Fermentation and Quality Characteristics of Kimchi Prepared Using Various Types of Maesil(Prumus mume Sieb. et Zucc) (매실의 첨가 형태에 따른 배추김치의 발효 및 품질 특성)

  • The fermentation and quality characteristics of kimchi prepared using various types of maesil, including freshly sliced maesil (FSM), freshly ground maesil (FGM),and dried maesil powder (MP), were investigated during 25-day fermentation at 10C. The pH value decreased rapidly in the control fermentation but only slowly in maesil-supplemented kimchi. Titratable acidity values changed in the same manner. The addition of maesil significantly inhibited the growth of total aerobes, and lactic acid bacteria during fermentation over 10 days. The color (L, a, and b values) of kimchi did not differ between control and maesil-supplemented kimchi samples regardless of maesil type or concentration. Kimchi hardness did not initially differ among the various samples but decreased during fermentation. However, the hardness of maesil-supplemented kimchi was higher than that of control samples. Thus, kimchi supplemented with either 6% (w/w) MP or 6% (w/w) FGM maintained optimal hardness during fermentation. The antioxidant activity of kimchi was increased in a dose-dependent manner by addition of maesil. Kimchi supplemented with 6% (w/w) FGM showed the highest antioxidant activity. The sensory qualities (taste, flavor, color, and overall acceptability) of FSM-supplemented kimchi were better than those of control samples.

Molecular Target Therapy of AKT and NF-kB Signaling Pathways and Multidrug Resistance by Specific Cell Penetrating Inhibitor Peptides in HL-60 Cells

  • Background: PI3/AKT and NF-kB signaling pathways are constitutively active in acute myeloid leukemia and cross-talk between the two has been shown in various cancers. However, their role in acute myeloid leukemia has not been completely explored. We therefore used cell penetrating inhibitor peptides to define the contributions of AKT and NF-kB to survival and multi drug resistance (MDR) in HL-60 cells. Materials and Methods: Inhibition of AKT and NF-kB activity by AKT inhibitor peptide and NBD inhibitor peptide, respectively, resulted in decreased expression of mRNA for the MDR1 gene as assessed by real time PCR. In addition, treatment of HL-60 cells with AKT and NBD inhibitor peptides led to inhibition of cell viability and induction of apoptosis in a dose dependent manner as detected by flow cytometer. Results: Finally, co-treatment of HL-60 cells with sub-optimal doses of AKT and NBD inhibitor peptides led to synergistic apoptotic responses in AML cells. Conclusions: These data support a strong biological link between NF-kB and PI3-kinase/AKT pathways in the modulation of antiapoptotic and multi drug resistant effects in AML cells. Synergistic targeting of these pathways using NF-kB and PI3-kinase/AK inhibitor peptides may have a therapeutic potential for AML and possibly other malignancies with constitutive activation of these pathways.

  • The effect of ${\gamma}$-ray irradiation on the color of nucleus and cultured pearls was investigated. After ($^{60}Co$) ${\gamma}$-ray irradiation on the pearl nucleus, its color was altered from brownish to blackish brown or gray depending on irradiation dose. It was clearly found that the all samples are composed of aragonite. ESR spectra were based on the measurement of the paramagnetic species $CO_2$-radicals by the interaction with the ${\gamma}$-ray irradiation in $Mn^{2+}$ containing $CaCO_3$. In this study, we investigated optimal detection method of ${\gamma}$-ray irradiation to establish destructive or non-destructive test for pearl nucleus.

Image Quality Evaluation in Computed Tomography Using Super-resolution Convolutional Neural Network (Super-resolution Convolutional Neural Network를 이용한 전산화단층상의 화질 평가)

  • High-quality computed tomography (CT) images enable precise lesion detection and accurate diagnosis. A lot of studies have been performed to improve CT image quality while reducing radiation dose. Recently, deep learning-based techniques for improving CT image quality have been developed and show superior performance compared to conventional techniques. In this study, a super-resolution convolutional neural network (SRCNN) model was used to improve the spatial resolution of CT images, and image quality according to the hyperparameters, which determine the performance of the SRCNN model, was evaluated in order to verify the effect of hyperparameters on the SRCNN model. Profile, structural similarity (SSIM), peak signal-to-noise ratio (PSNR), and full-width at half-maximum (FWHM) were measured to evaluate the performance of the SRCNN model. The results showed that the performance of the SRCNN model was improved with an increase of the numbers of epochs and training sets, and the learning rate needed to be optimized for obtaining acceptable image quality. Therefore, the SRCNN model with optimal hyperparameters is able to improve CT image quality.

Comparison of Measured and Predicted $^3H$ Concentrations in Environmental Media around the Wolsung Site for the Validation of INDAC Code (주면피폭선량 평가코드(INDAC)의 검증을 위한 월성원전 주면 삼중수소 농도 실측치와 예측치의 비교 평가)

  • The predicted results of INDAC code were compared with measured $^3H$ concentrations in air and pine-needle around the Wolsung site. The optimal sets of input data to INDAC were in addition selected by comparing the measured values with the predicted values of INDAC based on various conditions such as the release modes of effluents into the environment, the classification of wind classes, and the consideration of terrain. The predicted $^3H$ concentrations in air and pine-needle were shown to have good agreement with measured values, although there are some limitations such as uncertainties in measured values, complex topology around the site, and the land-sea breeze effects. The assumption on the $^3H$ behavior in vegetables or plants that the ratio of $^3H$ concentration in plant water to $^3H$ concentration in atmospheric water is 1/2 was shown to be conservative in terms of the audit calculation performed by the regulator. It was also found that data sets based on mixed mode and no terrain data were not appropriate for the audit calculation ensuring the compliance with regulations. Thus, if the mixed mode is considered as the release mode of effluents into the environment, meteorological data measured at 58 m height and terrain data should be used to evaluate the atmospheric dispersion factor.

Changes in Anti-inflammatory Effect of Pyungwi-san Decoction According to the Preservation Temperature and Period (평위산 전탕액의 보관 온도 및 기간에 따른 항염증 효능 비교)

  • Objectives : To provide the information of preservation method for herbal decoction, we evaluated the anti-inflammatory effects according to preservation temperature and period of Pyungwi-san (PWS, Ping wei-san) decoction. Methods : The anti-inflammatory activity of PWS was investigated by carrageenin-induced paw edema in SD rats. At 0 month, PWS extract was administrated 100, 300 and 1000 mg/kg/day orally for seven days prior to induction of edema. Edema was induced by subcutaneous injection of 1% carrageenin into the right hind paw. The paw volume was measured at 4 hr following carrageenin-induced paw edema in rats. At 6 and 12 months, PWS extracts according to preservation temperatures (room temperature, $4^{\circ}C$ and $-20^{\circ}C$) were administrated in rats and then carrageenin-induced paw edema volume was measured. Results : PWS showed inhibitory effect on carrageenin-induced paw edema in rats and the optimal dose was 1000 mg/kg/day at 0 month of storage. At 6 month of storage, PWS storaged at $4^{\circ}C$ and $-20^{\circ}C$ were inhibited paw edema but that storaged at room temperature was not reduced paw edema in rats. Conclusions : These results suggest that PWS decoction pouch has anti-inflammatory activities at both $4^{\circ}C$ and $-20^{\circ}C$ during 6 month storage.

Distribution of Microflora in Powdered Raw Grains and Vegetables and Improvement of Hygienic Quality by Gamma Irradiation (유통 생식제품의 미생물 분포 및 감마선 조사를 이용한 위생화)

  • Improvement of hygienic quality of powdered raw grains and vegetables by gamma irradiation was investigated. Five products of powdered raw grains and vegetables were collected in a local market and analysed. The total viable cell counts of Bacillus were 10$^4$~10$^{7}$ cfu/g, filamentous fungi l0$^2$~10$^3$cfu/g, coliform 10$^1$~10$^4$cfu/g, enteric bacteria on SS agar plate 10$^1$~10$^3$cfu/g. Coliform, enteric bacteria on SS agar plate and filamentous fungi were eliminated by 3 kGy of gamma irradiation. The D values of coliform, enteric bacteria on SS agar plate, Bacillus, and filamentous fungi were 0.68~0.80 kGy, 0.59~0.74 kGy, 1.84~2.18 kGy and 0.36~0.57 kGy, respectively. It was considered that optimal irradiation dose for radappertization was about 25 kGy, while 3~5 kGy of gamma irradiation was effective for radicidation.

3-Level Response Surface Design by Using Expanded Spherical Experimental Region (확장된 구형설계를 이용한 반응표면설계)

  • Response surface methodology(RSM) is a very useful statistical techniques for improving and optimizing the product process. By this reason, RSM has been utilized extensively in the industrial world, particularly in the circumstances where several product variables potentially influence some quality characteristic of the product. In order to estimate the optimal condition of product variables, an experiment is being conducted defining appropriate experimental region. However, this experimental region can vary with the experimental circumstances and choice of a researcher. Response surface designs can be classified, according to the shape of the experimental region, into spherical and cuboidal. In the spherical case, the design is either rotatable or very near-rotatable. The central composite design(CCD)s widely used in RSM is an example of 5-level and spherical design. The cuboidal CCDs(CCDs with ${\alpha}=1$) is appropriate when an experimental region is cuboidal but this design dose not satisfy the rotatability as it is not spherical. Practically, a 3-level spherical design is often required in the industrial world where various level of experiments are not available. Box-Behnken design(BBD)s are a most popular 3-level spherical designs for fitting second-order response surfaces and satisfy the rotatability but the experimental region does not vary with the number of variables. The new experimental design with expanded experimental region can be considered if the predicting response at the extremes are interested. This paper proposes a new 3-level spherical RSM which are constructed to expand the experimental region together with number of product variables.

Quality Characteristics of Jochung Containing Various Level of Steamed Garlic Powder (증숙마늘 분말 첨가 조청의 품질특성에 관한 연구)

  • This study was performed to investigate the effect of steamed garlic powder (SGP) on the quality characteristics of Jochung, saccharide by barley melt. Physicochemical properties and sensory quality were determined in Jochungs were prepared that 5% (SGP 5), 10% (SGP 10), 20% (SGP 20) and 30% (SGP 30) of SGP to sikhe for making jochung and hard boiled (W/W). Turbidity and viscosity of SGP added groups increased than control, significantly. The viscosity was $133{\pm}10^3$ cps in control group, it higher than SGP added groups ($140{\pm}11^3{\sim}272{\pm}2^3$ cps). The pH decreased with increasing SGP concentration. Reducing sugar was significantly higher in SGP added groups than control group, especially 30% SGP addition group in $44.66{\pm}0.26$ mg/100 g. HMF (Hydroxymethyl-furfural) content increased with increasing of SGP concentration, and 30% SGP addition group was higher in 48% than control group. In HPLC determination, free sugars content increased with amounts of SGP addition level. Fructose and glucose contents increased while maltose and raffinose contents decreased. Fructose content was $0.84{\pm}0.02$ mg/100g in control group but $1.36{\pm}0.03{\sim}1.62{\pm}0.01$ mg/100 g in SGP added groups. In the sensory evaluation, the color of Jochung decreased dose dependently with the SGP. Garlic flavor was significantly greater in SGP added groups compared to that of control group. The overall acceptability had not significantly difference among control, SGP 5 and 10 group. Preference was decreased in contain more than 20% of SGP added groups. The optimal concentration of SGP was found in the range of less than 10%.

The Algicidal Activity of Pseudoalteromonas sp. NH-12 against the Toxic Dinoflagellate Alexandrium catenella (유독성 와편모류 Alexandrium catenella에 대한 Pseudoalteromonas sp. NH-12의 살조능)

  • BACKGROUND: The aim of this study was to isolate and identify algicidal bacterium that tends to kill the toxic dinoflagellate Alexandrium catenella, and to determine the algicidal activity. METHODS AND RESULTS: Among of four algicidal bacteria isolated in this study, NH-12 isolate was the strongest algicidal activity against A. catenella. NH-12 isolate was identified on the basis of biochemical characteristics and analysis of 16S rRNA gene sequences. The isolate showed 97.67% homology with Pseudoalteromonas prydzensis ACAM $620^T$ (U85855), and was designated Pseudoalteromonas sp. NH-12. The optimal culture conditions of this isolate were $25^{\circ}C$, initial pH 8.0, and 3.0% (w/v) NaCl concentration. The algicidal activity of NH-12 was significantly increased to maximum value in the late of logarithmic phase of bacterial culture. As a result of 'cell culture insert' experiment, NH-12 is assumed to produce secondary metabolites, as an indirect attacker. When 10% culture filtrate of NH-12 was applied to A. catenella, over 99% of algal cells were destroyed within 24 h. In addition, the killing effects were increased in dose and time dependent manners. CONCLUSION(S): Taken together, our results suggest that Pseudoalteromonas sp. NH-12 could be a candidate for controlling of toxic algal blooms.
