채소즙이란 생채소를 마쇄하여 인체가 영양소를 흡수하기 쉬운 상태로 제조된 즙으로 생채소의 영양 섭취 효율을 높일 수 있는 식품이라 할 수 있다. 본 연구에서는 재배방법에 따라 생산된 유기농 및 일반농 케일을 착즙하여 다양한 실험을 통하여 항산화 활성을 비교하였다. DPPH radical, NO, $O_2{^-}$, ${\cdot}OH$ 라디칼 소거능에서 유기농 케일 착즙액은 일반농케일 착즙액보다 더 우수한 효과를 나타내었다. LLC-PK1 세포를 이용하여 산화적 스트레스 개선효과에서 유기농 케일 착즙액은 일반농 케일 착즙액에서보다 NO, $O_2{^-}$와 $ONOO^-$에 의해 유발된 산화적 스트레스에 대한 세포 생존율을 증가시키고, 지질과산화물을 억제시켜 유리라디칼에 대한 보호효과를 유의적으로 나타내었다. 위의 결과로 보아 유기농케일 착즙액은 일반농 케일 착즙액보다 산화적 스트레스에 대한 개선효과가 뛰어난 것으로 사료되며 재배방법의 차이에 따라 그 효과도 차이가 있었다.
Seventeen 30% EtOH extracts from the Compositae plants collected in Gangwon-do, Korea during autumn season were analyzed by HPLC using three standard caffeoylquinic acids (chlorogenic acid, 3,5-di-O-caffeoylquinic acid, 3,5-di-Omuco-quinic acid) and six flavonoids (rutin, isoquercitrin, astragalin, quercitrin, quercetin and kaempferol) to find the composition of phenolic compounds and also assayed to evaluate the peroxynitrite (ONOO$^-$) scavenging effect. The extracts with $IC_{50}$ values less than 2.0 ${\mu}g/ml$ were as follows: Aster tartaricus ($IC_{50}$, $1.26{\pm}0.10\;{\mu}g/ml$), A. maaki ($1.45{\pm}0.03\;{\mu}g/ml$), Solidago virga-aurea, ($1.45{\pm}0.03\;{\mu}g/ml$), Picris hierraciodes var. glabrescens ($1.45{\pm}0.04 \;{\mu}g/ml$), Lactuca triangulata ($1.50{\pm}0.09\;{\mu}g/ml$), Chrysanthemum zawadskii ssp. acutilobum, ($1.79{\pm}0.14\;{\mu}g/ml$). Particularly, the proportion of total phenolic compounds measured in the extract of L. triangulata was highest as the value 54.51%.
A bacterium CGH18 exhibiting antioxidizing and chitin-degrading activities in the colloidal chitin culture medium was isolated from salt-fermented crab. This strain was identified as Bacillus idriensis based on 16S rDNA sequence homology search. Its crude extract was partitioned between n-BuOH and $H_2O$. The organic layer was further partitioned between $CH_2$$Cl_2$ and $H_2O$. Antioxidant activities of crude extract and its solvent fractions were evaluated using five different bioassay methods, including the degree of occurrence of intracellular reactive oxygen species (ROS), peroxynitrite scavenging (ONOO), and oxidative damage of genomic DNA. All fractions exhibited significant antioxidant activity in bioassay systems used.
Objectives: This study is experimental comparison of brown rice (BR) and germinated brown rice (GBR) on upper gastrointestinal diseases animal models.Methods: The ICR mice were divided randomly into four groups of six animals each (Normal mice, gastritis mice, gastritis mice treated with BR, gastritis mice treated with 48h GBR). Gastritis was induced by administration of 0.5 mL 150 mM HCl-60% ethanol. Six-week-old male Sprague-Dawley (SD) rats were randomly divided into 7 groups after 1 week adaptation. (Normal rat, reflux esophagitis (RE) rat, RE rat treated with BR, RE rat treated with 24,30,36,48h GBR). Reflux esophagitis was induced by ligation with a 2-0 silk thread both the pylorus and the transitional junction between the forestomach and the corpus in SD rats.Results: HCl/ethanol-induced gastric mucosal injury mice were ameliorated mucosal damage upon histological evaluation by treatment of 48h GBR than BR. Optical changes such as hyperemia and multiple erosions were observed in the rats with RE and damage to the normal rats was not apparent. The oral administration of GBR significantly diminished against gross mucosal damage in a germination time-dependent manner. Also, the administration of GBR suppressed the biomarker of oxidative stress, reactive oxygen species (ROS) and produces peroxynitrite (ONOO-) in serum. However, the administration of GBR could not affect to the pH level secreted from stomach when compared with Control group.Conclusions: These findings suggest that GBR could have improving effects on upper gastrointestinal diseases in a germination time-dependent manner.
The study was conducted to investigate candidate materials as anti-inflammation agent from plant resources. Activities of 33 plant parts extracts with the final concentration of 5${\mu}g/ml$ were evaluated on the several inflammation-related markers such as the release of proinflammatoty cytokine [tumor necrosis factor-alpha (TNF-${\alpha}$) & interleukin-6 (IL-6)], nitric oxide (NO), the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and inhibitor of nuclear factor kappa-B alpha ($I{\kappa}-B{\alpha}$) in lipopolysaccharide (LPS)-treated RAW 264.7 cells. The extracts in the final concentration of 10 ${\mu}g/ml$ were also screened on peroxynitrite (ONOO$^-$) scavenging activity. Eleven extracts selected from the screening assay were verified on the inhibition activity on peroxynitrite and total reactive species oxygen (ROS) in the several concentrations. As results, Alpinia officinarum Hance (rhizome), Inula britannica var. chinensis Regel (flower), Ulmus arvifolia Jacq (trunk peel) and Aster scaber Thunb. (aerial part) showed comparatively potent anti-inflammatory activities in vitro cells or chemical level systems, and then these four plant parts should be studied on the antiinflammatory mechanism by further studies.
This study noted that the actual mechanism of N-nitrosodimethylamine (NDMA) photodecomposition in the presence of $H_2O_2$ is missing from the previous works. This study investigated a key unknown reactive species (URS) enhanced by the addition of $H_2O_2$ during the photolysis of NDMA with $H_2O_2$, not hydroxyl radicals. In order to provide experimental evidences in support of URS formation, we have mainly used p-nitrosodimethylaniline, methanol, and benzoic acid as well-known probes of ${\cdot}OH$ in this study. Both loss of PNDA and formation of hydroxybenzoic acids were dependent on NDMA concentrations during the photolysis in a constant concentration of $H_2O_2$. In particular, competition kinetics showed that the relative reactivity of an URS was at least identical with ${\cdot}OH$-like reactivity. In addition, the decay of NDMA was estimated to be about 65% by the direct UV light and about 35% by the reactive species or URS generated through the photolysis of NDMA and $H_2O_2$. Therefore, our data suggest that a highly oxidizing URS is formed in the photolysis of NDMA with $H_2O_2$, which could be peroxynitrite ($ONOO^-$) as a potent oxidant by itself as well as a source of ${\cdot}OH$.
Objective : This study was designed to evaluate the protective effect of Rehmanniae Radix Crudus (RC) in 150 mM HCl/ethanol induced acute gastritis mice. Methods : ICR mice were divided into 5 groups (normal group, control group, 10 mg/kg sucralfate treated group, 50 mg/kg RC treated group, 100 mg/kg RC treated group, n=8). Normal group was not take any treatment. Control group induced gastritis 1 hour after ingestion of distilled water. 10 mg/kg sucralfate induced group was induced gastritis 1 hour after ingestion of distilled of sucralfate 10 mg/kg. 50 mg/kg and 100 mg/kg RC treated groups were induced gastritis 1 hour after ingestion of distilled of RC 50 mg/kg and 100 mg/kg. After 1 hour of gastritis induction, removed the stomach tissue. We examined histological observations, oxidative stress biomarkers, antioxidant proteins, inflammatory mediators and cytokines. Results : In this study, the RC treatment group showed gastritis and gastric ulcer inhibition, and the area of injury decreased. The oxidative stress biomarkers such as reactive oxygen species (ROS) and peroxy nitrite ($ONOO^-$) in the serum were reduced in the RC treated group. Inaddition, antioxidant proteins (nuclear factor erythroid 2-related factor 2, Heme oxygenase 1) were increased in RC treated group, and the expression of inflammatory mediators and cytokines induced by nuclear factor-kappa B activation was inhibited. Conclusion : According to the results, RC may have an excellent inhibitory effect on acute gastritis and gastric ulcer.
Objective : Alcoholic liver disease (ALD) is caused by excess alcohol intake. In the liver, alcohol breakdown results formation of toxic byproducts that lead to damage to tissue. This study is to investigate the therapeutic effects of Paeoniae Radix Alba and Puerariae Radix combination (PP) on ALD. Methods : PP was analyzed for polyphenolic compounds and free radical scavenging activity. ALD mouse model was induced by feeding ethanol and water (Control), silymarin (50 mg/kg), low-dose (PP: 100 mg/kg) or high-dose (PP: 200 mg/kg) was orally administrated to ALD mice for 14 days. The serum was assessed with levels of AST, ALT, total bilirubin, total cholesterol, and triglyceride. Liver tissues were evaluated for ROS levels, degree of liver damage and protein expression. Results : The 3:1 (Paeoniae Radix Alba:Puerariae Radix) ratio showed the best antioxidant values for the experiment. In ALD model, levels of AST, ALT, total bilirubin, total cholesterol, and triglyceride were significantly increased in the Control and the levels were decreased by treatment of PP. In addition, increased ROS, ONOO- and MDA levels in the Control were reduced in the PP groups. Western blot analysis figured out that proteins related to ROS and cholesterol metabolism were higher in ALD than in PP-treated ALD. Antioxidant enzyme expression was low in the control group and increased by PP treatment. Conclusion : Our results suggest that PP has the potential to be a medicine in ALD in terms of regulating oxidative stress and adjusting lipid metabolism.
본 연구에서는 국내산 법제 하수오 메탄올 추출물로부터 생리활성물질을 확인하였고, 추출물의 라디칼 소거능과 LLC-$PK_1$ cell을 이용한 산화적 스트레스 개선 효과를 살펴보았다. HPLC 분석 결과 4종의 생리활성물질인 2,3,5,4'-tetrahydroxystilbene 2-O-${\beta}$-D-glucoside, emodin, chrysophanol 및 rhein을 확인하였다. 특히 주요 화합물인 2,3, 5,4'-tetrahydroxystilbene 2-O-${\beta}$-D-glucoside는 115.02 mg/kg이었다. 법제 하수오 추출물의 DPPH, ABTS, hydroxyl 라디칼 소거능은 농도 의존적으로 증가하였다. 추출물의 $50{\mu}g/mL$ 처리 시 DPPH 라디칼 소거능은 48.4%, ABTS 라디칼 소거능은 57.9% 및 hydroxyl 라디칼 소거능은 81.2%로 나타내었다. 한편 LLC-$PK_1$ cell에서 각각의 NO, $O_2{^-}$, 및 $ONOO^-$ 생성물질인 pyrogallol, sodium nitroprusside(SNP) 및 morpholinosydnonimine(SIN-1) 처리에 의해 유도된 산화적 스트레스 상에서 세포 생존율은 감소하였다. 그러나 법제 하수오 메탄올 추출물은 농도 의존적으로 세포 독성을 저해하였다. 추출물의 $50{\mu}g/mL$ 처리 시 세포 생존율은 각각 82.1%(pyrogallol), 89.1%(SNP) 및 77.6%(SIN-1)였다.
