말똥성게 (Hemicentrotus pulcherrimus)의 수정 및 배 발생률을 이용한 해양배출 폐기물의 독성평가 (Toxicity Assessment of Ocean Dumping Wastes Using Fertilization and Embryo Development Rates in the Sea Urchin (Hemicentrotus pulcherrimus))

  • Toxicity assessment of ocean dumping wastes (dye waste, urban sewage, food waste) were examined in the fertilization and embryo development rates of the Sea Urchin, Hemicentrotus pulcherrimus. Spawning was induced by injecting 1 mL of 0.5 M KCl into coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. Experiments were began within 30 min the collection of both gametes. The fertilization and embryo development rates test were performed for 10 min and 64 h after fertilization, respectively. The fertilization and embryo development rates in the control condition (not including ocean dumping wastes sludge elutriate) were greater than 90%, but suddenly decreased with increasing of ocean dumping waste sludge elutriate concentrations. The fertilization and normal embryogenesis rates were significantly inhibited in all waste sludge elutriate from dye waste ($EC_{50}$=4.37; $EC_{50}$=1.76), urban sewage ($EC_{50}$=5.79; $EC_{50}$=2.00) and food waste ($EC_{50}$=7.68; $EC_{50}$=2.16), respectively. The NOEC (<3.13) and LOEC (3.13) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest that biological assay using the fertilization and embryo development rates of H. pulcherrimus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.

The NOEC(>3.13%) and LOEC(3.13%) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest biological assay using the fertilization and embryo development rates of S. nudus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.

Optimal Conditions for Artificial Fertilization, Embryonic Development, and Larval Growth of the Purple Clam, Saxidomus purpuratus from Southern Coast of Korea

  • To obtain the basic information on culture conditions for the larvae of Saxidomus purpuratus, experiments were conducted on the population from southern coast for (1) the success in fertilization and development from artificial fertilization among different months of a year, (2) the viability of sperms after exposure to seawater, (3) and the effects of temperature, salinity, and food organism on the survival and growth of larvae. Gametes obtained from dissection showed high rate of fertilization at all months. But the rate of development was higher only May-July. Developmental success seemed to be related with the quality of eggs at the time of fertilization. Developmental times for 2-cell, 4-cell, 8-cell, blastula, trochophore larva, and veliger larva at 20$^{\circ}C$ were 1.5, 2, 4, 18, 24, and 32 hr, respectively. Sperms could survive for more than 8 hr, however, actively swimming sperms could be found within 1 hr after exposure to seawater. It is recommended that sperms should be used for fertilization as soon as possible when they are exposed to seawater. At temperature of 35$^{\circ}C$, all the larvae died during 48 hr. Larval survival decreased when salinity was either lower than 20 psu or higher than 40 psu, and was 0% when salinity was 10 psu. Optimal range of temperature and salinity for rearing larvae of S. purpuratus were 20-25$^{\circ}C$ and 20-40 psu, respectively. Larvae grew from 111.5 to 235.3 ${\mu}$m during 21 days. Larvae fed mixed diets grew faster than unialgal diets. The fastest growth was observed when larvae were fed on the mixture of Isochrysis galbana and Nannochloris oculata.

Ecological and Biogeochemical Response of Antarctic Ecosystems to Iron Fertilization and Implications on Global Carbon Cycle

  • The European Iron Fertilization Experiment EIFEX studied the growth and decline of a phytoplankton bloom stimulated by fertilising $10km^2$ in the core of a mesoscale $(80{\times}120km)$ cyclonic eddy south of the Antarctic Polar Front with about 2 times 7 tonnes of iron sulphate. The phytoplankton accumulation induced by iron fertilization did not exceed $3{\mu}g\;chl\;a\;l^{-1}$ despite a draw down of $5{\mu}M$ of nitrate that should have resulted in at least double to triple the amount of phytoplankton biomass assuming regular Redfield-ratios for draw down after phytoplankton growth in the Southern Ocean. During EIFEX the fertilized core of the mesoscale eddy evolved to a hotspot for a variety of small and medium sized mesozooplankton copepods. In contrast to copepods, the biomass of salps (Salpa thompson)) that dominated zooplankton biomass before the onset of our experiment decreased to nearly extinction. Most of the species of the rnosozooplankton community showed extremely hiか feeding rates compared to literature values from Southern Ocean summer communities. At the end of the experiment, massive phytoplankton sedimentation reached the sea floor at about 3800m water depth.

진주배말(Cellana grata)의 수정 및 후기 피면자 유생의 착저단계에 미치는 저염분 영향 (Low Salinity Effects on the Fertilization and Settlement of Post Veliger Larvae in the Limpet Cellana grata)

  • The effects of low salinity (fertilization success and larval survival) on the limpet Cellana grata were studied at early stages of development using the marine bioassay technique. It was shown that, under normal conditions for development from fertilization to the post veliger stage, the salinity must be not less than 20.0~35.0 psu. However, the fertilization rate and larval survival of C. grata was obviously reduced at 5.0 psu and 10.0 psu, respectively. Mass mortality was estimated to occur at <20.0 psu (48-h $EC_{50}=19.54psu$) and the survival rate of normal veliger larvae decreased with experimental time during exposure. No observed effective concentration (NOEC) and lowest observed effect concentration (LOEC) of post veliger were estimated at 30.0 psu and 25.0 psu, respectively, during 48-h exposure. The tolerance limits of the test species to salinity revealed various concentration ranges of salinity, which may reflect the physiology and ecology of the initial development stages of C. grata. These results demonstrate that reduced salinity is detrimental to the reproductive success and larval survival of C. grata, and if salinity is lowered by natural or anthropogenic sources during spawning, this would lead to decreased reproductive success and larval settlement.

해양 철분 시비(施肥)사업의 국제 관리체제 예비 분석 (A Preliminary Analysis on the International Management System for the Ocean fertilization with Iron at High Seas)

  • 우리인류는 생존과 번영을 위해 상당량의 에너지를 화석연료를 사용하는 연소공정에 의존하고 있다. 이 공정의 주된 폐기물은 이산화탄소로서 대기로 주로 배출한다. 특히 산업혁명 이후, 급격한 인구증가와 번영은 화석연료의 사용의 증대로 가능하였다. 그러나 이로 인하여 대기 중 이산화탄소함량은 급속히 증가하였고, 그 결과 지구 온난화가 가속되고 또 해양표면이 산성화되는 등 환경에 대한 부정적 영향이 심화되고 있다. 이러한 문제를 해결하기 위한 국제적 노력의 일환으로 1990년대에 유엔기후변화협약과 교토의정서 등의 다자간 환경협정이 체결되었고, 또한 대기 중 이산화탄소의 감축을 위한 다양한 과학 기술적 방안이 개발되거나 검토 되기 시작하였다. 이와 관련하여, 몇몇 기업들이 탄소 거래 시장에 진입하기 위해 해양 철분 시비 사업을 추진하고 있다. 해양 철분 시비 사업이란, 농경지에 비료를 살포하는 것과 마찬가지로, 철분이란 영양분이 결핍된 해양에 인위적으로 철분을 첨가함으로써 식물성 플랑크톤을 대규모로 번식시키고, 이를 통해 대기 중 이산화탄소를 감축하여 획득한 탄소 크레디트를 판매하려는 것이다. 이러한 해양 시비의 잠재적 해양환경영향과 관리방안에 대해 2007년부터 폐기물 및 기타 물질의 투기로 인한 해양오염방지에 관한 국제협약에서 논의하기 시작되었다. 본 소고는 이러한 시비 사업의 동향을 살펴보고 당해 활동에 대한 국제 관리체제의 발전을 모색하기 위하여 해양 시비의 원리 및 관련 기업의 사업 추진 현황을 검토하고, UN 해양법협약, 런던협약 및 의정서 등 관련 국제법적 체제를 영해, 배타적 경제수역, 공해 별로 고찰하였다. 국제사회는 기존의 런던협약/의정서를 중심으로 하고 UN총회 산하의 유엔 해양 및 해양법협약 비공식 공개협의체를 통해 해양철분시비사업을 관리할 것으로 전망된다.

해양심층수에 의한 해역 비옥화 효과에 관한 기초 연구 (A Fundamental Study on the Effect of Ocean Fertilization by Deep Sea Water)

  • 본 연구에서 식물플랑크톤 뿐 아니라 동물플랑크톤을 포함하는 생태계 모델을 이용하여 해양 심층수의 해역 비옥화의 효과를 검증하였다. 모델은 2종의 식물플랑크톤, 3종의 동물플랑크톤, 질산염, 암모늄염, 규산염, 입자성 유기질소, 용존 유기질소, 입자성 규소 총 11개 요소로 구성된 NEMURO에 기초하고 있다. NEMURO의 11개 요소 외에도 두 종의 식물플랑크톤내 질소 셀 쿼터와 대형 식물플랑크톤내 규산염 셀 쿼터를 추가하였다. 해역 비옥화 효과를 조사하기 위해서 이즈 오시마 섬에서 실험을 실시하였지만, 본 실험에서 심층수의 유무에 의한 명확한 차이를 확인할 수 없었다. 이 실험 결과의 원인들을 모델 시뮬레이션에 의해 조사하였으며, 그 원인의 하나는 실험에 사용된 표층수의 높은 영양염 농도이다. 다른 원인은 무기질소 섭취율이 $NH_4$에 대한 $NO_3$의 비율에 관련되어 있으므로 무기질소의 총 농도의 증가가 광합성 속도를 반드시 증진시키지 않는다는 것이다. 이 모델로 실험 결과들을 재현할 수 있었기 때문에 우리는 이 모델을 이용하여 해양 심층수에 의한 해역 비옥화의 효과를 검증하였다. 해양 심층수의 공급 간격이 너무 짧거나 해양 심층수의 양이 너무 많을 때 해역 비옥화 효과는 나타나기 어려운 것으로 밝혀졌다. 해양 심층수의 첨가가 너무 비번하거나 너무 많이 첨가하면 플랑크톤 농도의 희석이 식물플랑크톤의 광합성 증진 효과를 초과해 버린 것으로 추측된다.

Embryonic Development and Hatchability in the Reciprocal Hybrids of Kelp Grouper (Epinephelus bruneus) and Red-Spotted Grouper (Epinephelus akaara)

  • In this study, the embryonic development and hatchability of eggs fertilized by the reciprocal hybrids of kelp grouper (Epinephelus bruneus) and red-spotted grouper (E. akaara) were evaluated with the goal of establishing a novel hybrid with enhanced growth and viability during the farming period in the temperate waters of Korea. The fertilization rates were lower for hybrids than for maternal purebreds and were significantly higher in the red-spotted grouper ♀ × kelp grouper ♂ hybrid (RGKG, 89.61 ± 1.58%) than in the kelp grouper ♀ × red-spotted grouper ♂ hybrid (KGRG, 74.82 ± 4.23%, p < 0.05). Unlike the fertilization rates, the hatching rates of fertilized eggs were similar between hybrids and maternal purebreds and did not differ significantly between KGRG and RGKG (72.74 ± 3.60% vs. 75.23 ± 2.20%, respectively, p > 0.05). The embryonic development of the hybrids was similar to that of maternal purebreds; however, irregular cleavage and asymmetric blastoderm were noticeable in the developing eggs of KGRG hybrids. The deformity rates of newly hatched larvae were higher in hybrids than in maternal purebreds and were significantly higher in KGRG than in RGKG (17.47 ± 1.28% vs. 7.11 ± 0.54%, respectively, p < 0.05). These results demonstrate the potential to produce viable larvae from these two hybrids. Although the production efficiency of KGRG was lower than that of RGKG, the fertilization, hatching, and deformity rates make both hybrids useful for further comparative studies regarding economic aspects.

그물에 부착한 청어 (Clupea pallasii) 어란의 자연 부화유도 및 관리방안 (Natural hatching-induced and management for Pacific herring Clupea pallasii eggs attached to the gill net)

  • Pacific herring Clupea pallasii eggs were attached to the gill net in Korean coastal waters off Busan. To conservation and management the eggs attached to the gill net, we followed the natural hatching in coastal waters from five days after fertilization to the hatching finished, where the temperature was $9^{\circ}C$. The diameter of fertilized eggs was 1.24-1.55 mm (n= 50), and had a segmented pale yellow yolk, no oil globule. Natural hatching had started by 10 days after fertilization. Mass hatching occurred around 11-12 days after fertilization, most of eggs separated from the gill net. Therefore, when the Pacific herring eggs attached in gill net, should be natural hatching-induced in coastal waters during 12-14 days.

Transient Expression of Transgene Introduced by Lipofected Sperm in Olive Flounder(Paralichthys olivaceus)

  • The successful gene transfer and transient expression was demonstrated in olive flounder embryos using lipofected sperm. Olive flounder sperm interacted with foregn plasmid DNA encapsidated by positively charged liposome. The maximum plasmid copy number that associated with the sperm was 5 copies/sperm based on the examination of DNA blot assay. The foreign DNA was transferred into fertilized eggs without any adverse effect on fertilization and survival of embryos (P>0.05) and retained in embryos until at least 42 hours with successful expression. The maximal expression was detected in 18 hours after fertilization at 18$^{\cird}C$ and gradually decreased with development of embryo. Most of DNA transferred into embryos persisted extrachromosomally without significant sign of integration or replication.
