The Measuring Experiment of Irrigation Water for Spreading Varieties of Rice Plant in Chuncheon Area (춘천지역의 장려수도품종의 용수량 측정시험)

  • This Study was carried out at the experimental Plot of Kang-Weon Province, Institute of Agriculture experiment, to find out Irrigation Water requirement and suitable calculating formula of evapotranspiration on Spreading Varieties of rice plant such as Seul Oak, Bokkwang and Teaback in Chuncheon Area. The evapotranspiration, infiltration, and consumptive use of water were measured by Micro Lysimeter for four years from '86yr. Also, yield of rice was investigated during same period. With the Kc Value taken from experimental value, evapotranspiration was calculated by methods of Blaney & criddle, Penman, Hargreaves and Thornthwaite by Computer using meteorological data in Chuncheon Area for twenty one yrs from '66yr to '86yr. All analyses were conducted based on average value of experiment for four years and the results are summarized as follows : 1) The yield by varieties through this experiment showed 1.06 times in Seul Oak, 1.94 times in Bokkwang and 1.89 times in Teaback more than Standard Yield. 2) The consumptive use of water including infiltration were 1.068.4mm in Seul Oak, 1,102. 6mm in Bokkwang and 1,195.6mm in Teaback 3) The evapotranspiration by Actual measurement presented 520.lmm in Seul Oak, 540.lmm in Bokkwang and 598.4mm in Teaback 4)The ratio of evapotranspiration and infiltration over Panevaporation showed 1.2 to 1.4. 5) The irrigation water reguirement by water balance were shown to be irrigated more than 584 mm / yr in average during 21 years from '66yr to '86yr for all Varieties and those for loyr frequency 693 mm in Seul Oak, 712 mm in Bokkwang and 728 mm in Teaback respectively. 6) Crop Coefficient (Ke Value) of the tested rice plant during the period were shown as Table 10. 7) Penman Method was the formula the most close to experiment Value among four different methods of Blaney & Griddle Penman, Hargreaves, and Thornthwaite.

Study on Clinical Effects of Oak-Mushroom Herbal-Acupuncture on Osteoarthritis in Knee Joint (표고버섯 약침의 퇴행성 슬관절염 치료효과에 관한 임상연구)

  • Objective : This study is designed to find out the effects of Oak-mushroom Herbal acupuncture on osteoarthritis in knee joint. Methods : We investigated 33 cases of patients with osteoarthritis in knee joint that are visited in Hospital of Dong-Seo oriental medicine. We evaluated knee joint function that before and after its treatment by Lysholm score and satisfaction of treatment by Baumgaertner nine point scale and visual analog scale. Results : These results showed that female patients are 26 cases and these are more 19 cases than male patients. On the age of patients, above 61 are mainly distributed as 17 cases. According to Ahlaback's grading system, grade Ⅲ are more than others as 11 cases. We estimated effects of Oak-mushroom Herbal-acupuncture by Lysholm score, those results showed that the mean of Lysholm score before treatment is 51.42, and after treatment is 76.55. That showed statistically meaningful increase. In evaluating of satisfaction, excellent are 11 cases and good are 15 cases. The percentage of above good is 78.8%. Conclusion : According to the results, Oak-mushroom Herbal-acupuncture may be effects on the increase of knee joint function and the decrease of pain in osteoarthritis in knee joint.

In vivo Pathogenicity Test of Oak Wilt Fungus (Raffaelea quercus-mongolicae) on Oriental Chestnut Oak (Quercus acutissima)

  • Since the first report of the oak wilt disease at 2004 in Korea, the disease distributed over Korean peninsula and are still giving severe damages. The management of oak wilt disease in Korea has mainly focused on the control of insect vector, Platypus koryoensis. Neverthless the effective method for evaluating the pathogenicity of the pathogen, Raffaelea quercus-mongolicae (Rqm), and for screening chemical or biological agents with strong inhibitory activity against the pathogen, is absolutely necessary, an reliable method is not available so far. This study was conducted to develop the effective method for evaluating the pathogenicity of Rqm in oak trees. The culture suspensions of Rqm were artificially injected to the saplings of Quercus acutissima by using ChemJet tree injector. Three months after treatments, the treated saplings were cut and dipped into 1% fuchsin acid solution. There were significant differences in non-conductive area (%), discoloration area (%) and vertical discoloration length between the pathogen-injected and distilled water-injected control treatments. These results indicated that the pathogen is the causal agent for the dysfunction of water conductive tissue, which will finally result in wilt symptom. Re-isolation of the pathogen and PCR detection using specific primers for the pathogen also confirmed the presence of Rqm in the sapwood chips of the pathogen-injected saplings. These observations would be greatly applied to other related researches for evaluating the pathogenicity of tree wilt pathogens and biocontrol efficacy of the selected antagonistic microorganisms, in case that the wilt symptom is not easily shown by artificial inoculation of the causal agent.

Optimization of Sawdust Media Composition and Culture Conditions for the Mycelial Growth and Primordia Formation of Agrocybe cylindracea (버들송이(Agrocybe cylindracea)의 균사생장 및 원기형성을 위한 톱밥 배지조성(培地組成) 및 배양조건(培養條件)의 최적화(最適化))

  • Some factors effecting on the mycelial growth and primordia formation of Agrocybe cylindracea were investigated. Among supplements added into sawdust, wheat bran was the best for the mycelial growth, and its appropriate concentration was 30%. A variety of sawdust preparations was tested singly and in combinations. Oak sawdust and the combination of oak and larch sawdust were the most effective for the mycelial growth. The optimal moisture content was 65%, and the range of optimal temperature was $25\:{\sim}\:28^{\circ}C$. The favorable conditions for the primordia formation appeared to be similar to those of the mycelial growth. Among supplements added into sawdust, wheat bran 30% and the combination of wheat bran and rice bran 30% were the best for the primordia formation. Oak sawdust, and the oak plus larch sawdust and oak plus poplar sawdust were the best for the primordia formation. The optimal temperature for the primordia formation was $17{\pm}2^{\circ}C$.

  • Nondestructive evaluation methods play an important role in ensuring component integrity and safety in many industries. Operator fatigue can play a critical role in the reliability of such methods. This is important for inspecting high value assets or assets with a high consequence of failure, such as aerospace and nuclear components. Recent advances in convolution neural networks can support and automate these inspection efforts. This paper proposes using residual neural networks (ResNets) for real-time detection of corrosion, including iron oxide discoloration, pitting and stress corrosion cracking, in dry storage stainless steel canisters housing used nuclear fuel. The proposed approach crops nuclear canister images into smaller tiles, trains a ResNet on these tiles, and classifies images as corroded or intact using the per-image count of tiles predicted as corroded by the ResNet. The results demonstrate that such a deep learning approach allows to detect the locus of corrosion via smaller tiles, and at the same time to infer with high accuracy whether an image comes from a corroded canister. Thereby, the proposed approach holds promise to automate and speed up nuclear fuel canister inspections, to minimize inspection costs, and to partially replace human-conducted onsite inspections, thus reducing radiation doses to personnel.

Fungal Community Analyses of Endophytic Fungi from Two Oak Species, Quercus mongolica and Quercus serrata, in Korea

  • Fungal endophytes have been recorded in various plant species with a richness of diversity, and their presence plays an essential role in host plant protection against biotic and abiotic stresses. This study applied the Illumina MiSeq sequencing platform based on the amplification of fungal ribosomal ITS2 region to analyze fungal endophytic communities of two oak species (Quercus mongolica and Q. serrata) with different oak wilt disease susceptibilities in Korea. The results showed a total of 230,768 sequencing reads were obtained and clustered at a 97% similarity threshold into 709 operational taxonomic units (OTUs). The OTUs of Q. serrata were higher than that of Q. mongolica with the number of 617 OTUs and 512 OTUs, respectively. Shannon index also showed that Q. serrata had a significantly higher level of fungal diversity than Q. mongolica. Total of OTUs were assigned into 5 fungal phyla, 17 classes, 60 orders, 133 families, 195 genera, and 280 species. Ascomycota was the dominant phylum with 75.11% relative abundance, followed by Basidiomycota with 5.28%. Leptosillia, Aureobasidium and Acanthostigma were the most abundant genera detected in Q. serrata with the average relative abundance of 2.85, 2.76, and 2.19%, respectively. On the other hand, Peltaster, Cladosporium and Monochaetia were the most common genera detected in Q. mongolica with the average relative abundance of 4.83, 3.03, and 2.87%, respectively. Our results indicated that fungal endophytic communities were significantly different between two oak species and these differences could influence responses of host trees to oak wilt disease caused by Raffaelea quercus-mongolicae.

  • The magnetism of$Fe_3O_4$ nanoparticles was applied to magnetic bio-panning process for finding specific sequences against $Fe_3O_4$ crystal phase. Vibrating sample magnetometer (VSM) measurement showed that the coercivity of 30 Oe and the saturation magnetization of 55 emu/g were sufficient in controlling particle movement and magnetizing particles in the media, respectively. This ferrimagnetism of nanoparticles practically enhanced panning efficiency by exaggerating centrifuge step and preventing particle loss. Sequencing results showed that histidine which was commonly found in peptide sequences played an important role in the binding onto $Fe_3O_4$ nanoparticle surface. However, various possible motifs were also observed from several neighboring amino acids of histidine.

  • Peptides with specific sequences against $LaPO_4$ and $TiO_2$ nanoparticles were discovered through peptide phage display technique as an application to biomolecular recognition of inorganic materials. Sequencing results showed that a motif consisting of serine and proline was commonly expressed in specific sequences. It was postulated that serine directly bound to nanoparticles using its terminal hydroxyl (OH) group. In this sense, oxygen atom seemed to work as a ligand to metal ions and hydrogen atom as a H-bond donor, was thought to bind to the oxygen atoms or the hydroxyl groups on particle surface. Also, it was expected that proline assists serine to make an ideal van der Waals contact between serine and nanoparticles, which optimizes the binding of peptide onto surface.

Effects of Horsetail, Alfalfa, Ortie, Chêne and Aleppo oak as Potential Hemostatic Agents on Laboratory Coagulation Tests

  • This study investigated the effect of ethanol extracts of horsetail, alfalfa, ortie, chêne and aleppo oak on blood coagulation in vitro. Extraction was performed by the maceration method. Extracts were mixed with platelet and plasma, then prothrombin time (PT), activated partial thromboplastin time (APTT) and platelet aggregation tests were conducted. Alfalfa extract had a dose-dependent effect on the PT. Ortie, and horsetail, reduced the PT significantly compared to control group. Alfalfa, horsetail, and ortie reduced the APTT, but their effect was insignificant compared to the control group. The pooled extract showed the highest effect compared to the single extracts in a dose-dependent manner. Horsetail and alfalfa induced platelet aggregation in response to arachidonic acid but not in response to collagen. In the case of ortie, no aggregation occurred regarding the arachidonic acid, and incomplete was observed in response to collagen. Interestingly, blood clotting occurred immediately after adding the chêne, aleppo oak and the pooled extract, and therefore platelet poor plasma (PPP) and platelet rich plasma (PRP) became jelly. Generally, chêne and aleppo oak, as well as pooled extract, were more effective in inducing both primary and secondary coagulation pathways via shortening the PT and APTT, and induction of platelet aggregation.

Phylogeny, Morphology and Pathogenicity of Biscogniauxia mediterranea Causing Charcoal Canker Disease on Quercus brantii in Southern Iran

  • Charcoal canker of oak, which has recently increased in southern Iran, could pose a serious threat to the entire forest ecosystem in the near future. In addition, it seems that climate change and its consequences, such as drought in the southern regions of Iran, have exacerbated this phenomenon. Consequently, the objective of this study was to identify the fungal pathogens that could cause charcoal canker disease in the oak forests of South Zagros. It was also sought to find associations between changes in the occurrence/exacerbation of charcoal canker disease under non and intense drought stress in non-inoculated or inoculated Quercus brantii seedlings. In total, 120 isolates were obtained from eight oak forests located in the Zagros Mountains of Southern Iran, Kohgiluyeh & Boyer-Ahmad and Fars provinces, which were classified as Biscogniauxia mediterranea based on morphological assessment. Subsequently, molecular assay confirmed the result by phylogenetic inference of internal transcribed spacer-rDNA regions, α-actin, and β-tubulin genes. The results of the pathogenicity test showed that the response of isolates of B. mediterranea (Iran-G1 and Iran-M70) was varied in different environments for the measured necrotic lesion length. In comparison with the control moisture treatments (non-stress), the necrotic lesion length in inoculated treatments increased under intense drought stress. In general, inoculated oak seedlings' exposure to water-deficient stress by the pathogen of B. mediterranea could affect the spread/severity of the charcoal canker disease.
