  • The purpose of this work was to perform the characterization of NAD(P)H-nitroreductase isolated from Stenotrophomonas sp. OK-5 capable of degrading 2,4,6-trinitrotoluene (TNT). Initially, NADP(H)-nitroreductase by a series of purification processes including ammonium sulfate precipitation, DEAE-sepharose, andQ-sepharose was prepared. From samples harvested from fraction collector, three different fractions (I, II & III)having the enzyme activity of NAD(P)H-itroreductase were detected. Specific activities of three fractions I, II,and III of NAD(P)H-nitroreductase were determined to approximately 5.06 unit/mg, 4.95 unit/mg and 4.86 unit/mg, and concentrated to 10.5, 9.8, and 8.9-fold compared to crude extract, respectively. Among these three fractions, the fraction I of NAD(P)H-nitroreductase demonstrated the highest specific activity in this experiment. Several factors affecting on the enzyme activity of NAD(P)H-nitroreductase (fractions I, II & III) were investigated. The optimum temperature of all NAD(P)H-nitroreductase (fractions I, II & III) was 30oC, and the optimal pH was approximately 7.5. Metal ions such as Ag+, Cu2+, Hg2+ inhibited approximately 80% enzyme activity of all NAD(P)H-nitroreductase, and the enzyme activities were decreased about 30-40% inhibition in the presence of Mn2+ or Ca2+. However, Fe3+ showed stimulatory effect on the enzyme activity. The molecular weights of NAD(P)H-nitroreductase (fractions I, II & III) were measured about 27 kDa on the SDS-PAGE.

  • A new silkworm variety SooOkJam for spring rearing season is F1 hybrid between Jam157, a japanese race bred from introduction breeding and Jam158, a chinese race from introduction breeding. In the local adaptability test performed at 8 local areas in spring of 2011, the hatchability rate of SooOkJam was recorded 2% higher than the authorized silkworm variety, DaePoongJam. The larval period was recorded 12 hour shorter than DaePoongJam. The pupation percentage was recorded 0.8% higher than DaePoongJam. Single cocoon weight (2.40 g), Cocoon yield (22.6 kg) and best cocoon rate (95.5%) was similar to DaePoongJam. The concentration of DNJ was measured 4.12 mg in SooOkJam, lower than DaePoongJam (4.45 mg). Test of the artificial diet of SooOkJam showed excellent adaptability of the artificial diet for larval period from 1st to 3rd, similar to KumOkJam. In the Paecilomyces tenuipes production ability test, the pupal weight of SooOkJam was 1.61 g/individual, similar to DaePoongJam (1.65 g/individual).

  • The studies were conducted to investigate the anti-diabetic activities on the hyperglycemia induced by streptozotocin in rats, anti-hypertensive activities in SHR, anti-fatigue and decrease of body weight activities in mice by Kyung Ok-Ko water extract and drink, which is a traditional preparation in Korea. 1. The blood glucose levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease glucose levels comparing with control group. 2. The serum total cholesterol levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease total cholesterol levels comparing with control group. 3. The serum triglyceride levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease triglyceride levels comparing with control group. 4. The blood pressure in SHRs were dose-dependently lower descended by administration of Kyung Ok-Ko water extract 300, 600, and 1200 mg/kg, respectively. 5. The swimming time of Kyung Ok-Ko water extract(100, 200, 400 mg/kg) and drink(0.7, 1.4, 2.1 ml/kg) were significantly extended to all experimental group dose-dependantly. 6. The decrease of body weight of Kyung Ok-Ko water extract(100, 200, 400 mg/kg) and drink(0.7, 1.4, 2.1 ml/kg) were significantly evaluated dose-dependently in all experimental group.

  • The reaction of the heterobimetallic anion, ${(OC)_5CrMn(CO)_5^-M^+ (M^+=Na^+, PPN^+)\;with\;CH_3I$ was proven to be overall 1st order with respect to $[(OC)_5CrMn(CO)_5^-]$ This reaction mechanism may be described in terms of the consecutive reaction pathway in which Cr$(CO)_5$(THF) may be an important intermediate, leading to the corresponding products such as MeMn$(CO)_5\;and\;ICr(CO)_5^-$, accordingly. The counterion effect on this reaction was also elucidated.

  • A male 17-year-old boy was tranferred from a private hospital for persistent bilateral pleural effusion. The effusion was confirmed as bilateral chylothorax by chest CT and lymphangiography. Persistent accumulation of the chylothorax was uncontrollable more 1000cc daily ever after pleuroperitoneal shunt operation and thoracoscopic thoracic duct ligation at Rt. side. Chemical pleurodesis with OK 432 into pleural cavity through thoracostomy tube was attempted as 1.5 KE-3 KE diluted in 50ml of normal saline for 3 consecutive days resulted dramatic reduction of the drainage amount. Chemical pleurodesis with OK 432 appeared to be very helpful for management idiopathic bilateral chylothorax.
