Role of Non-Thermal DBD Plasma on Cell Migration and Cell Proliferation in Wound Healing

  • Plasma technology isbeing developed for a range of medical applications including wound healing. However, the effect of plasma on many cells and tissues is unclear. Cell migration and cell proliferation are very important biological processes which are affected by plasma exposure and might be a potential target for plasma therapy during wound healing treatment. In this study, we confirmed the plasma exposure time and incubation time after plasma treatment in skin fibroblast (L-929 cells) to evaluate the optimal conditions forplasma exposure to the cell in-vitro. In addition, we used a scratch method to generate artificial wound for evaluating the cell migration by plasma treatment. Where, the cells were treated with plasma and migration rate was observed by live-cell imaging device. To find the cell proliferation, cell viability assay was executed. The results of this study indicate the increased cell proliferation and migration on mild plasma treatment. The mechanisms for cell migration and cell proliferation after plasma treatment for future studies will be discussed.

Effects of the Incidence Angle and Temperature on the Performance of a Thin-Film CIGS Solar Cell for Solar Powered UAVs (태양광무인기를 위한 박막형 태양전지의 입사각 및 온도에 따른 성능분석)

  • This research aims to study the effects of the incidence angle and surface temperature on the power generation performance of a thin-film CIGS solar cell for solar powered unmanned aerial vehicles (UAVs). The test rig consists of a unit CIGS solar cell is installed on a table whose angle is controlled manually. A K-type thermocouple is attached to the solar cell surface for temperature measurements. A solar module analyzer measures the voltage and current generated from the test solar cell. The solar module analyzer also calculates the maximum solar power and efficiency of the solar cell. All test data are acquired in a PC. Test results show that the solar cell efficiency decreases significantly with increasing incidence angle and increasing surface temperature in general. As the incidence angle increases from 0 degree to 90 degree, the solar cell efficiency decreases by 60%. The solar cell efficiency decreases by 10% with increasing solar cell surface temperature from $20^{\circ}C$ to $30^{\circ}C$, for exmaple. The direct cooling method of the solar cell using dry ice decreases dramatically the solar cell surface temperature, thus increasing the solar cell efficiency by 15%.

Bio-inspired Cell Deformability Monitoring Chips Based on Strain Dependent Digital Lysis Rates (미소유로의 길이에 따른 통과세포의 파괴율을 바탕으로 한 생체모사 세포 변형성 검사칩에 관한 연구)

  • We present a novel cell deformability monitoring chip based on the digitally measured cell lysis rate which is dependent on the areal strain of the cell membrane. This method offers simple cell deformability monitoring by automated high-throughput testing system. We suggest the filter design considering the areal strain imposed on the cell membrane passing through the filter array having gradually increased orifice length. In the experiment using erythrocytes, we characterized the cell deformability in terms of average fracture areal strain which was $0.24{\pm}0.014\;and\;0.21{\pm}0.002$ for normal and chemically treated erythrocytes, respectively. We also verified that the areal strain of 0.15 effectively discriminates the deformability difference of normal and chemically treated erythrocytes, which can be applied to the clinical situation. We compared the lysis rates and their difference for the samples from different donors and found that the present chips can be commonly used without any calibration process. The experimental results demonstrate the simple structure and high performance of the present cell deformability monitoring chips, applicable to simple and cost-effective cell aging process monitoring.

The effect of the stem of Spatholobus suberectus Dunn on the proliferation and gene expression related apoptosis in human cervical cancer cells (계혈등(鷄血藤)이 자궁경부암세포의 세포자멸사 유도와 관련 유전자 발현에 미치는 영향)

  • Objective : Spatholobus Suberectus Dunn stems, Chinese vine plants, have been used for the relief of menstrual disorders and rheumatic arthralgia. In this study, we investigated the antitumor effect of Spatholobus Suberectus Dunn on cervical cancer in vitro. Methods : HeLA cervical cancer cell lines were used as targets. We examined the effect of water extract from Spatholobus Suberectus Dunn on cell proliferation, cell cycle regulation and cell cycle-regulating gene expression. Further, we investigated the apoptotic effects of Spatholobus Suberectus Dunn on cervical cancer cell lines. Results : Spatholobus Suberectus Dunn significantly inhibited the proliferation of cervical cancer cell lines in a dose-dependent and time dependent manner. Fluorescence activated cell sorter (FACS) analysis indicated that Spatholobus Suberectus Dunn induced G1 cell cycle arrest. Spatholobus Suberectus Dunn enhanced the expression of $p21^{waf1}$ and $p27^{kip1}$ with cell cycle arrest. Further, Spatholobus Suberectus Dunn stimulated apoptosis via caspase3 pathway. Conclusions : These findings suggest that Spatholobus Suberectus Dunn is a candidate agent for the treatment of cervical cancer. p21waf1 and $p21^{waf1}$ and $p27^{kip1}$ may play an important role in Spatholobus Suberectus Dunn-induced cell cycle arrest and cell growth inhibition.

Morphological observation on the subependymal layer of swine brain (자돈뇌실(仔豚腦室) 주위의 subependymal layer의 형태(形態)에 관한 관찰(觀察))

  • Normal swine brains at 1 to 70 days after birth were used to investigate the presence and morphology of the subependymal layer (SL) in the ventricle walls. The brain samples were taken from 27 pigs of 4 swine breeds. The results were summarized as follows: 1. SLs were observed on the walls of the lateral ventricle (LV) but none were observed on the walls of the 3th and 4th ventricles. 2. SLs of the LV walls were composed of mainly 3-to 10-cell layers in thickness. The thinest region of SLs was composed of only 1-to 2-cell thick on the dorsal and ventral walls, and the thickest region was composed of 250-to 300-cell thick on extension region of the SLs into the angle between the corpus callosum and caudate nucleus. 3. Of the LV parts observed, the SL thickness were 25-to 45-cell thick on the anterior horn, 3-to 10-cell thick on the body, 100-to 220-cell thick on the angle region between the corpus callosum and caudate nucleus, and 3-to 5-cell thick the superior walls of the posterior horn. Also the SL thickness was more thick on the anterior region than those on the posterior region. 4. SLs may be classified as three types by the cell distribution; one type of them is closely arranged cell region with the distinctive lateral margin from the periventricular white matter, the other type is loosely arranged cell region with the undistinctive lateral margin, and another type is two-subdivided region as the loosely and closely arranged cell layers in a layer. 5. SLs were extensively thick in young age but gradually decreased in size and cell number with age after 20-day age. SL layers were composed of mainly oligodendrocytes, astrocytes and immature cells of them. Morphological differences of SL in different breeds of pigs were not observed.

Ammonium Ion Effects and Its In Situ Removal by Using Immobilized Adsorbent in Hybridoma Cell Culture (하이브리도마 세포배양에서 암모늄 이온의 영향 및 고정화 흡착제에 의한 암모늄 이온의 동시제거)

  • The effects of ammonium ion on cell growth kinetics, monoclonal antibody productivity, and cell metabolism of hybridoma cells were investigated. The mouse-mouse hybridoma cell line VlIIH-8 producing mouse IgG2a was used as a model system. Ammonium ion showed an inhibitory effect on cell growth and monoclonal antibody production. New immobilized adsorbents were developed for the reduction of the inhibitory effect of ammonium ion. The ammonium ion selective zeolite, Phillipsite-Gismondine was entrapped in calcium alginate bead or in dialysis membrane and applied to the hybridoma cell culture system for the in situ removal of ammonium ion from culture media. The effects of ammonium the both serum supplemented and serum free media on the cell growth were studied by applying immobilized adsorbents of calcium alginate bead type. The results demonstrated a substantial enhancement in cell growth. Applying immobilized adsorbents of dialysis membrane type to serum supplemented media also resulted in the stimulation of cell growth, cell viability and monoclonal antibody production.

  • This study explores how the main variables of youth going through a transition period (parents' nurturing behavior, the youth's self-esteem and ego resilience, the peer group relationships) affect the youth's school adjustment. It also analyzes the effects of theses variables on the youth's dependence on cell phones, which are considered a necessity in modern society. This research is verified by using a structure model verification method based on data from the Korean Children Youth Panel Study. According to the research, first, 1st-year students in middle school show better adjustment to their school life and less dependence on cell phones when they have a stronger sense of self-esteem and ego resilience; further, it is revealed that cell phone dependence has an indirect influence on the school life of these children. Second, they tend to show better adjustment to their school life and less dependence on cell phones when their parents' nurturing behaviors are more positive. Third, these children tend to depend more on cell phones when they have a positive peer group relationship, Moreover, cell phone dependence has an indirect influence on their school life. When considering the impact of cell phone to youth in modern society, need to prepare proper training program for correct uses and preventing from addiction.

AN EXPERIMENTAL STUDY ON THE RADIOSENSITIVITY AND CHEMOSENSITIVITY OF B16 CELL LINE (B16세포주의 방사선 및 항암제감수성에 관한 실험적 연구)

  • The purpose of this study was to aid in the prediction of tumor cell tolerance to radiotherapy and/or chemotherapy. For this study, cell surviving curves were obtained for murine melanoma Bl6 cell line using semiautomated M1T assay. 2,4,6,8, 10Gy were irradiated at a dose rate of 210cGy/min using /sup 60/Co Irradiator ALOORADO 8. After irradiatior, B16 cell lines(2.5×10⁴ cells/ml) were exposed to bleomycin and cisplatin at concentration of 0.2㎍/㎖, 2㎍/㎖ and 20㎍/㎖ for I hour respectively. The viable cells were determined for each radiation dose and/or each concentration of drug. And they were compared to control values. The obtained results were as follows : 1. There was significant difference of surviving fraction at 4, 6, 8, 10Gy on B16 cell line(P<0.05). 2. There was significant difference of cytotoxicity between bleomycin and cisplatin at concentration of 0.2㎍/㎖ and 2㎍/㎖(P<0.05) on B16 cell line, but there was no significant difference of cytotoxicity at concentration of 20㎍/㎖ on B16 cell line. 3. There was significant difference of cytotoxicity of bleomycin after irradiation of 2Gy and 10Gy on B16 cell line(P<0.01). 4. There was significant difference of cytotoxicity of cisplatin at concentration of 20㎍/㎖ after irradiation on B16 cell line.

Effects of Feeder Cells on the Primary Culture of Ovarian Cell Populations from Adult Japanese Medaka (Oryzias latipes)

  • Fish ovarian germline stem cells (OGSCs) that have the abilities to self-renew and differentiate into functional gametes can be used in various researches and applications. A main issue to be solved for effective utilization of fish OGSCs is the development of their stable in vitro culture condition, but only few researches about fish OGSC culture have been reported so far. In this study, in order to find the clues to develop the culture condition for OGSCs from Japanese medaka (Oryzias latipes), we tried to establish somatic cell lines as a candidate for the feeder cells and evaluated its supporting effects on the culture of ovarian cell populations from O. latipes. As the results, the somatic cell lines could be established only from the embryonic tissues among three tissues derived from embryos, fins and ovaries. Three embryonic cell lines were tested as a feeder cell for the culture of ovarian cell population and all three cell lines induced cell aggregation formation of the cultured ovarian cells whereas the feeder-free condition did not. Furthermore, a significant cellular proliferation was observed in the ovarian cells cultured on two of three cell lines. As a trial to increase the capacity of the cell lines as a feeder cell that supports the proliferation of the cultured ovarian cells, we subsequently established a stable line that expresses the foreign O. latipes fibroblast growth factor 2 (FGF2) from an embryonic cell line and evaluated its effectiveness as a feeder cell. The ovarian cells cultured on FGF2 expressing feeder cells still formed cell aggregates but did not show a significant increase in cellular proliferation compared to those cultured on non-transformed feeder cells. The results from this study will provide the fundamental information for in vitro culture of medaka OGSCs.

Embryonic Stem Cell-Preconditioned Microenvironment Effects on Epidermoid Carcinoma

  • Embryonic stem cell-preconditioned microenvironment is important for cancer cells properitities by change cell morphology and proliferation. This microenvironment induces cancer cell reprogramming and results in a change in cancer cell properties such as differentiation and migration. The cancer microenvironment affects cancer cell proliferation and growth. However, the mechanism has not been clarified yet. Using the ES-preconditioned 3-D microenvironment model, we provide evidence showing that the ES microenvironment inhibits proliferation and reduces oncogenic gene expression. But ES microenvironment has no effect on telomerase activity, cell viability, cellular senescence, and methylation on Oct4 promoter region. Furthermore, methylation of Nanog was increase on ES-preconditioned microenvironment and supports results that no difference on RNA expression levels. Taken together, these results demonstrated that in the ES-preconditioned 3-D microenvironment is a crucial role for cancer cell proliferation not senescence.
