  • The use of Compaction Grouting evolved in 1950's to correct structural settlement of buildings. Over the almost 50 years, the technology has been developed and is currently used in wide range of applications. Compaction Grouting, the injection of a very stiff, 'zero-slump' mortar grout under relatively high pressure, displaces and compacts soils. It can effectively repair natural or man-made soil strength deficiencies in variety of soil formations. Major applications of Compaction Grouting include densifying loose soils or fill voids caused by sinkholes, poorly compacted fills, broken utilities, improper dewatering, or soft ground tunneling excavation. Other applications include preventing liquefation, re-leveling settled structures, and using compaction grout bulbs as structural elements of minipiles or underpinning. In this paper, on the basis of the case history constructed in this year, a study has been performed to analyze the basic mechanism of the Compaction Grouting. Also, the effectiveness of the ground improvement and the bearing capacity of the Compaction Pile has been verified by the Cone Penetration Test(CPT) and Load Test. Relatively uniform Compaction grouting column could be maintained by planning the Quality Control in the course of grouting. And, the Quality Control Plan has been conceived using grout pressure, volume of grout and drilling depth.

A Study on the Mixing Characteristics in Complex Turbulent Flow by a Laser Induced Fluorescence Method (레이저 형광여기법(LIF)를 이용한 복잡 난류유동장의 혼합특성에 관한 연구)

  • A non-intrusive Planar Laser-Induced Fluorescence(PLIF) technique was applied to study the turbulent mixing process in a Rushton turbine reactor. Instantaneous and ensemble averaged concentration fields are obtained by measuring the fluorescence intensity of Rhodamine B tracer excited by a thin Nd:Yag laser sheet illuminating the whole center plane of the stirred tank. The gray level images captured by a 14-bit cooled CCD camera can be transformed to the local concentration values using a calibration matrix. The dye injection point was selected at the tank wall with three quarter height (3/4H) from the tank bottom to observe the mixing characteristics in upper bulk flow region. There exist distinct two time scales: the rapid decay of mean concentration in each region after the dye infusion reflects the large scale mixing while the followed slow decay reveals the small scale mixing. The temporal change of concentration probability functions conjectures the two sequential processes in the batch type mixing. An inactive column of water existed above the impeller disk, in which the fluid rotates with the shaft but is isolated from the mean bulk flow.

  • A study on the feasibility of using improved computer-controlled HPLC and GC systems was carried out to shorten the time needed for measuring levels of the substrates (glucose, maltose, and glycerol) and products (acetone, butanol ethanol, acetic acid, and butyric acid) produced by Clostridium saccharobutylicum DSM 13864 during direct fermentation of sago starch to solvent. The use of HPLC system with a single injection to analyse the composition of culture broth (substrates and products) during solvent fermentation was achieved by raising the column temperature to $80^{\circ}C$. Although good separation of the components in the mixture was achieved, a slight overlap was observed in the peaks for butyric acid and acetone. The shape of the peak obtained and the analysis time of 26.66 min were satisfactory at a fixed flow rate of 0.8mL/min. An improved GC system was developed, that was able to measure the products of solvent fermentation (acetone, butanol, ethanol, acetic acid, and butyric acid) within 19.28 min. Excellent resolution for each peak was achieved by adjusting the oven temperature to $65^{\circ}C$.

  • Zingiberis Rhizoma (ZR, Zingiberaceae) is one of the foods for a long time and contains various biological activities including anti-cancer, anti-inflammatory, anti-oxidant, and anti-platelet activities. The present study was to compare the amount of 6-gingerol in the 70% ethanol extracts of non-processed ZR and processed ZR by roasting using a high-performance liquid chromatography equipped with photodiode array detector. The 6-gingerol was separated on a Gemini $C_{18}$ analytical column ($5{\mu}m$, $4.6{\times}250mm$) using two mobile phase consisting of distilled water and acetonitrile. The flow rate was 1.0 mL/min and injection volume was $10{\mu}L$. In non-processed ZR sample, the amount of the 6-gingerol was 0.43% and the amount of the marker compound in processed ZR samples by roasting were 0.16-0.55%.

Isolation and Characterization of Mucinase Complex Secreted from Vibrio parahaemolyticus

  • Mucinase complex from V. parahaemolyticus ATCC 17802 was purified 6-fold with 0.4% yield by two sequential steps of Q-Sepharose and Superdex 200HR column chromatographies. Partially purified mucinase complex showed at least 8 times higher mucin-degrading activity than the culture filtrates. The mucinase complex also showed gelatin-and-casein-hydrolyzing activities, which demonstrates that the protein is a complex compound containing several proteases. The optimum pH and temperature of partially purified mucinase complex for mucin degradation was 8.0 and $35^{\circ}C$, respectively. The partially purified mucinase complex showed high cytotoxic activity on vero cells when examined by MTT assay and microscopic observations. Cytotoxicity was significantly increased in proportion to the concentration of the mucinase complex. Mouse experiments revealed that the jejunum, ileum. and large intestinal tissues were damaged by the injection of the mucinase complex. In particular, the reduction of the goblet cells in the large intestine was remarkable. Collectively, these data suggest that the mucinase complex partially purified from V. parahaemolyticus ATCC 17802 contributes to the adhesion and invasion of V. parahaemolyticus into the host intestinal tract.

Transient Spray Structures of Supersonic Liquid Jet Injected by Projectile Impact Systems (발사체 충격 방식을 사용한 초음속 액체 제트의 과도 분무 형상에 관한 연구)

  • The effects of projectile impact system on the transient spray characteristic which is supersonic liquid tip velocity were studied by experimentally. Supersonic liquid jets were generated by impact of a high speed projectile driven by a Two-stage light gas gun. A high speed camera and schlieren optical system were used to capture the spray structures of the supersonic liquid jets. In a case of nozzle assembly Type-A, expansion gases accelerate a projectile which has a mass of 6 grams from 250 m/s at the exit of the launch tube. Accelerated projectile collides with the liquid storage part, then supersonic liquid jets are injected with instantaneous spray tip velocity from 617.78 m/s to 982.54 m/s with various nozzle L/d. However, In a case of nozzle assembly Type-B which has a heavier projectile (60 grams) and lower impact velocity (182 m/s), an impact pressure was decreased. Thus the liquid jet injected at 210 m/s of the maximum velocity did not penetrate a shock wave and fast break-up was occurred. Pulsed injection of liquid column generated second shock wave and multiple shock wave.

Isomeric Effects on Volatilization of 1,3-Dichloropropene Fumigant in Soil

  • The fumigant 1,3-dichloropropene (1,3-D) was recently proposed as a direct replacement for methyl bromide ($CH_3Br$) in soil fumigation. This study was conducted to better understand behavior phase partitioning, diffusion and volatilization of 1,3-D as affected by isomer. The Henry's law constant(KH) of cis-1,3-D and trans-1,3-D was 0.058 and 0.037 at $20^{\circ}C$, respectively. $K_H$ of cis form of 1,3-D was higher than that of trans form of 1,3-D. To compare with volatilization of 1,3-D isomer, soil column [70 cm (length)${\times}$12 cm (i.d.)] included a shank injection at 30 cm with 300 kg $ha^{-1}$. Maximum cis-1,3-D and trans-1,3-D concentration reached 57 mg $L^{-1}$ and 39 mg $L^{-1}$ at 30 cm depth at 1h after application. Cumulatively, after 10 days, 51.8% and 43.57% of applied cis-1,3-D and trans-1,3-D was emitted via volatilization, respectively. The total losses of cis-1,3-D were significantly greater than that of trans-1,3-D. Finally, cis-1,3-D and trans-1,3-D, such as isomer are dominant of 1,3-D fates in soil.

Quantitative Analysis of the Twenty Marker Components in Gwakhyangjeonggi-san using Ultra-Performance Liquid Chromatography with Mass Spectrometer (LC-MS/MS를 이용한 곽향정기산(藿香正氣散) 추출물 중 20종 성분의 함량분석)

  • Generally, Gwakhyangjeonggi-san has been used for treatment of diarrhea-predominant irritable bowel syndrome. In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometer method was established for the simultaneous quantification of marker compounds 1-20 in Gwakhyangjeonggi-san water extract. All analytes were separated by gradient elution using two mobile phases on a UPLC BEH $C_{18}$ ($100{\times}2.1mm$, $1.7{\mu}m$) column and maintained at $45^{\circ}C$. The injection volume was $2.0{\mu}L$ and the flow rate was 0.3 mL/min with detection at mass spectrometer. Regression equations of the compounds 1-20 were acquired with $r^2$ values ${\geq}0.9950$. The values of limit of detection and quantification of all analytes were 0.01-2.79 ng/mL and 0.03-8.37 ng/mL, respectively. The amounts of the compounds 1-20 in Gwakhyangjeonggi-san water extract were not detected $-3,236.67{\mu}g/g$. The established LC-MS/MS methods will be valuable to improve quality control of traditional herbal formula, Gwakhyangjeonggi-san.

Flatfish Vitellogenin Detection Using Optical Waveguide Lightmode Spectroscopy-based Immunosensor

  • A sensitive optical waveguide lightmode spectroscopy-based immunosensor was developed to detect vitellogenin in seawater flatfish (Paralichthys olivaceus). For this purpose, anion-exchange column chromatography with DE-52 resin was used to purify flatfish vitellogenin from flatfish serum containing vitellogenin that had been induced using an intraperitoneal $17{\beta}$-estradiol injection. The anti-flatfish vitellogenin antibody used as the biological component of the above immunosensor was prepared using the purified flatfish vitellogenin. The change in the incoupling angle according to the complexation between the flatfish vitellogenin and its antibody, immobilized over an optical grating coupler sensor chip, was measured to calculate the sensor response. The immunosensor was quite specific to flatfish vitellogenin binding, based on no sensor response in the case of bovine serum albumin immobilization. When plotted using double-logarithmic scales, the sensor responses increased linearly in flatfish vitellogenin concentrations of 0.00675-67.5 nM, with a detection limit of 0.0675 nM. The reusability during seven repetitive measurements was reasonably fair for the preliminary screening of flatfish vitellogenin.
