Periplasmic glucans isolated from Proteobacteria

  • Periplasmic glucans (PGs) are general constituents in the periplasmic space of Proteobacteria. PGs from bacterial strains are found in larger amounts during growth on medium with low osmolarity and thus are often been specified as osmoregulated periplasmic glucans (OPGs). Furthermore, they appear to play crucial roles in pathogenesis and symbiosis. PGs have been classified into four families based on the structural features of their backbones, and they can be modified by a variety of non-sugar substituents. It has also recently been confirmed that novel PGs with various degrees of polymerization (DPs) and/or different substituents are produced under different growth conditions among Proteobacteria. In addition to their biological functions as regulators of low osmolarity, PGs have a variety of physico-chemical properties due to their inherent three-dimensional structures, hydrogen-bonding and complex-forming abilities. Thus, much attention has recently been focused on their physico-chemical applications. In this review, we provide an updated classification of PGs, as well as a description of the occurrences of novel PGs with substituents under various bacterial growth environments, the genes involved in PG biosynthesis and the various physico-chemical properties of PGs.

i-PGS 기반 선회베어링의 접촉피로강도 설계 (Contact Fatigue Strength Design of a Slewing Bearing Based on i-PGS)

  • To overcome the large ring gear manufacturing problems seen in slewing bearings and girth gears, pin gear drive units have been developed. Among them, a novel slewing bearing with an internal pinwheel gear set (i-PGS) is introduced in this paper. First, we consider the exact cam pinion profile of i-PGS with the introduction of a profile shift coefficient. Furthermore, a new root relief profile modification for the i-PGS cam pinion is presented. Then, the contact stresses are investigated to determine the characteristics of the surface fatigue by varying the shape design parameters. The results show that the contact stresses of i-PGS can be reduced significantly by increasing the profile shift coefficient. In addition, the contact ratio, a measure of teeth overlapping action, decreases with the decrease of the allowable pressure angle.

Phylogenetics and Gene Structure Dynamics of Polygalacturonase Genes in Aspergillus and Neurospora crassa

  • Polygalacturonase (PG) gene is a typical gene family present in eukaryotes. Forty-nine PGs were mined from the genomes of Neurospora crassa and five Aspergillus species. The PGs were classified into 3 clades such as clade 1 for rhamno-PGs, clade 2 for exo-PGs and clade 3 for exo- and endo-PGs, which were further grouped into 13 sub-clades based on the polypeptide sequence similarity. In gene structure analysis, a total of 124 introns were present in 44 genes and five genes lacked introns to give an average of 2.5 introns per gene. Intron phase distribution was 64.5% for phase 0, 21.8% for phase 1, and 13.7% for phase 2, respectively. The introns varied in their sequences and their lengths ranged from 20 bp to 424 bp with an average of 65.9 bp, which is approximately half the size of introns in other fungal genes. There were 29 homologous intron blocks and 26 of those were sub-clade specific. Intron losses were counted in 18 introns in which no obvious phase preference for intron loss was observed. Eighteen introns were placed at novel positions, which is considerably higher than those of plant PGs. In an evolutionary sense both intron loss and gain must have taken place for shaping the current PGs in these fungi. Together with the small intron size, low conservation of homologous intron blocks and higher number of novel introns, PGs of fungal species seem to have recently undergone highly dynamic evolution.

핀 휠을 구비한 외륜형 선회베어링의 면압강도 (Contact Stress of Slewing Ring Bearing with External Pinwheel Gear Set)

  • The pin-gear drive is a special form of fixed-axle gear mechanism. A large wheel with cylindrical pin teeth is called a pinwheel. As pinwheels are rounded, they have a simple structure, easy processing, low cost, and easy overhaul compared with general gears. They are also suitable for low-speed, heavy-duty mechanical transmission and for occasions with more dust, poor lubrication, etc. This paper introduces a novel slewing ring bearing with an external pinwheel gear set (e-PGS). First, we consider the exact cam pinion profile of the e-PGS with the introduction of a profile shift. Then, the contact stresses are investigated to determine the characteristics of the surface fatigue by varying the shape design parameters. The results show that the contact stresses of the e-PGS can be lowered significantly by increasing the profile shift coefficient.

Production of Prostaglandin $E_2$ and $I_2$ is Coupled with Cyclooxygenase-2 in Human Follicular Dendritic Cells

  • Background: Prostaglandins (PGs) play pathogenic and protective roles in inflammatory diseases. The novel concept of PGs as immune modulators is being documented by several investigators. By establishing an in vitro experimental model containing human follicular dendritic cell-like cells, HK cells, we reported that HK cells produce prostaglandin $E_2$ ($PGE_2$) and prostaglandin $I_2$ ($PGI_2$) and that these PGs regulate biological functions of T and B cells. Methods: To investigate the respective contribution of cyclooxygenase-1 (COX-1) and COX-2 to $PGE_2$ and $PGI_2$ production in HK cells, we performed siRNA technology to knock down COX enzymes and examined the effect on PG production. Results: Both $PGE_2$ and $PGI_2$ productions were almost completely inhibited by the depletion of COX-2. In contrast, COX-1 knockdown did not significantly affect PG production induced by lipopolysaccharide (LPS). Conclusion: The current results suggest that mPGES-1 and PGIS are coupled with COX-2 but not with COX-1 in human follicular dendritic cell (FDC) and may help understand the potential effects of selective COX inhibitors on the humoral immunity.

The physiological and pharmacological roles of prostaglandins in hair growth

  • Hair loss is a common status found among people of all ages. Since the role of hair is much more related to culture and individual identity, hair loss can have a great influence on well-being and quality of life. It is a disorder that is observed in only scalp patients with androgenetic alopecia (AGA) or alopecia areata caused by stress or immune response abnormalities. Food and Drug Administration (FDA)-approved therapeutic medicines such as finasteride, and minoxidil improve hair loss temporarily, but when they stop, they have a limitation in that hair loss occurs again. As an alternative strategy for improving hair growth, many studies reported that there is a relationship between the expression levels of prostaglandins (PGs) and hair growth. Four major PGs such as prostaglandin D2 (PGD22), prostaglandin I2 (PGI2), prostaglandin E2 (PGE2), and prostaglandin F2 alpha (PGF) are spatiotemporally expressed in hair follicles and are implicated in hair loss. This review investigated the physiological roles and pharmacological interventions of the PGs in the pathogenesis of hair loss and provided these novel insights for clinical therapeutics for patients suffering from alopecia.

곤충 면역반응을 중개하는 프로스타글란딘의 분자적 기작과 해충방제 응용 (Molecular Action of Prostaglandin to Mediate Insect Immunity and Its Application to Develop Novel Insect Control Techniques)

  • 척추동물과 유사하게 곤충도 인지질분해효소(phospholipase A2)의 촉매 작용으로 다양한 아이코사노이드를 합성한다. 그러나 일련의 아이코사노이드 생합성과정은 척추동물과 차이를 보이는데, 이는 곤충의 인지질에는 전구물질인 아라키도닉산의 함량이 낮기 때문이다. 대신에 비교적 풍부하게 존재하는 다가불포화지방산인 리놀레익산을 기반으로 사슬 연장 및 불포화반응으로 아라키도닉산을 합성하여 척추동물과 같이 아이코사노이드 전구물질로 이용하는 것 같다. 이렇게 해서 형성된 아라키도닉산은 다시 척추동물의 cyclooxygenase와 유사한 peroxynectin이 PGH2 형태의 프로스타글란딘(prostaglandin: PG) 전구물질을 형성하게 된다. 이후 여러 이성체 효소들의 특이적 반응에 의해 PGA2, PGD2, PGE2, PGI2, TXB2의 다양한 PG가 생성된다. 반면에 또 다른 형태의 아이코사노이드인 에폭시아이코사트리에노익산(epoxyeicosatrienoic acid: EET)은 척추동물과 유사한 단일산화효소의 산화반응으로 아라키도닉산을 전구물질로 5,6-EET, 8,9-EET, 11,12-EET, 14,15-EET를 형성하게 된다. 그러나 세 번째 아이코사노이드 부류인 류코트리엔(leukotriene)의 경우 곤충 체내 존재는 확인되었지만 생합성 과정은 아직 밝혀지지 않았다. 이들 아이코사노이드가 곤충의 대사, 배설, 면역 및 생식에 관여하는 생리작용을 중개한다. 따라서 아이코사노이드 생합성 과정을 교란하는 물질 탐색은 새로운 살충제 개발 전략이 된다. 본 종설은 이 가운데 PG의 곤충 면역 중개 기작을 소개한다.

Surface-Displayed Porcine IFN-λ3 in Lactobacillus plantarum Inhibits Porcine Enteric Coronavirus Infection of Porcine Intestinal Epithelial Cells

  • Interferon (IFN)-λ plays an essential role in mucosal cells which exhibit strong antiviral activity. Lactobacillus plantarum (L. plantarum) has substantial application potential in the food and medical industries because of its probiotic properties. Alphacoronaviruses, especially porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV), cause high morbidity and mortality in piglets resulting in economic loss. Co-infection by these two viruses is becoming increasingly frequent. Therefore, it is particularly important to develop a new drug to prevent diarrhea infected with mixed viruses in piglets. In this study, we first constructed an anchored expression vector with CWA (C-terminal cell wall anchor) on L. plantarum. Second, we constructed two recombinant L. plantarum strains that anchored IFN-λ3 via pgsA (N-terminal transmembrane anchor) and CWA. Third, we demonstrated that both recombinant strains possess strong antiviral effects against coronavirus infection in the intestinal porcine epithelial cell line J2 (IPEC-J2). However, recombinant L. plantarum with the CWA anchor exhibited a more powerful antiviral effect than recombinant L. plantarum with pgsA. Consistent with this finding, Lb.plantarum-pSIP-409-IFN-λ3-CWA enhanced the expression levels of IFN-stimulated genes (ISGs) (ISG15, OASL, and Mx1) in IPEC-J2 cells more than did recombinant Lb.plantarum-pSIP-409-pgsA'-IFN-λ3. Our study verifies that recombinant L. plantarum inhibits PEDV and TGEV infection in IPEC-J2 cells, which may offer great potential for use as a novel oral antiviral agent in therapeutic applications for combating porcine epidemic diarrhea and transmissible gastroenteritis. This study is the first to show that recombinant L. plantarum suppresses PEDV and TGEV infection of IPEC-J2 cells.

  • Phospholipase A$_2$(PLA$_2$) catalyzes the hydrolysis of the sn-2 position of membrane glycerophospholipids to liberate arachidonic acid(AA), a precursor of eicosanoids including prostaglandins(PGs) and leukotrienes (LTs). The same reaction also produces lyso-phospholipids. So far, at least 19 enzymes that possess PLA2 activity have been identified, consists of low-molecular-weight, Ca$\^$2+/-requiring, secretory enzymes that have been implicated in a number of biological processes, such as modification of eicosanoid generation, inflammation, host defense, and atherosclerosis. Suppressive effects on the expression of cyclooxygenase-2 and inducible nitric oxide synthase by a natural sesquiterpenoid in lipopolysaccharide-stimulated mouse macrophage cells

  • Prostaglandins (PGs) and nitric oxide (NO) produced by inducible cyclooygenase (COX-2) and nitric oxide synthase (iNOS), respectively, have been implicated as important mediators in the process of inflammation and carcinogenesis. On this line, the potential COX-2 or iNOS inhibitors have been considered as anti-inflammatory and cancer chemopreventive agents. In our continuing efforts of searching for novel cancer chemopreventive agents from natural products, we isolated natural sesquiterpenoids as potential COX-2 and iNOS inhibitors in cultured lipopolysaccharide (LPS)-activated mouse macrophage RAW 264.7 cells. Alantolactone, a natural eudesmane-type sesquiterpenoid, exhibited a potent inhibition of COX-2 (IC50 = 0.4 $\mu\textrm{g}$/$m\ell$) and iNOS activity (IC50 = 0.08 $\mu\textrm{g}$/$m\ell$) in the assay system determined by PGE2 and NO accumulation, respectively. The inhibitory potential of alantolactone on the PGE2 and NO production was well coincided with the suppression of COX-2 and iNOS protein and mRNA expression in LPS-induced macrophages. Furthermore, alantolactone inhibited NF-kB but not AP-l binding activity on nuclear extracts evoked by LPS-stimulated macrophage cells, suggesting the possible involvement of NF-kB in the regulation of COX-2 and iNOS expression. In further study with COX-2-expressing human colon HT-29 cells, alantolactone inhibited the cell proliferation, down-regulated COX-2, and inhibited the ERK phosphorylation in the early time. These results suggest that a natural sesquiterpenoid alantolactone might be a potential lead candidate for further developing COX-2 or iNOS inhibitor possessing cancer chemopreventive or anti-inflammatory activity
