Biogenic Mn oxides are expected to have great potential in the control of water pollution due to their high catalytic activity, although information on biological Mn oxidation is not currently sufficient. In this study, the growth of a Mn oxidizing microorganism, Pseudomonas putida MnB1, was examined, with the Mn oxides formed by this strain characterized. The growth of P. putida MnB1 was not significantly influenced by Mn(II), but showed a slightly decreased growth rate in the presence of Pb(II) and EE2, indicating their insignificant adsorption onto the cell surface. Mn oxides were formed by P. putida MnB1, but the liquid growth medium and resulting biogenic solids were poorly crystalline, nano-sized particles. Biogenic Mn oxidation by P. putida MnB1 followed Michaelis-Menten kinetics, with stoichiometric amounts of Mn oxides formed, which corresponded with the initial Mn(II) concentration. However, the formation of Mn oxides was inhibited at high initial Mn(II) concentration, suggesting mass transfer obstruction of Mn(II) due to the accumulation of Mn oxides on the extracellular layer. Mn oxidation by P. putida MnB1 was very sensitive to pH and temperature, showing sharp decreases in the Mn oxidation rates outside of the optimum ranges, i.e. pH 7.43-8.22 and around 20-$26^{\circ}C$.
억새즙액을 이용하여 유용한 미생물의 대량배양을 위한 새로운 배지를 개발하고자 본 연구를 수행하였다. 억새즙액의 성분을 조사한 결과 칼륨 (37,952 mg/L), 질소 (14,000 mg/L), 인산 (6,800mg/L), 마그네슘 (5,969 mg/L), 칼슘 (5,910 mg/L) 등이 주성분을 차지하였다. 본 연구를 위하여 먼저 식물병원성균인 뿌리썩음병 원인균인 Rhizoctonia solani, 잿빛곰팡이 병을 일으키는 Botrytis cinerea에 대하여 강력한 저해 활성을 갖는 길항균 G-74를 토양으로부터 분리하였으며, 이 균의 형태관찰과 MIDI 분석을 통하여 Bacillus lentimorbus G-74 균주로 최종 동정하였다. 이 균의 억새즙액 배지에 대한 최적 배양조건을 조사한 결과 30% (V/V) 희석배지, $35^{\circ}C$ 배양온도, 5.0의 초기 배지 pH에서 가장 좋은 성장을 기록하였다. 또한 이 배지에 탄소원으로서 2.0% (W/V)의 corn starch 및 질소 원으로서 1.0% (W/V)의 yeast extract를 추가적으로 첨가하면 최상의 성장을 보여주었으며, 이 경우 대조구인 LB 배지보다도 균체성장이 66% 이상 증가하였다.
속성수로서 현재까지 이용연이 그리 넓지 못한 활엽수(闊葉樹)를 이용(利用)한 당화액(糖化液)으로 단세포단백질(單細胞蛋白質) 생산(生産)에 의(依)한 사료화(飼料化) 전환(轉換)에 관(關)한 기초적(基礎的)인 몇가지 실험(實驗)을 한 결과(結果)는 아래와 같다. Aspergillus niger sm-10, Irpex lacteus, Tricoderma viride sm-6 3균주(菌株)의 셀룰라아제 역가(力價)를 비교(比較)해 본 결과(結果) Tricoderma viride sm6 의 역가(力價)가 가장 높았으며, 이들을 combination하였을 때에는 상조에 의한 역가상승(力價上昇)(Synergistic effect)은 보이지 않았다. T. viride sm-6의 배양학적(培養學的)인 면을 살펴본 결과(結果) 밀기울배지에 질소원으로서 proteose peptone, 유도제 (inducer)로서 Tween 80, cellulose를 첨가(添加)했을때 역가증가(力價增加) 현상(現象)이 나타났으며 이와 같은 배지에서 배양시(培養時) C.M. Cellulose와 Filter paper 분해 효소역가(酵素力價)는 120hr에 최고에 도달(到澾)했다. 기질(基質)에 효소(酵素)를 반응(反應)시켜 당화액(糖化液)을 생성(生成)하는데 있어서 전처리(前處理)를 한 기질(基質)은 대조주에 비해 약 40%의 환원당 증가(增加)를 보였고, 또한 기질입자(基質粒子)가 작을수록 환원당 증가현상(增加現象)을 보였으며 글루코스 등의 당(糖)은 반응액중(反應液中) 적은 농도(濃度)에서도 커다란 feed back inhibition현상을 나타냈다. 기질(基質)을 rehydralysis시에는 약 31%의 환원당 전환율(轉換率)을 보였다. 이때 당화액(糖化液)을 정량해 본 결과(結果) glucose : xylose의 양이 1.77:1로서 glucose의 양이 많았다. 여기에 단세포(單細胞) 단백질(蛋白質)을 생산(生産)할 목적(目的)으로 부식토, 퇴비, 수액, 토양등의 42점의 시료로부터 xylose 자화성 효모(酵母) 13균주(菌株)를 선별하였으며 그때의 우수균주(優秀菌株)로서 3개균주(個菌株)를 선발(選拔)하였다. 이들 분리주의 배양학적(培養學的) 특징(特徵)을 살펴 본 결과(結果)는 다음과 같다. 1. 분리주중(分離株中) CHS-2, CHS-3, ST-40, CHS -12, CHS-13 균주(菌株)가 당화액(糖化液)을 잘 자화함을 알았다. 2. 분리균주중(分離菌株中) CHS-13 균주(菌株)의 생육도(生育度)가 가장 양호하였으며 초기 최적 pH는 4.4이었으며 최적온도(最適溫度)는 $30^{\circ}C$이었다. 3. CHS-3 균주(菌株)의 specific growth rate는 $0.23\;h^{-1}$, generation time 은 3.01h이었다. 4. CHS -13균주(菌株)의 기질소비율(基質消費率)은 81%이었다. 5. CHS-13 균주(菌株)의 형태학적(形態學的) 배양학적(培養學的) 특성(特性)을 조사(調査)한 결과(結果) Candida guilliermondii var. guilliermondii 로 동정(同定)되었다. 6. CHS-13 균주(菌株)를 당화액(糖化液)에 배양(培養)시켰을 때 Lowry-protein 함량은 $0.72\;mg/m{\ell}$ 이었으며 , yeast extract 첨가(添加)하여 배양시(培養時)는 yeast extract 농도(濃度)가 증가(增加)함에 따라 단백질(蛋白質) 함량(含量)도 증가(增加)하였다. 7. CHS-13 균주(菌株)의 RNA 함량(含量)은 $4.92{\times}10^{-2 }\;mg/m{\ell}$이었으며 yeast extract 농도(濃度)가 증가(增加)함에 따라 증가(增加)하다가 농도(濃度) 0.2%에서 최대함량(最大含量)을 나타내고 그후는 감소(減少)하였다.
무염조건에서는 생육할 수 없고 2 M의 NaCl 존재하에서 가장 잘 생육하는 중도 호염성균이며 alkaline protease를 생산하는 ES 10균주를 멸치젓에서 분리하여 Halomonas속 균으로 동정하였다. 이 균은 합성배지인 TSM배지에 DL-alanine의 첨가로 생육이 촉진되고 L-proline의 첨가로 생육이 저해되었다. 이 균의 세포내 $Na^+$함량은 Bacillus subtilis나 E. coli보다 5배 정도 많았으며 $K^+$함량은 25배, $Mg^{2+}$함량은 38배 정도 많았다. 이 균의 Protease 생산은 NaCl 1 M첨가된 Norberg와 Hofsten배지에서 $20^{\circ}C$로 배양했을 때 가장 양호하였다.
Recently, it is on the increase interest for Al alloy with new material for ship application to substitute for FRP ship. The reason is thatAl alloy ship has beneficial characteristics such as high sea speed, increase of loadage and easy to recycle compared with FRP ship. In this paper, mechanical and electrochemical properties are investigated by slow strain rate test experiment in various applied potential condition. These results will provide as reference data to design ship by deciding optimum protection potential regard to hydrogen embrittlement and stress corrosion cracking. In general, Al and Al alloys are not corroded with forming film which has the corrosion resistance property in neutral solution. However, it was observed that formation and destruction of passive film by $Cl^-$ ion in sea water environment. At comparison of current density after 1200 sec in potentiostatic experiment, the current density in the potential range of -0.68 $\~$-1.5 V is shown low value. The low current density means protection potential range. Elongation in applied potential of 0 V was high. However, the corrosion protection application in this condition is impossible potential because the toughness is low value by decreasing strength by active dissolution reaction at parallel part of specimen. The film composed with $CaCO_3$ and $Mg(OH)_2$ has a corrosion resistance property. However, the uniform electrodeposition coating at below -1.6 V potential is not formed since the time to form the uniform electrodeposition coating is short. Therefore, it is concluded that mechanical property is poor because effect by hydrogen gas generation is larger than that of electrodeposition coating. It is concluded that the optimum protection potential range from comparison of_maxim urn tensile strength, elongation and time to fracture is -1.3$\~$0.7 V (SSCE).
Covalent cross-links between a number of proteins and peptides explain why transglutaminase may be widely used by food processing industries. The objective of this work was optimization of the fermentation process to produce transglutaminase from a new microbial source, the Streptomyces sp. P20. The strategy adopted to modify the usual literature media was: (1) fractional factorial design (FFD) to elucidate the key medium ingredients, (2) central composite design (CCD) to optimise the concentration of the key components. Optimization of the medium resulted in not only an 86% increase in microbial transglutaminase activity as compared to the media cited in the literature, but also a reduction in the production cost. Optimal fermentation conditions - namely temperature and agitation rate - were also studied, using CCD methodology. Usual conditions of $30^{\circ}C$ and 100 rpm were within the optimal area. All other parameters for enzyme production were experimentally proven to be optimum fermentation conditions.
In order to produce vinegar using onions, 12 acetic acid bacteria were screened from the juice of fallen peaches, and a strain showing the highest acetic acid productivity among them was selected and identified as Acetobacter tropicalis No. 22. The culture broth containing 2.5%(w/v) of initial sugar concentration showed maximum acetic production after 10 days of cultivation, and the acetic acid was produced at the highest rate and reached the maximum acidity after 2 to 6 days of cultivation when the residual sugar and the ethanol concentration were in the range of 1.6 to 2%(w/v) and 0.6 to 1.8%(v/v), respectively. Also optimum conditions for acetic acid production by response surface method using the fractional factorial design with 3 variables and 5 levels were involved with initial ethanol content of 4.67%(v/v), initial acidity of 0.03%, and initial glucose concentration of 2.35%(w/v) and predicted level of acetic acid production at these conditions was 3.77%.
Hot deformation behavior of a carbon steel (A350 LF2) was characterized by compression tests in the temperature range of $800-1250^{\circ}C$ and the strain rate range of $0.001-10s^{-1}$, The microstructural evolution during hot compression was investigated and deformation mechanisms were analyzed by constructing a deformation processing map. Processing maps were generated using the combination of dynamic material model (DMM) and flow instability theories based on the flow stability criteria and Ziegler's instability criterion. In order to evaluate the reliability of the map, the mirostructural characteristics of the hot compressed specimens were correlated with test conditions in the stable and unstable regime. The combined microstructural and processing map of A350 LF2 was applied to predict an optimum condition and unstable regions for hot forming.
Five microalgal species isolated from the Jeju coast and four microalgal stock strains in hatchery were cultured in order to investigate their adapation to extreme changes in environmental factors such as salinity, water temperatue, adn nutrients. In case of salinity variation, Nitzschia sp. of Bacillariophyceae, Isochrysis galbana of Haptophyceae and Tetraselmis gracilis of Prasinophyceae showed optimum growth at the low salinity of 20 and 25 psu. Amphora coffeaeformis and Chetoceros simplex of Bacillariophyceae, and Pavlova lutheri of Haptophyceae adapted well at the relatively high salinities of 30 and 35 psu. However Phaeodactylum tricornutum of Bacillariophyceae and Chlorella sp. of Chlorophyceae showed euryhaline property In case of water temperature variation, most of all the species studied wer inhibited at 10℃. C. simplex, Nitzschia sp., p. tricornutum, Chlorella sp. and T. gracilis grew well at above 20℃. A. coffeaeformis, I. galbana and P. lutheri adapted also at the high temperature of 30℃. Each microalgal strain showed different growth rates and its maximum biomass. Generally microalgal populations from the Jeju coast grow well in relatively high salinity and high water temperature. Their growth were inhibited at low water temperature, but not likely affected at low salinity. This study indicates that the microalgal populations could not be affected by abnormally low salinity phenomena, which have happened occasionally around the west Jeju coast in summer and have led macrobenthic animals to mass mortality.
As a component for a recirculating aquaculture system, a new strain of denitrifying bacterium was isolated from municipal sewage. The isolate was motile by means of one polar flagellum, catalase-positive, and a Gram-negative rod-shaped cell measuring $0.5-0.6{\mu}m$ in width and $1.3-1.9{\mu}m$ in length. The isolate was identified as Pseudomonas fluorescens and produced dinitrogen gas via the reduction of nitrate. The optimal growth conditions (pH, temperature, carbon source, and C/N ratio) of the isolate were found to be 6.8, $30^{\circ}C$, malate, and 3, respectively. Under optimal growth conditions of P. fluorescens, dinitrogen gas was first detected in the exponential growth phase, then a small amount of nitrite was developed and converted to dinitrogen gas in the stationary phase. Pseudomonas fluorescens cells were immobilized in modified polyvinyl alcohol (PVA) gel beads, and the maximum denitrification rate was measured as $36.6 {\mu}lN_2h^-1$ per bead with an optimum cell loading of $20mg {\mu}l^-1$ and $2\%$ sodium alginate added to the PVA gel. The operating stability of the modified PVA gel beads remained unchanged for up to 43 repeated batches.
