Comparison of Ablation Performance between Octopus Multipurpose Electrode and Conventional Octopus Electrode

  • Objective: To compare Octopus multipurpose (MP) electrodes, which are capable of saline instillation and direct tissue temperature measurement, and conventional electrodes for radiofrequency ablation (RFA) in porcine livers in vivo. Materials and Methods: Sixteen pigs were used in this study. In the first experiment, RFA was performed in the liver for 6 minutes using Octopus MP electrodes (n = 15 ablation zones) and conventional electrodes (n = 12 ablation zones) to investigate the effect of saline instillation. The ablation energy, electrical impedance, and ablation volume of the two electrodes were compared. In the second experiment, RFA was performed near the gallbladder (GB) and colon using Octopus MP electrodes (n = 12 ablation zones for each) with direct tissue temperature monitoring and conventional electrodes (n = 11 ablation zones for each). RFA was discontinued when the temperature increased to > 60℃ in the Octopus MP electrode group, whereas RFA was performed for a total of 6 minutes in the conventional electrode group. Thermal injury was assessed and compared between the two groups by pathological examination. Results: In the first experiment, the ablation volume and total energy delivered in the Octopus MP electrode group were significantly larger than those in the conventional electrode group (15.7 ± 4.26 cm3 vs. 12.5 ± 2.14 cm3, p = 0.027; 5.48 ± 0.49 Kcal vs. 5.04 ± 0.49 Kcal, p = 0.029). In the second experiment, thermal injury to the GB and colon was less frequently noted in the Octopus MP electrode group than that in the conventional electrode group (16.7% [2/12] vs. 90.9% [10/11] for GB and 8.3% [1/12] vs. 90.9% [10/11] for colon, p < 0.001 for all). The total energy delivered around the GB (2.65 ± 1.07 Kcal vs. 5.04 ± 0.66 Kcal) and colon (2.58 ± 0.57 Kcal vs. 5.17 ± 0.90 Kcal) were significantly lower in the Octopus MP electrode group than that in the conventional electrode group (p < 0.001 for all). Conclusion: RFA using the Octopus MP electrodes induced a larger ablation volume and resulted in less thermal injury to the adjacent organs compared with conventional electrodes.

The Standardized Recipe and Nutrient Analysis of Stir-fried Whip-arm Octopus in Busan (부산지역 낙지볶음의 표준조리법 개발 및 영양소 분석)

  • The purpose of this study was to develop the standardized recipe and to analyze the nutrients of stir-fried whip-arm octopus as one of the kind of traditional local foods in Busan. The standardized recipe was developed by using cookbooks, home recipes, and the recipes by commercial food restaurants. The nutrient was analyzed by using an AOAC method. Sensory evaluations were made on nine sensory attributes by a 12-member panel. As the results, the whip-arm octopus was recorded as a food source for health, rejuvenation in Jasanobo. Since early 1900, the stir-fried method has been used for cooking with the ship-arm octopus. The main ingredients were whip-arm octopus, onion, large green onion as the local stir-fried whip-arm octopus in Busan. The ingredients of soup were shrimp, little neck clam, and water. Seasoning sauce was mixed with red pepper powder, soy sauce, sesame oil, chopped garlic, chopped onion, and sugar. The seasoning sauce was fermented for three days. The results of sensory scores were salty seasoning 3.66/5.00, taste 3.75/5.00, thickness 3.84/5.00, and smell 4.09. Nutrient retention per 100g of the stir-fried whip-arm octopus was 67.54kcal, protein 6.43g, fat 1.66g, Ca 28.06mg, Fe 2.56mg, cholesterol 68mg, and taurin 51mg. Fatty acid consists of various unsaturated fatty acids such as linoleic acid(46.24%) and oleic acid(33.67%).

  • In order to investigate the influence of sea condition on the catch fluctuation of long line for common octopus, octopus variabilis. the oceanographic factors. I .e., the water temperature, the salinity and the density in the coastal waters of Yosu from Jan 11 to Jul. 25 in 1997 and compared with the catches of common octopus, octopus variabilis by long line. The results obtained summerized as follows ; 1) The range of water temperature, salinity and density from Jan. to Jul. at the sea area were 4.1∼23.6℃. 31.7∼34.5‰ and 21.30∼27.81. respectively, and the catch of common octopus, octopus variabilis was high respectively at the temperature of 17∼20℃ at the salinity of 33.0∼33.5‰ and at the density of σ/sub t/ 22.5∼24.0. 2) The catch of common octopus. octopus variabilis was lowest from Jan. to Feb. and highest from May to June.

Concentration of Polybrominated Diphenyl Ethers and Their Composition in Octopus minor Collected from Seosan Intertidal Zone (낙지(Octopus minor)에서의 브롬계화합물(Polybrominated diphenyl ethers, PBDEs)의 잔류농도와 조성특성)

  • Octopus(Octopus minor), benthic cephalopod, were collected from intertidal zone in Seosan, Choongnam and analyzed for polybrominted diphenyl ethers(PBDEs). PBDEs concentrations ranged from 29 to 109 ng/g lipid wt(mean; 54 ng/g) in mantle and from 12 to 89 ng/g lipid wt(mean; 48 ng/g) in internal organ. PBDEs concentrations in octopus internal organ were lower about two times than that in common squid collected in Yellow Sea, indicating Seosan is relatively less contaminated with PBDEs. Major congener was BDE 206, occupying 72% and 49% of total PBDE concentration in mantle and in internal organ, respectively, which is very different from PBDE composition in common squid with major congeners of BDE 47 and 99. From PBDE composition, octopus seemed to concentrate higher brominated BDE rather than lower brominated BDE. This PBDE composition in octopus may be affected by sediment with extremely high contribution of deca-BDE to total PBDE concentration.

A study on the status of octopus pot fishery in the coastal seas of Gyeongbuk, Korea (우리나라 경북 연안해역 문어통발 어업실태에 관한 연구)

  • In this study, the fishery status of the octopus pot fishery in the east coastal sea were investigated, and the fishing performance of each pot shape was compared and analyzed. The fishery status survey was conducted through listening surveys at Jukbyeon Port, Uljin Port and Pohang Daebo Port in Gyeongbuk Province, and the amount of fishing gear used, fishing method, size and loss of octopus pot fishery was investigated. On the east coastal sea, octopus is one of the commercially important fish stocks and is caught in inshore pots, inshore combos, inshore gillnets and offshore pots. Among these fishing methods, pot fishing yields the highest catch. The shape of the pot differs depending on the region. In Uljin (Jukbyeon Port and Hupo Port), Gyeongbuk, rectangular type net pots are mainly used, and in Pohang (Daebo Port) in Gyeongbuk, drum-type pots are mostly used. Enteroctopus dofleini accounts for more than 90% of the catch of octopus. For the octopus fishing performance test by trap type, three types of traps (rectangular pot, drum pot and cylinder pot) were used on the coastal sea of Pohang Daebo. As a result, the total catch by pot shape was shown in the order of rectangular-type pot > drum-type pot > cylinder-type pot. The catch of octopus, the target species, was in the following order: rectangular-type pot > drum-type pot > cylinder-type pot. Such result shows a significant difference (Mann-Whitney test, p<0.05).

Entering behavior and fishing efficiency of common octopus, Octopus minor to cylindric trap (통발에 대한 낙지의 입망 행동과 어획성능)

  • The behavior patterns of common octopus, Octopus minor to a cylindric trap were examined in the water tank using a video camera in order to know entering mechanism to the trap and to improve it. Fishing efficiency by 2 kinds of traps(A: 3 entrances, B: 2 entrances) was investigated in the coastal area of Deugryang Bay from May to July, 2005. Common octopus tends to approach by swimming more than walking towards trap. When they approached to the trap, they showed much more behaviors that sate at the around than upper part of it. Approaching behaviors of common octopus was more vigorous at nighttime than daytime on the trap, they showed the most vigorous action between 2 am and 4 am of the day. The rate of staying 30 seconds over around the trap was 41.5% in the nighttime. CPUE(g/trap) of common octopus caught by A type trap was 21.4% higher than the B type trap but there was no difference on the significance level of 5% by the ANOVA. Catch rate of common octopus and by-catch species caught by the A type trap were 97.2%, three and 98.7%, two for B type trap, respectively.

Color effect of the environment-friendly sinker for octopus drift-line (문어흘림낚시용 친환경 봇돌의 색 효과)

  • The choice behavior of the octopus in response to the environment-friendly colored sinker for octopus driftline and the sinker's fishing effect were studied under experimental conditions in the water tank and the field. The colors of the sinkers used for the experiment are white, black, yellow and green. Artificial baits are attached to the sinkers in order to attract the octopuses in the experiment. In the water tank experiment, two sinkers of two different colors are placed in a compartmentalized corner of the rectangular tank, and a CCD camera records the choice behaviors of the octopuses to the colored sinkers. In the field experiment, the catch investigation of octopus for each colored sinker was conducted 14 times in total by using 2(A, B) commercial fishing boats at the coast of Gangneung within 30m of depth in 2006. The number of colored sinkers per operation was a total of 24-40 pieces with the same number of sinkers individually for four colors. As a result, it was found that the octopus selected white the most followed by black and yellow in their choice of colored sinkers in the water tank experiment, and green was the lowest in their choice. Even in the field experiment, the sinkers of white and black showed a higher catch of octopus than the sinkers of yellow and green.

Comparison of the Exopeptidase Activity of Fractions from Crude Extracts of Octopus Octopus vulgaris Cuvier Hepatopancreas Using Different Fractionation Methods

  • This study was performed to identify the optimum fractionation method and conditions to obtain exopeptidase-active fractions from octopus hepatopancreas (HP) crude extracts (CEs) using four techniques: solid ammonium sulfate fractionation, polyethylene glycol (PEG) fractionation, anion exchange chromatography, and gel filtration chromatography. The fractions with the highest total activity toward L-leucine-p-nitroanilide (Leu-pNA) were fraction IV from the ammonium sulfate and PEG fractionation, and fraction II in ion exchange and gel filtration chromatography. The total exoprotease activity of these fractions was highest in fraction IV (4,050.20 U) of ammonium sulfate fractionation, followed by fraction II (3,600.28 U) from gel filtration chromatography, fraction IV (2,861.30 U) from PEG fractionation, and fraction II (2,576.28 U) from ion exchange chromatography. These results suggest that ammonium sulfate fractionation using 60-80% ammonium sulfate was the most efficient method for separating the exoprotease active fractions from CEs of octopus HP.

Rapid and Direct Detection of Vibrio vulnificus in Small Octopus (Octopus variabilis) Using Polymerase Chain Reaction

  • The cells of Vibrio vulnificus can be induced to the viable but nonculturable (VBNC) state by natural environmental parameters. The V. vulnificus cells in the VBNC state can not be recovered by ordinary laboratory techniques. This nonculturability could often hamper development of effective processing strategies to minimize the number of V. vulnificus in seafoods. Even with V. vulnificus cells in a culturable state, the length of time required to identify the bacteria in contaminated food by phenotyphic characterization may prevent appropriate in-time responses by public health agencies to infections of the bacteria. In the present study, we used polymerase chain reaction (PCR) to develop a rapid and direct detection method for V. vulnificus in small octopus (Octopus variabilis) which is consumed as a raw food in Korea. The region targeted was a 704-base pair (bp) portion of the hemolysin gene, vvhA, of V. vulnificus. The primers designed for PCR amplification were specific for all V. vulnificus sp. tested. Several methods were examined to extract total DNA directly from V. vulnificus seeded into the octopus homogenate and the guanidine isothiocyanate (CITC) method appeared to be most effective. From the octopus homogenate seeded by V. vulnificus at an initial level of $10^2$ CFU/ml of the homogenate and then incubated for 12 h, the targeted sequence was successfully amplified by PCR and the 704-bp DNA fragment was observed by gel electrophoresis. The total completion of this assay requires less than one day.
