Spore Display Using Bacillus thuringiensis Exosporium Protein InhA

  • A new spore display method is presented that enables recombinant proteins to be displayed on the surface of Bacillus spores via fusion with InhA, an exosporium component of Bacillus thuringiensis. The green fluorescent protein and $\beta$-galactosidase as model proteins were fused to the C-terminal region of InhA, respectively. The surface expression of the proteins on the spores was confirmed by flow cytometry, confocal laser scanning microscopy, measurement of the enzyme activity, and an immunogold electron microscopy analysis. InhA-mediated anchoring of foreign proteins in the exosporium of Bacillus spores can provide a new method of microbial display, thereby broadening the potential for novel applications of microbial display.

  • This paper proposes an automatic tracking control algorithm for efficiency improvement of photovoltaic generation. Increasing the power of PV systems should improve the efficiency of solar cells or the power condition system. The normal alignment of the PV module always have to run perpendicular to the sun's rays. The solar tracking system, able to improve the efficiency of the PV system, was initiated by applying that to the PV power plant. The tracking system of conventional PV power plant has been studied with regard to the tracking accuracy of the solar cells. Power generation efficiency were increased by aligning the cells for maximum exposure to the sun's rays. Using a perpendicular position facilitated optimum condition. However, there is a problem about the reliability of tracking systems unable to not track the sun correctly during environmental variations. Therefore, a novel control algorithm needs to improve the generation efficiency of the PV systems and reduce the loss of generation. This control algorithm is the proposed automatic tracking algorithm in this paper. Automatic tracking control is combined the sensor and program method for robust control in environment changing condition. This tracking system includes the insolation, rain sensor and anemometer for climate environment changing. Proposed algorithm in this paper, is compared to performance of conventional tracking control algorithm in variative insolation condition. And prove the validity of proposed algorithm through the experimental data.

  • In this study, we developed a dispersion analysis program of the gas-generator cycle liquid propellant rocket engine by expanding the mode analysis software(GEMAT). The performance dispersions of an engine that are arisen from the internal dispersion factors of engine's sub-components were formulated and solved to find the effects of each dispersion factor. We were also able to present the calculation method to find the required pressure margin for the compensation of those dispersion to satisfy the required performances of engine. Using this method, we could propose a novel procedure of compensating during the ground firing test which would induce the performance improvement by lessening the pumps discharge pressures or augmenting the combustion chamber pressure.

  • We fabricated a novel simple and rapid method of three dimensional nanoporous gold thin film (NPGF) onto a Au substrate using electrochemical deposition method. The NPGF-modified electrode analysis by scanning electron microscope and reveals the formation of nanopores, approximately 30~50 nm in diameter. differential pulse voltammetry was measured for the determination of ${\gamma}$-aminobutiric acid in the concentration range of ($10{\sim}100{\mu}M$ using a NPGF. The high sensitivity feature of NPGF is expected to be applied for real sample biosensor applications.

Mutations in Streptomycin Resistance Genes and Their Relationship to Streptomycin Resistance and Lineage of Mycobacterium tuberculosis Thai Isolates

  • Background: Streptomycin (SM) is recommended by the World Health Organization (WHO) as a part of standard regimens for retreating multidrug-resistant tuberculosis (MDR-TB) cases. The incidence of MDR-TB in retreatment cases was 19% in Thailand. To date, information on SM resistance (SMR) gene mutations correlated to the SMR of Mycobacterium tuberculosis Thai isolates is limited. In this study, the mutations in rpsL, rrs, gidB, and whiB7 were investigated and their association to SMR and the lineage of M. tuberculosis were explored. Methods: The lineages of 287 M. tuberculosis collected from 2007 to 2011 were identified by spoligotyping. Drug susceptibility profiles were evaluated by the absolute concentration method. Mutations in SMR genes of 46 SM-resistant and 55 SM-susceptible isolates were examined by DNA sequencing. Results: Three rpsL (Lys43Arg, Lys88Arg, and Lys88Thr) and two gidB (Trp45Ter and Gly69Asp) mutations were present exclusively in the SM resistant M. tuberculosis. Lys43Arg rpsL was the most predominant SMR mutations (69.6%) and prevailed among Beijing isolates (p<0.001). No SMR-related mutation in was found rrs. The combination of rpsL and gidB mutations provided 76.1% sensitivity for detecting SMR in M. tuberculosis Thai isolates. whiB7 was not responsible for SMR in SM resistant isolates lacking rpsL and rrs mutations. The significance of the three gidB mutations, 276A>C, 615A>G, and 330G>T, as lineage signatures for Beijing and EAI were underscored. This study identified 423G>A gidB as a novel sub-lineage marker for EAI6-BGD1. Conclusion: Our study suggested that the majority of SMR in M. tuberculosis Thai isolates were responsible by rpsL and gidB polymorphisms constantly providing the novel lineage specific makers.

Post-tuning of Sample Position in Common-path Swept-source Optical Coherence Tomography

  • Common-path interferometers are widely used for endoscopic optical coherence tomography (OCT) because an arbitrary arm length can be chosen for the endoscopic imaging probe. However, the scheme suffers from the limited range of the sample position distance from the end of the imaging probe because the position between the reference reflector and the sample is limited by the optical path-length difference (OPD) to induce an interference signal. In this study, we developed a novel method for compensating the arbitrary sample position in common-path swept-source OCT by adding an extra Mach-Zehnder interferometer in the post-path of the interfered optical signal. Theoretical analysis and an experimental demonstration of imaging depth tuning for the flexible sample position of an endoscopic OCT image are discussed. After post-tuning of sample position distance, the positioning limitation between the reference reflector and the sample can be solved for various sample positions over a range of 26 mm for the cross-sectional images of a fish eye sample.

An Integrated Expert Model for Delay Management in Construction Projects

  • Delay claim should actually be supported by a set of proper information so that the contractors could prove their validity. The so-called information should be able to clarify the relationship between delay events and how they impact on the whole project. Therefore, exploiting an integrated system by people who are involved in construction business would certainly prove helpful. In the present study, delay analysis methods have been investigated along with selecting a relatively comprehensive method which has been modified, and eventually, a novel model and its required modules have been proposed for evaluating delay claims. The suggested integrated model is formed to identify delayed events, to classify delays, to measure the impacts of delays on the project scheduling, and finally to estimate the damages which were caused by those so-called delays. A decision support system (DSS) model which is related to the integrated system is actually extracted from Iran's general contract conditions, that is, 4311 magazine (equivalent to red FIDIC book). It is then programmed and coded by C# program. This DSS model can be used as an input of Easy Plan program. In addition, at the end of this research, the coded DSS has been used along with the so-called program so that a modified and developed model could be generated.

Applied 2D equivalent linear program to analyze seismic ground motion: Real case study and parametric investigations

  • Seismic ground response evaluation is one of the main issues in geotechnical earthquake engineering. These analyses are subsequently divided into one-, two- and three-dimensional methods, and each of which can perform in time or frequency domain. In this study, a novel approach is proposed to assess the seismic site response using two-dimensional transfer functions in frequency domain analysis. Using the proposed formulation, a program is written in MATLAB environment and then promoted utilizing the equivalent linear approach. The accuracy of the written program is evaluated by comparing the obtained results with those of actual recorded data in the Gilroy region during Loma Prieta (1989) and Coyote Lake (1979) earthquakes. In order to precise comparison, acceleration time histories, Fourier amplitude spectra and acceleration response spectra diagrams of calculated and recorded data are presented. The proposed 2D transfer function diagrams are also obtained using mentioned earthquakes which show the amount of amplification or attenuation of the input motion at different frequencies while passing through the soil layer. The results of the proposed method confirm its accuracy and efficiency to evaluate ground motion during earthquakes using two-dimensional model. Then, studies on irregular topographies are carried out, and diagrams of amplification factors are shown.

  • Being a cost-efficient solution for alleviating the traffic congestion in airspace, Air Traffic Flow Management (ATFM) has drawn more attentions from not only air traffic controllers but also researchers in the field of Air Traffic Management (ATM). Among other ATFM initiatives, it is believed that Ground Delay Program (GDP) could be effectively applied to reduce the congestion particularly in the relatively small airspace with dense traffic demand. This paper introduces a novel way that suggests flights to be delayed on the departing airports together with amount of the delays (in time) for those flights to be delayed. Adopting a fast-time simulation for predicting airspace delay of each flight for a given flight plan, the method is designed to iteratively and incrementally adjust the departure times in the plan towards reducing total airspace delays. Applying the method to Jeju airport with a hypothetically high demand, the paper demonstrates the airspace delay could be significantly reduced by applying GDP at Gimpo airport where more than 60% of Juju-bound flights departure. Although the simulation model needs to be calibrated and validated for the real-world application, the results clearly shows that the approach can possibly implemented as a tool for preparing the daily plan at the pre-tactical stage defined in the ICAO ATFM manual.

  • Fungal particles have been known to aggravate indoor air quality. To develop fungal particle cleaning devices requires a well-controlled generator of fungal aerosol particles. In this study, a novel fungal aerosol generator was designed and tested for anti-fungal experiment. Cladosporium cladosporioides was selected as test fungal particle. After aerosolization, the number concentration and the size of particles were measured by aerodynamic particle sizer. The number concentration depended on the vibration strength and vibration period of the designed fungal aerosol generator. For the vibration strength of 10volt and the period of 10 sec (5 sec on and 5 sec off), the stable particle generation with concentration of 10#/cm3 was maintained during 35 minutes.
