Effects of native and RS-3 type resistant starches prepared from autoclaved-cooled amylomaize VII(AVII) and normal maize starches(NMS) on the rheological and baking properties of wheat flour dough and quality of breads were investigated. In farinogram, water absorption and dough development time were increased, but stability was reduced by the addition of RS. The addition of native starches or resistant starch made from AVII to wheat flour improved the total volume and specific loaf volume of bread compared with the control(no addition), but the addition of NMS-RS reduced those. During the storage of bread at room temperature, the moisture content of control was decreased but those of native starch- or RS-added breads remained constantly. AVII-RS- or NMS-RS-added bread was evaluated to have good overall acceptability compared with control by elementary school students. The addition of enzyme-resistant starch to bread regardless of botanical sources of starch not only improved the overall acceptability and nutritional benefits but also improved the sensory acceptability.
A food frequency questionnaire (FFQ), which can be used in studies investigating relationship between diet and chronic diseases in a rural area of Korea, was developed and validated. Food items were selected from two sources ; (1) preliminary survey in a rural area by 24-hour recalls and (2) National Nutritional Survey of 1991. Sixty-five food items were finally selected based on the frequency of consumption and contributions to major nutrient intakes. Portion size of each food item was determined considering the mean and median values of the amounts consumed by subjects in the preliminary survey. Frequency of consumption was asked in nine categories ranging from 'more than three times a day' to 'almost never'. The newly developed FFQ was administered in 24-hour recalls conducted in different seasons in the same area after the administration of FFQ. Sixty-one subjects completed both FFQ and all three repeated 24-hour recalls. The results of the two different survey methods showed that mean daily intake levels of energy, carbohydrate, vitamin A and vitamin C were significantly higher in FFQ compared to 24-hour recalls(p<0.05). Intake levels of energy, protein, fat, carbohydrate and iron were significantly correlated by Pearson's correlation coefficients(p0.05). Ranking on nutrient intake of the subjects by two method were significantly correlated(Spearman's correlation coefficients) in all above nutrients plus Ca and vitamin A. Percentage of subjects in the lowest or in the highest quintile by 24-hour recalls who belong to the nearest two categories by FFQ ranged from 46% to 83%, while the percentage falling into the opposite category were below 10% in most of the nutrients. The proportion of subjects classfied into the same quintiles by the two methods were between 23% and 33%, with an average of 28%. From the results, FFQ developed in this study seems to be useful in evaluating nutrient intake pattern of 1 year in adults living in rural area of Kyonggi province.
Although nutrition education for pregnant women is important, few such programs have been carefully examined from the participants' perspective in Korea. Focus groups were used to identify 1) perceived eating behaviors during pregnancy and lactation, 2) factors associated with healthy eating behaviors, and 3) needs for nutrition education programs. Using a trained moderator, we conducted 7 focus group interviews with 44 pregnant women over a four-month period. Focus group discussions were video - and audio - taped, transcribed and categorized by major themes. Participants expressed interest in receiving nutrition education regarding healthy eating, weight control after delivery, weaning foods and health management, yet they showed little interest in breastfeeding. The majority of them said that meal balance and meal regularity were the most important components of good health during pregnancy. They were less likely to be confident about taking dietary supplements, including Oriental medicines. life stress and poor appetite associated with pregnancy were major barriers to healthy eating habits during pregnancy. The most important sources of nutrition and health information were friends and family members, especially those who had become pregnant recently. Qualified educators and reliable information appeared to be the most important aspects of program development. Regarding types of nutrition education, participants tended to prefer a combination of individual counseling and small group education with hands-on materials and interactive formats. The use of Web-based nutrition education was well received Major concerns about Web-based nutrition education were authenticity, tailored messages and interactive formats for sharing information. These results offer useful information for designing nutrition education programs for pregnant and lactating women in Korea for health promotion.
To assess zinc status by dietary intake and urinary excretion of preschool children in Busan and to evaluate the relationship of intakes of food and nutrient with urinary zinc excretion, zinc food frequencies of 40 common foods affecting intakes of zinc by food fequency method, nutrient intake by 24hr recall and 24hr urinary zinc excretion were measured with 97 preschool children. The mean zinc intake was 4.29 mg and 43.0% of RDA. The mean zinc intake per 1,000 kcal was 3.09 mg.97.9% of subjects had zinc intake less than 75% of RDA. Grains food group was the primary source of zinc intake and supplied 38.9% of the total daily zinc intake. Altogether, plant food products supplied 49.7% of zinc intake. The mean urinary zinc excretion and zinc excretion per gram of creatinine were 0.19 mg and 1.00 mg respectively. The urinary zinc excretion showed positive significant correlations with height and weight (p < 0.05, p < 0.05) , urine volume and urinary creatinine excretion (p < 0.05, p < 0.001) , urinary zinc excretion per creatinine (p < 0.001) , urinary zinc excretion per weight (p < 0.001) , intakes of energy and carbohydrate (p < 0.05, p < 0.01) and usual intake of zinc from eggs food group (p < 0.05) . In conclusion, these results show that the zinc intake of preschool children is low and that sources of dietary zinc are mainly plant foods, suggesting low bioavailability. So nutritional education is needed in order to inc-rease usual intake of animal food group. Interpretation of urinary zinc excretion data is complicated by current uncertainty about "normal" zinc level at this age group. Further studies are needed to obtain extensive data on urinary zinc excretion for this age group.age group.
Seed storage proteins are used as carbon and nitrogen sources for the nutritional improvement of seeds. Since the composition of proteins from the Korean cultivars of proso millet is unknown, this study was conducted to obtain a reference map of millet seed proteins and identify the functional characteristics of the identified proteins. Proteins extracted from proso millet seeds of various cultivars were investigated using proteomic techniques such as 2-D electrophoresis coupled with mass fingerprinting; 1152 (differentially expressed) protein spots were detected on the 2-D gels. Among them, 26 reproducible protein spots were analyzed using matrix-assisted laser desorption/ionization time-of-flight/time-of-flight mass spectrometry. Out of the 26 proteins, 2 proteins were upregulated in all the millet cultivars, while 13 proteins were upregulated and 11 proteins were downregulated in 2 cultivars. Abundance of most of the identified protein species associated with polysaccharide and starch metabolism, transcription, and pathogenesis was significantly enhanced, while that of other protein species involved in glycolysis, stress response, and transduction was severely reduced. Taken together, the results suggest that the differential expression of the proteins from the four millet cultivars may be cultivar-specific. By conducting a proteomic investigation of millet seeds from different cultivars, we sought to better understand the functional categorization of individual proteins on the basis of their molecular functions. We believe that the identified proteins may help in investigating genetic variations in millet cultivars.

Commercial application of laccase is often hampered by insufficient enzyme stocks, with very low yields obtained from natural sources. This study aimed to improve laccase production by mutation of a Coriolopsis gallica strain and to determine the biological properties of the mutant. The high-yield laccase strain C. gallica TCK was treated with N-methyl-N-nitro-N-nitrosoguanidine and ultraviolet light. Among the mutants isolated, T906 was found to be a high-production strain of laccases. The mutant strain T906 was stabilized via dozens of passages, and the selected ones were further processed for optimization of metallic ion, inducers, and nutritional requirements, which resulted in the optimized liquid fermentation medium MF9. The incubation temperature and pH were optimized to be 30℃ and 4.5, respectively. The mutant strain T906 showed 3-times higher laccase activity than the original strain TCK under optimized conditions, and the maximum laccase production (303 U/ml) was accomplished after 13 days. The extracellular laccase isoenzyme 1 was purified and characterized from the two strains, respectively, and their cDNA sequence was determined. Of note, the laccase isoenzyme 1 transcription levels were overtly increased in T906 mycelia compared with values obtained for strain TCK. These findings provide a basis for C. gallica modification for the production of high laccase amounts.
The survival and growth of marine benthic invertebrate larvae such as abalone depend on the nutritional value of micro algae. However, it is difficult to determine the dietary value of the many microalgal species used for food by benthic larvae. Therefore, we tested the benthic copepod, Tigriopus japonicus, which grazes microalgae on substrata in a manner similar to abalone larvae. It also has short generation time and is easy to rear which makes to be easier to examine the dietary value of each micro algal species. We measured the daily production of nauplii from gravid females of T. japonicus fed 26 microalgal species separately. Amino acid and fatty acid content of the micro algae and the copepod was also analyzed. The nauplii production of T. japonicus was the highest (10.7) when they were fed Navicula sp. (B-394) and the lowest (0.8) when they were fed Scrippsiella trochoidea. In Tetraselmis suecica the nauplii production was so high (8.2), which was not significantly different with the diatom group. We determined that Navicula sp. (B-394), Rhaphoneis sp. and T. suecica were good sources of food for T. japonicus. We suggest that a diet of with a mixture of these three micro algal species may be also good for invertebrate larvae such as abalone.
In order to independently examine the effects of long-chain saturated fatty acids and dietary cholesterol levels on plasma and hepatic cholesterol concentrations, six different diets were fed to male Mongolian gerbils (14 gerbils pes grcup) for an 8-week period. Purified diets contained 36% energy as fat (each saturated fatty acid tested comprised about 20% of the total fat energy) and 0.06% (w/w) cholesterol, corresponding to typical human consumption patterns in Western diets. Fat blends were formulated with natural fat sources. To determine the effects of different saturated fatty acids on plasma and liver cholesterol levels, four of the six diets contained constant levels of all nutrients except for the amounts of lauric acid (12 0), myristic acid (14 : 0), palmitic acid (16 0), and stearic acid (18 : 0). Dietary cholesterol effects were tested using 16 : 0-enriched diets containing 0, 0.006, and 0.06% (w/w) cholesterol. None of the plasma lipids were influenced by fatty acid treatment, including triglycerides, plasma total-, VLDL+LDL-, and HDL-cholesterol. However, hepatic esterified cholesterol concentrations were increased in the palmitic and stearic arid diet groups compared to the lauric and myristic acid diet group. The molar ratios of hepatic EC/FC were the highest in the palmitic acid diet (12.2 $\pm$0.6) and the lowest in the myristic acid diet (6.4$\pm$0.2). Dietary cholesterol significantly (p<0.001) increased the plasma total cholesterol which was due to the increase of both HDL-and VLDL+LDL-cholesterol. In the absence of dietary cholesterol and compared to other species, the gerbil exhibited a high molar ratio of hepatic EC/FC, which was funker elevated by dietary cholesterol feeding (0.06%). The results from this study indicate that hepatic cholesterol concentrations are sensitive to both low levels of dietary cholesterol and saturated fatty acid chain length and also, that plasma cholesterol concentrations are sensitive to low levels of dietary cholesterol .
Zinc nutriture in South Koreans was evaluated by estimating Zn, Ca, and phytate intake, and the molar ratios of phytate : Zn and the millimolar ratios of phytate $\times$ Ca Zn. food consumption data from the 95 National Nutrition Survey was used. For the present study, data from the nationwide, large city and urban area level were used. No standard deviation measures were provided in the 95 National Nutrition Survey : only mean values were reported. Nationwide daily intake of Zn and Ca were 10.1 mg/day and 426.5 mg/day, respectively. The estimated daily phytate intake was 1676.6 mg/day nationwide. The molar ratio of phvtate : Zn, the millimolar ratio of phytate $\times$ Ca : Zn and the phytate $\times$ Ca Zn mmol per 4.2 MJ (1000 kcal) were 15.9, 168.9 and 91.8 in nationwide, respectively. The major food groups for zinc intake were meat, poultry products (43%), and cereals and grain products (18%). Sixty two percent of Zinc was from animal food sources. Cereal and grain products supplied most of the phytate intake (46%), followed by seasonings, fruits, and legume products. The major food source of phytate was rice (39%) . The results of the present study suggest that Zn status of Koreans maybe influenced by high dietary intake of phytate and high molar ratios of phytate : Zn and phytate $\times$ Ca : Zn. These results raise concerns about Zn status of Koreans, who consume a diet higher in phytate than Western diets. further research is necessary to confirm whether such poorly available dietary Zn has any impact on the health of Koreans.
The objective of this study is to collect basic data which can lead effective direction in performing nutritional education, by investigating 273 homemakers' knowledge and attitudes about nutrition. This study was performed using questionnaire about the knowledge and the attitudes of nutrition according to the age, education level, occupation, food cost, and total income. Results were summarized as follows; 1) sources in the knowledge of nutrition appeared by order of T.V., cookbooks, magazines newspapers, mothers, friends, lecturers in cooking school, and nutritionist. 2) A level in the knowledge of nutrition which scord an average marks 154.7(the highest marks 210) was higher compared to other studies. Informations about food composition scord lower marks than other studies. 3) Attitudes about nutrition showed high score in the aspects that nutrition has an important role in health, and desirable eating habits were formed in childhood. 4) The knowledge of nutrition had no significant differences with age, occupation, and food cost whereas it had significant correlation with education level and total income. The higher the level in education and total income, the higher the level in the knowledge of nutrition. 5) The attitudes about nutrition had no correlation with age, occupation, but it had a significant correlation with education level, food cost and total income. 6) There was an significant correlation between the knowledge and attitudes about nutrition. In order to promote children's nutrition status, nutrition education for mothers must be accomplished in the first place. Development of T.V. program for children's nutrition education is desirable.
