구아바 잎 열수추출물의 당류분해효소 저해 효과 및 OGTT에 미치는 효과 (Effect of Aqueous Extract of Guava (Psidium guajava L.) Leaves on the Oral Glucose Tolerance Test and Inhibition of Glycoside Hydrolase)

  • Guava ($Psidium$ $guajava$) contain a great deal of polyphenol compound and work on the treatment of $Diabetes$ $mellitus$ effectively. In this study, the bioactivities of aqueous extract (GLEx) of guava leaf were investigated. Total phenolic contents of GLEx was 266.9 mg tan/g. The effects of GLEx on digestive enzymes, ${\alpha}$-amylase, maltase and sucrase were investigated. $IC_{50}$ values of GLEx against ${\alpha}$-amylase, maltase and sucrase were 0.65 mg/$m{\ell}$, 2.0 mg/$m{\ell}$ and 3.5 mg/$m{\ell}$ respectively. The effect of GLEx on oral glucose tolerance test (OGTT) was performed in normal ICR mouse, control (dstilled water) and GLEx (aqueous extract of Guava leaf) were co-administered orally with glucose, showed reducing effect on the blood glucose level. The guava is likely to useful for prevention or improvement of hyperglycemia by lowering the blood glucose level and inhibiting glycoside hydrolase activity.

본초 귀경에 따른 상소한약의 당내성 유발 마우스에서의 개선 효능 비교 연구 (Comparison of the efficacy of the herbs for upper medication on glucose tolerance induced by high fat/high sucrose feeding-induced mice)

  • Objectives : To prove the channel-tropism theory of herbal medicines on diabetes mellitus as emaciation-thirst disease in Korean Medicine Theory, we investigated the selective therapeutic effects of Mori Cortex Radidus (MCR), Schisandrae Fructus (SF), Anemarrhenae Rhizoma (AR) for the upper emaciation on different organs in high fat and high sucrose (HF/HS) feeding-induced prediabetic mice. Methods : Diabetes in C57BL/6 mice was induced by the administration of high fat (45 kal%) and high sucrose (32 kal%) for 8 weeks, and them treated with each extract at 250 or 500 mg/kg body weight for 4 weeks (once a day). Oral glucose tolerance test and body weight was measured once a week. Insulin, total cholesterol, triglyceride, ${\gamma}GTP$, GOT and GPT were measured in the sera of all mice. Histopathological changes of different organs, lung, heart, pancreas, stomach, liver, and kidney were observed by H&E staining. Results : The results revealed that MCR extract inhibited the impaired glucose tolerance and lung damage, and increased serum insulin levels in HF/HS-induced prediabetic mice. SF extract inhibited the impaired glucose tolerance and lung damage, increased serum insulin levels, and decreased serum triglycerige levels. Meanwhile, AR extract inhibited the impaired glucose tolerance and lung damage, and decreased serum levels of insulin, total cholesterol and triglycerige levels. Conclusions : These results demonstrated that MCR, SF, and AR extract as the upper emaciation herbal medicines were followed their channel-tropism theory like a lung, and may have a selective therapeutic potential for control of diabetic stage.

하엽(荷葉) 및 하엽옥천산(荷葉玉泉散)이 streptozotocin으로 유발된 당뇨(糖尿)에 미치는 영향 (Effects of Nelumbo Folium and Hayopokchun-san on Diabetes Mellitus in Streptozotocin Treated Rats)

  • The therapeutic anti-diabetic effect of Nelumbo Folium and a poly herbal formula (NOC) was evaluated in the streptozotocin (STZ 60 mg/kg, I.p.) induced diabetic rats. For this study, test articles were orally administrated once a day from 7 d after STZ-injection for 3 weeks. The fasting blood glucose, body weight, pancreas weight changes, oral glucose tolerance test, hemoglobin Alc (Hb A1c) level changes and the histopathological changes were observed. WNL-treated rat had lower fasting blood glucose levels compared to control group and NOC-treated rat had significantly lower fasting blood glucose levels, compared to control group (p < 0.05). After 21 days of extract treatment, body weight in control was reduced (p < 0.001). WNL and NOC groups were also reduced compared to control group but insignificantly. Pancreas weight of control group was reduced (p < 0.001), but the weight of NOC group were increased (P< 0.05). During the 2 h oral glucose tolerance test (OGTT), WNL group were improved compared to control group (P < 0.05). NOC group were improved compared to control group but insignificantly. WNL and NOC groups had lower Hb Alc level compared to control group. In addition, atrophy of islet and decrease of insulin-producing cells were detected in STZ induced diabetes rats. However, these diabetic changes were decreased in WNL and NOC groups. These results suggest that N. Folium and NOC have favorable effects to inhibit the changes on the fasting blood glucose levels, pancreas weight, glucose tolerance, hemoglobin Alc and the histopathological changes of pancreas in STZ-induced diabetes.

Glucose-lowering Effect of Powder Formulation of African Black Tea Extract in $KK-A^y/TaJcl$ Diabetic Mouse

  • We observed the suppressive effect of a powder formulation of African black tea extract prepared from the leaves of Camellia sinensis on type 2 non-insulin dependent diabetic mice, $KK-A^y/TaJcl$. Black tea extract significantly showed suppressive effect of the elevation of blood glucose on oral glucose tolerance test of 8 week-old $KK-A^y/TaJcl$ mice (p<0.05). Long-term treatment with black tea extract showed significant suppression of post-prandial blood glucose and obesity (p<0.05). The weight of the intestine of mice treated with black tea extract was significantly reduced (p<0.05). From these results, African black tea used in this study showed a suppressive effect on the elevation of blood glucose during food intake and the body weight.

삼귀용탕(蔘歸茸湯)이 정상 흰쥐 및 Streptozotocin으로 유도된 당뇨병 흰쥐의 혈당에 대한 영향 (Effects of Samguiyong-tang on the Blood Glucose Levels in Normal Rats or Diabetic Rats Induced by Streptozotocin)

  • Objectives : In order to evaluate the effect of Samguiyong-tang (SGYT) on diabetes, we prepared two types of Samguiyong-tang (Type-I and -II) which was composed of three kinds of oriental drug such as Ginseng, Angelica gigantis radix and Deer antler. Type I was traditional hot-water extract prepared from three kinds of drug, and Type II was the mixture of ethanol-extract of ginseng and hot-water extract prepared from the other two drugs. Methods : We tested the effects of SGYT on the blood glucose levels in normal rats by the method of glucose tolerance test. And also examined the effects of SGYT on the levels in normal rats or diabetic rats induced by Streptozotocin during 20 days. Results : 1. In the course of oral glucose tolerance test, the blood glucose level decreased by administration of SGYT I or II in normal rats. 2. In the course administration of SGYT during 20 days in normal rats, the blood glucose levels decreased until day 4 by Type I or Type II, but thereafter the level was recovered to the normal. 3. In the course administration of SGYT during 20 days in the diabetic rats induced by streptozotocin, Type I (SGYT) had some effect on the blood glucose levels only at 12 day, and Type II (SGYT) decreased the levels from 6th day and so on, significantly. Conclusions : The results suggested that SGYT II had some decreasing effects on the blood glucose levels in diabetic rats induced by Streptozotocin.

가감생혈윤부음(加減生血潤膚飮)의 당뇨병 치료효과 확인을 위한 생리활성성분 분석과 경구포도당부하 연구 (Studies about the bioactive component analysis and an oral glucose tolerance test of Add-Omit-Saenghyeoryunbu-eum(AO-SHU) for confirmation of diabetes therapy)

  • Objectives : Instrumental chemical analysis was utilized to investigate the effect of Add-Omit-Saenghyeoryunbu-eum(AO-SHU) on diabetic treatment. One of the most exciting, yet also controversial, arguments is the safety and biological mechanisms of the natural medicine on human body. Therefore, the aim of this study is to provide a better understanding on bioactive chemical components, hazards of heavy metal contamination and biological mechanism of the diabetic medicine composed of 12 different natural herbs. Methods : To study bioactive compound and metallic component in the diabetic medicine in detail, LC-MS/MS (Liquid Chromatography-Mass/Mass), GC (Gas Chromatography) and ICP (Inductively Coupled Plasma) were utilized to characterize the extract of the diabetic medicine and the result was compared with 18 marker substances selected from literature survey. In addition, in vitro assay experiments including GPR 119 activity and human DGAT-1 inhibition, and OGTT (Oral Glucose Tolerance Test) were performed to verify the effectiveness of this medicine on diabetic treatment. Results : Out of 18 marker substances, 9 bioactive compounds were identified from LC-MS/MS analysis which include Citruline, Catalpol, Berberine, Ginsenoside Rb1, Ginsenoside Rg1, Oleanolic acid, β-Sitosterol, Mangiferin, and Schizandrin. ICP study on 245 residual pesticides revealed that 239 species were not detected but 6 species, Dimethomorph, Trifloxystrobin, Pyraclostrobin, Isoprocarb, Carbaryl and Flubendiamide, while the amounts are trace levels, below permitted concentrations. The biological activity was observed in vitro assay and Oral Glucose Tolerance Test(OGTT), which are consistent with a preliminary clinical test result, a drop in blood sugar level after taking this herbal medicine. Conclusions : Instrumental chemical analysis using LC-MS/MS, GC, and ICP was conducted successfully to identify bioactive compounds in AO-SHU for the treatment of diabetes, finding 9 bioactive compounds. Furthermore, in vitro assay experiments and OGTT show that AO-SHU has its biological activities, which imply that it can be a candidate for the future diabetes remedy.

The Hypoglycemic Effects of Acarviosine-Glucose Modulate Hepatic and Intestinal Glucose Transporters In vivo

  • Acarviosine-glucose (AcvGlc) is an ${\alpha}$-glucosidase inhibitor and has similar inhibitory activity to acarbose in vitro. We synthesized AcvGlc by treating acarbose with Bacillus stearothermophilus maltogenic amylase and fed C57BL/6J and db/db mice with diets containing purified AcvGlc and acarbose for 1 week. AcvGlc (50 and 100 mg/100 g diet) significantly reduced plasma glucose and triglyceride levels in db/db mice by 42 and 51 %, respectively (p<0.0001). The hypoglycemic and hypotriglyceridemic effects of AcvGlc were slightly, but significantly, greater than those seen with acarbose treatment (p<0.0001) in C57BL/6J mice. In an oral glucose tolerance test, glucose tolerance was significantly improved at all time points (p<0.01). The expression of two novel glucose transporters (GLUTs), GLUT10 and GLUT12, were examined by Western blot analysis. GLUT10 was markedly increased in the db/db livers. After AcvGlc treatment, the expression of hepatic GLUT10 was decreased whereas intestinal GLUT12 was significantly increased in both strains of mice. Our results show that AcvGlc improves plasma lipid and glucose metabolism slightly more than acarbose. Regulation of hepatic GLUT10 and intestinal GLUT12 may be important in controlling blood glucose levels.

고지방식이로 유발된 비만 백서에서 가미감비제습탕이 비만 유발에 미치는 영향 및 기전 연구 (Anti-obesity Effects and Mechanism of Original and Modified Gambejaeseup-tang in Female Rats with Diet-induced Obesity)

  • Gambejaeseup-tang (GBJST) have recently been used as an anti-obesity herbal medicine but their effect and mechanism of action have not been studied. We modified ingredients of GBJST based on the previous experiments about exploring herbs to suppress triglyceride accumulation in 3T3-L1 adipocytes. We investigated the effects of modified GBJST on energy, glucose and lipid homeostasis using female rats with diet-induced obesity and their action mechanism was also determined. Rats fed a high-fat diet (HFD) were divided into 3 groups: rats in each group received 0.2 or 2 g water extracts of modified GBJST (L-GBJST or H-GBJST) or 2 g cellulose per kg body weight (a negative control) on a daily basis. A further group was fed a low-fat diet (LFD) as a positive control. We found that modified GBJST dose-dependently decreased body weight and mesenteric and retroperitoneal fat more than the control. This decrease was due to the reduction in energy intake and the increase of energy expenditure. HFD increased fat oxidation more than LFD and modified GBJST further increased fat oxidation as a major energy source more than the control in a dose-dependent manner. In addition, H-GBJST improved glucose tolerance without changing serum insulin levels during an oral glucose tolerance test. H-GBJST also suppressed the increase of serum total and LDL cholesterol and triglyceride levels by HFD. In conclusion, modified GBJST have a good anti-obesity effect by decreasing energy intake and increasing energy expenditure mainly as fat in female rats with diet-induced obesity. It also improves glucose tolerance and lipid metabolism.

인동등지골피탕(忍冬藤地骨皮湯)이 Streptozotocin으로 유발된 흰쥐에서의 항당뇨 효과에 대한 연구 (Anti-diabetic Effect of Indongdeungjikolpi-tang in Streptozotocin-induced Diabetic Rats)

  • Objectives: Indongdeungjikolpi-tang(IJT) is used as a traditional treatment of diabetes in oriental clinincs. This study aimed to evaluate the anti-diabetic effect of Indongdeungjikolpi-tang(IJT) in streptozotocin(STZ)-induced diabetic rats. Methods: Diabetes was induced by intraperitoneal injection of streptozotocin(STZ; 60 mg/kg BW) to Sprague-Dawley male rats. Experimental animals(six per group), were treated by oral administration of IJT(100 and 500 mg/kg BW) and glibendimide(3 mg/kg), a known antidiabetic drug for comparison, during 4 weeks. We measured the levels of glucose, insuline, triglyceride, creatinine and urea in sera of each group. An oral glucose tolerance test(OGTT) was also performed in all groups. Results: IJT(100 and 500 mg/kg) significantly reduced blood glucose levels and increased plasma insulin levels in STZ-induced diabetic rats. IJT also significantly reduced the plasma levels of tryglyceride, creatinine and urea in STZ-induced diabetic rats. The OGTT results showed a significant improvement in glucose tolerance in IJT-administrated rats. Conclusions: These data indicate that IJT may improve glocose homeostasis in STZ-induced diabetes, which could be associated with stimulation of insulin secretion.

Antidiabetic Effects of Corni Fructus Extract on Blood Glucose and Insulin Resistance in db/db Mice

  • This study investigated the effect of Corni Fructus (Cornus officinalis Sieb. et Zucc.) extract on blood glucose and insulin resistance in db/db mice. Seven weeks old male mice were divided into normal control group (NC), diabetic control group (DC) and Corni Fructus treated diabetic group (DCF). Over an 8-week experimental period, Corni Fructus extract was administered orally at 500 mg/kg BW/day. Corni Fructus inhibited increase in blood glucose level during the OGTT (oral glucose tolerance test). At 8 weeks after beginning of the experiment, blood glucose level in the DCF group was significantly lower (p<0.01) than the DC group. Final fasting serum glucose and triglyceride in the DCF group were significantly lower (p<0.05) than the DC group by 32% and 41% respectively. Serum insulin did not differ among the NC, DC and DCF groups. The mRNA expression of adiponectin, GLUT 4 and PPAR-$\gamma$ in adipose tissue in the DC group were significantly lower than the NC group and they were higher in the DCF group than the DC group by 76%, 130% (p<0.05) and 43%, respectively. In conclusion, these results indicated that Corni Fructus would have antidiabetic effects via improving insulin resistance in favor of higher glucose utilization and reducing blood glucose level in db/db mice.
