본 연구에서는 천연물 유래 구강 건강소재로써 인도감나무 줄기 추출물의 이용 가능성을 평가하기 위해 TLC, TLC-bioautography, HPLC, ESI-MS 등을 이용하여 인도감나무 줄기 추출물로부터 S. mutans KCTC3065에 대해 항균활성이 있는 항균물질을 분리하고 주사전자현미경과 real-time PCR을 이용하여 추출물이 S. mutans의 생물막에 미치는 영향을 조사하였다. S. mutans에 대한 인도감나무 줄기 추출물의 항균활성은 극성이 낮은 n-hexane 분획물에서 확인되었고 TLC, TLC-bioautography, HPLC에 의해 분리된 항균물질의 분자량은 ESI-MS분석 결과 188로 추정되었다. 인도감나무 줄기 추출물(1 mg/ml) 처리에 따른 S. mutans의 바이오필름 바이오매스 변화는 주사전자현미경으로 관찰하였으며 추출물을 처리하지 않은 대조구는 추출물 처리구에 비해 세포가 군집을 이루고 모여 있었으며 세포 주변에서 바이오필름이 관찰되었지만 추출물을 처리한 처리구의 세포 주변에서는 바이오필름을 관찰할 수 없었다. Real-time PCR을 이용하여 바이오필름 생성 과정에서 치면 부착에 필수적인 GTFs의 발현 양상을 조사한 결과, 인도감나무 추출물이 0.2-1.0 mg/ml의 농도로 처리된 배양액에서 gtfB 유전자 발현은 추출물의 농도에 따라 큰 변화가 없었지만 gtfC 유전자 발현은 추출물의 농도가 높아질수록 감소하는 경향을 나타내었다. 이상의 결과를 종합하면 인도감나무 줄기 추출물은 바이오필름 형성 단계에서 치아우식증 원인균인 S. mutans의 초기 치면 부착을 저해함으로서 바이오필름 생성을 억제할 수 있는 천연물 유래 구강 건강소재로써 이용 가능성이 높을 것으로 판단된다.

Periodontitis results from the activation of host immune and inflammatory defense responses to subgingival plaque bacteria, most of which are gram-negative rods with lipopoly-saccharides (LPSs) in their cell walls. LPSs have been known to induce proinflammatory responses and recently it was reported also that they induce the expression of microRNAs (miRNAs) in host cells. In our current study therefore, we aimed to examine and compare the miRNA expression patterns induced by the LPSs of major periodontopathogens in the human gingival epithelial cell line, Ca9-22. The cells were treated with 1 ${\mu}g$/ml of E. coli (Ec) LPS or 5 ${\mu}g$/ml of an LPS preparations from four periodontopathogens Porphyromonas gingivalis (Pg), Prevotella intermedia (Pi), Aggregatibacter actinomycetemcomitans (Aa), and Fusobacterium nucleatum (Fn) for 24 h. After small RNA extraction from the treated cells, miRNA microarray analysis was carried out and characteristic expression profiles were observed. Fn LPS most actively induced miRNAs related to inflammation, followed by Aa LPS, Pi LPS, and Ec LPS. In contrast, Pg LPS only weakly activated miRNAs related to inflammation. Among the miRNAs induced by each LPS, miR-875-3p, miR-449b, and miR-520d-3p were found to be commonly up-regulated by all five LPS preparations, although at different levels. When we further compared the miRNA expression patterns induced by each LPS, Ec LPS and Pi LPS were the most similar although Fn LPS and Aa LPS also induced a similar miRNA expression pattern. In contrast, the miRNA profile induced by Pg LPS was quite distinctive compared with the other bacteria. In conclusion, miR-875-3p, miR-449b, and miR-520d-3p miRNAs are potential targets for the diagnosis and treatment of periodontal inflammation induced by subgingival plaque biofilms. Furthermore, the observations in our current study provide new insights into the inflammatory miRNA response to periodontitis.

We have developed an aptamer that specifically binds to Porphyromonas gingivalis to reduce the cellular damage caused by P. gingivalis infection and applied it as a biosensor. P. gingivalis is one of the major pathogens causing destructive periodontal disease among the periodontal microorganisms constituting complex biofilms. Porphyromonas gingivalis G-protein (PGP) known to play an important role in the transmission of germs was used as a target protein for the screening of aptamer. The aptamer that has binds to the G-protein of P. gingivalis, was screened and developed through the Systemic Evolution of Ligands by Exponential Energy (SELEX) method. Modified-Western blot analysis was performed with the aptamer which consisted of 38 single-stranded DNA to confirm the selectivity. ELONA (enzyme linked oligonucleotide assay) used to confirm that the aptamer was sensitive to PGP even at low concentration of 1 ㎍/ml. For the rapid detection of P. gingivalis, we constructed a surface plasmon resonance biosensor with SPREETA using the PGP aptamer. It was confirmed that PGP could be detected as low concentration as at 0.1 pM, which is the minimum concentration of aptamer sensor within 5 min. Based on these results, we have constructed a SPREETA biosensor based on aptamer that can bind to P. gingivalis G-protein. It can be used as an infection diagnosis system to rapidly diagnose and analyze oral diseases caused by P. gingivalis.
Purpose: An implant-supported prosthesis consists of an implant fixture, an abutment, an internal screw that connects the abutment to the implant fixture, and the upper prosthesis. Numerous studies have investigated the microorganisms present on the implant surface, surrounding tissues, and the subgingival microflora associated with peri-implantitis. However, there is limited information regarding the microbiome within the internal screw space. In this study, microbial samples were collected from the supragingival surfaces of natural teeth, the peri-implant sulcus, and the implant-abutment screw hole, in order to characterize the microbiome of the internal screw space in healthy subjects. Methods: Samples were obtained from the supragingival region of natural teeth, the peri-implant sulcus, and the implant screw hole in 20 healthy subjects. DNA was extracted, and the V3-V4 region of the 16S ribosomal RNA was sequenced for microbiome analysis. Alpha diversity, beta diversity, linear discriminant analysis effect size (LEfSe), and network analysis were employed to compare the characteristics of the microbiomes. Results: We observed significant differences in beta diversity among the samples. Upon analyzing the significant taxa using LEfSe, the microbial composition of the implant-abutment screw hole's microbiome was found to be similar to that of the other sampling sites' microbiomes. Moreover, the microbiome network analysis revealed a unique network complexity in samples obtained from the implant screw hole compared to those from the other sampling sites. Conclusions: The bacterial composition of the biofilm collected from the implant-abutment screw hole exhibited significant differences compared to the supra-structure of the implant. Therefore, long-term monitoring and management of not only the peri-implant tissue but also the implant screw are necessary.
Objectives: Cetylpyridinium chloride CPC-based mouthwashes are well known to have no harmful ingredients in the mouth and can be used for a long time. The purpose of this study was to evaluate the effect of using CPC-based mouthwashes to suppress the biofilm formation and antibiotics for handling orthodontic appliances. Methods: To measure the antibacterial effect, Streptococcus mutans (S. mutans) cultured orthodontic appliances were precipitated in Gargreen and Polident for 5 minutes, incubated at 37℃ for 24 hours(h). In order to measure the biofilm removal effect, the degree of biofilm formation on the orthodontic appliances was stained with a methylene blue and the difference before and after was compared using image J software program (NIH Image J; NIH, Bethesda, MD). Results: The viability of S. mutans according to the concentration showed that Gargreen and Polident inhibited colony formation compared to the control, respectively (p<0.01). The degree of biofilm formation was significantly higher in the control, however both Gargreen and Polident significantly reduced it compared to the before and after condition on removable orthodontic appliances (p<0.01). Conclusions: This study suggests that the use of Gargreen, a cetylpyridinium chloride based oral cleaning cleanser, could be replaced by Polident for antibacterial effect and biofilm formation on removable orthodontic appliances.
Background: Dental caries is one of several prevalent oral diseases caused by dental plaque biofilms. This study evaluated the anti-cariogenic effects of a bamboo salt (BS) and sodium fluoride (NaF) mixture on oral bacteria. Methods: The effects of several mixtures of NaF and BS on acid production, growth, and adhesion to glass beads of Streptococcus mutans, and their anti-cariogenic properties were investigated. The growth of S. mutans was measured according to optical density at 3, 6, 9, 12, 15, 18, and 24 hours after treatment using spectrophotometry at a wavelength of 600 nm, while pH was measured using a pH meter. Adhesion of S. mutans was measured according to the weight of glass beads from each group before and after incubation. Gene expression was measured using real-time polymerase chain reaction. Acid production and growth patterns of S. mutans were compared using repeated measures analysis of variance, followed by Scheffe's post-hoc test. The Kruskal-Wallis test was used to compare adhesion, followed by the Mann-Whitney test. Gene expression in the experimental and control samples was compared using the Student's t-test. Results: Growth, acid production, and adhesion of S. mutans were inhibited in all experimental groups. Expression of gft and fructosyltransferase in S. mutans was inhibited in all groups. A mixture of NaF and BS significantly reduced growth, acid production, adhesion, and gene expression of S. mutans compared with the other groups. Conclusion: Results of the present study demonstrated that a mixture of NaF and BS was useful as a mouth rinse in preventing dental caries.
Several medicinal plants are ethnomedically used in Korea as agents for treating infection, anti-inflammation, and pain relief. However, beyond typical inhibitory effects on cell growth, little is known about the potential anti-biofilm activity of these herbs, which may help to prevent cavities and maintain good oral health. This study aimed to investigate the antimicrobial and anti-biofilm activities of the methanol extracts of 37 Korean medicinal plants against dental pathogens Streptococcus mutans and Candida albicans, which synergize their virulence so as to induce the formation of plaque biofilms in the oral cavity. The antimicrobial activities were investigated by broth dilution and disk diffusion assay. The anti-biofilm and antioxidant activities were evaluated based on the inhibitory effect against glucosyltransferase (GTase) and the DPPH assay, respectively. Among 37 herbs, eight plant extracts presented growth and biofilm inhibitory activities against both etiologic bacteria. Among them, the methanol extracts (1.0 mg/ml) from Camellia japonica and Thuja orientalis significantly inhibited the growth of both bacteria by over 76% and over 83% in liquid media, respectively. Minimum inhibitory concentration (MIC) values of these methanol extracts were determined to be 0.5 mg/ml using a disk diffusion assay on solid agar media. Biofilm formation was inhibited by more than 92.4% and 98.0%, respectively, using the same concentration of each extract. The present results demonstrate that the medicinal plants C. japonica and T. orientalis are potentially useful as antimicrobial and anti-biofilm agents in preventing dental diseases.
DUWL에 형성된 바이오필름 제거를 위한 효과적인 소독제의 제시와 새로운 소독제의 개발을 위해 DUWL의 실험실 모델의 확립이 필요하다. 따라서 본 연구에서는 실험실에서 쉽게 구할 수 있는 장비들로 실험실 모델을 제작하여, DUWL 바이오필름을 재현하기 위한 새로운 실험실 모델을 확립하고자 하였다. 사용 중인 DUWL을 통해 수집한 물에서 세균을 모은 후, R2A 액체 배지에서 10일 동안 배양시켰다. 10일 배양시킨 세균액을 $-70^{\circ}C$에 보관하여 사용하였다. $-70^{\circ}C$에 저장한 세균 stock은 R2A 액체배지에 5일 동안 회분 배양시킨 배양액은 모델에서 바이오필름을 형성시키기 위해 사용되었다. 바이오필름 형성 모델은 실험실 내 장비인 1 L 비커에 폴리우레탄 튜빙이 부착된 20 cm 유리막대를 꽂아서 제작하였다. 모델을 멸균시킨 후 R2A 액체배지 300 ml와 5일 동안 회분 배양한 세균액 50 ml을 넣고 stir plate에서 $25^{\circ}C$로 배양시켰다. 배양 2일마다 R2A 액체배지를 교체해주었다. 임상의 상황과 유사한 조건에서 바이오필름을 형성하기 위해 와류상태는 오전 9시에서 오후 6시까지 적용시키고 그 이외의 시간에는(약 15시간) 정체상태로 배양시켰다. 바이오필름 형성은 4일 동안 진행하였으며, 그 후 바이오필름의 두께, 바이오필름을 구성하는 세균의 분포 및 형태학적 특징을 SEM과 CLSM을 사용하여 분석하였다. 4일 바이오필름 형성 후 평균 바이오필름 축적량은 $4.68{\times}10^4CFU/cm^2$였고, 바이오필름의 두께는 $10{\sim}14{\mu}m$였다. 또한 바이오필름을 구성하는 세균들이 부분적으로 응집되어 덩어리를 이루고 있는 양상을 확인할 수 있었다. 본 연구에서 제작한 실험실 모델을 대상으로 차아염소산나트륨, 과산화수소 그리고 클로르헥시딘과 같은 소독제의 효과를 확인하였다. 그 결과 적용된 소독제의 농도가 낮을수록 바이오필름 내 생존한 세균의 수가 많았다. 따라서 우리의 실험실 모델에서 형성시킨 바이오필름은 소독제의 효과를 비교하기 위해 적절한 것으로 판단된다. 우리의 실험실 모델은 향후 DUWL 소독을 위한 새로운 방법의 개발을 위해 유용하게 사용될 것으로 예상된다.
이 연구는 강릉원주대학교 치과대학 학생들의 임상실습을 위해 사용되고 있는 DCU에서 배출되는 물 속 종속영양세균의 수준을 평가하면서 사용빈도에 따른 세균 오염수준의 차이를 확인하고 기회 감염성 병원균의 존재를 분자생물학적 방법을 사용하여 확인하였다. 임상 실습실에서 사용되는 DCU 36개를 대상으로 초음파치석제거기에서 물 시료를 수집하여 평균 CFU/ml를 조사하고 초음파치석제거기의 한달 사용빈도에 따라 DCU를 세 집단으로 분류하여 세균오염수준을 비교하였다. 또한 수집한 물 시료에서 세균의 genomic DNA를 추출한 후 PCR 분석을 통해 기회감염성 병원균의 존재를 확인한 결과는 다음과 같다. 학생 실습에 사용한 DCU에서 수집한 물 시료의 평균 종속영양세균수준은 16,095 CFU/ml로 ADA에서 권장하는 200 CFU/ml 이하의 수준에 적합하지 않은 것을 확인하였다. 초음파 치석제거기의 한 달 사용빈도에 따라 3집단으로 나누어 CFU/ml를 조사하였을 때, 초음파치석제거기를 한 달에 1번 이상 3번 미만 사용한 DCU에서 평균 CFU/ml가 20,070 CFU/ml로 가장 높게 나타났으며, 3번 이상 사용한 유니트는 CFU/ml 평균이 8,420 CFU/ml로 가장 적게 나타났다. 3개군의 CFU/ml 차이는 통계학적으로 유의성이 있는 것을 보여주었고(p<0.05), 그 중 사용빈도가 가장 높은 군에서 유의하게 낮은 CFU/ml를 보여주었다. 치과에서 사용하는 DCU에 존재하는 기회감염성 병원균이 학생실습에 사용하는 DCU에서도 분리되었다. 36개의 genomic DNA 시료 중 1개의 시료에서 Pseudomonas species가 검출되었고, 2개의 시료에서 비결핵성 Mycobacterium species가 검출되었다. 따라서 학생실습용으로 사용되는 DCU는 학생들과 대상자에게 잠재적 감염의 원인이 될 수 있으며, 실습 전 학생들의 보호장비 착용과 실습 후 수관관리가 필요하다.
본 연구는 Silver diamine fluoride (SDF)와 요오드화 칼륨(KI)이 이차 우식에 노출된 상아질의 내산성에 미치는 영향을 실험실 환경에서 평가하는 것이다. 소의 절치를 이용하여 상아질 시편을 제작한 뒤 인공적으로 초기 우식병소를 형성하여 음성 대조군, 양성 대조군, SDF 도포, SDF 도포 후 KI 도포(SDFKI)의 4개 군으로 나누었다. 음성 대조군을 제외한 각 군 당 4개의 시편 상면에 Streptococcus mutans, Lactobacillus casei 및 Candida albicans를 포함하는 다종 우식원성 세균을 접종하고 28일 간 배양하였다. 음성 대조군은 초기 우식병소 형성 뒤 인산 완충 용액에 담가 28일 간 보관하였다. 모든 시편의 탈회 정도는 micro-CT를 사용하여 분석하여 SDF와 KI가 탈회 상아질의 내산성에 미치는 영향을 비교, 평가하였다. 실험 결과 SDF와 SDFKI군에서는 양성 대조군에 비해 유의하게 탈회 깊이가 감소하여 SDF와 SDFKI의 도포가 탈회 상아질의 내산성을 증가시켜 이차 우식에 대한 저항성을 향상시킨 것을 확인할 수 있었다. KI 도포는 SDF의 항균 효과에 대해 유의한 영향을 미치지 않았다. 본 연구 결과 SDF와 SDFKI 도포는 탈회 상아질의 내산성을 증가시켜 이차 우식의 예방에 효과가 있었다. 또한 KI는 SDF의 항균 효과에 유의한 영향을 미치지 않으면서 검은 변색의 가능성을 줄일 수 있기 때문에 임상에서 효과적으로 사용될 수 있을 것으로 생각된다.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
