Glucosylation of Isoflavonoids in Engineered Escherichia coli

  • A glycosyltransferase, YjiC, from Bacillus licheniformis has been used for the modification of the commercially available isoflavonoids genistein, daidzein, biochanin A and formononetin. The in vitro glycosylation reaction, using UDP-${\alpha}$-D-glucose as a donor for the glucose moiety and aforementioned four acceptor molecules, showed the prominent glycosylation at 4' and 7 hydroxyl groups, but not at the $5^{th}$ hydroxyl group of the A-ring, resulting in the production of genistein 4'-O-${\beta}$-D-glucoside, genistein 7-O-${\beta}$-D-glucoside (genistin), genistein 4',7-O-${\beta}$-D-diglucoside, biochanin A-7-O-${\beta}$-D-glucoside (sissotrin), daidzein 4'-O-${\beta}$-D-glucoside, daidzein 7-O-${\beta}$-D-glucoside (daidzin), daidzein 4', 7-O-${\beta}$-D-diglucoside, and formononetin 7-O-${\beta}$-D-glucoside (ononin). The structures of all the products were elucidated using high performance liquid chromatography-photo diode array and high resolution quadrupole time-of-flight electrospray ionization mass spectrometry (HR QTOF-ESI/MS) analysis, and were compared with commercially available standard compounds. Significantly higher bioconversion rates of all four isoflavonoids was observed in both in vitro as well as in vivo bioconversion reactions. The in vivo fermentation of the isoflavonoids by applying engineered E. coli $BL21(DE3)/{\Delta}pgi{\Delta}zwf{\Delta}ushA$ overexpressing phosphoglucomutase (pgm) and glucose 1-phosphate uridyltransferase (galU), along with YjiC, found more than 60% average conversion of $200{\mu}M$ of supplemented isoflavonoids, without any additional UDP-${\alpha}$-D-glucose added in fermentation medium, which could be very beneficial to large scale industrial production of isoflavonoid glucosides.

  • In this work was produced and analyzed a z-cut Ti:LiNbO$_{3}$ optical waveguide which applies for various optical devices.A waveguide channel with a thickness 8 .mu.m and a length 66,000.mu.m and a mach-zehnder interferometer type waveguide were fabricated at a diffusion temperature 1050.deg. C for 6-8hours in a wet $O_{2}$ environment. The resulting Ti:LiNbO$_{3}$ optical waveguide was measured to have a Ti-strip thickness of 950.angs. and low loss. Surfaces and cross-sections of a fabricated waveguide were analysed. The mode pattern anaysis revealed that the waveguide showed a single mode at a 1550nm wavelength. The effective dimension of the waveguide was calculated by measuring a gaussian profile; Wx=10.95.mu. and Wy=9.14.mu.m. a propagation loss, of 0.50dB/cm for a TM mode and 0.45dB/cm for a TE mode, was low enough to be accepatable for optical devices.

사람의 O-linked N-acetyl-$\beta$-D-glucosaminidase 유전자의 분석과 재조합 발현 (Analysis of Human O-GlcNAcase Gene and the Expression of the Recombinant Gene.)

  • 세포질과 핵단백질의 serine과 threonine 잔기에 O-linked N-acetyl-$\beta$-glucosamine (O-GlcNAc)의 첨가는고등 진핵 세포에서 흔히 일어나는 번역 후 단백질의 변형 중 하나로서 단백질의 인산화와 유사한 세포 내 신호전달에 관여하는 것으로 보인다. O-GlcNAc의 첨가와 제거는 O-GlcNAc transferase (OGT)와 O-linked N-acetyl-$\beta$-D-glucos-aminidase (O-GlcNAcase) 효소에 의해 각각 촉매된다. 두가지 종류의 사람 유래 O-GlcNAcase 유전자(O-GlcNAcase, v-O-GlcNAcase)를cloning하고 세 가지의 융합단백질로 대장균에서 생산을 시도하였다. O-GlcNAcase의 기질 유사체 인 ${\rho}$-nitrophenyl-N-acetyl-$\beta$-D-g1ucosaminide (${\rho}$NP-$\beta$-D-GlcNAc)를 기질로 사용하여 효소활성을 측정 한 결과 v-O-GlcNAcase는 활성을 나타내지 않았다. 여러 종류의 amino sugar 기질 유사체를 사용하여 O-GlcNAcase의 활성을 측정하였으나 오직 ${\rho}$NP-$\beta$-D-GlcNAc만이 활성을 보였다. Blast검색으로 분석한 결과 아미노 말단의 hyaluronidase-like domain (hyaluronidase-유사 영역)과 카르복시 말단의 N-acetyltransferase 영역 두 곳의 conserved domains 존재하였다. 효소촉매에 중요한 영역을 밝히기 위해 여러 deletion mutants(결손 변이체)를 제작한 후 효소활성을 측정하고 Western blot으로 분석하였다. Hyaluronidas-유사 영역, 유전자 내부와 N-acetyltransferase 영역을 제거할 경우 효소활성이 사라졌으나 아미노 말단의 55개 아미노산과 카르복시 말단의 truncation은 활성을 일부분 유지하였다. 위의 사실에 기초하여 hyaluronidas-유사 영역은 효소활성에 중요하고 카르복시 말단의 N-acetyltransferase 영역은 조절기능으로 작용하는 것으로 추정된다.

목통의 페놀성 화합물의 항산화 활성 및 HPLC-UV 동시 함량분석 (Anti-oxidant Activity of Phenolic Compounds and Simultaneous Contents Determination using HPLC-UV from the Akebiae caulis)

  • Akebiae caulis have been used in folk medicines for diuretic, menstrual pain, and diuretic pain. It has been also resolved nephritis and cystitis. In this study, we isolated three phenolic compounds from 70% methanol extract using the open column chromatography. These isolated compounds which were 2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside, 5-O-caffeoylquinic acid, and syringoylglycerol 2-O-${\beta}$-D-glucoside were determined by physico-chemical apparatus. Furthermore, we conducted DPPH and ABTS assay in order to screen the antioxidant activity of isolated three compounds. Also, we developed a rapidly HPLC-UV analysis method of two phenolic compounds (2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside, 5-O-caffeoylquinic acid) for evaluating the Akebiae Caulis collected 30 samples from different regions. From the experiments, all three isolated compounds showed the significant antioxidant activity. We suggested that the content criteria of marker compounds were shown by a simple and rapid HPLC-UV method. The contents respectively were 0.009% (2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside) and 0.036% (5-O-caffeoylquinic acid).

Orostachys japonicus DW and EtOH Extracts Induce Apoptosis in Cholangiocarcinoma Cell Line SNU-1079

  • Objectives: This study was performed to investigate the anti-tumor effect of O. japonicus extracts on intrahepatic cholangiocarcinoma cell line SNU-1079. Methods: Cholangiocarcinoma SNU-1079 cells were treated with various concentrations of O. japonicus DW and EtOH extracts ($0-300{\mu}g/ml$) for 24, 48 or 72 h. Cell viability was evaluated through a PMS/MTS assay, and the apoptosis rate was examined through ELISA assay and flow cytometry analysis. The mRNA expression of apoptosis- and cell cycle progression-related genes (Bcl-2, Mcl-1, Bax, Survivin, Cyclin D1, and p21) was evaluated using real-time PCR, and the caspase activity was examined using immunoblot analysis. Results: O. japonicus extracts inhibited cell proliferation and increased apoptosis rate in both ELISA assay and flow cytometry analysis. O. japonicus extracts decreased Bcl-2, Mcl-1, Survivin, and Cyclin D1 mRNA expression and increased Bax mRNA level. O. japonicus extracts also increased Caspase-3 activation. Overall, O. japonicus DW extracts were more effective than EtOH extracts. Conclusions: O. japonicus inhibited cell proliferation and induced apoptosis in SNU-1079 cells via mitochondria -mediated intrinsic pathway, which leads to Caspase-3 activation. The results indicate that O. japonicus is a potential therapeutic herb with anti-tumor effect against intrahepatic cholangiocarcinoma.

GaN 단결정 분말을 이용한 $Ga_2O_3$ 합성 및 구조 특성 ($Ga_2O_3$ synthesis using GaN mono-crystal powder and its structural properties)

  • $Ga_2O_3$ is associated with the fabrication of thin window layer of solar cell. Usually, $Ga_2O_3$ is synthesized from Ga-metal oxidation method and GaN mono-crystal heat treatment method. We synthesized $Ga_2O_3$ powder using two methods and analyzed powder using latter method compared with powder by former method. XPS, XRD, IR analysis are conducted. XPS result, surface of GaN powder is almost oxidized to $Ga_2O_3$ at $1124^{\circ}C$ heat treatment and XRD and IR result, the inside of GaN powder is dramatically oxidized at $1124^{\circ}C{\sim}1300^{\circ}C$.

송도국제도시 연결도로의 유고상황 발생에 따른 신도시 내부 영향 분석 (Analysis of the effect in the city due to the bridges incidents in Songdo International City)

  • The purpose of this study is to analysis the impact on the inside of the new city when an incidents occurs on the Songdo International City connecting road, which has a limited access. The analysis data used KTDB's O/D and network data of the Seoul metropolitan area. In addition, the scenario composition applied a method of reducing the number of lanes on the road according to the situation of incidents, targeting bridges advancing from Songdo International City to the outside in the morning peak hours. The analysis method analyzed the traffic volume, total travel time, total travel kilometer, and route change in the new city based on the results of the traffic allocation model. As a result of the analysis, the range of influence was shown to two types. First, of the seven bridges, Aam 3, Aam 2, and Aam 1 were analyzed to have an impact only in some areas of the northwestern part of the new city. On the other hand, the remaining bridges were analyzed to affect the new city as a whole. The analysis results of this study are expected to be used as basic data to establish the scope of internal road network management when similar cases occur in the future.

레일레이 페이딩 환경하에서 적응형 변조기를 적용한 다중 반송파 CDMA 시스템의 성능 분석 (Performance Analysis of Multicarrier CDMA System with Adaptive Modulators in Rayleigh Fading Channel)

  • 본 논문은 레일레이 페이딩 채널 환경하에서, 적응형 변조기와 적응형 부채널 할당 방식을 적용한 다중 반송파 CDMA 시스템의 성능을 분석한다. 제안한 적응형 변조기는 QPSK, 16 QAM, 64 QAM, 256 QAM으로 구성하고, 신호점은 그레이 코드(Gray Code)로 나타내며, 임계값은 비트 에러율(BER)이 1 %일 경우의 평균 Eb/No를 사용한다. 적응형 부채널 할당 방식을 적용한 다중 반송파 시스템은 사용자의 DS파형을 L개의 부채널 중 가장 큰 K개의 부 채널로 선택하여 전송한다. 제안한 시스템의 경우, 총 부 채널수가 4이고 이 가운데 두 개의 부 채널로 데이터를 전송한다면 각 채널의 임계값은 5.2 dB, 9 dB, 13.2 dB와 8.4 dB, 12.2 dB, 16.3 dB 나타났다. BER이 $10^{-3}$을 만족하는 평균 $E_{b/}$ $N_{o}$ 는 8.1 dB로 기존의 시스템에 비해 12.9 dB의 성능이 개선되었다. 평균 심볼당 비트수(BPS)는 7 bit의 경우 15 dB의 평균 $E_{b/}$ $N_{o}$ 가 필요하다. 부 채널 에러가 발생한다면 BER이 $10^{-3}$을 만족하는 평균 $E_{b/}$ $N_{o}$ 는 13.6 dB로 5.5 dB의 성능 저하가 일어났다. 일어났다.

한.양방 협진체계 개발에 관한 연구 (A Study on Cooperative Medical Treatment System between Oriental and Western Medical Practitioners)

  • A study on cooperative medical treatment system between oriental and western medical practitioners was conducted from March through August 1997 in order to determine a productive model of cooperation of two medical parts for treatment of patients. Questionnaires were distributed to 195 medical doctors(M.D.) and 195 doctors of oriental medicine(O.M.D.) working in clinics and hospitals in six major cities. Statistical analysis tools used for this study were frequency, t-test and multiple regression by using SPSS/P.C package. The results are summarized as follows: 1. The respondents were composed of male docotors(78%) and female doctors(22%) and 68.2% of M.D. and 97% of O.M.D. were interested in the other medical part. The doctors of both disciplines had some limitation on treatment of patients but they were reluctant to refer their patients to other doctors in different discipline. 2. M.D assumed that oriental medicine was more effective for chronic diseases, and O.M.D. thought that western medicine was more effective for acute diseases. 92.3% of O.M.D.s responded that even though they do not utilize western medical technology for diagnosis, they believed the results of diagnoses by western medical technology. 3. 60.5% of O.M. and 93.3% of O.M.D. said that cooperative medical treatment system could be necessary for patients and it would be effective for control of diseases and 69% of western medical doctors and 99% of oriental medical doctors agreed that oriental medical practice would be more effective for cerebellar vascular accidents than other diseases. 77.4% of western medical doctors and 92.3% of oriental medical doctors responded that the students of two different disciplines have to be taught on the other disciplines. 82.6% of western medical doctors and 83.3% oriental medical doctors agreed that it would be necessary to have collaborative research between scientists in two disciplines. 81.5% of M.D.s and 93.3% of O.M.D.s believed that they had prejudice and distrust on other discipline 4.90% of the doctors were not satisfied with the government medical policies on health insurance, legal matters, and health delivery system. 5. 75.4% of M.D.s and 50.2% of O.M.D.s said that two medical disciplines should be integrated, but they were skeptical on that. 75.3% of M.D.s thought that western hospitals should employ O.M.D.s Finally this study recommended that western medical students study oriental medicine and the vice versa, and M.D.s and O.M.D.s should have seminars and workshops to exchange knowledge, and experiences. It is also recommended that medical laws be revised to allow medical doctors refer the patients whom they can not handle to oriental medical doctors and the vice versa.

A New Flavonol Glycoside from Tristemma hirtum (Melastomataceae)

  • Chemical investigation of the plant Tristemma hirtum P. Beauv (Melastomataceae) resulted to the isolation of a new flavonol glycoside named quercetin-7-O-${\alpha}$-D-arabinofuranoside (1), together with nine known compounds including 3'-hexadecanoyl-2'-(9aZ)-tetradecanoyl-glycerol 1'-O-[${\beta}$-D-galactopyranosyl-(1'' ${\rightarrow}$ 6'')-${\alpha}$-D-galactopyranoside] (2), arjunolic acid (3), ${\beta}$-sitosterol-3-O-${\beta}$-D-glucopyranoside (4), terminolic acid (5), quercetin (6), asiatic acid (7), maslinic acid (8), $1{\beta}$-O-galloylpedunculagin (9) and 6-hydroxyapigenin 7-O-${\beta}$-D-glucopyranoside (10) from the methanol extract using normal and reversed phase column chromatography. The structures of these compounds were determined by comprehensive interpretation of their spectral data mainly including 1D- 2D-NMR ($^1H-^1H$ COSY, HSQC, and HMBC) spectroscopic and ESI-TOF-MS mass spectrometric analysis.
