The Experimental Study on Anti-inflammatory Effects of Eungapbang (EGB) (은갑방(銀甲方)이 염증 관련 cytokines의 유전자 발현과 생성량에 미치는 영향)

  • Purpose: This study was performed to evaluate the anti-inflammatory effect of Eungapbang extract (EGB). Methods: To evaluate the anti-inflammatory effects of EGB, we nourished RAW 264.7 cell lines in the laboratory dish. Next, inflammatory cytokine concentrations were analyzed. Then, sera were prepared from blood after lipopolysaccharide (LPS) injection in chemically induced mouse models of intestinal inflammation, and Interleukin-1${\beta}$ (IL-1${\beta}$), interleukin-6 (IL-6) and tumour necrosis factor alpha (TNF-${\alpha}$) were measured using ELISA kits. Results: 1. EGB significantly suppressed the expression levels of IL-1${\beta}$ and NOS-II genes at 100, 50 and 10 ${\mu}g/m{\ell}$ concentrations, and IL-6, TNF-${\alpha}$ and COX-2 mRNAs at 100 and 50 ${\mu}g/m{\ell}$ concentrations. 2. EGB significantly reduced the production level of IL-1${\beta}$ and TNF-${\alpha}$ at 100${\mu}g/m{\ell}$ concentrations, and IL-6 at 100 and 50 ${\mu}g/m{\ell}$ concentrations. 3. EGB significantly decreased the production level of IL-1${\beta}$ and IL-6 in sera of acute inflammation induced mice. 4. EGB could suppress the expression level of IL-1${\beta}$ and IL-6 mRNA in spleen tissues in acute inflammation induced mice. Conclusion: On the basis of the above results, it is confirmed that the anti-inflammatory effects of EGB were recognized. Therefore, EGB is recommended as promising therapy for treatment of such ailments as pelvic inflammatory disease.

Safety and Antioxydative effects of Cuscuta chinensis Lam. in PC12 Cell (PC12 Cell에 대한 토사자(?絲子)의 안정성 및 항산화작용에 대한 연구)

  • Purpose : Cuscuta chinensis Lam. is utilized extensively as important medicines for threatened abortion habitual abortion. However, objective data related to an embryo is not existed until now. Therefore, this study is focused to find out stability of Cuscuta chinensis Lam. about an embryo during pregnancy based on data related to stability of nerve cell and antioxydative effect by using neural cell line, PC 12 cell. Methods : Experimentation concerns cytotoxic effects and antioxydative effect through methods such as MTT ssay, western botting after abstracting an undiluted solution from domestic Cuscuta chinensis Lam. Results : 1. As a result of experimentation on MTT assay according to each magnification from Cuscuta chinensis Lam. extraction solution with different abstraction methods, cytotoxic effect is not observed to all extract except an undiluted solution which is abstracted from MeOH stiring. Also, an undiluted solution in stiring with MeOH could not confirm whether come from Cuscuta chinensis Lam. or not. 2. As a result of revelation of Bax and GSK-3${\beta}$ which is responsive to the first stage from general stress in order to observe antioxydative effects of Cuscuta chinensis Lam. revelation of Bax by Cuscuta chinensis Lam. appeared to decrease. Conclusion : Cytotoxic effects with Cuscuta chinensis Lam. about PC12 cell is not discovered and it assume that it would be anti apoptotic effect by ROS as Bax and GSK-3${\beta}$ inviable effect. In the future, this study could be used as basic data for additional research on Cuscuta chinensis Lam. and effect and stability of complicated prescriptions for keeping pregnancy.

The Effect of Yukmijihwangtang -gagambang (YMG) on Melanin Synthesis and Gene Expression (육미지황탕가감방(六味地黃湯加減方)이 멜라닌 생성 및 유전자발현에 미치는 영향)

  • Purpose: This study was performed to elucidate the inhibitory effect of Yukmijihwangtang-gagambang (YMG) on melanin synthesis in B16F10 mouse melanoma cell. Methods: To demonstrate the inhibitory effects of YMG on melanin synthesis, we measured the amount of released and produced melanin in B16F10 melanoma cell. Also, we evaluated tyrosinase-activity in vitro as well as in B16F10 melanoma cell. And to investigate the action mechanism we assessed the gene expressions of tyrosinase, TRP-1, TRP-2, MMP-2, PKA, PKC${\beta}$, ERK-1 ERK-2, AKT-1 and MITF in B16F10 melanoma cells. Results: 1. YMG decreased the release and production of melanin in B16F10 melanoma cells. 2. YMG decreased tyrosinase activity in vitro and in B16F10 melanoma cells. 3. YMG decreased the expression of tyrosinase, TRP-1, TRP-2, PKA, PKC${\beta}$ and MMP-2 in B16F10 melanoma cells. 4. YMG increased the expression of ERK-1, ERK-2, and AKT-1 in B16F10 melanoma cells. 5. YMG decreased the expression of MITF in B16F10 melanoma cells. Conclusion: From these results, we suggest that YMG inhibit melanin synthesis via tyrosinase inhibition and regulation of the gene expression in B16F10 melanoma cells.

The Experimental Study on Anti-thrombotic and Anti-inflammatory Effect of BokbangHongdeungPaejangSan (BHPS) (복방홍등패장산(復方紅藤敗醬散)의 항혈전(抗血栓) 및 항염작용(抗炎作用)에 대한 실험적(實驗的) 연구(硏究))

  • Purpose : This study was performed to evaluate anti-thrombotic and anti-inflammatory effects of BokbangHongdeungPaejangSan water extract (BHPS). Methods : BHPS was investigated using cultured cells and a murine models. As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were determined in mouse lung fibroblast cells (mLFC) and RAW 264.7 cells. Results : In experiment of anti-thrombotic effect, BHPS inhibited the platelet aggregation induced by ADP and epinephrine, and inhibited pulmonary embolism induced by collagen and epinephrine. BHPS increased Platelet number and fibrinogen amount, and shortened PT and APTT in thrombus model induced by dextran. In experiment of anti-inflammatory effect, BHPS inhibited IL-1${\beta}$, IL-6, TNF-${\alpha}$, COX-2 and NOS-II mRNA expression in a concentration-dependent manner in RAW 264.7 cell line, and inhibited significantly NO production at 50, 100 ${\mu}g/ml$, and also inhibited ROS production in a concentration-dependent manner. BHPS inhibited IL-1${\beta}$, IL-6 and TNF-${\alpha}$ production significantly in serum of acute inflammation-induced Balb/c mice, and decreased IL-1${\beta}$, IL-6 and TNF-${\alpha}$ production in spleen tissue, but increased IL-1${\beta}$, IL-6 and TNF-${\alpha}$ production in liver tissue. BHPS increased survival rate at the 3th day in ICR mice with lethal endotoxemia induced by LPS. Conclusion : These results suggest that BHPS can be used for treating diverse female diseases caused by thrombosis and inflammation such as endometriosis, pelvic pain, cervicitis, pelvic inflammatory disease and pelvic tuberculosis and so forth.

  • Purpose: There has not been decisional consensus in the concepts and the ways of postpartum cares between Western and non-Western cultures though the importance of postpartum period has been emphasized by the multitude of health professionals. In this study, we investigated the differences in recognition concerning postpartum care among Koreans, Korean residents in America, and Americans (hereafter, K, KA, A). Methods: We used questionnaire of women who had given birth from last 6 weeks to less than 13 years. The women consisted of three groups;104 K, 44 KA, and 44 A. Results: In the perception survey concerning postpartum care, every three group thought that postpartum care was important. However, there were differences in understanding how it would affect women's health, and what they did in the period. K significantly agreed with the content and the importance of postpartum care. The investigation concerning seven categories of postpartum care showed that K group performed the 6 categories except consuming medication beneficial for the postpartum care. KA group performed in a low rate and A group performed two categories in a significantly low rate two categories; avoiding cold stimulation and keeping the body warm. Conclusion: Based on this study, it should be suggested that there were common points and differences among the recognition concerning postnatal care in three groups. Therefore, health professionals make efforts in understanding diversities on postnatal care and providing the adequate services reflecting each mother's needs and recognition.

The Effect of Kamijoaguiem on Osteoporosis Induced by Ovariectomy in Rats (가미좌귀음(加味左歸飮)이 난소적출(卵巢摘出)로 유도(誘導)된 백서(白鼠)의 골다공증(骨多孔症) 미치는 영향(影響))

  • Purpose : This study was peformed to evaluate the effect of Kamijoaguiem(JGE) on the bone mass and its related factors. Methods : We used ovariectomized rat as an estrogen-deficient animal model. The model rats of osteoporosis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 7th day after operating ovariectomy, rats were administered with JGE per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of JGE. Results : Bone density, bone ash density, bone calcium, magnesium and phosphorus was decreased in osteoporotic rats. JGE improved the decreased bone density, bone ash density and the decreased bone magnesium, but JGE didn't improve the decreased bone calcium and phosphorus in osteoporotic rats. Osteocalcin in serum and hydroxy-proline excretion in urine were increased in osteoporotic rats. Their levels were decreased when JGE was administered. ALP activity in serum was increased in osteoporotic rats. JGE didn't induce any significant changes. JGE showed significant increase in serum calcium level, total protein level, albumin level, BUN level, serum LDH activity. JGE didn't show significant increase in serum T-cholesterol density, triglyceride density, HDL-cholesterol density. JGE didn't show significant increase in RBC number, hemoglobin level, platelet number, hematocrit level. JGE showed inhibitory effect on the degradation of bone-matrix in osteoporotic rats, in histological examination to Hematoxylin-eosin stain. Conclusion : JGE might improve bone density due to inhibition of bone resolution in osteoporotic rats. It suggest that JGE may be useful prescription in osteoporosis.

  • Purpose: This study was performed to evaluate anti-oxidant activities and anti-inflammatory effects of Sungyoutanggagambang(SYTG). Methods: In the study of anti-oxidant activities. SYTG was investigated by DPPH radical scavenger activity. superoxide dismutase activity and superoxide anion radical scavenger activity. In the study of anti-inflammatory effects. SYTG was investigated using cultured cells and murine models. As for the parameters of inflammation. levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were measured in mouse lung fibroblast cells(mLFCs) and RAW264.7 cells. Results: Prior to the experiment. we investigated the security of SYTG by measuring GOT and GPT in serum. 1. SYTG showed high antioxidant activity in a concentration-dependent degree by measured scavenging activity of DPPH free radical, superoxide dismutase and superoxide anion radical. 2. SYTG inhibited IL-1$\beta$, IL-6. TNF-$\alpha$, COX-2 and NOS-II mRNA expression as compared with the control group in a concentration-dependent degree in RAW264.7 cell line. 3. SYTG inhibited IL-1$\beta$, IL-6 production significantly at 100 ${\mu}g/ml$ and TNF-$\alpha$ production significantly at 50, 100 ${\mu}g/ml$ as compared with the control group in RA W264.7 cell line. 4. SYTG inhibited IL-1$\beta$, and IL-6 production significantly as compared with the control group in serum of acute inflammation-induced mice. and decreased IL-1$\beta$, IL-6 production in spleen tissue. and also decreased IL-1$\beta$, IL-6 production in liver tissue. Conclusion: These results suggest that SYTG can be useful in treating diverse female diseases caused by inflammation such as endometrosis, myoma, pelvic congestion. chronic cervicitis, chronic pelvic inflammatory disease and so on.

JGTG in RAW 264.7 cell decreased IL-$1{\beta}$ mRNA expression, IL-6 mRNA expression, TNF-${\alpha}$ mRNA expression at 50, $100{\mu}g/m{\ell}$ and also decreased NOS-II mRNA expression at $100{\mu}g/m{\ell}$, and decreased COX-2 mRNA expression at 10, 50, $100{\mu}g/m{\ell}$. 4. JGTG in RAW 264.7 cell decreased significantly IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ at 50, $100{\mu}g/m{\ell}$. 5. JGTG inhibited significantly IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ production in serum of acute inflammation-induced mice. 6. JGTG decreased significantly IL-$1{\beta}$ mRNA production in spleen tissue. Conclusion: These results suggest that JGTG can be used for treating diverse female diseases caused by inflammation

  • Purpose: This study was performed to evaluate anti-thrombotic and anti-inflammatory effects of NeungaSoJeokTang water extract (NSJT). Methods: In the study of anti-inflammatory effects, NSJT was investigated using cultured cells and murine models. As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were determined in mouse lung fibroblast cells(mLFC) and RAW 264.7 cells. Results: Prior to the experiment, we evaluated sGOT, sGPT, BUN and creatine after the treatment. As a result, NSJT was innoxious on liver and kidney. In experiment of anti-thrombotic effect, NSJT inhibited the platelet aggregation induced by ADP and epinephrine, and inhibited pulmonary embolism induced by collagen and epinephrine. NSJT did not affect significantly the blood flow rate both in vitro and in vivo. NSJT increased platelet number and fibrinogen amount, and NSJT shortened PT and APTT in thrombus model induced by dextran. In experiment of anti-inflammatory effect, NSJT inhibited $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, COX-2 and NOS-II mRNA expression in a concentration-dependent manner in RAW 264.7 cell line, and inhibited significantly NO production at 50, 100 ${\mu}g/ml$, and also inhibited ROS production in a concentration-dependent manner.
