with water soluble chitosan (WSC) by electro-ionic interaction. The shape of the chitosomes observed by transmission electron microscopy (TEM) was spherical in all the formulations and the coating layer by WSC could be founded through TEM images. The mean size and the zeta potential values of the chitosomes increased significantly with depending on the content of WSC added for coating the liposomes. The stability of the chitosomes in the GI tract was confirmed through the change of relative turbidity of the liposomal suspension. The plain liposomes (plasomes) suspension without adding WSC clearly showed the change of relatively turbidity in simulated gastric fluid (SGF), while the change degree of turbidity of the chitosomes in the SGF decreased as increasing of WSC content added for coating liposome. In the 5-CF release study from the plasomes and chitosomes, the plasomes released >90% of the initial 5-CF content at 4 h of release measurement. In contrast, the chitosomes released below 40% of initial content of 5-CF. In conclusion, these results indicate that the chitosomes can be used as a potential carrier for effective oral drug delivery.
In order to search for anticancer agent as therapy of oral squamous cell carcinoma (OSCC) and malignant pleural mesothelioma (MPM) from Korean traditional prescriptions, we selected 58 traditional prescriptions based on a review of the Korean traditional medicine books. Among the selected traditional prescriptions, only water extracts of paedoksan (敗毒散) showed relatively good cytotoxicity at the concentration of $50{\mu}g/ml$. Additionally, we evaluated cytotoxicity for various paedoksans and each herbal ingredient in paedoksans. The root of Anthriscus sylvestris exhibited more cytotoxic effect than any other ingredients in paedoksans. Bioactivity-guided fractionation of the MC layer of Anthriscus sylvestris led to the isolation of deoxypodophyllotoxin (DPT). DPT exhibited dose-dependently significant cytotoxicity against OSCC and MPM cell with nM range. Therefore, DPT from A. sylvestris might be a potential candidate as an effective anticancer therapeutic agent for OSCC and MPM.

Objectives: Papilloma frequently develops as a benign tumor of the head and neck area, but its potential for malignant transformation has yet to be studied. This study aims to provide basic information for papillomas using the immunohistochemical staining of matrix metalloproteinase 2 (MMP-2) and human papilloma virus (HPV) 16 and 18. Materials and Methods: To evaluate the malignant transformation of papillomas, the selected tissue samples were serially diagnosed with pre-cancerous papilloma (with epithelial dysplasia, pseudo-epitheliomatous hyperplasia) or malignant lesion (squamous cell carcinoma, SCC) after the first diagnosis (squamous papilloma, inverted papilloma). The selected tissues were stained with an antibody to MMP-2 and HPV 16-E7, HPV 18-L1. A statistical analysis was performed according to each transformation step. Results: The epithelial layer of papilloma and pre-cancerous papilloma lesions had a similar MMP-2 expression, but that of the malignant lesion had a significantly increased MMP-2 expression. HPV 16 and 18 infection rates were 28.6%, 33.3% and 63.6% in papillomas, pre-cancerous papilloma lesions, and SCC. Conclusions: A relatively high MMP-2 expression and HPV 16 or 18 infection of papillomas may be associated with early events in the multistep processes of malignant transformation of papillomas.

The purpose of this study was to investigate chronic radiation effects on the basal cell of the rat tongue epithelium according to different irradiation timing. Forty-two female rats were devided into 5 experimental groups according to different irradiation timing and were irradiated single dose of 396cGy by MK cell irradiator using Cs-137. Experimental rats were sacrificed at the 2nd week, 4th week and 6th week after birth. The specimens were examined with light microscope and transmission electron microscope. The following results were obtained. 1. The first changes after irraditation were vacuoles. The vacuoles were chiefly observed in the cytoplasm, perinuclei area, and nuclei. 2. The most severe degenerative changes in the basal cell layer were observed in all experimental groups. ; cellular disarrangement, vacuole formation, widening of intercellular space, enlarged mitochondria & rER, and chromatin clumping were seen. 3. The cellular degenerative changes were most severe at the 4th week after birth in all experimental group, and the basal cell hyperplasia was seen at the 6th week in the most of experimental groups 4. The experimental groups 3 and 4 show more severe and more prolonged cellular degeneration than experimental groups 1 and 2, which were irradiated in pregnancy, and experimental group 5, which was irradiated after tongue maturation.

It is known that radiation therapy is a kind of treatment choices of the maxillofacial tumors. This study is designed to investigate the effects of irradiation on rat's tongue tissues as functional tissues which relate to taste, mastication, and pronunciation. 88 rats (Sprague Dawley branch, male) were divided into control group of 4 and experimental group of 84. Experimental group was singly exposed to Co-60 irradiation with 8, 13, 18 Gy in the head and neck region. Animals were sacrificed on 1 hour, 3 hours, 6 hours, 1day, 3 days, 7 days, and 28 days after the irradiation. The specimens were observed by histopathological examination employing H-E stain and Van-Gieson stain. The results were follows; 1. The tongue tissue were severely swollen on the 1 hour after irradiation, but gradually decreased in course of time. 2. The basal cells of epithelium of tongue proliferated at initial stage of irradiation, but gradually decreased. The Keratin layer were gradually increased. 3. The muscles within the tongue were severely degenerated at initial stage of irradiation, but gradually recovered almost normally. 4. The tissue changes after irradiation were gradually increased by the degree of irradiation.

The objective of this study was to examine the expression pattern of Kelch-like ECH-associated protein 1 (Keap1) in the maxillary $2^{nd}$ molar germs of rats. We used the maxillary $2^{nd}$ molar germs in rats' pup at postnatal day 3 (bell stage), 6 (crown formation stage) and 9 (root formation stage). The investigation on mRNA and protein levels were done using reverse transcription - polymerase chain reaction and western blot. Localization of Keap 1 in the maxillary $2^{nd}$ molar germs were revealed through immunofluorescence staining. Keap1 from the maxillary 2nd molar germs were mostly manifested on postnatal day 3 and dramatically decreased on postnatal day 6 and 9 at mRNA and protein levels, while amelogenin and ameloblastin increased during the development of maxillary 2nd molar germs. During immunofluorescence analysis, the strong immunoreactivity against Keap1 was detected in the apical side of ameloblasts at the presecretory and secretory stages. However, Keap1 expression was hardly observed in the ameloblasts at the maturation stage. These results shows that Keap1 is strongly expressed in the presecretory and secretory ameloblasts of amelogenesis, and suggest that Keap1 may be a crucial molecule for the regulatory mechanisms tasked with the formation of enamel layer.
As a selective ${\alpha}_{1A}-adrenoreceptor$ antagonist, tamsulosin has been used clinically for urinary obstructed patients with benign prostatic hyperplasia. The single and multi-layered pellets containing tamsulosin hydrochloride were prepared in an effort to control the drug release, avoiding dose-dependent side effects of tamsulosin hydrochloride upon oral administration. The drug release from multi-layered pellets was substantially controlled, compared with single layered pellets. The drug release from coated pellets with single or multi layer was affected by the nature of coating agent, the percentage of coating level and the presence of hydrophilic material in coating layer. In conclusion, the controlled release oral delivery system using multi-layered pellet is very useful for tamsulosin hydrochloride, resulting in improvement of patient compliance and therapeutic drug levels for a longer period of time.
The pellets with multiple drug release system (MDRS) of Diltiazem. HCl which consist of immediate drug release layer, drug reservoir layer and controlled release rate membrane, were prepared by using CF-Coater. As main factors for more effective MDRS of Diltiazem. HCl, ethylcellulose was used for the controlling drug release rate, and diethylphthalate was used for plasticizer, respectively. In vitro evaluation study was performed by comparative dissolution test between our test MDRS and reference Diltiazem. HCl preparation. The physical tests were performed using FT-IR and SEM. In vivo evaluation was also performed by observing the behavior of a plasma drug concentration after oral administration. The bioavailability was determined by analyzing the blood sample after oral administration to healthy, male volunteers once a day. As a result, there were no significant differences in bioequivalence parameters $(AUC_{\infty},\;C_{max},\;t_{1/2})$ between two systems. It might be concluded that our MDRS of Diltiazem. HCl could be an alternative delivery system to reference drug preparation.

The author observed the effects of /sup 60/Co irradiation on the development and subcellular structure of tongue tissue of the fetal rats. The lower left abdomen of mothers were exposed to radiation on 15½th day of gestation with 300R. The fetuses were removed on the 6hr, 14hr, 24hr, 48hr and 72hr after irradiation and the light microscopic and electron microscopic observations of the lingual epithelium, lamina propria and muscle layer were carried out. The results were as follows: 1. The irradiated fetuses showed the retardation of filiform papillae formation. 2. Epithelial cells revealed fusion and myelination of mitochondria, large autolysosomes, increased lipid droplets, retardation of tonofilaments and desmosome formation. 3. In the lamina propria, undifferentiated cells showed bleb formation of nuclear membrane, pyknosis and fragmentation of nucleus, edema of cytoplasm I and nucleus, increased auto-lysosomes, dilatation of cell membrane and cell necrosis. Also, collagenous fibril formation was inhibited by irradiation. 4. In the muscle layer, growth of myotubes was inhibited. Myotubes showed swelling of mitochondria, loss of mitochondrial cristae, autolysosomes, retardation of myofibril formation, and large vacuoles. Undifferentiated cells adjacent myotube contained pyknotic nucleus and autolysosomes. 5. Among the various tissues of tongue, it seems that mesenchymal cells were most radiosensitive.
