Optimization of Ethanol Extraction Conditions from Glasswort (Salicornia herbacea) Using Response Surface Methodology (반응표면분석법을 이용한 퉁퉁마디 에탄올 추출조건의 최적화)

  • Response surface methodology (RSM) was used to monitor the characteristics of ethanol extracts from glasswort (Salicornia herbacea). A central composite design was used to investigate the effects of the independent variables of sample ratio, extraction temperature, and ethanol concentration on the dependent variables color, sugar, salinity, yield, electron donating ability, and total polyphenol content of extracts. The maximum $^{\circ}Brix$ (8.46) was obtained under specific extraction conditions, with a sample ratio of 7.04 g/100 mL, an extraction temperature of $89.01^{\circ}C$, and an ethanol concentration of 34.29% v/v. At a sample ratio, extraction temperature, and ethanol concentration of 7.00 g/100 mL, $89.15^{\circ}C$, and 34.14% v/v, respectively, the salinity was 7.35%. When the sample ratio, extraction temperature, and ethanol concentration were 5.56 g/100 mL, $68.61^{\circ}C$, and 99.14% v/v, respectively, the maximum electron donating ability was 86.10%. A maximized total polyphenol content of 1,140.15 mg/100 g was found with the following conditions: sample ratio of 8.6 g/100 mL, extraction temperature of $64.19^{\circ}C$, and ethanol concentration of 71.74% v/v. Overall, the optimal ranges of extraction conditions for effective components of glasswort were 3.38.5.33 g/100 mL sample ratio, $55.87-76.96^{\circ}C$, and 25.00.67.31% v/v ethanol.

Antioxidant and Neuroprotective Effects of Green Tea Seed Shell Ethanol Extracts (녹차씨껍질 에탄올 추출물의 항산화 활성 및 신경세포 보호 효과)

  • The objective of this study was to evaluate the antioxidant activity of green tea seed shell as an industrial byproduct. Green tea seed shell extract (GTSSE) was obtained by ethanol extraction, and the yield was $1.4{\pm}0.22%$. The radical scavenging activities [1,1-diphenyl-picrylhydrazyl and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)], xanthine oxidase inhibition activity, and reducing power of GTSSE dose-dependently increased. To estimate the neuroprotective effect of GTSSE, viability was tested in HT22 mouse hippocampal cells. GTSSE treatment induced cytotoxicity at a concentration higher than $100{\mu}g/mL$ but not at a concentration lower than $50{\mu}g/mL$. Using this optimal concentration range, GTSSE treatment significantly increased cell viability in $H_2O_2$-treated HT22 cells. Further, GTSSE treatment increased superoxide dismutase activity and decreased the malonaldehyde level, a product of lipid peroxidation, in HT22 cells. Therefore, these results indicate that green tea seed shell extract may be useful for the development of antioxidant materials and have potential activity to prevent and treat neuro-degenerative diseases such as Alzheimer's disease.

Effects of Crop Loads on Vine Growth and Fruit Quality of 'Jinok' Grape in Unheated Plastic House (포도 '진옥' 품종의 무가온 하우스 재배시 착과량이 수체생육 및 과실품질에 미치는 영향)

  • This study was conducted to investigate optimal crop loads of 'Jinok' grape for unheated plastic house culture. The crop loads of 'Jinok' grapes were managed to be 1.3, 1.8, 2.2, 2.4, and 2.6t per 10a from 2012 to 2014. We measured vine growth and berry qualities. Crop loads were not significantly affected on plant height, trunk diameter, shoot length, and the internode length of 'Jinok'. However, the maturation of berries was delayed when the crop load was higher. And the harvest date was earlier about three weeks in an unheated plastic house compared to in an open field. The average berry weight was decreased by the higher crop load although higher crop loads made higher yields showing the lowest weight at 342g with the treatment of 2.6t per 10a and the highest weight at 363g with the treatment of 1.3t per 10a. Also, the soluble solids content showed a tendency that higher crop loads brought to lower degree Brix. The contents of P, K, Ca, and Mg in grape leaves and shoots were not significantly different by crop loads. To sum up, when crop loads were under the 2.4t per 10a, the berries were harvested as a marketable fruit having $15^{\circ}Brix$ in the cultivar 'Jinok' grape. This result could help to increase grower's benefit having improved quality of fruit for the sustainable production by the established cultivation techniques for the newly developed cultivar 'Jinok'.

Effects of LED(Light Emitting Diode) Photoperiod and Light Intensity on Growth and Yield of Taraxacum coreanum Nakai in a Plant Factory (식물공장 내 광주기 및 광도가 흰민들레의 생육과 수량에 미치는 영향)

  • The objective of this study was to examine the effect of photoperiod and light intensity of RBW LED (red:blue:white = 2:1:1) on the growth of Taraxacum coreanum Nakai in a fully artificial light type plant factory. 3 photoperiods and 4 light intensity were used respectively in a fully artificial light type plant production system. Plants were cultured with three photoperiods and four light intensity regimes (conditions) for 270 and 120 days, respectively, using nutrient film technique (NFT) or aeroponics culture methods. For each photoperiod, the total leaves per plant harvested 8 times in all cultivation period was 224 in the 16/8(day/light) photoperiod that had no significant difference from 220 in the 12/12 photoperiod and the lowest number of leaves was 151 occurred in the 8/16 photoperiod, which means that the longer photoperiod, the more leaves harvest. Total fresh weight of above ground was the high in order of in 16/8 photoperiod as 125g, 12/12 photoperiod as 91g, 8/16 photoperiod as 56g. For each light intensity, the total leaves per plant harvested 4 times in all cultivation period was the great in order of $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 123, $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 107, $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 95, $50{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 56 which was the smallest number of total leaves harvest. Total fresh weight of above ground per plant was the high in order of $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 43.6g, $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 34.6g, $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 32.2g, $50{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 18.2g. From these results, it was concluded that photoperiod of 16/8 and light intensity at $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ can be used as the light condition of RBW LED (red:blue:white = 2:1:1) for optimal growth of Taraxacum coreanum Nakai in a fully artificial light type plant factory.

Development of medium for Pleurotus eryngii cultivation using Kojongsi persimmon peels (감과피를 이용한 큰느타리버섯 배지 개발)

  • This study was carried out to investigated the avaiability of dried Koojongsi persimmon peels (KPP) as a useful mushroom medium using Pleurotus eryngii ASI 2312. Mushroom cultivation medium used in this study was mixed with medium mixture, corn cob and sawdust (220:65:15, v/v). Dried KPP was replaced mushroom cultivation medium (control) with 5, 10, 15, 20, 30, 40, 50% dried KPP. The T-N content of dried KPP treatments decreased to increase replaced ratio of the dried KPP and C/N ratio was increased to increase replaced ratio of the dried KPP. But T-C content of dried KPP treatments was similar to untreated control. The average cultivating periods of mycelium on dried KPP treatments was delayed to increase replaced ratio of the dried KPP and cultivating periods was delayed over 30% dried KPP treatments. The length of stipe of dried KPP treatments was longer than that of the untreated control to increase replaced ratio of the dried KPP and thickness of stipes was tend to be thinner than that of the untreated control to increase replaced ratio of the dried KPP. The moisture, carbohydrate, crude protein and crude ash content of mycelial were similar to untreated control, but crude fatty acid was increased to increase replaced ratio of the dried KPP. The ${\beta}-glucan$ content of 10% and 15% treatments were higher than untreatment control. The results based on cultivation yield and ${\beta}-glucan$ content indicated that optimal mixture ratio dried KPP was 15%.

Simultaneous Treatment of Carbon Dioxide and Ammonia by Microalgal Culture (조류배양을 통한 이산화탄소 및 암모니아의 동시처리)

  • A green microalga, Chlorella vulgaris UTX 259, was cultivated in a bench-scale raceway pond. During the culture, 15%(v/v) $CO_2$ was supplied and industrial wastewater discharged from a steel-making plant was used as a culture medium. In a small scale culture bottle, the microalga grew up to 1.8 g $dm^{-3}$ of cell concentration and ammonia was completely removed from the wastewater with an yield coefficient of 25.7 g dry cell weight $g^{-1}\;NH_3-N$. During the bottle-culture, microalga was dominant over heterotrophic microorganisms in the culture medium. Therefore, the amount of carbon dioxide fixation could be estimated from the change of dry cell weight. In a semi-continuous operation of raceway pond with intermittent lighting (12 h light and 12 h dark), increase of dilution rate resulted in increase of the ammonia removal rate as well as the $CO_2$ fixation rate but the ammonia removal efficiency decreased. Ammonia was not completely removed from the medium (wastewater) of raceway pond which was operated in a batch mode under a light intensity up to 20 klux. The incomplete removal of ammonia was believed due to insufficient light supply. A mathematical model, capable of predicting experimental data, was developed in order to simulate the performance of the raceway pond under the light intensity of sun during a bright daytime. Simulation results showed that the rates of $CO_2$ fixation and ammonia removal could be enhanced by increasing light intensity. According to the simulation, 80 mg $dm^{-3}$ of ammonia in the medium could be completely removed if the light intensity was over 60 klux with a continuous lighting. Under the optimal operating condition determined by the simulation, the rates of carbon dioxide fixation and ammonia removal in the outdoor operation of raceway pond were estimated as high as $24.7 g m^{-2} day^{-1}$ and $0.52 g NH_3-N m^{-2} day^{-1}$, respectively.

Growth Performance, Carcass Characteristics and Plasma Mineral Chemistry as Affected by Dietary Chloride and Chloride Salts Fed to Broiler Chickens Reared under Phase Feeding System

  • Requirements of dietary chloride (dCl) and chloride salts were determined by using $4{\times}2$ factorial arrangement under four phase feeding program. Four levels (0.31, 0.45, 0.59 and 0.73%) and two sources ($NH_4Cl$ and $CaCl_2$) of the dCl were allocated to 1,472 chicks in eight dietary treatments in which each treatment was replicated four times with 46 birds per replicate. The four phase feeding program was comprised of four dietary phases: Prestarter (d 1 to 10), Starter (d 11 to 20), Grower (d 21 to 33) and Finisher (d 34 to 42); and diets were separately prepared for each phase. The cations, anions, pH, dissolved oxygen (DO), temperature, electrical conductivity (EC), total dissolved solids (TDS) and salinity were analyzed in drinking water and were not affected by dietary treatments. BW gain (BWG; $p{\leq}0.009$) and feed:gain (FG; $p{\leq}0.03$) were improved in $CaCl_2$ supplemented diets during d 1 to 10. The maximum response of BWG and FG was observed at 0.38% and 0.42% dCl, respectively, for d 34 to 42. However, the level of dCl for BWG during d 21 to 33 ($p{\leq}0.04$) and d 34 to 42 ($p{\leq}0.009$) was optimized at 0.60% and 0.42%, respectively. The level of dCl for optimized feed intake (FI; $p{\leq}0.006$), FG ($p{\leq}0.007$) and litter moisture (LM; $p{\leq}0.001$) was observed at 0.60%, 0.38% and 0.73%, respectively, for d 1 to 42. Water intake (DWI) was not affected by increasing dCl supplementation (p>0.05); however, the ratio between DWI and FI (DWI:FI) was found highest at 0.73% dCl during d 1 to 10 ($p{\leq}0.05$) and d 21 to 33 ($p{\leq}0.009$). Except for d 34 to 42 ($p{\leq}0.006$), the increasing level of dCl did not result in a significant difference in mortality during any phase. Blood pH and glucose, and breast and thigh weights (percentage of dressed weight) were improved while dressing percentage (DP) and gastrointestinal health were exacerbated with $NH_4Cl$ as compared to $CaCl_2$ supplemented diets ($p{\leq}0.001$). Higher plasma $Na^+$ and $HCO_3{^-}$ and lower $Cl^-$ and $Ca^{{+}{+}}$ were observed in $NH_4Cl$ supplemented diets ($p{\leq}0.001$). Increasing supplementation of dCl increased plasma $Cl^-$ ($p{\leq}0.04$; quadratically) and linearly reduced plasma $K^+$ ($p{\leq}0.001$), $Ca^{{+}{+}}$ ($p{\leq}0.003$), $HCO_3{^-}$ ($p{\leq}0.001$), and $Na^+$ ($p{\leq}0.001$; quadratically). Consequently, higher requirements of dietary chloride are suggested for feed intake; nevertheless, lower levels of dietary chloride are sufficient to support optimal BWG and FG with increasing age. The $NH_4Cl$ supplemented diets ameliorate breast and thigh meat yield along with overall energy balance (glucose).

Analysis of growth environment for precision cultivation management of the oyster mushroom 'Suhan' (병재배 느타리버섯 '수한'의 정밀재배관리를 위한 생육환경 분석)

  • In this study, we analyze the growth environment using smart farm technology in order to develop the optimal growth model for the precision cultivation of the bottle-grown oyster mushroom 'Suhan'. Experimental farmers used $88m^2$ of bed area, 2 rows and 5 columns of shelf shape, 5 hp refrigerator, 100T of sandwich panel for insulation, 2 ultrasonic humidifiers, 12 kW of heating, and 5,000 bottles for cultivation. Data on parameters such as temperature, humidity, carbon dioxide concentration, and illumination, which directly affect mushroom growth, were collected from the environmental sensor part installed at the oyster mushroom cultivator and analyzed. It was found that the initial temperature at the time of granulation was $22^{\circ}C$ after the scraping, and the mushroom was produced and maintained at about $25^{\circ}C$ until the bottle was flipped. On fruiting body formation, mushrooms were harvested while maintaining the temperature between $13^{\circ}C$ and $15^{\circ}C$. Humidity was approximately 100% throughout the growth stage. Carbon dioxide concentration gradually increased until 3 days after the beginning of cultivation, and then increased rapidly to approximately 2,600 ppm. From the 6th day, $CO_2$ concentration was gradually decreased through ventilation and maintained at 1,000 ppm during the harvest. Light was not provided at the initial stage of oyster mushroom cultivation. On the $3^{rd}$ and $4^{th}$ day, mushrooms were irradiated by 17 lux light. Subsequently, the light intensity was increased to 115-120 lux as the growth progressed. Fruiting body characteristics of 'Suhan' cultivated in a farmhouse were as follows: Pileus diameter was 30.9 mm and thickness was 4.5 mm; stipe thickness was 11.0 mm and length was 76.0 mm; stipe and pileus hardness was 0.8 g/mm and 2.8 g/mm, respectively; L values of the stipe and pileus were 79.9 and 52.3, respectively. The fruiting body yield was 160.2 g/850 ml, and the individual weight was 12.8 g/10 unit.

Influences of Feeding Seleniferous Whole Crop Barley on Growth Performance, Blood and Carcass Characteristics, and Tissue Selenium Deposition in Finishing Barrows (셀레늄함유 청보리 급여가 거세비육돈의 생산성, 혈액 및 도체특성, 조직 내 셀레늄 축적에 미치는 영향)

  • The present study has been conducted to investigate the effects of feeding seleniferous whole crop barley (WCB) to finishing pigs on their growth performance, blood and carcass characteristics as well as on tissue selenium deposition. A total of 40 cross-bred barrows ((Landrace${\times}$Yorkshire)${\times}$Duroc) were allotted to five replicates of four treatments. Each replicate was arranged to 2 pigs per pen; the experimental period lasted for 6 weeks. The finishing pigs were fed diets containing 0.1 (non-seleniferous WCB as a control), 0.2, 0.4 and 0.6 ppm of selenium (Se) by supplementing the diets with seleniferous WCB. The isonitrogenous and isocaloric diets containing 5% non-seleniferous or seleniferous WCB were formulated. Feeding seleniferous WCB did not affect (p<0.05) the feed intake and BW gain. Total blood lipid concentration was significantly (p<0.05) decreased with increasing Se levels. Total blood cholesterol concentration for the control was significantly (p<0.05) higher than that for 0.4 and 0.6 ppm of Se treatments. Increasing the Se levels in WCB significantly (p<0.05) decreased blood triglyceride concentration; however, the levels increased immunoglobulin G and selenium concentrations. Feeding seleniferous WCB did not affect the carcass rate, backfat thickness and meat quality as well as yield grades. The Se concentration in the kidney, liver and loin were significantly (p<0.05) increased with increasing levels of seleniferous WCB. The results indicated that feeding seleniferous WCB may improve the blood characteristics related to lipid metabolism and thus, could produce selenium-fortified pork. Moreover, it is shown that the dietary optimal selenium level to depose selenium in porcine tissues by utilizing seleniferous WCB would be 0.4 mg of Se/kg of ration. Moreover, when 100 g of pork produced from pigs raised under such condition is served to consumers, it meets the minimum recommended daily requirements (40 ${\mu}g$) of dietary selenium proposed by the World Health Organization (1996).

Comparison of I-131 Scintigraphy, T1-201 Scintigraphy, and Serum Thyroglobulin in the Postoperative Follow-Up of Differentiated Thyroid Cancer (분화된 갑상선암의 수술후 경과관찰에서 I-131 스캔, T1-201 스캔 및 혈청 갑상선 글로불린 농도의 비교)

  • To evaluate the utility of I-131, T1-201 scintigraphy, and serum thyroglobulin(Tg) in the follow-up of differentiated thyroid cancer, we compared retrospectively the data from 33 patients who underwent total or subtotal thyroidectomy. I-131 scintigraphy was performed after optimal endogenous TSH stimulation ($TSH>50{\mu}U/ml$). Total 41 cases of I-131 and T1-201 scintigraphy pairs were examined. Concomitant serum thyroglobulin levels were measured for 41 pairs of scan. Tg-off levels(that measured after discontinuation of the thyroid hormone) higher than 40ng/m1 were considered positive, and Tg-on levels(that measured during the thyroid hormone replacement) higher than 5ng/ml were considered positive. The concordance rates between I-131 therapeutic scintigraphy and T1-201 scintigraphy was 48% in the 38 case of total scan pairs(59% in the 17 cases of postoperative preablation group, and 38% in the 21 cases of postoperative postablation group). Of 17 studies before the I-131 ablation therapy(preablation group), 7 showed positive I-131 therapeutic scintigraphy despite of negative T1-201 scintigraphy. Among patients with negative I-131 therapeutic scintigraphy, no patients had abnormal T1-201 uptake. However, of 21 studies which were done after radioiodine therapy(postablation group) 6 had abnormal uptake on T1-201 scintigraphy which were not seen on I-131 therapeutic scintigraphy, and Tg-off levels also elevated in this 4 of 6 cases. As a result, I-131 therapeutic scintigraphy showed highest positive rate at postoperative preablation follow-up study in differentiated thyroid cancer patients. T1-201 scintigraphy may be useful in postablation studies, and the use of the combined modalities(T1-201 and Tg levels) provides a higher diagnostic yield.
