Conditional lethal mutation nup49-1 of a nuclear pore complex component gene was constructed in Saccharomyces cerevisiae. This mutation deleted one third of the essential NUP49 gene at the carboxy-terminal, but retained 13 repeats of the highly conserved GLFG domain. The nup49-1 mutant strain was viable with a slow-growth phenotype, indicating that the C-terminal is dispensable at normal growth temperature. This strain exhibited both temperature-sensitivity at 37.deg.C and cold-sensitivity at 16.deg.C. Temperature shift experiments revealed that the arrest phenotype at 37.deg.C was random in the cell division cycle. The nup49-1 mutation was tested to be recessive and is expected to be useful for the functional analysis of nuclear pore complex proteins as well as for studies of nuclear transport systems.
To construct transformed Bm5 cells, Autographa californica nuclear polyhedrosis virus (AcNPV)IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% uncleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/$m\ell$ G418 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistnt cells was isolated and characterized by PCR using AcNPV IE1 gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. moridrived Bm5 cells as well as Spodaptera frugiperda-derived Sf9 cells to produce stably-transformed insect cells.
목적: 조직 특이 프로모터를 이용하면 특정 암조직내에서만 원하는 치료유전자를 발현시킬 수 있다. 나트륨 옥소 공동 수송체(sodium iodide symporter: NIS) 유전자는 옥소를 섭취하는 특성을 가져 방사성옥소를 이용한 치료용 유전자로 사용될 수 있다. 광학 영상용 유전자인 luciferase (Luc) 유전자를 세포에 이입하면 비침습적으로 유전자가 이입된 세포의 상태를 평가할 수 있다. 본 연구는 간암 특이성을 나타내는 AFP 프로모터에 의해 발현이 조절되는 NIS유전자와 CMV프로모터에 의해 발현되는 Luc유전자를 간암세포에 이입하여 NIS유전자 이입에 의한 방사성옥소 유전자치료의 효과를 알아보고, 종양사멸 정도를 광학 리포터 유전자 발현으로 알아보고자 하였다. 대상 및 방법: AFP enhancer와 GSTP 프로모터를 연결하여 AFP프로모터를 제작하였으며 이를 NIS유전자와 연결하였다. 또한 CMV 프로모터에 조절 받는 Luc 유전자를 동시에 삽입하여 AFP-NIS-CMV-Luc 유전자 발현 벡터를 생산하였다. 실험 대상 세포주로는 간암세포주인 HepG2와 Huh-7 세포와 사람 대장암세포주인 HCT-15 세포를 이용하였다. AFP-NIS-CMV-Luc 발현벡터를 Liposome을 이용해 실험대상 세포주 내로 이입하였으며, 방사성옥소 섭취율과 방사성옥소의 유출량을 측정하였다. 또한 Luciferase 발현 정도를 luminometer로 측정하였으며, clonogenic assay를 통하여 I-131에 대한 세포주에 따른 사멸효과 차이를 알아보았다. AFP-NIS-CMV-Luc 유전자 이입 세포주를 누드마우스에 대퇴부 피하에 주입하여 I-131 축적여부를 감마카메라 영상을 획득하였다. 결과: AFP-NIS-CMV-Luc 유전자 발현 벡터를 제작하였다. AFP-NIS-CMV-Luc 유전자가 이입된 HepG2와 Huh-7 세포의 방사성옥소 섭취율은 유전자 이입이 되지 않은 대조군 HepG2와 Huh-7 세포에 비하여 높았으며, $KClO_4$를 처리시 옥소 섭취가 저해되었다. 대장암 세포주인 HCT-15세포에 AFP-NIS-CMV-Luc유전자를 이입 시 방사성옥소의 섭취률은 증가되지 않았다. 30분간 방사성옥소를 섭취시킨 AFP-NIS-Luc 유전자가 이입된 HepG2와 Huh-7 세포에서의 방사성옥소의 유출반감기는 약 4분과 6분으로 각각 나타났다. AFP-NIS-CMV-Luc 유전자가 이입된 HepG2, Huh-7세포의 Luc 유전자의 발현은 241, 441 $RLU/2\;{\times}\;10^5$ cells로 나타났으며, 대조군 HepG2와 Huh구세포에서의 Luc 유전자의 발현은 74, $RLU/2\;{\times}\;10^5$ cells로 나타났다. HCT-15 세포는 AFP-NIS-CMV-Luc 유전자 이입에 따라 I-131에 의한 세포 사멸능이 증가되지 않았으나, HepG2 및 Huh-7 세포는 FP-NIS-CMV-Luc 유전자 이 입에 따라 I-131에 의한 세포 사멸능이 증가되었으며, Huh-7세포의 경우 0.5mCi의 I-131을 투여한 경우 모든 세포가 사멸하였다. AFP-NIS-CMV-Luc 유전자가 이입된 Huh-7 세포수가 많을수록 방사성옥소 섭취율이 증가하며 luciferase활성도도 높게 나타났다. AFP-NIS-CMV-Luc 유전자가 이입된 Huh-7 세포를 이식한 누드마우스에 I-131 감마카메라 영상에서 종양이식부위에 방사능 축적을 관찰 할 수 있었다. 결론: AFP프로모터의 의하여 NIS유전자가 발현되며, CMV프로모터에 의한 Luc 유전자가 발현되는 벡터를 제작하였으며, 이 벡터를 이입한 경우 간암세포에서만 I-131의 세포 독성이 증가하는 효과를 나타내었다. 또한 Luc유전자를 이용하여 비침습적인 광학 영상으로 세포사멸 효과를 확인할 수 있었다. 간암특이 프로모터에 조절되는 치료 유전자와 광학리포터 유전자를 한 벡터에 동시에 이입하면 간암 특이 유전자 치료와 그 치료효과를 비침습적으로 평가할 수 있을 것으로 생각된다.
Even though success in birth of live offspring from nuclear transfer(NT) using somatic cells in many species, detailed information on processes or mechanisms of development are not well known. Cytoplasm of bovine oocyte has been known to support the development of nuclear transferred embryos using nuclear donor cells from different species. Therefore, interspecies NT might be used to find answers of some questions in basic aspect of nuclear transfer In this study, we examined the developmental potential of reconstructed embryos when bovine oocyte as a cytoplasm recipient and mouse embryonic fibroblast as a nuclear donor were used. The nuclear transfer units were aliocated in Group 1 (murine block media and normal media) and Group 2. (bovine block media and normal media). NT units were not blocked at 2-cell stage regardless of types of medium. On mouse media, poor development of interspecies NT units was observed compared to bovine media. However, as NT units cultured in bovine normal medium, embryos developed over 8-cell stage. Further studies performed to increase the developmental rate in condition of antioxidant treatment. Despite low development, bovine-murine interspecies nuclear transferred embryos could develop to blastocysts and they showed that blastocyts rate of antioxidant group was superior to those of non-antioxidant group. Next, we investigated gene expression pattern which is carried out for zygotic activation. The Xist gene is expressed in female mouse embryo after zygotic activation of 4-cell stage. But interspecies nuclear transferred embryos do not express Xist gene at 4-cell stage. As a result, it is suggested that the bovine cytoplasm controls the early preimplantation development in interspecies NT However, the development of later stages might require genomic control from transferred donor nucleus. Therefore, even though the involvement of several other factors such as mitochondrial incompatibility, effective development of embryos produced by interspecies NT requires proper genomic activation of donor nucleus after overcoming the cytoplasmic control of recipient oocytes.

We have constructed a novel recombinant Bombyx mori nuclear polyhedrosis virus (BmNPV) producing the green fluorescent polyhedra. For the production of the fluorescent polyhedra, partial polyhedrin gene containing KRKK as nuclear localization site from the BmNPV polyhedrin gene and the green fluorescent protein (gfp) gene were introduced under the control of p10 promoter of BmNPV. The recombinant BmNPV was stably produced fluorescent polyhedra in the infected Bm5 cells and the morphology of the fluorescent polyhedra was similar to that of wild-type BmNPV. The fluorescent polyhedra had 32 kDa native polyhedrin and 41 kDa fusion protein. From these data, we have further developed a novel BmNPV p10-based transfer vector producing recombinant polyhedra with foreign gene Product. The novel BmNPV P10-based transfer vector is composed of partial polyhedrin gene, factor Xa, and multiple cloning sites.

We have cloned and characterized an immediate early-1 gene, iel, which is activated immediately upon entrance of the viral genome into the cell nucleus, from Bombyx mori nuclear polyhedrosis virus (BmNPV) K1 strain. This gene encodes a protein 584 amino acids with a predicted molecular weight of 67 kDa. The promoter and coding regions of BmNPV-K1 ie1 showed high homology with Autographa californica nuclear polyhedrosis virus and BmNPV T3 strain. The BmNPV-K1 ie1 was different from amino acid sequence at 4 positions in BmNPV T3. The location of ie1 gene in the BmNPV-K1 genome was confirmed by Southern blot analysis and its expression patterns at the transcriptional level in the infected cells were confirmed by Nerthern hybridization analysis.

We have cloned and characterized an antiapoptotic gene, p35, which blocks apoptosis, from Bombyx mori nuclear polyhedrosis virus (BmNPV) K1 strain. The 897 bp p35 has an open reading frame of 299 amino acids. The BmNPV-K1 p35 showed a high identity to Autographa californica nuclear polyhedrosis virus and BmNPV T3 strain. The BmNPV-K1 p35 was different from the amino acid sequences of BmNPV T3 at 6 positions. The p35 gene of BmNPV-K1 was 99.2% identical at the nucleotide level and 98% identical at the amino acid level to BmNPV T3. The location of p35 gene in the BmNPV-K1 genome was confirmed by Southern blot analysis and its expression patterns at the transcriptional level in the infected cells were con- firmed by Northern hybridization analysis.
We demonstrate that radicicol, a macrocyclic antifungal antibiotic originally isolated from Monosporium bonorden, inhibits LPS-induced expression of iNOS gene in RAW 264.7 cells. Treatment of peritoneal macrophages and RAW 264.7 cells with radicicol inhibited LPS-stimulated nitric oxide production in a dose-related manner. Immunohistochemical staining of iNOS and RTPCR analysis showed that the decrease of NO was due to the inhibition of iNOS gene expression in RAW 264.7 cells. Immunostaining of p65, EMSA, and reporter gene assay showed that radicicol inhibited $NF-\kappa/Rel$ nuclear translocation. DNA binding, and transcriptional activation, respectively. Collectively, these series of experiments indicate that radicicol inhibits iNOS gene expression by blocking $NF-\kappa/Rel$ nuclear translocation. Due to the critical role that NO release plays in mediating inflammatory responses, the inhibitory effects of radicicol on iNOS suggest that radicicol may represent a useful anti-inflammatory agent.
Recent progress in the development of non-invasive imaging technologies continues to strengthen the role of biomedical research. These tools have been validated recently in variety of research models, and have born shown to provide continuous quantitative monitoring of the location(s), magnitude, and time-variation of gene delivery and/or expression. This article reviews the use of PET technologies as they have been used in imaging biological processes for molecular imaging applications. The studies published to date demonstrate that noninvasive imaging tools will help to accelerate pre-clinical model validation as well as allow for clinical monitoring of human diseases.
A positive selectable marker system was adapted for transformation of mature embryo-derived calli of Indica rice (Oryza sativa L.) utilizing the PMI gene encoding for phosphomannose-isomerase that converts mannose-6-phosphate to fructose-6-phosphate. The transformed cells grew on medium supplemented with 3% mannose as carbon source and calli were selected on media containing various concentrations of mannose. Molecular analyses showed that the transformed plants contained the PMI gene. The results indicate that the mannose selection system can be used for Agrobacterium-mediated transformation of mature embryo in rice to substitute the use of conventional selectable markers in genetic transformation.
