Chemical and Bacteriological Water Qualify of the Sonakdong River (서낙동강 강수의 화학적${\cdot}$세균학적 수질)

  • The Sonakdong river is very important water source not only for agricultural water of Kimhae field but also for fishery water. Recently the middle and lower areas of the river have the tendency to be heavily contaminated by domestic sewage and agricultural chemicals. Fifty six water samples were collected from 8 stations from July to December in 1985 for the experiment (Fig. 1). To evaluate the water quality, pH, water temperature, electrical conductivity, chloride ion, nutrients ($NO^{-}_{2}-N,\;NO^{-}_{3}-N,\;NH^{+}_{4}-N,\;PO^{3-}_{4}-P,\;SiO_2-Si$), total coliform, fecal coliform, and fecal streptococcus were determined. Range and mean value of the samples were as follows; pH $6.3{\sim}9.4$, 7.91; water temperature $6.1{\sim}34.8^{\circ}C,\;23.88^{\circ}C$; electrical conductivity (from St. A to G) $1.575{\times}10^2{\sim}30.50{\times}10^2{\mu}{\mho}/cm,\;6.57{\times}10^2{\mu}{\mho}/cm$; chloride ion $23.5{\sim}14,300mg/l$, 770.0mg/l; nitrite-nitrogen $0.007{\sim}0.110mg/l$, 0.053mg/l; nitrate-nitrogen $0.001{\sim}1.638mg/l$, 0.649 mg/l; ammonia-nitrogen $0.017{\sim}4.200mg/l$, 0.497mg/l; phosphate-phoshorus $0.011{\sim}0.281mg/l$, 0.086mg/l; and silicate-silicious $2.4{\sim}6.5mg/l$, 4.43mg/l. Electrical conductivity and chloride ion of the station F(Chomanpo) were $2.676{\times}10^2{\mu}{\Omega}cm$ and 123.99mg/l which were lower than those of others. Among the analyzed nutrients, silicate-silicious concentration was the highest through all the samples. The bacterial density of the samples ranged $36{\sim}110,000/100ml$ for total coliform, $15{\sim}46,000/100ml$ for fecal coliform and $3.6{\sim}15,000/100ml$ for fecal streptococcus. The range and the mean of the TC/FC ratio were $3.0{\sim}9.6$, 5.51 and those of the FC/FS ratio were $1.1{\sim}9.2$, 6.19, respectively. On the other hand, fecal coliform was not detected in about $78\%$ of the water samples examined. Composition of coliform was $52\%$ Escherichia coli group, $3\%$ Citrobacter freundii group, $13\%$ Enterobacter aerogenes group and $31\%$ others.

Superiority of Modifiled University of Wiscinsin Solution in the Prolonged Preservation of Isolated Rat Heart (적출 쥐 심장의 장시간 보존에 있어서 University of Wisconsin 수정 용액의 우수성)

  • The paucity of donor hearts for transplantation can be remedied by distant heart procurement. Prolonging donor heart preservation is essential for successful clinical cardiac transplantation. Thirty-two isolated rat hearts were perfused with Krebs-Henseleit buffer solution for 15 minutes, arrested and preserved at 4 oC for 4 hours, and then reperfused for 25 minutes. The following three groups were prepared and hemodynamic changes, creatine kinase-MB isoenzyme levels and ultrastructural changes of the myocardium were analysed before and after cardiac arrest. ; Group I : the heart was arrested with the cardioplegic solution [Plegisol, potassium : 16 mM, sodium : 120 mM] and then stored in a solution with ionic compositions of the extracellular fluid [Hartman, potassium : 4 mM, sodium : 130 mM] ; Group II : the heart was arrested with the cardioplegic solution and stored in a solution with ionic compositions of the intracellular fluid [Modified Euro-Collins, potassium : 108 mM, sodium : 10 mM] ; Group III : the heart was arrested with the cardioplegic solution containing adenosine 20 uM, and then stored in a solution with ionic compositions of the intracellular fluid [Modified University of Wisconsin solution, potassium : 119 mM, sodium: 23 mM]. Left ventricular developed pressure at 20 minutes of the reperfusion was significantly higher in group III [64.3 $\pm$ 3.12 mmHg, p<0.01] and group II [58.3 $\pm$ 1.55 mmHg, p<0.05] as compared with group I [51.4$\pm$ 2.78 mmHg]. The time to induce cardiac arrest after infusion of cardioplegic solution with adenosine 20 uM [5.3 $\pm$ 0.30 second, p<0.005] was significantly shorter than without adenosine [10.6$\pm$ 0.55 second]. Coronary flow at 20 minutes of the reperfusion was augmented significantly in group III [9.6$\pm$ 0.50 ml/min, p<0.05, p<0.05] as compared with group I [8.0 $\pm$ 0.41 ml/min] and group II [8.1$\pm$ 0.51 ml/min]. Percentage recovery of left ventricular developed pressure at 20 minutes of the reperfusion was significantly higher in group III [94.6$\pm$ 2.51 %, p<0.005] as compared with group II and in group II [83.1 $\pm$ 1.22 %, p<0.005] as compared with group I [69.9 $\pm$ 1.73 %], and also percentage recovery of coronary flow at 20 minutes of the reperfusion was significantly higher in group III [82.3 $\pm$ 3.86 %, p<0.05] as compared with group II [71.4 $\pm$ 3.46 %] but there was no significant difference between group I and group II. Measured level of creatine kinase-MB isoenzyme at 15 minutes of the reperfusion was significantly lower in group III [1.23 $\pm$ 0.16 ng/ml, p<0.025] and group II [1.42$\pm$ 0.10 ng/ml, p<0.05] as compared with group I [1.79 0.14 ng/ml]. In the semiquantitative evaluation of the ultrastructural changes of the myocardium, mitochondrial score was lower in group III [0.7 $\pm$ 0.21] than in group I [3.1$\pm$ 0.28] and group II [1.7 $\pm$ 0.19], and also the other structural score was lower in group III [2.7$\pm$ 0.99] than in group I [7.9 $\pm$ 0.89] and group II [5.0 $\pm$ 1.22]. In conclusion, the solution with ionic compositions of the intracellular fluid is appropriate for prolonged cardiac preservation, and it appears to be better preserving method for distant procurement when the donor heart is rapidly arrested with cardioplegic solution containing adenosine 20 uM, and then stored with Modified University of Wisconsin solution.

Influences of Roasted or Non-roasted Brown Rice Addition on the Nutritional and Sensory Properties and Oxidative Stability of Sunsik, Korean Heated Cereal Powder (현미의 볶음 처리 유무와 첨가량이 선식의 영양적 관능적 특성 및 산화안정성에 미치는 영향)

  • Six kinds of sunsik containing different contents of brown rice(BR; 20, 30, and 50%) were prepared and subjected to various processing conditions(with or without roasting at $200^{\circ}C$ for 20 min e.g., designated as RBR50 or BR50) to assess their functionality as ready-to-eat foods. They were also assessed for their nutritional and sensory properties and oxidative stability. Dietary fiber contents were proportionate to the levels of added BR. Protein was highest in RBR50 (p<0.001), which also had the highest amounts of free and structural amino acids. The amount of free amino acids tended to increase with roasting, although most amino acids were present in structural form. Oleic acid and linoleic acid were the predominant fatty acids in all prepared sunsik, and RBR50 presented noticeably higher peroxidability index due to its higher amount of linoleic acid(p<0.05). Nevertheless, RBR50 showed good oxidative stability; this phenomenon was observed in all sunsik with roasted BR but not in those with non-roasted BR. It is implied that potential antioxidants might have been newly formed or converted from their precursors while BR was roasted. Roasting process also had an impact on the sensory properties of sunsik, e.g., sunsik with added roasted BR showed lower dissolution and darker color intensity compared to its counterpart sunsik.

Blood Picture of Army Pack Horses(Jeju Horses) (군용타마(軍用馱馬)(제주마(濟州馬))의 혈액상(血液像))

  • Although considerable research has been done on the blood picture of the horse, hot-blooded and cold-blooded, little work has been made of the blood picture of the army pack horse, Jeju horse. The object of the present investigation was to make good this deficiency, and to suggest standard for the blood picture of army pack horses kept under the regular military training and the ideal feeding in the heart of a mountain. Blood samples were drawn from the jugular vein through a 15-gauge bleeding needle from 41 males and 28 females, aging 3 to 9 years old. It was taken between seven and nine o'clock in the morning. Animals were handled as quietly as possible to avoid any excitation. No restraint other than a halter was used. Enumeration of erythrocyte, total and differential leukocyte count, determination of hemoglobin in blood, and the value of packed cell volume were male in the usual manner, and erythrocytic constant was calculated by the method of Wintrobe. Erythrocyte count was $7.83{\pm}0.20(4.95{\sim}11.05){\times}10^6/mm^3$(SE). This value was much lower than hot-horses, but slightly higer than the values of cold-horse reported from foreign country. Concentration of hemoglobin in blood was $13.0{\pm}0.33(9.5{\sim}17.8)g/100ml$. This value was much higher than that of cold-horses observed by the other authors, approaching to the values of hot-horses. Packed cell volume was $32.1{\pm}0.92(22{\sim}42)ml/100ml$. This vague was a little higher than that of the other cold-horses. Mean corpuscular volume was $41.5{\pm}1.20(26.6{\sim}59.3){\mu}m^3$. This value matched so well with the other results recorded by various investigators. Mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration were $16.9{\pm}0.43(12.3{\sim}25)$ pg and $41.0{\pm}0.45(29.1{\sim}51.1)g/100ml$, respectively. These values were significantly higher than the values found by the other investigations. Total leukocyte enumeration was $10.5{\pm}0.41(5.6{\sim}17.9){\times}10^3/mm^3$, being considered as normal. And differential leukocyte count of neutrophil was $44.5{\pm}2.23(15{\sim}76)%$, $5,527{\pm}234(2,231{\sim}9,144)/mm^3$, of lymphocyte $50.5{\pm}1.19(19{\sim}77)%$, $4,307{\pm}125(1,456{\sim}11,098)/mm^3$, of monocytel (0~4)%, $105(0{\sim}352)/mm^3$, of eosiophil 3.2(0~14)%, $340(0{\sim}1,232)/mm^3$ and of basophil 0.25(0~3)%, $23(0{\sim}236)/mm^3$. The percentage of the differential count obtained from the present work showed a good agreement with the results of various authors. Of the horses examined monocyte was found from 42 horses, eosinophil from 62 horses and basophil from 10 horses. No significant differences recognized between male and female horses, and the effect of age was not observed between three to nine years old. Judging from the blood picture of the present investigation, it could be stated that the army pack horses on training were kept better than the average farming conditions.

Effects of Embryo Density on Development of In Vitro Produced Bovine Embryos (수정란의 밀도가 소 체외수정란의 체외발달에 미치는 효과)

  • This study was carried out to improve of effective culture system on development of IVM/IVF/IVC bovine embryos. The cumulus-oocyte-complexes (COCs) collected from Korean cattle ovaries harvested at a local abattoir were matured in 50 ${mu}ell$ of TCM199 supplemented with 10% fetal bovine serum (FBS) and hormones (35 $\mu\textrm{g}$/$m\ell$ FSH, 10 $\mu\textrm{g}$/$m\ell$ LH, 1$\mu\textrm{g}$/$m\ell$ estradiol 17 $\beta$ under paraffin oil at 39$^{\circ}C$ in a humidified atmosphere of 5% $CO_2$in air. At 24 hrs after culture, matured oocytes were fertilized in vitro for 22~24 hrs with motile semen in which obtained by centrifugation of a frozen thawed semen on Percoll-density gradients (45% vs. 90%) at 500 g for 20 min. The presumptive zygotes were divided into three experimental groups. Single egg (Group 1), 25 (Group 2) or 50 eggs (Group 3) were cultured on cumulus cell in 50 ${mu}ell$ TCM199 supplement with 10% FBS for 6~9 days after fertilization. In vitro developmental rates into the blastocysts in the groups 2 and 3 were significantly (P<0.05) higher than those of group 1 (37,27 vs. 6%, respectively). Cell number of blastocysts obtained in groups 2 and 3 at day 8 were significantly (P${mu}ell$) resulted in higher developmental competence and cell number of bovine blastocysts produced in vitro than those the culture of single embryos with cumulus cells.

Species Diversity of Riparian Vegetation by Soil Chemical Properties and Water Quality in the Upper Stream of Mankyeong River (만경강 상류 수질 및 식생분포와 토양환경에 따른 하천식생의 종 다양성)

  • This study was conducted to evaluate influence of chemical properties in the riparian on the species diversity and to get plant information for enhancement of natural purification in Mankyeong River. The concentration of total nitrogen was high in Jeonju and Sam stream, while that of total nitrogen showed the highest peak in Winter. Concentrations of $NH_4-N$ was $0.01{\sim}0.06\;mg/L$ in Gosan and Soyang stream. The water quality of upstream along with Mankyeong River was suitable for the irrigation source. The riparian vegetation was investigated by Zurich-Montpellier school's method from June, 2001 to September, 2002. The number of riparian plants were 59 families, 129 genera, 165 species, 20 varieties in Gosancheon, on the while 53 families, 111 genera, 141 species, 19 varieties in Soyangcheon. The number of riparian plants in Bari basin was higher than that of other sites namely, 73 families, 134 genera, 218 species, 33 varieties. Riparian vegetation was consisted of 12 plant communities. The contents of organic matter, total nitrogen and electrical conductivity had negative relationship with species diversity (Species richness index, Heterogeneity index, Species evenness index Species number). On the while, species diversity had positive relationship with soil pH. Species diversify of the plant communities were affected by topography and disturbance.

THE COMPARATIVE STUDY FOR THE SHEAR BOND STRENGTH OF DENTAL ADHESIVES CURED WITH VARIOUS LIGHT SOURCES (다양한 광원으로 중합한 치과용 접착제의 전단강도에 관한 비교 연구)

  • The objective of this study was to compare the shear bond strengths of five dentin adhesive systems cured with three different light curing sources. Seventy five noncarious permanent teeth were collected and stored in an 0.1% thymol solution at room temperature after extraction. The tested adhesives were: Adper Scotchbond Multi-purpose Plus Adhesive (SM) Adper Single bond 2 (SB), Clearfil SE Bond (SE), Adper Prompt L-Pop (PL), G-Bond (GB). And three light curing unit systems were used: Elipar Free light 2(LED), OptiLux 501 (Halogen), Flipo (PAC). For the shear bonding test, the labial and lingual surfaces of permanent teeth were used. To obtain a flat dentin surface, the labial and lingual surfaces of the teeth were sanded on SiO2 with number 600 grit and then divided into 15 groups of 10 surfaces each. All samples were theromocycled in water $5^{\circ}C$ and $55^{\circ}C$ for 1000 cycles. The results were as follows: 1. When cured with Freelight 2, the shear bond strength of SM was significantly higher than that of PL, GB (p<0.05), whereas no significant difference was found among those of any other bonding agents. 2. When cured with Optilux 501, the shear bond strength of SM was significantly higher than those of any other bonding agents (p<0.05), whereas no singnificant difference was found among those of andy other bonding agents. 3. When cured with Flipo, the shear bond strength of SM was significantly higher than those of SB, SE, GB (p<0.05), whereas no significant differences was found among those of any other bonding agents. 4. For comparison according to three different light cure unit system, except SB and GB, each three dentin bonding agents showed no significant difference. For SB, only Freelight 2 was significantly higher than the others, with no significant difference between Optilux 501 and Flip. For GB, Statistically significant difference was found only between Freelight and Flipo.

Evaluation of free radical scavenging and pancreatic lipase inhibitory effects of Aquilaria agallocha extracts (침향 추출물의 라디칼 및 췌장 지방분해 효소저해 활성 평가)

  • In a continuing screening of selected medicinal plants native to South Korea, the antioxidant and pancreatic lipase inhibitory activities of an aqueous methanolic extract from the heartwood of Aquilaria agallocha were investigated. Eighty percent of the methanolic extract of A. agallocha was further divided into $CH_2Cl_2$, EtOAc and n-BuOH in order to yield four solvent-soluble portions, namely $CH_2Cl_2$-soluble, EtOAc-soluble, n-BuOH-soluble and $H_2O$ residue. The antioxidant properties were evaluated by employing radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals, while the anti-obesity efficacy of A. agallocha extracts and solvent-soluble portions were tested by porcine pancreatic lipase assay. All tested samples showed dose-dependent radical scavenging and pancreatic lipase inhibitory activities. Among the tested extracts and solvent-soluble portions, the $CH_2Cl_2$-soluble portion showed much higher radical scavenging activity and pancreatic lipase inhibitory properties when compared with other solvent-soluble portions. This result suggested that there was a significant relationship between the total phenolic content and biological efficacies, and A. agallocha extract might be considered as a new potential source of natural antioxidants and as a pancreatic lipase inhibitory source. A more systematic investigation of this biomass will be performed for further investigation of activity against antioxidative and anti-obesity effects.

  • Thiobencarb retarded the growth of new leaves in only barnyardgrass under dry condition while under water condition the shoot growth of broadcast rice, and both shoot and root growth of barnyardgrass was considerably inhibited. Root elongation of rice and barnyardgrass was severely inhibited only under water condition, while that of transplanted rice was slightly inhibited. Inhibition of shoot and root growth in broadcast, drilled rice and barnyardgrass under water condition was much higher than that under dry condition, whereas the inhibition was less in transplanted rice than direct seeded rice. Microscopically, thiobencarb severely inhibited shoot growth and development of barnyardgrass by inducing tubular-like leaves. The cells of the shoot apices of treated barnyardgrass seedling under dry condition was vacuolated and irregularly arranged. Under water condition, leaf primordia of broadcast rice was constricted, barnyardgrass showed tubular-like leaves, inhibited apices elongation and vacuolated cells(visually lack cytoplasm).

Effects of Natural Complex Food on Specific Enzymes of Serum and Liver and Liver Microstructure of Rats Fed a High Fat Diet (지방간 환자를 위한 생식용 천연복합식품이 고지방식이를 급여한 흰쥐의 혈청, 간장의 효소 및 간조직 구조에 미치는 영향)

  • In order to design and develop a product that can treat the fatty liver, natural complex food with all natural ingredients was developed and supplemented to rats with high fat diet to induce fatty liver. As a result, when the amount of natural complex food was increased in diet of subjects, the activities of the blood serum AST, ALT, ALP, 3-GT and LDH were decreased. The total protein concentration levels of the 30% and the 50% natural complex food groups did not show changes in respect to the control group, but the 100% natural complex food groups showed significant decrease (p<0.05). Likewise, the amount of blood serum albumin in the 30% and the 50% natural complex food groups did not show improvement, but the 100% natural complex food did showed significant changes (p<0.05). The amount of blood serum triglyceride decreased as the amount of natural complex food was increased. In order to investigate the appearances of the accumulated fat in the liver, the animals were dissected. Livers of the control group (no natural complex food) were appeared as a white color, which means serious fat accumulation. However, all the natural complex food groups (30,50 and 100% natural complex food) showed noticeable decrease of fat content. Even the histology showed that livers of the control group had expansion of the fat, but a11 the natural complex food groups had e decreased as the contents and continued to show destroyed fatty cells. By observing the biological numeric data, the physical appearance and the history of the fatty liver, it is highly expected that natural complex food is very effective in treating the liver damaged -by the to fat and the cholesterol.
