A review: Synthetic strategy control of magnetite nanoparticles production

  • Iron oxide nanoparticles excite researcher interest in biomedical applications due to their low cost, biocompatibility and superparamagnetism properties. Magnetic iron oxide especially magnetite ($Fe_3O_4$) possessed a superparamagnetic behaviour at certain nanosize which beneficial for drug and gene delivery, diagnosis and imaging. The properties of nanoparticles mainly depend on their synthesis procedure. There has been a massive effort in developing the best synthetic strategies to yield appropriate physico-chemical properties namely co-precipitation, thermal decomposition, microemulsions, hydrothermal and sol-gel. In this review, it is discovered that magnetite nanoparticles are best yielded by co-precipitation method owing to their simplicity and large production. However, its magnetic saturation is within range of 70-80 emu/g which is lower than thermal decomposition and hydrothermal methods (80-90 emu/g) at 100 nm. Dimension wise, less than 100 nm is produced by co-precipitation method at $70^{\circ}C-80^{\circ}C$ while thermal decomposition and hydrothermal methods could produce less than 50 nm but at very high temperature ranging between $200^{\circ}C$ and $300^{\circ}C$. Thus, co-precipitation is the optimum method for pre-compliance magnetite nanoparticles preparation (e.g., 100 nm is fit enough for biomedical applications) since thermal decomposition and hydrothermal required more sophisticated facilities.

Optimization Studies for the Production of Microbial Transglutaminase from a Newly Isolated Strain of Streptomyces sp.

  • Covalent cross-links between a number of proteins and peptides explain why transglutaminase may be widely used by food processing industries. The objective of this work was optimization of the fermentation process to produce transglutaminase from a new microbial source, the Streptomyces sp. P20. The strategy adopted to modify the usual literature media was: (1) fractional factorial design (FFD) to elucidate the key medium ingredients, (2) central composite design (CCD) to optimise the concentration of the key components. Optimization of the medium resulted in not only an 86% increase in microbial transglutaminase activity as compared to the media cited in the literature, but also a reduction in the production cost. Optimal fermentation conditions - namely temperature and agitation rate - were also studied, using CCD methodology. Usual conditions of $30^{\circ}C$ and 100 rpm were within the optimal area. All other parameters for enzyme production were experimentally proven to be optimum fermentation conditions.

Problem and Optimum Operational Strategy of Multipurpose Reservoir in Korea (우리나라 다목적 Dam 운영의 문제점과 개선방안)

  • The number of visiters to Bukhan Mt. national park, generation quantity of solid waste and collection system were researched to consider a counterplan for the pollution control of the national park and study for developing the effective treatment of solid waste was tried through the proximate analysis of each component containing. Results obtained in this study were summerized as follows; The great part of visitors go on an excursion to the Bukhan Mt. national park during July and August and also, the solid waste was generated nearly a half of the total amount at the same period. The major collection facilities in the national park were waste basket and incineration box. But the incineration box was too large in volume and very far in distance, and its collection period was irregular, so it was cause to the congestion of solid waste and bad smell and dirty. Therefore, to complete collection of solid waste, we must set up the waste basket which able to find within 40~50m from the origination place of solid waste and induce the visitors to throw the solid waste. It was obtained as moisture content: 48.5 wt%, volatile solid: 28.4wt%, fixed solid: 23.1 wt%, lower heating value: 1,320kca1/kg from experimental analysis of solid waste. According to this analysis, the incineration operation is possible, but the generation quantity of solid waste was too small to construct incineration plant for heat recovery. It was found that it is suitable for the aerobic composting by mixing with the night soil which generate in the national park after the recovery of resources such as metals, glasses and plastics.

  • Recently, a variable valve timing system has been widely adopted in internal combustion engine in order to improve the fuel economy and torque at low engine speed. In addition, it is known that varying valve timing according to the various engine operations could reduce exhaust gas, especially NOx, because of residual gas by valve overlap. In this study, to improve the low exhaust gas and fuel economy at part load condition, the residual gas and back flow of exhaust gas due to valve overlap were calculated computationally. Moreover, the characteristics of engine performances and NOx formations were investigated with the experiment of combination of intake and exhaust valve timing condition. Under these various valve operating conditions, the effects of both the positive valve overlap and negative valve overlap(valve underlap) were examined simultaneously. Finally, the characteristics of cyclic THC emission were analyzed by using Fast Response FID(FR-FID) in the cylinder, intake port and exhaust port positions. Besides, the effect of the different gradients of the valve timing change on engine performance was investigated and an optimum control strategy was suggested.

Desalting of tobacco extract using electrodialysis

  • Papermaking reconstituted tobacco is an important strategy for recycling the waste tobacco residues. To indentify the influences of the inorganic components on harmful components delivery in cigarette smoke, a self-made electrodialysis stack was assembled to desalt the tobacco extract. The influences of the applied current and extract content on the removal rate of the inorganic ions were investigated. Results indicated that the applied current was a dominant impact on the desalination performance. High currents lower than the limiting current density could accelerate the desalting efficiency but cause higher energy consumption. A current of 2 A, or current density of ${\sim}11mA{\cdot}cm^{-2}$, was an optimal choice by considering both the energy consumption and desalting efficiency. A 20% tobacco extract was an appropriate content for the electrodialysis process. More than 90% of inorganic ions could be removed under the optimum condition. The preliminary result indicated that removal of inorganic components was beneficial to decrease the harmful component delivery in cigarette smoke. Naturally, ED is an environmentally friendly and high-effective technology for desalting the tobacco extract.

Design and Expression of Recombinant Antihypertensive Peptide Multimer Gene in Escherichia coli BL21

  • The design and expression of an antihypertensive peptide multimer (AHPM), a common precursor of 11 kinds of antihypertensive peptides (AHPs) tandemly linked up according to the restriction sites of gastrointestinal proteases, were explored. The DNA fragment encoding the AHPM was chemically synthesized and cloned into expression vector pGEX-3X. After an optimum induction with IPTG, the recombinant AHPM fused with glutathione S-transferase (GST-AHPM) was expressed mostly as inclusion body in Escherichia coli BL21 and reached the maximal production, 35% of total intracellular protein. The inclusion body was washed, dissolved, and purified by cation-exchange chromatography under denaturing conditions, followed by refolding together with size-exclusion chromatography and gradual dialysis. The resulting yield of the soluble GSTAHPM (34 kDa) with a purity of 95% reached 399 mg/l culture. The release of high active fragments from the AHPM was confirmed by the simulated gastrointestinal digestion. The results suggest that the design strategy and production method of the AHPM will be useful to obtain a large quantity of recombinant AHPs at a low cost.

  • We have developed a program for generating shotgun data sets from known genome sequences. Generation of synthetic data sets by computer program is a useful alternative to real data to which students and researchers have limited access. Uniformly-distributed-sampling clones that were adopted by previous programs cannot account for the real situation where sampled reads tend to come from particular regions of the target genome. To reflect such situation, a probabilistic model for biased sampling distribution was developed by using an experimental data set derived from a microbial genome project. Among the experimental parameters tested (varied fragment or read lengths, chimerism, and sequencing error), the extent of sequencing error was the most critical factor that hampered sequence assembly. We propose that an optimum sequencing strategy employing different insert lengths and redundancy can be established by performing a variety of simulations.

An Approach for Lactulose Production Using the CotX-Mediated Spore-Displayed β-Galactosidase as a Biocatalyst

  • Currently, enzymatic synthesis of lactulose, a synthetic prebiotic disaccharide, is commonly performed with glycosyl hydrolases. In this work, a new type of lactulose-producing biocatalyst was developed by displaying β-galactosidase from Bacillus stearothermophilus IAM11001 (Bs-β-Gal) on the surface of Bacillus subtilis 168 spores. Localization of β-Gal on the spore surface as a fusion to CotX was verified by western blot analysis, immunofluorescence microscopy, and flow cytometry. The optimum pH and temperature for the resulting spore-displayed β-Gal was 6.0 and 75℃, respectively. Under optimal conditions, it showed maximum activity of 0.42 U/mg spores (dry weight). Moreover, the spore-displayed CotX-β-Gal was employed as a whole cell biocatalyst to produce lactulose, yielding 8.8 g/l from 200 g/l lactose and 100 g/l fructose. Reusability tests showed that the spore-displayed CotX-β-Gal retained around 30.3% of its initial activity after eight successive conversion cycles. These results suggest that the CotX-mediated spore-displayed β-Gal may provide a promising strategy for lactulose production.

  • This paper presents a novel three-phase current-fed Pulse Width Modulation converter with switched capacitor type resonant DC link commutation circuit operating PWM pattern strategy under a design consideration of low-pass filter, which can operate on the basis of the principle of zero current soft switching commutation. In the first place, the steady state operating principle of this converter with a new resonant DC link snubber circuit is described in connection with the equivalent operation circuit, together with the practical design procedure of the switched-capacitor type resonant DC link circuit is discussed from a theoretical viewpoint on the basis of a design example for high-power applications. The actively delayed time correction method to compensate distorted currents due to a relatively long resonant commutation time is newly implemented in the open loop control scheme so as to acquire the new optimum PWM pattern. Finally, the experiment of set-up in laboratory system of this converter is concretely demonstrated herein to confirm a zero current soft-switching commutation of this converter. The comparative evaluations between current-fed hard switching PWM and soft-switching PWM converters are carried out from a viewpoint of their PWM converter characteristics.

The Seed Germination Response of Rumex acetocella by Distribution Area (분포지역에 따른 애기수영(Rumex acetocella) 종자의 발아반응)

  • To study the relationship between the germination response and geographical distribution, the seeds of Rumex acetocella were collected from nine sites (Taegwallyong, Alps lodge, Kans'ong, Sokch'o, Chumunjin, Suwon, Taechon, Sukgok, Mokpo) of different latitudes and altitudes. The populations of Rumex acetocella were subdivided into four groups based on the characteristics of the seed germination. The first group of Rumex acetocella (Taegwallyong, Alps lodge) germinated rapidly in early spring, the second group (Suwon, Teachon) in late summer through early autumn, and the third group(Sukgok, Mokpo) slowly in late autuman with various conditions. The thermal time values needed for the germination rate of 10% to 60% for these three groups indicated that the germination rate was low at the lower latitude, while it was increased as the latitude elevated. The population of the fouth group (Kansong, Sokcho, Chumunjin) showed the narrow optimum temperature range of germination response and the highest thermal time value to germinate 10% to 30% of seeds. The seeds in the high latitude region with relatively mild winter germinated synchronously in early autumn. These different germination responses of Rumex acetocella to geographic variations were thought to be an ecological strategy to survive in different environmental conditions.
