Effects of Dietary Octacosanol on Growth Performance, Carcass Characteristics and Meat Quality of Broiler Chicks

  • Octacosanol, which has prominent physiological activities and functions, has been recognized as a potential growth promoter in animals. A total of 392 1-d-old male Arbor Acres broiler chicks with similar body weight were randomly distributed into four dietary groups of seven replicates with 14 birds each supplemented with 0, 12, 24, or 36 mg octacosanol (extracted from rice bran, purity >92%)/kg feed. The feeding trial lasted for six weeks and was divided into the starter (day 1 to 21) and the grower (day 22 to 42) phases. The results showed that the feed conversion ratio (FCR) was significantly improved in broilers fed a diet containing 24 mg/kg octacosanol compared with those fed the control diet in the overall phase (day 1 to 42, p = 0.042). The average daily gain and FCR both showed linear effects in response to dietary supplementation of octacosanol during the overall phase (p = 0.031 and 0.018, respectively). Broilers fed with 24 or 36 mg/kg octacosanol diet showed a higher eviscerated yield, which increased by 5.88% and 4.26% respectively, than those fed the control diet (p = 0.030). The breast muscle yield of broilers fed with 24 mg/kg octacosanol diet increased significantly by 12.15% compared with those fed the control diet (p = 0.047). Eviscerated and breast muscle yield increased linearly with the increase in dietary octacosanol supplementation (p = 0.013 and 0.021, respectively). Broilers fed with 24 or 36 mg/kg octacosanol diet had a greater (p = 0.021) $pH_{45min}$ value in the breast muscle, which was maintained linearly in response to dietary octacosanol supplementation (p = 0.003). There was a significant decrease (p = 0.007) in drip loss value between the octacosanol-added and the control groups. The drip loss showed linear (p = 0.004) and quadratic (p = 0.041) responses with dietary supplementation of octacosanol. These studies indicate that octacosanol is a potentially effective and safe feed additive which may improve feed efficiency and meat quality, and increase eviscerated and breast muscle yield, in broiler chicks. Dietary supplementation of octacosanol at 24 mg/kg diet is regarded as the recommended dosage in the broilers' diet.

  • The purpose of this study was to examine the effects of octacosanol and Taekwondo program on the fatigue substance, immunoglobulin after short term weight loss 21 male Taekwondo players. They were stay in a camp training together 6 days from same exercise and Octacosanol group and weight loss group is reduced over 5% weight loss by diet calorie restriction decrease gradually. Octacosanol intaked 2 times 20 mg after a meal. Control and weight loss group intaked placebo drug. The conclusions obtained from the above were as follow: Blood Fatigue Substance As lactate, The intra-group comparison decreased octacosanol group after loss weight. The between groups comparison, octacosanol group was superior to control and weight loss group. As lactate dehydrogenase, control and weight loss group increased after weight loss. As ammonia, The intra- group comparison decreased octacosanol group after loss weight. The between groups comparison, octacosanol group was superior to control and weight loss group. Immunoglobulin As IgA, IgG The intra-group comparison increased all group. As IgM, The were no significant differences between group. In conclusion, these results indicate that intake octacosanol after loss weight positive effects on the fatigue substance, Immunoglobulin for male Taekwondo players. Mter this study, We need to study about multilateral research for weight players.

Effect on Endurance Improving of Mixture of the Extracts of Watermelon and Octacosanol (Octacosanol과 박과식물 추출물을 주성분으로 하는 혼합물의 지구력 증진 효과)

  • Octacosanol is known to enhance endurance activities, control cholesterol in body and improve the function of cardiopulmonary. Citrulline, which is main compound of watermelon, is known to improve angiectasia through stimulating production of nitric oxide. To improve endurance activity swimming test on rats was carried out using four samples such as 1$\%$ octacosanol, citrulline, the extracts of barks of watermelon and products, mixture of 1$\%$ octacosanol and the extracts of barks of watermelon (6 : 4). Biochemical assays on the liver and serum of tested rats were also performed using commercial analysis kits. In result, it was shown that swimming time of III group increased by 26$\%$ and that of V group was increased by 22$\%$ at the swimming test. As a result of biological assays on the liver and serum of tested rats it was possible to confirm stability of toxicity When compared with creatine kinase of control group (549.11$\pm$39.15 U/l) citrulline (644.11 $\pm$50.67 U/l) and products group (646.00$\pm$46.99 U/l) were largely increased. When compared with inorganic phosphate of control group (12.01$\pm$0.75 mg/이), citrulline (13.03$\pm$0.94 mg/dl) and products group (12.90$\pm$0.55 mg/dl) showed similar results. Also, when compared with lactic acid and glucose of control group (152.91 $\pm$ 13.45, 103.00$\pm$ 8.69 mg/dl), citrulline (125.53$\pm$15.54, 83.75$\pm$7.29 mg/dl) and products group (135.26$\pm$11.50, 78.57$\pm$9.79 mg/dl) were largely decreased. As these test results, it was determined that 1$\%$ octacosanol and extracts of barks of watermelon had some effect of improving endurance activity. Futhermore, it was thought that it could be used as source of functional food.

Effects of Octacosanol Extracted from Rice Bran on the Laying Performance, Egg Quality and Blood Metabolites of Laying Hens

  • A 42-d study with 384 Hy-line brown laying hens was conducted to assess the effects of dietary octacosanol supplementation on laying performance, egg quality and blood metabolites of laying hens. Hens were randomly allocated into 4 dietary groups of 8 cages each, which were fed basal diet supplemented with 0 (Control), 9 (OCT9), 18 (OCT18), and 27 (OCT27) mg/kg diet of octacosanol isolated from rice bran, respectively. The experiment was conducted in an environmental controlled house and hens were fed twice daily for ad libitum intake. Laying performance was determined over the 42-d period, and egg quality as well as blood metabolites were estimated on d 21 and d 42. Diets in OCT18 and OCT27 increased (p<0.05) laying rate, egg weight, egg mass, egg albumen height, Haugh unit and eggshell strength on d 42, but decreased (p<0.05) feed conversion rate and levels of total cholesterol, triglyceride and low density lipoprotein cholesterol in the serum as compared to those of Control. Feed intake, yolk color, yolk diameter, eggshell thickness and high density lipoprotein cholesterol were similar (p>0.05) among treatments. Results demonstrate that supplementing 18 to 27 mg/kg diet of rice bran octacosanol can improve laying rate and egg quality and reduce blood lipid of laying hens.

  • This study was conducted to investigate the effect of dietary supplementation of octacosanol (OCT) extracted from triticale sprout on laying performance, egg quality, and blood parameters of laying hens. A total of 192, Hyline brown laying hens aged 43 weeks were divided into 4 dietary groups of 48 birds each and they were randomly subjected to one of the experimental diets containing OCT at the levels of none, 10, 20, and 30 mg/kg of diet. All birds were fed with isoenergetic and isonitrogenous mash diets for 6 weeks. The result showed that hens supplemented with 20 and 30 mg/kg OCT in diet significantly increased (p < 0.05) egg production than those fed with the basal diet. OCT concentration in the egg yolk of hens fed with 20 and 30 mg/kg OCT was significantly higher than in those fed the control diet. Hens fed 20 and 30 mg/kg OCT exhibited greater high-density lipoprotein (HDL) cholesterol and interleukin (IL) concentrations and reduced serum concentrations of cholesterol and triglyceride compared to those fed with 0 and 10 mg/kg OCT. This study indicates that supplementing the diet of laying hens with 20 and 30 mg/kg of OCT can improve the performance, egg quality, and health status of laying hens.

  • The Ministry of Food and Drug Safety (MFDS) is amending its test methods for health-functional foods (dietary food supplements) to establish regulatory standards and specifications in Korea. In this regard, we continue our research on developing analytical methods for the items. Octacosanol is the major component of polycosanol and is a high-molecular-mass primary fatty alcohol, obtained from sugar cane wax. Previous researchers have shown that octacosanol can lower cholesterol and has antiaggregatory properties, cytoprotective uses, and ergogenic properties for human health. Recently, octacosanol products have been actively introduced into the domestic market because of their functional biological activity. We have developed a sensitive and selective test method for octacosanol that the TMS derivatives by means of gas-chromatographic-tandem mass spectrometry (GC-MS). The trimethylsilyl ether derivative of the target analyte showed excellent chromatographic properties. The procedure was validated in the range of $12.5{\sim}200{\mu}g/L$. Standard calibration curves presented linearity with the correlation coefficient ($r^2$) > 0.999, and the limits of detection (LOD) and limits of quantitation (LOQ) were $4.5{\mu}g/L$ and $13.8{\mu}g/L$, respectively. The high recoveries (92.5 to 108.8%) and precision (1.8 to 2.4%) obtained are in accordance with the established validation criteria. Our research can provide scientific evidence to amend the octacosanol test method for the Health-Functional Food Code.

  • The methanol extract of Euphorbia milii exhibited strong inhibitory effect on platelet aggregation in the cource of our search for anti-platelet component from succulent plants. Two components, components 1 and 2 were isolated from this plant. 1 was the mixture of 72% of 1-octacosanol (1a) and 28% of 1-triacontanol (1b), and 2 was identified as ${\beta}-sitosterol$. 2 ($IC_{50}$: $195\;{\mu}M$, and $170\;{\mu}M$ respectively) was about two fold stronger than ASA ($IC_{50}$: $420\;{\mu}M$ and $340\;{\mu}M$ respectively) on both collagen and U46619 induced aggregation, while the effect of 1 to platelets was negligible.

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.

  • Besides chrysophanol and friedelin, mixtures of n-hydrocarbons [pentacosane (72.6%), heptacosane (14.6%), tetracosane (5.8%), nonacosane (4.1 %) and hexacosane (2.9%)], n-hydrocarbon alcohols [octacosanol (70.5%) and hexacosanol (29.5%)], l-monoacyl glycerols [acyl part; behenic acid (43.5%), lignoceric acid (32.4%), cerotic acid (9.3%), tricosanoic acid (8.9%), pentacosanoic acid (2.6%), octacosanoic acid (2.3%), heneicosanoic acid (1.0%)], wax esters [behenic acid (56.3%), lignoceric acid (23.0%), cerotic acid (19.8%), tricosanoic acid (4.6%), octacosanoic acid (4.0%), pentacosanoic acid (1.7%), triacontanoic acid (0.6%)/ octacosanol (33.7%), hexacosanol (21.0%), tetracosanol (15.6%), triacontanol (10.5%), docosanol (6.0%), tricosanol (6.0%), heptacosanol (4.2%), nonacosanol (3.0%)] and sterols [${\beta}-sitosterol$ (73.2%), stigmasterol (14.6%), campesterol (12.2%)] were isolated from the roots of Hemerocallis fulva. The acid : alcohol combinations of the major wax esters were $C_{48}$: 22 : 26, 24 : 24, $C_{46}$: 22 : 24, and $C_{44}$: 22 : 22. This is the first report of l-monoacyl glycerols and friedelin from this plant. All isolates were identified on the basis of spectral data and chemical reactions.

  • A new 3-indolyl propionate alkaloid; (-) abrine (1), was isolated from the alcohol extract of Erythrina caffra stem bark together with the alkaloid (-) hypaphorine (2). Phytochemical investigation of the chloroform extract led to the isolation of one fatty alcohol; 1-octacosanol, isolated for the first time from genus Erythrina, two dienoid alkaloids; erythraline and erysodine, and three isoflavonoids; 3S (+) 2'-O-methylphaseollidinisoflavan (3), (6aR, 11aR)- 3-hydroxy-10-dimethylallyl-9-methoxypterocarpan, known as sandwicensin (4) and 3R (-) rythbidin A (5). It is worth mentioning that this is the first report for the isolation of these isoflavonoids from E. caffra. The absolute configuration of 3S (+) 2'-O-methylphaseollidinisoflavan (3) was not previously reported. The isolated isoflavonoids showed promising antibacterial activity against Staphylococcus aureus.
