The Promotive Effect of NAA, IBA and Ethychlozate on Rooting Cuttings of Certain Ornamental Plants and Some Physiological Studies. (관상식물 삽목발근에 있어서 NAA, IBA 및 Ethychlozate의 발근촉진효과와 그 생리학적연구)

  • The present studies were undertaken to elucidate the influence of auxins, auxin-like substance-ethychlozate ("Figaron"),and pH and sort of rooting media on rooted propagation of certainornamental woody plant cuttings, and to see possible changes in internal compositions characterizing after root-promoting treatment as the cutting stage proceeded. The experimental check-up srevealed and summarized as seen in the following;I. Effect of three different auxin treatments on rooting cuttings: 1) Promotive influence of auxin varied according to different concentration levels, hours of dipping treatment of the auxins, and kind of plants. The greatest effect was obtained for Forsythia ksreana with NAA and IBA, for Ligustrurn obtusifolium var. variegatum with NAA and ethychlozate, for Hydrangea macrophylla, Magnolia kobus, and Magnolia liliflora with NAA, lBA and ethychlozate also. The most effective level of the promotive agents was found 200mg/l for NAA, 1000mg/l for IBA, and 200mg/l for ethychlozate. For Weigela florida and Gardenia jasminoides, range of the most effective level was shown relatively wide spread. 2) NAA was more effective at its optimal level of the rooting agent than ethychiozate for Weigela florida, Viburnum awabuki, Forsythia koreana, Acer palmatum 'Nomura', Bouga invillea glabra, Elaeagnus umbellata, Prunus tomentosa, Ligustrum obtusifolium, Pyracantha coccinea, Cestrum noctu rnum, Hydrangea macrophylla, Codiaeum variegatum, Rhododen dron lateritium, and Ilex crenata var. macrophylla, and yet ethychlozate was found either as equally as effective or more so than NAA for Zebrina pendula, Hibiscus syriacus, Fatshedera lizei, Schefflera arboricola, Campsis grandiflo ra, Ixora chinensis, Euonymus japonica, and Magnolia liliflora. On the contrary, no the auxin effect was noted with Lagerstroemia indica, Trachelospermum asiaticum, and Syringa vulgaris. This probably indicates that these species are genetically different for the auxin response.II. Effect of different pH and sorts of cutting media on rooting cuttings: 1) Bougainvillea showed best in rooting for the number and dry weight at pH 6.5, more with ethychlozate than NAA, while Ligustrum did at pH 5.0 more with NAA than ethychlozate. pH 4.0 medium resulted in the best rooting for Rhododendron with NAA, more than ethychlozate. 2) Use of cutting medium with peat: perlite: vermiculite = 1:1:1 showed to give the greatest rooting percent and dry weight, apart from considering the number of roots. This apparently meant the fact that cutting medium has more to do with root growth than root differentiation. Rhododendron yet showed results with cutting media that use of peat: perlite = 2:1 mixed is more effective on rooting than using peat alone.III. Effect of auxinic treatments on rooting cuttings and change in some cutting compositions: 1) Under the climatic conditions of July having temperature $26.3\pm$$2.4^{\circ}C$for cutting bed, new roots of Magnolia started to show up generally 20 days after the cutting was made, whereas Cestrum did much earlier than that, namely 14 days after. 2) Although total carbohydrate content of Magnolia cuttings showed no marked change without auxin treatment, it did so with the treatment, especially 30 days after the start of cutting. Cestrum cuttings demonstrated a gradual in crease in total carbohydrate content as rooting took place, and the content became reduced more with auxin than with out, just about when rooting proceeded to 14 days after the start of cutting. 3) Magnolia generally showed an increase in total nitrogen content as rooting proceeded more, and Cestrum showed a decrease in total nitrogen of cuttings. The auxin treatment exhibited no pertinent relation with change in plant nitro gen when rooting is promoted with auxin treatment. 4) An abrupt drop of total sugar and reducing sugar was noticed as Magnolia rooting started, and this reduction was parti cularly outstanding with auxin treatment. Starch content also was decreased in the later stage of cutting with auxin treatment, and was rather increased without auxin. Although sugar content soon increased as cutting started with auxin treatment in the case of Cestrum, it became reduced after rooting took place. 5) Total phenol content increased with rooting, and this was especially true when rooting started. This increase was reversed somehow regardless of auxin treatment. A decrease in phenol of Magnolia was found more striking with auxin than without in the later stage of the cutting period. 6)Avena coleoptile test for auxin-like substances presented the physiologically active factor is more in easy-to-root Magnolia liliflora than hard-to-root Magnolia kobus, and the activity of auxin-like substances was much increased with auxin treatment. The increase in the growth promoting substances was markedly pronounced when rooting just started. The active growth substances decreased in the later stage of cutting, and certain inhibitory substances started appearing. Cestrum also showed physiologically similar growth promoting substances accompanying auxin-like active substances if auxin is treated, and some strong inhibitory substances seemed to appear in the later stage of cutting. 7) Mung-bean-rooting test indicated biologically that endogenous growth substances in Magnolia all promoted mung-bean rooting, and activity of the growth substances apparently stimulated mung-bean rooting with auxin more than without. Here auxin treatment seemed to give a rise to an increased activity of endogenous growth substances in cuttings. This activity was found much greater with either NAA or IBA than ethychlozate, and showed its peak of the activity when rooting first started taking place. Certain inhibitory substances for Avena coleoptile growth strongly promoted mung-bean rooting, and it was also much like in the case of Cestrum.

Somatic Cell Analysis and Cobalamin Responsiveness Study in Ten Korean Patients with Methylmalonic Aciduria (한국 메틸말로닌산혈증 환아 10례에서 Somatic Cell 분석과 cobalamin 반응성 연구)

  • Purpose: Isolated methylmalonic acidemia (MMA) is an autosomal recessive inherited disorder of propionate metabolism. There are two subtypes of MMUT gene defects. $Mut^0$ represents complete loss of methylmalonyl-CoA mutase (MCM) activity while mut- is associated with residual MCM activity, which can be stimulated by hydroxocobalamin (OHCbl) supplementation. The objective of this study is to investigate cobalamin responsiveness and mutations present in Korean MMA population. Methods: We evaluated 10 MMA patients using somatic cell complementation analysis on their fibroblasts to measure MCM activity and vitamin B12 responsiveness for the optimal treatment. MMUT gene was sequenced to identify the MMA mutations. Results: For all patients, the incorporation of $[^{14}C]-propionate$ was low, and there was no response to OHCbl. The incorporation of $[^{14}C]-methyltetrahydrofolate$ and $[^{57}Co]-CNCbl$ fell within the normal range. There was adequate synthesis of methylcobalamin while the synthesis of adenosylcobalamin was low. The complementation analysis showed all patients were $mut^0$. The sequence analysis identified 12 different MMUT mutations, including 2 novel mutations, p.Gln267Ter and p.Ile697Phe, were identified. All the patients in this study had neonatal onset of symptoms, belonged to $mut^0$ complementation class, and as a result, showed no cobalamin responsiveness. Conclusion: No Korean MMA patient showed cobalamin responsiveness.

Analysis of the Characteristics of Bulking Agents Used in Livestock Manure Composting (축산분뇨 퇴비화에 이용되는 수분조절제의 특성 분석)

  • This study analyzed the characteristics of 6 recyclable bulking agents which can replace sawdust and rice hulls that are commonly used in livestock manure composting. The content of all the hazardous chemicals found in the bulking agents used in this experiment was between 0.0 and 34.1 ppm, which was noticeably lesser than the maximum content levels of hazardous substances 5.0~900.0 ppm prescribed by the Rural Development Administration(RDA), which means that the bulking agents would be able to replace sawdusts and rice husks. The water content of the bulking agents ranged from a minimum of 12.4% to a maximum of 16.6% in the original state, which was much less than 60%, the optimal moisture content. These results indicate that they would be able to function fully as bulking agents. Their water absorption rate was in the range of minimum 31.9% ~ maximum 600.0%, which showed huge differences among the types of bulking agents. The most appropriate bulking agent in terms of water absorption rate was wood pellets. It was the highest in the wood briquette manufactured by A, which was followed by C and B in the order. The shear strength of the 5 types of bulking agents in the original state was 271.7 N on an average, the highest figure of all, except for the oak briquette whose shear strength could not be measured and the wood pellets in the absorption state. The shear strength in the dry and absorption states then followed, and were 78.0 N and 27.7 N on an average, respectively. The wood briquette of A recorded considerably lower shear strength than that of B and C. Overall, shear strength tended to increase according to lower water absorption rates. Since pine wood chips and oak cuts have relatively greater shear strength even in the absorbing state than the other materials, they will be able to endure some power in the case of stirring with pig droppings.

Development of Stand Yield Table Based on Current Growth Characteristics of Chamaecyparis obtusa Stands (현실임분 생장특성에 의한 편백 임분수확표 개발)

  • We constructed a stand yield table for Chamaecyparis obtusa based on data from an actual forest. The previous stand yield table had a number of disadvantages because it was based on actual forest information. In the present study we used data from more than 200 sampling plots in a stand of Chamaecyparis obtusa. The analysis included theestimation, recovery and prediction of the distribution of values for diameter at breast height (DBH), and the result is a valuable process for the preparation ofstand yield tables. The DBH distribution model uses a Weibull function, and the site index (base age: 30 years), the standard for assessing forest productivity, was derived using the Chapman-Richards formula. Several estimation formulas for the preparation of the stand yield table were considered for the fitness index, and the optimal formula was chosen. The analysis shows that the site index is in the range of 10 to 18 in the Chamaecyparis obtusa stand. The estimated stand volume of each sample plot was found to have an accuracy of 62%. According to the residuals analysis, the stands showed even distribution around zero, which indicates that the results are useful in the field. Comparing the table constructed in this study to the existing stand yield table, we found that our table yielded comparatively higher values for growth. This is probably because the existing analysis data used a small amount of research data that did not properly reflect. We hope that the stand yield table of Chamaecyparis obtusa, a representative species of southern regions, will be widely used for forest management. As these forests stabilize and growth progresses, we plan to construct an additional yield table applicable to the production of developed stands.

Development and Performance Evaluation of Multi-sensor Module for Use in Disaster Sites of Mobile Robot (조사로봇의 재난현장 활용을 위한 다중센서모듈 개발 및 성능평가에 관한 연구)

  • Disasters that occur unexpectedly are difficult to predict. In addition, the scale and damage are increasing compared to the past. Sometimes one disaster can develop into another disaster. Among the four stages of disaster management, search and rescue are carried out in the response stage when an emergency occurs. Therefore, personnel such as firefighters who are put into the scene are put in at a lot of risk. In this respect, in the initial response process at the disaster site, robots are a technology with high potential to reduce damage to human life and property. In addition, Light Detection And Ranging (LiDAR) can acquire a relatively wide range of 3D information using a laser. Due to its high accuracy and precision, it is a very useful sensor when considering the characteristics of a disaster site. Therefore, in this study, development and experiments were conducted so that the robot could perform real-time monitoring at the disaster site. Multi-sensor module was developed by combining LiDAR, Inertial Measurement Unit (IMU) sensor, and computing board. Then, this module was mounted on the robot, and a customized Simultaneous Localization and Mapping (SLAM) algorithm was developed. A method for stably mounting a multi-sensor module to a robot to maintain optimal accuracy at disaster sites was studied. And to check the performance of the module, SLAM was tested inside the disaster building, and various SLAM algorithms and distance comparisons were performed. As a result, PackSLAM developed in this study showed lower error compared to other algorithms, showing the possibility of application in disaster sites. In the future, in order to further enhance usability at disaster sites, various experiments will be conducted by establishing a rough terrain environment with many obstacles.

Ecological Study on Poisonous Snake and Investigation of the Venom Characteristics, Snakebiting Frequenty in Korea (한국산 독사의 생태학적 특징 및 독성, 교상빈도에 관한 조사, 연구)

  • Four species(25%) of Viperidae(Agkistrodon brevicaudus, Agkistrodon ussuriensis, Agkistrodon saxatilis) and Cloubridae(Rhabdophis tigrenus tigrenus) were Korean poisonous snake. Copulation season of these species was from July to August. Reproduction mode of genus Agkistrodon species was ovoviviparous but Rhabdophis tigrinus tigrinus was the other pattern of oviparous. Optimal movement temperature range was from 20$\circ $C to 29$\circ $C(March~September). Wjen atmosphere temperature was below 10$\circ $C, at that time they hibernate at the ground, rock bottom, stone wall and embankment around the end of a field. The venom of these snakes consist mainly Hematoxin, Cytolysin, Neurotoxin and Cardiotoxin of poisonous liquids. These material injection to animal cause systemic syndrome such as Dizziness(25.7%), Vomitting(23.1%), Fever(22%), Visual trouble(18%), Headace(17.7%), Dyspnoea(17.6%) and bring about other local syndrome such as Discoloration(54.2%), Bleeding(20.2%), Bullae(10.7%) and Skin ulcer(!0.8%). The annual distribution was appeared to decrease 1972 after 1992 and average snakebiting patients was 25.6 per year, but practically total estimated snakebiting was 2,700 per year. The seasonal distribution was most frequent in August(25%), and mortality was 1.8%(26 per 1,430). The sex ratio was 2:1 and according to age distribution, it was most prevalent at one's fifties(19%). The most frequent place where the accident happened was the field(48.2%) and most predilection site of the body for victim were hand(47.8%) and foot(39.5%), Commonly bite snake were Agkistrodon ussuriensis(27.1%), Agkistrodon brevicaudus(22.6%) and Agkistrodon saxatilis(9.6%) but 40.7% of species could not be identified. Treatment of antivenin patient was 75.9% (1,068/1,407).

Growth and Quality Characteristics of Korean Bread Wheat in Response to Elevated Temperature during their Growing Season (밀 재배기간 온도상승이 빵용 밀의 생육 및 품질 특성에 미치는 영향)

  • Wheat (Triticum aestivum L.) is a major staple foods and is in increasing demand in the world. The elevated temperature caused by changes in climate and environmental conditions is a major factor affecting wheat development and grain quality. The optimal temperature range for winter wheat is between 15 and 25℃, and it is necessary to study the physiological characteristic of wheat according to elevated temperatures. This study presents the effect of elevated temperature on the yield and quality of two Korean bread wheat (Baekkang and Jokyoung) in temperature gradient tunnels (TGT). Two bread wheat cultivars were grown in TGT at four different temperature conditions: T0 (control, near ambient temperature), T1 (T0+1℃), T2 (T0+2℃), (T0+2℃), T3 (T0+3℃). The period from sowing to heading stage accelerated and the number of grains per spike and grain yield reduced under T3 condition compared with those under T0 condition. Grain filling rate and grain maturity also accelerated with elevated temperature (T3). The increase in temperature led to the increase in protein contents, whereas decreased the total starch contents. These results are consistent with the decreased expression of starch synthesis genes and increased gliadin synthesis or gluten metabolism genes during the late grain filling stage. Taken together, our results suggest that the increase in temperature (T3) led to the decrease in grain yield by regulating the number of grains/spike, whereas increased the protein content by regulating the expression of starch and gliadin-related genes or gluten metabolism process genes expression. In addition, our results provide a useful physiological information on the response of wheat to heat stress.

Evaluation of Efficient Pb Removal from Aqueous Solutions using Biochar Beads (바이오차 비드를 이용한 수용액에서 Pb의 효율적인 처리효율 평가)

  • The fine particulate structure of biochar limits its use as a heavy metal adsorbent, and makes separation of the biochar from the solution technically challenging, thereby reducing recovery of the heavy metals. To address this issue, this study prepared biochar beads under various mixing conditions and investigated their efficiency in removing Pb from aqueous solutions using adsorption models. The biochar beads were produced by mixing alginate and biochar at different ratios: alginate bead (AB), 1% biochar + bead (1-BB), 2.5% biochar + bead (2.5-BB), and 5% biochar + bead (5-BB). The results revealed that the Freundlich isothermal adsorption pattern of the biochar beads to Pb was of the L-type. The highest Langmuir isothermal adsorption capacity (28.736 mg/g) was observed in the 2.5-BB treatment. The dominant mechanism among the kinetic adsorption characteristics of biochar beads for Pb was chemical adsorption. Additionally, the optimal pH range for Pb adsorption was found to be between 4 and 5.5. The highest Pb removal efficiency (97.9%) was achieved when 26.6 g/L of biochar beads were used. These findings suggest that biochar beads are an economical and highly efficient adsorbent that enables separation and recovery of fine biochar particles.

Selection of Forage Corn Varieties Adapted to High Latitude (The South of Mt. Suyang) (고위도 기후대 재배 적합 국산 사료용 옥수수 품종 선발)

  • Since maize (Zea mays L.) originated in central and south America, it requires warm climate conditions throughout its growing season. Growth halts when night-time temperatures drop below 10℃, and the plant may die if temperature reach -1.7℃. Thus, temperature should be maintained between 10 and 30℃ from seeding to maturity. The germination temperature for maize should be at least 8-11℃, whit an optimal range 32-34℃. Since temperature significantly affects the germination rate and period, it plays a crucial role in maize growth. In this study, we evaluated the quantity and feed value of 11 major varieties to determine those best suited for maize cultivation as feed in higher latitude, specifically in Democratic People's of Republic of Korea, below 38 degrees north. A cultivation test was also conducted in Suwon in Republic of Korea, to assess adaptability in areas south of Mt. Suyang. Among the varieties tested, Shinhwangok2 reached silking the fastest, in 65 days, while Gwangpyeongok took the longest at 75 days. The stem length of all varieties exceeded 230 cm. Gwangpyeongok had the tallest stems, while Daanok and Shinhwangok2ho displayed the highest ear ratios. Dacheongok presented the highest values in both dry matter and TDN quantity, with 31,420 kg/ha and 21,66 kg/ha respectively. Pyeonggangok had the highest crude protein content at 8.0%. TDN (%) ranged from 57-68%, with Hwangdaok reaching up to 68%. Based on these findings, Dacheongok and Pyeonggangok appear to be the most suitable varieties for cultivation in terms of both quantity and feed value.

Isolation and Characterization of Lactic acid bacteria Leuconostoc mesenteroides DB3 from Camellia japonica Flower (백꽃으로부터 분리한 Leuconostoc mesenteroides DB3의 특성)

  • Lactic acid bacteria (LAB) are widespread in a variety of environments including fermented dairy products, gastroinstetinal and urogenital tracts of human and animals, plant, soil and water. Leuconostoc mesenteroides DB3 was detected by the strongest antibacterial activities among 24 Leuconostoc strains isolated from Camellia japonica flowers. Acid tolerance of L. mesenteroides DB3 existed up to pH 2.5, but the resistance did not show at pH 2.0, which relatively excellent acid resistance existed. Bile acid tolerance was very stable within the test range to 1.2%. L. mesenteroides DB3 exhibited the optimal growth at 30℃, and showed a slight slow growth when compared with L. mesenteroides KCTC3505, which reached a stationary phase at 18 hr. The pH was changed along with the growth curve, but was maintained above pH 3.98. L. mesenteroides DB3 had higher initial antibacterial activities when compared to L. mesenteroides KCTC3505, but it showed similar activities with the standard strain after the latter part of the logarithmic growth phase. Although lactic acid production in L. mesenteroides DB3 was induced by lower amount in the initial part to the standard strain, it was exhibited by similar amounts after the late logarithmic growth phase. Muicin adhesion of L. mesenteroides DB-3 maintained superior to L. mesenteroides KCTC3505. Both strains showed excellent emulsification ability for kerosene. In summary, we evaluate that L. mesenteroides DB-3 has a high potential for application as probiotics owing to its excellent antibacterial activity, acid resistance, bile acid resistance, and muicin adhesion.
