토끼 핵이식에 의한 복제산자의 생산효율 향상에 관한 연구 (Production of Identical Rabbit Offspring by Nuclear Transplantation)

  • This study was carried out to improve a technique of cloned animal prodcution by preactivation of nuclear recipient oocytes with ionomycin and 6-dimethylaminopurine (6-DMAP) in rabbits. The oocytes were collected from the oviduct of superovulated rabbit at 19∼20 hours post hCG injection. The collected oocytes were preactivated and self-enucleated by treating 5 uM ionoycin for 5 min, and 2.0 mM 6-DMAP for two hours. Microsurgical removel of the chromation complex in the second polar bodies was effectively performd and single blastomere separated from 32-cell stage rabbit embryos was injected into the perivitelline space of the enculeated recipient oocyts. Follwoing electrofusion and in vitro culture for 18 hours, the nuclear transplant(NT) embryos were transferred into the uterine horns of naturally mated or synchronized recipient does. When 32 NT embryous reconstituted with preactivated oocytes were transferred to 2 recipient does, one foster doe delivered two offspring (6.3%), while not a offspring was delivered from three foster does which received 17 NT embryos reconstituted with non-preactivated oocytes. A total of 68 NT embryos reconstituted with preactivated oocytes were transferred into the uterine horns of 7 synchronized ecipient does. Among them, two recipients were pregnant and delivered three offspring(5.9%).

Effects of Maternal Nutrition during Pregnancy on the Body Weight, Muscle Fiber Number, Carcass Traits, and Pork Quality Traits of Offspring

  • The purpose of the current study was to examine the influence of different maternal nutrition treatments during pregnancy on body weight, muscle fiber number, carcass traits, and pork quality traits of offspring. A total of 18 crossbred sows (Landrace${\times}$Yorkshire${\times}$Duroc) were randomly assigned to one of three nutritional treatment groups; control, high energy, and high protein. The control group was fed a standard diet, the high energy group was fed a diet that contained 30% increased metabolizable energy, and the high protein group was fed a diet that contained 30% increased limiting amino acids compared to the control. The sows in each group were fed equal quantities of each diet (1.9 kg/d) for the entire gestational period. A total of 36 piglets from each sow were used to evaluate changes in body weight, muscle fiber number in the longissimus dorsi muscle at birth, carcass traits, and pork quality traits. Birth weight of offspring born to sows in the high energy diet group was significantly higher compared to the high protein diet group (p<0.05). However, body weight of offspring after birth was not significantly different between the groups. Muscle fiber number for the longissimus dorsi muscle at birth was not significantly different between the groups. In addition, there were no significant differences in carcass traits or pork quality traits between offspring born to sows in the control group and those born to sows that received high energy or high protein diets during pregnancy. Based on these results, it appears that maternal nutrition treatment during pregnancy, regardless of whether it is with high energy or high protein diets, does not have a significant effect on body weight, muscle fiber number at birth, carcass traits, or pork quality traits.

Comparison of three boosting methods in parent-offspring trios for genotype imputation using simulation study

  • Background: Genotype imputation is an important process of predicting unknown genotypes, which uses reference population with dense genotypes to predict missing genotypes for both human and animal genetic variations at a low cost. Machine learning methods specially boosting methods have been used in genetic studies to explore the underlying genetic profile of disease and build models capable of predicting missing values of a marker. Methods: In this study strategies and factors affecting the imputation accuracy of parent-offspring trios compared from lower-density SNP panels (5 K) to high density (10 K) SNP panel using three different Boosting methods namely TotalBoost (TB), LogitBoost (LB) and AdaBoost (AB). The methods employed using simulated data to impute the un-typed SNPs in parent-offspring trios. Four different datasets of G1 (100 trios with 5 k SNPs), G2 (100 trios with 10 k SNPs), G3 (500 trios with 5 k SNPs), and G4 (500 trio with 10 k SNPs) were simulated. In four datasets all parents were genotyped completely, and offspring genotyped with a lower density panel. Results: Comparison of the three methods for imputation showed that the LB outperformed AB and TB for imputation accuracy. The time of computation were different between methods. The AB was the fastest algorithm. The higher SNP densities resulted the increase of the accuracy of imputation. Larger trios (i.e. 500) was better for performance of LB and TB. Conclusions: The conclusion is that the three methods do well in terms of imputation accuracy also the dense chip is recommended for imputation of parent-offspring trios.

Effects of flaxseed oil supplementation on lactating sows and their offspring

  • This study was conducted on sows to evaluate the effect of flaxseed oil on the sows and their offspring's performance. Forty-eight (48) sows (Landrace × Yorkshire) and their offspring were assigned randomly to each treatment (Control and Control + flaxseed oil 0.5% [FX]). Body weight (BW), average daily feed intake (ADFI) and nutrient digestibility of the sows were estimated. Sow backfat thickness, days-to-estrus, and chest circumference were measured by different methods. In piglets, survival rate, the initial weight, weaning weight, and average daily gain (ADG) were calculated. Fecal scores of both the sows and piglets were recorded. Inclusion of FX did not influence (p > 0.05) the litter size, days-to-estrus, sow BW, ADFI and digestibility of nutrient throughout the experiment. Reduction in (p < 0.05) BW loss was observed with the FX supplementation in sows. Only the backfat thickness during the weaning period showed an increase (p < 0.05) by FX supplementation. Chest circumference was decreased (p < 0.05) in the FX group compared to the control. Sows fed the FX diet had a reduced (p < 0.05) fecal score only at weaning. Piglets' fecal score showed no significant differences for FX supplementation. However, the inclusion of FX brought about a higher (p < 0.05) birth weight, weaning weight, and average daily gain (ADG). In conclusion, our study suggests that FX has positive supplementary effects on a sow's body condition and its offspring's performance as a source of n-3 fatty acid.

Maternal high-fructose intake during pregnancy and lactation induces metabolic syndrome in adult offspring

  • BACKGROUND/OBJECTIVES: Nutritional status and food intake during pregnancy and lactation can affect fetal programming. In the current metabolic syndrome epidemic, high-fructose diets have been strongly implicated. This study investigated the effect of maternal high-fructose intake during pregnancy and lactation on the development of metabolic syndrome in adult offspring. SUBJECTS/METHODS: Drinking water with or without 20% fructose was administered to female C57BL/6J mice over the course of their pregnancy and lactation periods. After weaning, pups ate regular chow. Accu-Chek Performa was used to measure glucose levels, and a tail-cuff method was used to examine systolic blood pressure. Animals were sacrificed at 7 months, their livers were excised, and sections were stained with Oil Red O and hematoxylin and eosin (H&E) staining. Kidneys were collected for gene expression analysis using quantitative real-time Polymerase chain reaction. RESULTS: Adult offspring exposed to maternal high-fructose intake during pregnancy and lactation presented with heavier body weights, fattier livers, and broader areas under the curve in glucose tolerance test values than control offspring. Serum levels of alanine aminotransferase, aspartate aminotransferase, glucose, triglycerides, and total cholesterol and systolic blood pressure in the maternal high-fructose group were higher than that in controls. However, there were no significant differences in mRNA expressions of renin-angiotensin-aldosterone system genes and sodium transporter genes. CONCLUSIONS: These results suggest that maternal high-fructose intake during pregnancy and lactation induces metabolic syndrome with hyperglycemia, hypertension, and dyslipidemia in adult offspring.

Association between polycystic ovary syndrome and risk of attention-deficit/hyperactivity disorder in offspring: a meta-analysis

  • Background: There is evidence of a relationship between prenatal excess androgen exposure and central nervous developmental problems and attention-deficit/hyperactivity disorder (ADHD) in the offspring of mothers with polycystic ovary syndrome (PCOS). Purpose: Here we aimed to use a meta-analysis to investigate whether the offspring of mothers with PCOS are at an increased chance of developing ADHD. Methods: Three main English databases were searched for articles published through December 2020. The Newcastle-Ottawa Scale was used to assess study quality. Study heterogeneity was determined using I2 statistics and publication bias was assessed using Begg and Egger tests. The results are presented as odds ratio (OR) and relative ratio (RR) estimates with 95% confidence intervals (CIs) using a random-effects model. Results: Six articles (3 cohort and 3 case-control studies; 401,413 total ADHD cases) met the study criteria. Maternal PCOS was associated with an increased risk of ADHD in the offspring based on OR and RR (OR, 1.42; 95% CI, 1.27-1.57) and (RR, 1.43; 95% CI, 1.35-1.51), respectively. There was no heterogeneity among the included articles based on OR (I2=0.0%, P=0.588) and RR (I2=0.0%, P=0.878). Conclusion: Our study showed that maternal PCOS is a risk factor for ADHD. Therefore, screening their offspring for ADHD should be considered part of the comprehensive clinical care of women with PCOS.

Cohort Study Protocol: A Cohort of Korean Atomic Bomb Survivors and Their Offspring

  • In 1945, atomic bombs were dropped on Hiroshima and Nagasaki. Approximately 70 000 Koreans are estimated to have been exposed to radiation from atomic bombs at that time. After Korea's Liberation Day, approximately 23 000 of these people returned to Korea. To investigate the long-term health and hereditary effects of atomic bomb exposure on the offspring, cohort studies have been conducted on atomic bomb survivors in Japan. This study is an ongoing cohort study to determine the health status of Korean atomic bomb survivors and investigate whether any health effects were inherited by their offspring. Atomic bomb survivors are defined by the Special Act On the Support for Korean Atomic Bomb Victims, and their offspring are identified by participating atomic bomb survivors. As of 2024, we plan to recruit 1500 atomic bomb survivors and their offspring, including 200 trios with more than 300 people. Questionnaires regarding socio-demographic factors, health behaviors, past medical history, laboratory tests, and pedigree information comprise the data collected to minimize survival bias. For the 200 trios, whole-genome analysis is planned to identify de novo mutations in atomic bomb survivors and to compare the prevalence of de novo mutations with trios in the general population. Active follow-up based on telephone surveys and passive follow-up with linkage to the Korean Red Cross, National Health Insurance Service, death registry, and Korea Central Cancer Registry data are ongoing. By combining pedigree information with the findings of trio-based whole-genome analysis, the results will elucidate the hereditary health effects of atomic bomb exposure.

Avian Somitic Cell Chimeras Using Surrogate Eggshell Technology

  • A classical technique to study somitic cell fate is to employ the cross-transplantation of quail somites into a chick host. The densely stained nucleoli of the quail cells makes it possible to assess the fate of the donor quail cells in the chick host. Classical somite transplantation techniques have been hampered by the necessity of a small opening in the chick eggshell, difficulty in hatching the offspring and interspecies post-hatch graft rejection. With the advent of transgenic chicken technology, it is now possible to use embryos from transgenic chickens expressing reporter genes in somite cross-transplantation techniques to remove any possibility of interspecies graft rejection. This report describes using a surrogate eggshell system in conjunction with transgenic chick:chick somitic cell cross-transplantation to generate viable chimeric embryos and offspring. Greater than 40% of manipulated embryos survive past 10 days of incubation, and ~80% of embryos successfully cultured past 10 days of incubation hatched to produce viable offspring.

Effect of anthelmintic drug in pregnancy

  • Pyrantal pamoate's anthelmintic activity is due to its action as a neuromuscular blocking agent. It is generally well tolerated. Transient rises in SGOT levels have been reported in the drug-treated patients. Decreased levels of serum alkaline phosphatase post treatment were found in yound dogs. The present study was performed to investigate the possible toxic effects of pyrantal pamoate in pregnant mice progenies. The drug was given in different doses to these mothers in the first, second and third trimester. Serum alkaline phosphatase, SGOT and SGPT of one or two month old offspring were monitored. SGOT levels showed an increase in some doses in one and two month old offspring where alkaline phosphatase showed a decrease in some doses in one and two month old offspring. The latter effect may be due to osteoblastic alkaline phosphatase inhibition. The effect on SGOT levels, however, was difficult to explain, but may be due to a toxic effect on liver cells or cardiac muscles.

Effective Multi-label Feature Selection based on Large Offspring Set created by Enhanced Evolutionary Search Process

  • Recent advancement in data gathering technique improves the capability of information collecting, thus allowing the learning process between gathered data patterns and application sub-tasks. A pattern can be associated with multiple labels, demanding multi-label learning capability, resulting in significant attention to multi-label feature selection since it can improve multi-label learning accuracy. However, existing evolutionary multi-label feature selection methods suffer from ineffective search process. In this study, we propose a evolutionary search process for the task of multi-label feature selection problem. The proposed method creates large set of offspring or new feature subsets and then retains the most promising feature subset. Experimental results demonstrate that the proposed method can identify feature subsets giving good multi-label classification accuracy much faster than conventional methods.
