  • Five flavonoids isolated from the ethyl acetate fraction of Cedrela sinensis A. Juss. were identified by high performance liquid chromatography. Separation was achieved by reversed phase chromatography on ${\mu}-bondapak$ C18 column with isocratic elution method. The content of the major flavonoid, quercitrin was about 9.48%(w/w) and 37.06%(w/w) for the methanol extract and ethyl acetate fraction, respectively.

  • Gene expression levels of choriogenin, vitellogenin and estrogen receptor were determined using Reverse transcription (RT)-PCR technique after exposure to estrogenic chemical bisphenol A in the Korean wild medaka (Oryzias sinensis). These genes have been known to be induced in male test fish when the fish are exposed to estrogenic chemicals. Therefore they can be suggested as a possible biomarker of endocrine disruption in fish, however, relatively little has been known about these genes expression by estrogenic chemicals in Korean wild fish. Mature male Oryzias sinensis were treated with bisphenol A at nominal concentrations of 0.02, 0.2 and 2 mg/L for 6 days and total RNA was extracted from the livers of treated fish for RT-PCR. When the five biomarker genes were amplified by RT-PCR in the same condition, mRNA induction level of each gene was elevated with different sensitivities. Conclusively, the results of this work indicated that measurement of vitellogenin and choriogenin using RT-PCR is effective as a simple tool for the screening of estrogenic chemicals and suggested that O. sinensis would be a suitable model fish for the environmental risk assessment of potential endocrine disruptors.

First reliable record of a stingray, Hemitrygon izuensis (Nishida and Nakaya, 1988) (Chondrichthyes: Dasyatidae) from Korea (한국산 색가오리과(Dasyatidae) 어류 1미기록종, Hemitrygon izuensis (Nishida and Nakaya, 1988))

  • Two specimens of Hemitrygon izuensis (395.8-471.5 mm in disc width), belonging to the family Dasyatidae (Myliobatiformes), were collected from the waters off north-eastern Jeju-do Island in June 2016 and Pohang in August 2019. This species was characterized by having no spots on dorsal side of disc, white ventral tail fold and short preorbital snout length (15.6-18.3% of disc width). This species is similar to H. akajei and H. sinensis reported in Korea, but differs in small denticles on mid-line of dorsal surface of disc (absent in H. izuensis vs. present in H. akajei and H. sinensis), the length of ventral tail fold (28.1-31.0% in H. izuensis vs. 48.0-48.6% in H. akajei vs. 45.0% in H. sinensis), color of ventral tail fold (white in H. izuensis vs. black in H. akajei and H. sinensis) and small tubercles on the posterior part of tail (absent in H. izuensis vs. present in H. akajei and H. sinensis). We suggest a Korean name for H. izuensis as "I-ju-gal-saeg-ga-o-li" following Kim et al. (2019).

Extract of Cedrela sinensis Leaves Protects Neuronal Cell Damage Induced by Hydrogen Peroxide in Cultured Rat Neurons (과산화수소수로 유도된 배양신경세포손상에 대한 참죽나무잎 추출물의 보호효과)

  • Dried leaves from Cedrela sinensis A. Juss. (CS), have been observed to possess various pharmacological activity and contain various antioxidant constituents. The protective effect of ethanol extract of CS on hydrogen peroxide $(H_2O_2)-induced$ neurotoxicity was examined using primary cultured rat cortical neurons in the present study. Exposure of cultured neurons to 100 ${\mu}M\;H_2O_2$ caused a significant neuronal death as assessed by a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and Hoechst 33342 staining. The addition of CS, over a concentration range of 10 to $50{\mu}g/m{\ell}$, concentration-dependently prevented the $H_2O_2-induced$ neuronal apoptotic death. CS $(50{\mu}g/m{\ell})$ significantly inhibited $H_2O_2-induced$ elevation of the cytosolic $Ca^{2+}$ concentration $([Ca^{2+}]_c)$, which was measured by a fluorescent dye, Fluo-4 AM. CS (30 and $50{\mu}g/m{\ell})$ inhibited glutamate release and generation of reactive oxygen species (ROS) induced by $100{\mu}M\;H_2O_2$. These results suggest that CS may mitigate the $H_2O_2-induced$ neurotoxiciy by interfering with the increase of $[Ca^{2+}]_c$, and then inhibiting glutamate release and generation of ROS in cultured neurons.

Effects of the Cedrela sinensis A. Juss. Leaves on the Alcohol-Induced Oxidative Stress in the Human Hepatic HepG2 Cells (알코올을 처리한 HepG2 세포에서 참죽나무 잎 추출물의 세포 보호 및 항산화 효과)

  • All the parts of the Cedrela sinensis A. Juss., including the seeds, roots, and leaves, have been known to exert medicinal effects. The C. sinensis and its major compound, quercetin, were previously reported to exhibit the anti-inflammatory and anti-oxidative activities. However, the hepatoprotective effects of the C. sinensis leaves against the alcohol-induced oxidative stress in the HepG2 cells have not been studied. In this study, we investigated the antioxidant activities and analyzed the flavonoid contents of the C. sinensis-leaf extract (CE). The total flavonoid contents of the CE is 1,874.5 mg/100 g dry weight (DW), while the total quercetin 3-O-rhamnoside (quercitrin) contents, which was identified as the major flavonol in the CE, is 1,456.0 mg/100 g DW. In the ethanol-stimulated HepG2 cells, the CE effectively prevented the cytotoxic effect and increased the gene expression of the antioxidant enzymes, such as the heme oxygenase-1 (HO-1) and the glutathion peroxide (GPx). The level of the reactive oxygen species (ROS) production was significantly decreased in the CE-treated HepG2 cells. In conclusion, the C. sinensis extract suppressed the alcohol-induced oxidative stress in the HepG2 cells via the induced GPx and HO-1 gene expressions. It is expected the CE positive effects will likely be attributed to the flavonoids, like the quercetin, within the CE.

  • The anti-inflammatory and analgesic action of the components isolated from some edible plants carried out. We isolated the quercitrin, compound 3 and o-counmaric acid from Cedrela sinensis, Oenanthe javanica and Artemisia princeps var. Orientalis respectively. O-coumaric acid showed the inhibitory effect on carrageenin-induced edema as well as vascular permeability in mice. And we also found the analgesic activity in all compounds isolated from these plants.

  • The seed of cowpea (Vigna sinensis K.) was extracted with 80% aqueous methanol (MeOH). And the concentrated extract was partitioned with ethyl acetate (EtOAc), n-butanol (n-BuOH) and $H_2O$, successively. The repeated silica gel and octadecyl silica gel (ODS) column chromatographic separations for the EtOAc and n-BuOH fractions led to isolation of four sterols. And the chemical structures of the compounds were determined as a mixture of stigmasterol and $\beta$-sitosterol with the ratio of 4 to 3 (1), 7-ketositosterol (2), and stigmasterol 3-O-$\beta$D-glucopyranoside (3) from the interpretation of spectroscopic data including nuclear magnetic resonance (NMR) spectrum metric, mass (MS) spectrum metric and infrared (IR) spectroscope. This study reports the first isolation of $\beta$-sitosterol, 7-ketositosterol, and stigmasterol 3-O-$\beta$-D-glucopyranoside from the seed of Vigna sinensis K. In addition, compound 2, 7-ketositosterol, is rarely occurred in natural source including plant.

  • 틸보꼬마풀게 (Hemigrapsus sinensis Rathbun) (바위 게 과, 참게 아과)의 유생을 부추에서부터 첫번째 게퇘까지 수온 20.2$^{\circ}C$에서 24.1$^{\circ}C$, 염분농도 32,9도에서 33.2%o에서 사육하고, 각 유생기의 형태적인 특징을 기술 및 도시하여, 같은 속내에 보고된 다른 7종의 유생들과 kk교하였다. 이 종은 5기의 Zoea와 1기의 Megalopa 유생을 거쳐 첫번째 게탰로 변태하였다. 제 1 Zoea기의 제 1촉각과 미절이 Aikawa의 기준에 의한 공형이며 갑측극은 있고 제 2소악내지의 강량식이 2.2인 점에서는 같은 속의 다른 7종과 일치하나, 제 1악각의 기절 및 내파와 제 2악각의 내지의 강모식에 약간의 차리가있다.

The Anthelmintic Principle of "O-Mae", the Roasted Fruits of Prunus mume, Against Clonorchis sinensis (오매의 간디스토마 살충성 물질에 관한 연구)

  • The anthelmintic substance of the roasted fruits of Prunas mume against Clonorchis sinensis was isolated and its structure was identified by chemical and physical methods. The results obtained from the experiments are as follows: 1) The methanol extract of the roasted fruits of P. mume was fractionated into four parts: petroleum ether, ethyl ether, ethyl acetate and water soluble part. Among these, etherial fraction was found to have strong wormicidal effect on liberated metacercaria of Clonorchis sinensis. 2) From the etherial fraction, the wormicidal substance was isolated by silica gel, polyamide and sephadex column chromatography and identified to be 5-hydroxymethylfurfural (5-HMF) by chemical and spectral data. 3) 5-HMF was synthesized and administered to the rabbits infected with C. sinensis. On the 2nd day after administration, the EPG (eggs per gram in feces) reached to the maximal value. Among the adult worms isolated from the bile duct of the treated animal, 84% of worms were damaged morphologically. 4) The content of 5-HMF in the fruits of P. mume which were roasted in an oven at $90-110^{\circ}C$ for 52 hours and that in the fresh fruits was evaluated by HPLC. The content of 5-HMF was 0.8% in the roasted fruits and 0.02% in the fresh ones.

Inhibitory Effects of Various Edible Plants and Flavonoids from the Leaves of Cedrela sinensis on Human Immunodeficiency Virus Type 1 Protease

  • The inhibitory effect of extracts from 15 edible plants on the protease of human immunodeficiency virus (HIV) type 1 was investigated. Protease activity was determined by incubating the extracts in a reaction mixture containing protease and substrate His-Lys-Ala-Arg-Val-Leu-(p-NO$_2$-Phe)-Glu-Ala-Nle-Ser-NH$_2$ to inhibit proteolytic cleavage. Of various plants tested, the leaves of Cedrela sinensis inhibited the HIV-1 protease by 42% at a concentration of 100$\mu\textrm{g}$/ml. A major flavonoid isolated from the leaves of C. sinensis, quercetin 3-O-$\alpha$-L-rhamnoside showed inhibitory activity of 19% at a concentration of 100$\mu$M.
