Influence of rutin on the effects of neonatal cigarette smoke exposure-induced exacerbated MMP-9 expression, Th17 cytokines and NF-κB/iNOS-mediated inflammatory responses in asthmatic mice model

  • Allergic asthma is one of the most enduring diseases of the airway. The T-helper cells and regulatory T-cells are critically involved in inflammatory responses, mucus hypersecretion, airway remodelling and in airway hyper-responsiveness. Cigarette smoke (CS) has been found to aggravate inflammatory responses in asthma. Though currently employed drugs are effective, associated side effects demand identification and development of novel drugs with negligible or no adverse effects. Rutin, plant-derived flavonoid has been found to possess antioxidant and anti-inflammatory effects. We investigated the ability of rutin to modulate T-cells and inhibit inflammation in experimentally-induced asthma in cigarette smoke exposed mice. Separate groups of neonatal mice were exposed to CS for 10 days from post-natal days 2 to 11. After 2 weeks, the mice were sensitized and challenged with ovalbumin (OVA). Treatment group were given rutin (37.5 or 75 mg/kg body weight) during OVA sensitization and challenge. Rutin treatment was found to significantly inhibit cellular infiltration in the airways and Th2 and Th17 cytokine levels as well. Flow cytometry revealed effectively raised $CD4^+CD25^+Fox3^+$ Treg cells and supressed Th17 cell population on rutin treatment. Airway hyper-responsiveness observed following CS and OVA challenge were inhibited by rutin. $NF-{\kappa}B$ and iNOS, chief regulators of inflammatory responses robustly activated by CS and OVA were down-regulated by rutin. Rutin also inhibited the expression of matrix metalloproteinase 9, thereby aiding in prevention of airway remodelling in asthma thereby revealing to be a potent candidate in asthma therapy.

  • Objectives : To clarify the effects of Actinidia polygama and CsA on OVA-induced asthma model, we examined the influence of Actinidia polygama fructus extract (APF) and CsA on the number of regulatory T cells, NKT cells and ${\gamma}{\delta}$ T cells in murine model of asthma. Methods : All mice were immunized on two different days (21 days and 7 days before inhalational exposure) by i.p. injections of OVA in PBS. Seven days after the second sensitization, mice were exposed to aerosolized ovalbumin for 30 min/day on 3 days/week for 12 weeks and APF (400, 40 mg/kg) were orally administered 3 times a week for 8 weeks. Results : The suppressive effects of APF on asthma model were demonstrated by the increase the number of regulatory T cells, ${\gamma}{\delta}$ T cells and by reducing the number of NK T cells. Conclusion : These results indicate that APF has a deep inhibitory effect on airway inflammation and hyperresponsiveness in murine model of asthma by increase the number of regulatory T cells, and ${\gamma}{\delta}$ T cells and by reducing the number of NK T cells.

  • To study the effects of EF(Eriobotryae Folium) herbal acupuncture on asthma, we injected EF-HAS into Jok-samni(ST36) of C57BL/6 mice Objective : The aim of this study was to investigate the effect of EF-HA(herbal acupuncture) at ST36 on ovalbumin-induced asthma in mice Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). Experimental groups were treated with concentrations(1%) of EF-HA at Jok-samni(ST36) for the later 8 weeks(3times/week). Result : 1. The weight and total cells of lung of the mice group treated with EF-HA decreased significantly compared with that of Control group. 2. Total Leukocytes and Eosinophils in BALF of the mice group treated with EF-HA decreased significantly compared with those of Control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with EF-HA decreased significantly compared with those of Control group. 4. The concentration of IgE, IL-4, IL-5 in BALF of the mice group treated with EF-HA decreased significantly compared with those of Control group. 5. The concentration of IL-4, IL-5, IL-13 in Serum of the mice group treated with EF-HA decreased significantly compared with those of Control group. 6. The number of $Gr-1^+/CD11b^+,\;CD3^-/CCR3^+,\;CD4^+,\;CD8^+,\;CD3e^+/CD69^+$ cells in the lungs of the mice group treated with EF-HA decreased significantly compared with those of Control group. 7. The cytokine's manifestation of mRNA of the mice group treated with EF-HA with RT-PCR decreased significantly compared with that of Control group. Conclusion : We conclude that EF-HA is effective on OVA-induced asthma of C57BL/6 mouse.

Effects of Inhalable Microparticles of Seonpyejeongcheon-Tang in an Asthma Mouse Model - Effects of Microparticles of SJT -

  • Objectives: Allergic asthma generally presents with symptoms of wheezing, coughing, breathlessness, and airway inflammation. Seonpyejeongcheon-tang (SJT) consists of 12 herbs. It originated from Jeong-cheon-tang (JT), also known as Ding-chuan-tang, composed of 7 herbs, in She-sheng-zhong-miao-fang. This study aimed to evaluate the effects of local delivery of SJT via inhalable microparticles in an asthma mouse model. Methods: Microparticles containing SJT were produced by spray-drying with leucine as an excipient. SJT microparticles were evaluated with respect to their aerodynamic properties, in vitro cytotoxicity, in vivo toxicity, and therapeutic effects on ovalbumin (OVA)-induced asthma in comparison with orally-administered SJT. Results: SJT microparticles provided desirable aerodynamic properties (fine particle fraction of $48.9%{\pm}6.4%$ and mass median aerodynamic diameter of $3.7{\pm}0.3{\mu}m$). SJT microparticles did not show any cytotoxicity against RAW 264.7 macrophages at concentrations of 0.01 - 3 mg/mL. Inhaled SJT microparticles decreased the levels of IL-4, IL-5, IL-13, IL-17A, eotaxin and OVA-IgE in bronchoalveolar lavage fluid (BALF) in mice with OVA-induced asthma. These effects were verified by histological evaluation of the levels of infiltration of inflammatory cells and collagen, destructions of alveoli and bronchioles, and hyperplasia of goblet cells in lung tissues. The effects of SJT microparticles in the asthma model were equivalent to those of orally-administered SJT extract. Conclusion: This study suggests that SJT is a promising agent for inhalation therapy for patients with asthma.

마황 추출물 투여가 Ovalbumin으로 유발된 마우스 알레르기성 천식에 미치는 영향 (Effects of Ephedra sinica (ES) Extract on the Ovalbumin-Induced Allergid Asthma in Mice)

  • Objective : Ephedra sinica (ES) has has been used as remedy of allergic diseases for a long time in Korea. In the present study, we investigated the anti-allergic effects of ES on experimental allergic asthma mouse model using ovalbumin (OVA). Methods : BALB/c mice were sensitized and challenged with 100 ug of OVA and 1 mg of aluminum potassium sulfate of 0.2 ml phosphate-buffered saline(PBS) intraperitoneally on day 1 and 15. mice were challenged on 3 consecutive days with 5% OVA and AHR was assessed 24 h after the last challenge. we examined the lung histology, airway hyper sensitivity, total inflammatory cell count in bronchoaveloar lavage fluid(BALF), Th2-associated cytokines production and IgE production. Results : ES potently inhibited the lung damage and the development of Penh. ES also reduced the number of BAL cells during OVA-induced allergic asthma. Furthermore, ES inhibited cytokines production such as IL-4, IL-13 productions, and IgE level of serum. Conclusion : These results suggest that ES may inhibit the production of IL-4, IL-13, IgE and infiltration of inflammatory cell and be beneficial oriental medicine for allergic asthma.

  • Background : Recently, so many people are suffered from the allergic or auto-immune disease, and the representative disease is just Allergic Asthma. It is because human immune function has been decreased. Many treatments were done to treat this disease, and many methods were studied to increase immune function and to suppress the asthma. But, the effect of asthma-suppression and improvement of immune response of EH-HA(Ephedrae Herba Herbal Acupuncture) has not been studied in detail. To study the effects of EH-HA, we injected EH-HA at Joksamni(ST36) of C57BL/6 mice. Objectives : The purpose of this study is to investigate the effect of asthma-suppression and improvement of immune response of EH-HA(Ephedrae Herba Herbal Acupuncture). EH-HA was done at Joksamni(ST36) of the mice with ovalbumin-induced asthma. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks. Two experimental groups were treated with different concentrations(1%, 0.1%) of EH-HA at Joksamni(ST36) for the later 8 weeks(3times/week). Results : 1. The lung weight of the group treated with EH-HA decreased significantly compared with that of control group. 2. The total cells in lung, total leukocytes and eosinophils in BALF of the group treated with EH-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the group treated with EH-HA in photomicrographs decreased significantly compared with those of control group. 4. The concentrations of IL-13, IgE, IL-4 in serum and IL-4 in BALF of the group treated with EH-HA decreased significantly compared with those of control group. 5. The numbers of $Gr-1^+/CD11b^+\;cells,\;CD3e^-/CCR3^+\;cells,\;CD4^+\;cells,\;CD8^+\;cells,\;CD3e^+/CD69^+\;cells\;and\;IgE^+/B220^+\;cells$ in lung of the group treated with EH-HA decreased significantly compared with those of control group. 6. In RT-PCR, the mRNA expression of IL-4, IL-5 and IL-13 in the group treated with EH-HA decreased compared with those of control group. Conclusion : These results suggested that EH-HA at Joksamni(ST36) in C57BL/6mice may be effective to OVA-induced asthma of C57BL/6 mice.

프랑킨센스 에센셜 오일 흡입이 OVA로 유도된 알러지성 천식 모델 생쥐에 미치는 영향 (The Effect of Nebulized Frankincense Essential Oil in an OVA-Induced Allergic Asthma Mouse Model)

  • 프랑킨센스 에센셜 오일(BSEO)이 알러지성 천식 생쥐 모델의 폐, BALF 및 PBMC 내 면역세포 변화에 미치는 영향을 조사하였다. 본 연구는 Balb/c 생쥐를 이용하여 실험 1일에 $100\;{\mu}g$ ovalbumin (OVA)을 복강 내 투여하고, 실험 14일에 $50\;{\mu}g$ OVA를 기도 내 투여하여 감작시킨 후 OVA를 반복 흡입시켜 천식을 유발하였다. 천식 유발 생쥐군에 0.3% BSEO를 8주간, 1주에 3회, 1회에 30분간 nebulizer로 흡입시켰고, 실험일로부터 총 12주 후에 살처분시켜 폐, 기관지 폐포 세척액 (bronchoalveolar lavage fluid; BALF), 말초혈액단핵세포 (peripheral blood mononuc1ear cell; PBMC)를 분리하여 조직 내의 면역세포 변화를 관찰하였다. 본 연구 결과 알러지성 천식 유발 생쥐에서 기관지 수축제에 대한 기도 과민성반응이 천연 아로마 에센셜 오일군인 Bergamot군, Eucalyptus군, Chamomile군, Marjoram군, 및 Frankincense군 중 Frankincense군에서 효과적으로 억제됨을 확인하였다. 또한 알러지성 천식유발로 증가한 폐무게, 총폐포수, BALF수, MNL, DLN수가 억제됨을 확인하였다. BSEO는 알러지성 천적 모델 생쥐의 폐 조직 내의 $CD3e^+/CD19^-$, $B220^+/CD23^+$, $CD11b^+/Gr-1^+$ 세포 비율을 억제하였으며 BALF 내의 $CD4^+/CD8^-$, $B220^+/CD23^+$, $CD3e^+/CCR3^+$ 세포비율을 억제하였다. 또한 PBMC내 $CD3e^+/CD19^-$, $CD3^+/CD69^+$, $B220^+/CD23^+$ 세포비율을 억제하였다. 이러한 결과는 BSEO가 각 조직내의 면역세포의 활성 억제를 통한 면역조절작용으로 알러지성 천식의 치료에 효과적일 것으로 사료되었다.

Anti-inflammatory Activity of Chihyo-san to Protect Respiratory Tissues from Asthmatic Damage

  • The present study was carried out to investigate the effect of Chihyo-san (CHS) administration on asthma induced by Alum/OVA treatment in the mice. In CHS-treated animal group, lung weight, which was increased after asthma induction, was significantly decreased, and total number of cells in the lung, peripheral lymph node (PLN) and spleen tissue was significantly decreased in CHS-treated group compared to the asthma control group. The number of immune cells including natural killer (NK) cells in asthmatic animals was largely regulated by CHS treatment, showing a similar pattern as that of CsA-treated positive control group. Levels of mRNAs encoding inflammatory cytokines IL-5, IL-13, $TNF-{\alpha}$, and eotaxin were determined by RT-PCR in the lung tissue and showed decreases in CHS-treated group to the similar levels of CsA-treated control group, Histamine level in the serum was significantly lower in CHS-treated group than asthma-induced control group. Both haematoxylin and eosin staining and Masson's trichrome staining results showed decreased number of inflammatory cells, reduced immune cell infiltration, and normalized epithelial cell layering in the bronchial tissue of CHS-treated mouse group. Thus, the present findings suggest that CHS may be useful for protecting bronchial tissues from consistent inflammatory damages that occur in asthma patients.

Effects of Ascaris suum Extract and Sulfamethoxazole on Allergic Airway Inflammation

  • Allergic asthma is complex inflammatory airway disorder caused by genetic and environmental factors. Sulfamethoxazole, a sulfonamide, is the cause of drug rash with eosinophilia and systemic symptoms syndrome. Parasites infection also related with eosinophilia and allergic diseases. In the present study, we investigated the modulating effects of parasitic derivative and sulfamethoxazole (SMX) on allergic airway inflammation in the ovalbumin (OVA)-induced murine asthma model. Histopathological changes, cytokine secretion, and total and allergen-specific IgE were investigated. BALB/c mice were treated with Ascaris suum extract or SMX for 4 weeks before sensitized and challenged to ovalbumin. Pre-treatment of Ascaris suum extract decreased allergic inflammation in lung tissue and IL-4, total IgE, and OVA-specific IgE levels in bronchoalveolar lavage fluid. However, pre-treatment of SMX did not show any effects on allergic airway inflammation. These results indicate that parasitic infection has protective effects on allergic asthma, but the sulfamamides may not relate with allergic asthma.
