Isolation and Identification of Lactic Acid Bacteria Isolated from a Traditional Jeotgal Product in Korea

  • Seventeen lactic acid bacterial strains (LAB) were isolated using MRS agar medium from Jeotgal, a Korean fermented food, purchased at the Jukdo market of Pohang. To identify the strains isolated, they were tested by examining their cell morphologies, gram-staining, catalase activity, arginine hydrolase activity, D-L lactate form and carbohydrate fermentation. According to the phenotypic characteristics, three strains were tentatively identified as Lactobacillus spp., ten were Enterococcus spp. (or Streptococcus spp., or Pediococcus spp.) and the rest were Leuconostoc spp. (or Weissella spp.). Five strains among 17 were chosen by preliminary bacteriocin activity test. Four bacterial strains which inhibited both indicator microorganisms were identified by 16S rRNA sequencing. The results are as follows; Leuconostoc mesenteroides (HK 4), Leuconostoc mesenteroides (HK 5), Leuconostoc mesenteroides(HK 11), Streptococcus salivarius(HK 8). In order to check LAB which are showing a high survival rate in gut, we investigated three strains inhibiting both indicator microorganisms in artificial gastric acid and bile juice -all except HK8. The three strains mentioned above grew in extreme low acid conditions.

  • Olive flounder and Black rockfish comprises around 90% of the total fish culture in Korea. Most of the olive flounder is consumed as raw fish meat “saengseonhoe”, as like a sashimi in Japan. Also, texture is particularly a important factor which determine the quality of raw fish meat in Korea. Many works have attempted to improve of texture such as arai, electrical stimulation and compulsory exercise of fishes. (omitted)

  • Mushroom-based vegan meat has thus far been used as a food for humans instead of pets. However, based on its texture and nutritional content, it is considered suitable for processing into pet treats. In the present study, we developed a prototype dog chew with a sweetening coating added to a fungal mycelium mat obtained by culturing the Basidiomycetous fungus Trametes orientalis. The palatable coating applied to the mycelium mat by plasticizing the mat with glycerol improved the taste and aroma of the existing mat, and the dog consumed it without difficulty. Future improvements may include a softening process to reduce the chewiness level and a procedure to reduce the crude fiber content. Mycelium-mat-based dog chews, manufactured using eco-friendly materials and processes that are not harmful to the environment are expected to enter the market as eco-friendly alternatives to conventional pet treats. Controlling their physical properties require further study.

A review on the Marine Healing Industry (해양치유 산업에 관한 고찰)

  • This study aims to understand marine therapy through an overall review of the marine therapy industry and provide basic data for policy establishment on marine therapy based on this. Talasotherapy refers to the medical use of marine climate, seawater, marine mud, algae, sand and other marine extracts, which are beneficial components of the ocean for preventive and therapeutic purposes. Centers in Taean, Goseong, Uljin, and other marine healing pilot projects should be well combined with programs within rehabilitation clinics so that local tourism and marine healing programs can be combined, and how well the existing marine infrastructure is established is a very important factor. Establish legal and institutional foundations, inter-ministerial and central-local cooperative governance by reorganizing laws and regulations related to marine healing and establishing a cooperative system between ministries and regions. It is necessary to expand the function of the marine therapy R&D project to play a role as a marine therapy center for each coastal region.

  • Salted anchovy, generally favored in Europe, is one of the traditional fermented fish products in southern European countries and raw anchovy are aged in 20∼30% NaCl solution for 6 months (Ishida et al., 1994). They are then steeped in oil in glass jar or can, and marketed. The salt-fermented fish sauce, such as anchovy sauce and shrimp sauce are tropical fermented seafood from fish in Korea. (omitted)

Evaluation of Optimal Performance of Hydraulic Barriers in Offshore Landfill using Seepage-Advection-Dispersion Analysis under Steady State Flow (정상류하 침투·이류 분산 해석을 이용한 폐기물 해상최종처리장 차수시스템의 최적 성능 평가)

  • This study was conducted to propose the optimum minimum requirement of cutoff system composed of the impermeable soil layer and vertical barrier in offshore landfill for prevention pollution leakage by seepage, advection, and dispersion numerical analyses under steady state. According to the study results, the minimum requirement of impermeable soil layer is below $1{\times}10^{-6}cm/s$ of hydraulic conductivity with more than 500 cm thickness or a system with equivalent cutoff effect. The minimum requirement of vertical barrier is below $1{\times}10^{-6}cm/s$ of hydraulic conductivity with more than 50 cm thickness or a system with equivalent cutoff effect. In addition, the vertical barrier should be embedded enough to seal securely with the impermeable soil layer for working cutoff effect.

Spatial Variability and Contents of Metals in the Surficial Sediments of Youngil Bay, East Coast of Korea (한국 동해안 영일만 표층 퇴적물의 금속 함량과 공간 변화 특성)

  • Bottom sediments from Youngil Bay, East Coast of Korea, were analyzed for grain composition as well as elemental compositions and total organic carbon (TOC) content in order to investigate the spatial variability and content of metal elements. Grain size distribution of the sediments seems to be controlled by anticlockwise current pattern with bottom topography of the study area. Spatial variability of TOC and all elemental contents reflects those of grain size, but an exception was found in the harbor area (Old-Port): their contents are high in the central part of the bay with the muddy sediment and decrease toward the sand-dominated coastal zone. However, contents of Ca, Sr, K are high in the sand-dominated coastal zone and contents of some heavy metals (Cd, Cu, Zn) are high in the Old-Port area and the mouth of Hyeongsan River. The correlation matrix and R-mode factor analyses reveal that four important factors controlling the distribution of metals in the bay are sediment grain size (or quartz dilution effect), the formation of sulfide minerals associated with decomposition of organic matters under anoxic geochemical environment, calcium carbonate (mainly shell fragments) and coarse-grained feldspar mineral. According to the metal content of labile fraction an CER (concentration enrichment ratio) value, high accumulation of some heavy metals in the harbor area seems to result not formed by early diagenetic processes under anoxic environment.

  • To monitor and predict the change of coastal environment according to the construction of Saemangeum sea dyke and the development of land reclamation, we have done real-time and periodic ocean observation and numerical simulation since 2002. Saemangeum coastal environmental data can be largely classified to marine meteorology, ocean physics and circulation, water quality, marine geology and marine ecosystem and each part of data has been generated continuously and accumulated over about 10 years. The collected coastal environmental data are huge amounts of heterogeneous dataset and have some characteristics of multi-dimension, multivariate and spatio-temporal distribution. Thus the implementation of information system possible to data collection, processing, management and service is necessary. In this study, through the implementation of Saemangeum coastal environmental information system using geographic information system, it enables the integral data collection and management and the data querying and analysis of enormous and high-complexity data through the design of intuitive and effective web user interface and scientific data visualization using statistical graphs and thematic cartography. Furthermore, through the quantitative analysis of trend changed over long-term by the geo-spatial analysis with geo- processing, it's being used as a tool for provide a scientific basis for sustainable development and decision support in Saemangeum coast. Moreover, for the effective web-based information service, multi-level map cache, multi-layer architecture and geospatial database were implemented together.

The Influence of Water Temperature and Body Weight on Metabolic Rate of Olive Flounder Paralichthys olivaceus (넙치 Paralichthys olivaceus 대사율에 미치는 수온과 체중의 영향)

  • The effect of water temperature and body weight on oxygen consumption by the fasted olive flounder Paralichthys olivaceus was investigated in order to assess the metabolic rate of this species under different conditions. The oxygen consumption rate (OCR) was measured at three different water temperatures (15, 20 and $25^{\circ}C$) and two different body weights [$9.1{\pm}1.2$ g (mean${\pm}$SD) for the juvenile group and $266.4{\pm}29.3$ g for the immature group] at an interval of 5 minutes for 24 hours using a closed flow-through respirometer. For each treatment condition, three replicates were set up and 135 fish in the juvenile group and 18 fish in the immature group were used. The OCRs exhibited a linear increase described by OCR=-82.06+28.30T ($r^2$=0.96, p<0.001) in the juvenile group and OCR=-52.52+14.73T ($r^2$=0.97, p<0.001) in the immature group. The OCRs decreased with increasing body weights at a given water temperature (p<0.001). The metabolic rate was related to the body weight of the fish as a power function with a weight exponent of between 0.77 and 0.82. $Q_{10}$ values ranged 1.67~2.28 when the temperature was between 15 and $20^{\circ}C$, 1.57~1.93 when the temperature was between 20 and $250^{\circ}C$, and 1.79~1.89 when the temperature was between 15 and $250^{\circ}C$. The energy expenditure by respiration increased with increasing water temperature and decreasing body weight (p<0.001). The mean energy loss rates at 15, 20 and $25^{\circ}C$ were 115.9, 149.8 and 208.2 kJ $kg^{-1}d^{-1}$ in the juvenile groups and 53.8, 81.2 and 101.9 kJ $kg^{-1}d^{-1}$ in the immature groups.

  • Voltammetric detection of the toxic Zn ion was investigated using a fluorine-doped graphite pencil electrode (FPE). It is notable from the study that pencils were used as reference and working electrodes. In all the experiments, a clean seawater electrolyte solution was used to yield good results. The analytical working range was attained to 10 ${\mu}gL^{-1}$. The optimized voltammetric condition was examined to maximize the effect of the detection of trace Zn. The developed sensor was applied to an earthworm's tissue cell. It was found that the methods can be applicable to in vivo fluid or agriculture soil and plant science.
