The changes of plasma renin activity, aldosterone concentration, serum sodium-, and potassium levels were studied before and after the water loading followed by diuretics injection. The materials were: 13 non-, 11 normal-, and 11 toxemic pregnancy cases. The plasma renin activity and aldosterone concentration of the cord and postpartum blood were also measured. Following were the results: 1. The plasma renin activity was elevated significantly in normal pregnancy, and slightly in toxemic pregnancy. The serum sodium levels were decreased in pregnancy. 2. The plasma aldosterone concentration was slightly decreased in normal pregnancy, and slightly increased in toxemic pregnancy, however, statistically insignificant. 3. The plasma renin activity of the cord and postpartum blood were lower than those of pregnancy cases. 4. The changes of plasma renin activity after the diuretic administration showed an initial increase, which recovered within 2 hours. These changes were the least in normal pregnancy, and the most in toxemic pregnancy. 5. The changes of plasma aldosterone concentration after the diuretic administration were similar to those of plasma renin activity, although the variations were not so wide.
We examined the effects of calcium intake levels on mineral metabolism during pregnancy using calcium-deficient young adult rats. Five week-old female Sprague-Dawley rats were fed normal Ca (NCa, 0.5%) and low Ca (LCa, 0.15%) diets for five weeks (pre-pregnancy). The low Ca intake group was then divided into three groups and fed low Ca (0.15%), normal Ca (0.5%) and high Ca (1.5%) diets for 3 weeks (pregnancy). All of the rats were mated with normal male rats. The control group was fed a consistently normal Ca (0.5%) diet during pre-pregnancy and pregnancy. On the day after delivery, dams and their pups were sacrificed. We measured total protein, albumin, alkaline phosphatase and mineral content in serum and weight, length, breaking force, ash and mineral content in the femur and lumbar (L2-L4) of the dams. Whole body mineral content was measured in the pups. There was no difference in weight gain and food intake among the groups. Serum total protein and albumin were in the normal range but a little lower during pregnancy. High Ca supplementation decreased serum Mg and Fe during pregnancy. Weight, ash and Ca of the femur and lumbar significantly decreased in rats fed a chronically low Ca diet during pre-pregnancy and pregnancy. Calcium supplementation levels were above normal during pregnancy and increased the bone weight and breaking force of rats fed the low Ca diet during pre-pregnancy. However, Ca supplementation did not increase the ash and Ca contents in the bones. High Ca supplementation during pregnancy significantly decreased Mg in the bones and increased Ca and P in the kidneys. Ash content of pups from dams fed the chronically low Ca diet decreased but there was no difference in whole body Ca among the groups. Mg and Fe in the whole body decreased in pups from dams fed the high Ca diet. Pregnancy performance was reduced in dams fed the low Ca diet. These results suggest that above normal Ca supplementation levels during pregnancy restored maternal bone status to some extent in rats fed the chronically low Ca diet. The same could not be said for mineral content. Also, high Ca supplementation during pregnancy may deteriorate mineral composition in bones and other tissues. Therefore, more detailed research is needed to facilitate sound recommendations on appropriate calcium intake during pregnancy. (Korean J Nutrition 36(5): 459∼469, 2003)
In order to keep the normal pregnancy, a number of gene products are required at the feto-matemal interface. We have isolated approximately 30 genes, involved in keeping the normal pregnancy, via subtractive hybridization and RT-PCR analyses of cDNAs from the chorionic villi of normal and RPL patients. Characterizing their functions will help us to understand the process of establishing and maintaining pregnancy. In addition, more detailed studies of their expression in normal and RPL patients are required to evaluate their clinical relevance. Further identification of genes aberrantly expressed in RPL patients will help the prognosis of the pregnancy, identifying pregnancies with a high risk of miscarriage and enabling management of those pregnancies.
In order to estimate the human placental lactogen levels and its value as an indicator of placental function during the first half of normal pregnancy, we defermined HPL levels. in normal snbjects (No=40) from the 6th week to the 20th week of gestation For the determination of human placental lactogen in serum of normal pregnant women during the first half of prognancy, radioimmunoassay have been performed using Phadebas HPL Test kit. The range of 25ng to 800ng HPL/ml. were determined by the procedures using Phadbas HPL Test kit, which correspond to levels recorded throughout the first half of normal pregnancy. HPL was detected in the earliest pregnancy samples collected at 6 weeks but was generally present in minute amount during the first trimester of pregnancy, when the HPL level ranged from 25 ng/ml. to 0.86 ${\mu}g$/ml. The mean concentration of HPL increased gradually from only 0.03 ${\mu}g$/ml, at 6 weeks to 0.25 ${\mu}g$/ml, at 12 weeks of gestation, Subsequently the mean HPL value rose steeply from 0.25 ${\mu}g$/ml, at 12 weeks to 1.65 ${\mu}g$/ml, at 20 weeks of gestation.
The purpose of this study is to present the reference values for cardiac size and cardiothoracic ratio change, normal range and diagnosis of cardiovascular disease in pregnancy and non pregnancy of normal Korean women using chest X-ray. The subjects of this study were 58 women, who were read as normal by chest radiologist, had chest radiography taken on both last month of pregnancy, pre-pregnancy and within 2 years following delivery. In this study, we defined the last month of pregnancy as pregnancy and before or post pregnancy as non-pregnancy. CS and CTR were measured by two radiological technologist who had clinical experience more ten years with Danzer's method. Statistical methods were paired t-test and one-way ANOVA. Significance level ${\alpha}$ was 0.05 and p-value 0.05 or less was statistically significant. For pregnancy, the mean of left and right cardiac size was $40.11{\pm}8.73mm$ and $89.51{\pm}11.9mm$, CS was $128.60{\pm}13.15mm$, CTR was $44.51{\pm}4.21%$. In non pregnancy, $36.50{\pm}8.18mm$ and $77.68{\pm}13.1mm$. CS and CTR were $114.18{\pm}14.28mm$ and $42.03{\pm}4.04%$. Both pregnancy and non pregnancy, the difference of the mean value in left and right cardiac size, CS and CTR were statistically significant (p<0.01). but comparing mean on age, height and weight, the difference of the mean value between groups was not (p>0.05). In the result of this study, the mean size of CS increased by 12.6% in pregnancy($128.60{\pm}13.15mm$) compared to the non pregnancy($114.18{\pm}14.28mm$), and increased by 9.8% in the right side of the heart and 15.2% in the left side. The mean size of CTR increased about 5.9% in pregnancy ($44.5{\pm}4.21%$) compared to non pregnancy($42.03{\pm}4.04%$).
Every month of pregnancy, vaginal smears were obtained from 42 dairy cows, and an attempt was made to correlate the cell picture with the stage of pregnancy. The results obtained in this work were summerized as follows: 1. At early pregnancy, most of the cells in the smears were the cells with normal nucleus and cytoplasm, and leucocytes. After the first month of pregnancy there was a progressive decrease in the rate of appearance of the cells with normal nucleus and cytoplasm, which dropped to the lowest value at the third month of pregnancy, thereafter, showing steady state. 2. After the first month of prgnancy the rate of appearance of the cells with karyolysis began to increase gradually, and reached a maximum at the fifth month of pregnancy, returning towards initial levels. 3. The rate of appearance of the cells with karyopyknosis was much lower at early pregnancy, but at the third month of pregnancy there was a rapid increase, thereafter, showing steady state (ranging from 20 to 50 percent). 4. The non-nucleated cornified cells, the cells with vacuolized cytoplasm, and the cells with swollen nucleus and cytoplasm were appeared much fewer or not, and significant changes were not recognized during pregnancy. 5. At early pregnancy there appeared many of leucocytes but dropped intensively at the third month of pregnancy, and, thereafter, appeared much fewer or not.
We performed this study to evaluate the role of Interleukin-17 (IL-17) and Interleukin-18 (IL-18) in insulin resistance during normal pregnancy. This descriptive cross sectional study was carried out on 97 healthy pregnant women including 32, 25, and 40 individuals in the first, second, and third trimesters, respectively, and on 28 healthy non pregnant women between the autumn of 2012 and the spring of 2013. We analyzed the serum concentrations of IL-17 and IL-18 by using the enzyme linked immunosorbent assay (ELISA). Insulin resistance was measured by homeostasis model assessment of insulin resistance equation. No significant differences between the demographic data of the pregnant and non pregnant groups were observed. Insulin resistant in pregnant women was significantly higher than the controls (p=0.006). Serum IL-17 concentration was significantly different in non pregnant women and pregnant women in all gestational ages (p<0.05). Serum IL-18 level was significantly lower in subjects with first, second, and third trimesters of pregnancy in compared to non pregnant women (p<0.05). No significant correlations were found between serum IL-17 and IL-18 levels with insulin resistance (r=0.08, p=0.34 vs. r=0.01, p=0.91, respectively). Our data suggested that IL-17 and IL-18 do not appear to attribute greatly to pregnancy deduced insulin resistance during normal pregnancy.

Objective: This study was conducted to investigate the impact of previous delivery mode on pregnancy outcomes in patients with secondary infertility after frozen-thawed embryo transfer. Methods: This prospective observational study included 140 patients experiencing secondary infertility. Of these, 70 patients had a previous cesarean delivery (CD), while the remaining 70 patients had a previous normal vaginal delivery (NVD). The primary outcome was the implantation rate. The secondary outcomes included rates of clinical pregnancy, chemical pregnancy, miscarriage, and ectopic pregnancy. Results: The comparison of all fertility outcomes between the two groups revealed no statistically significant differences. The implantation rate was 40.4% in the CD group and 41.7% in the NVD group (p=0.842). The clinical pregnancy rate was 50% in the CD group and 49.3% in the NVD group (p=0.932), while the chemical pregnancy rate was 14.6% in the CD group and 19% in the NVD group (p=0.591). The miscarriage rates in the CD and NVD groups were 20% and 17.6%, respectively (p=0.803). One case of tubal ectopic pregnancy occurred in the NVD group (1.4%). Conclusion: The mode of prior delivery did not significantly impact pregnancy outcomes following frozen-thawed embryo transfer.
The effect of splenectomy on the concentration of progesterone in pregnant wistar rats showing 4-day regular estrous cycles more than two cycles were investigated. The day after mating with the same male animal was designated day 0 of pregnancy. Splenectomy was conducted on day 0 (early-S), 6 (middle-S) and 13 (late-S) of pregnant rat, respectively. Blood sample were collected at day 1, 7, 14 and 21 of pregnancy. Concentraton of serum progesterone was determined by emzyme-linked immunosorbent assay (ELISA) using specific antibodies and emzyme conjugate(progesterone peroxidase labeled). In normal pregnant rats, serum progesterone concentrations were increased significantly (P<0.05) on day 7 and 14 of pregnancy compared with on day 1 of pregnancy and then dramatically decreased on day 21. However in early and middle splenectomized rats, progesterone concentrations were significantly (P<0.05) decreased on day 7 and 14 of pregnancy compared with same day of normal rats. These observations indicate that luteotropic action and steroidogenesis by corpus luteum of ovary may be regulated by splenocytes in rat.
Decorin (DCN) is a member of small leucine-rich proteoglycans which are ubiquitous components of the extracellular matrix. It regulates many physiological processes, such as matrix formation, collagen fibrillogenesis, angiogenesis, cancer growth, and cardiovascular diseases. It has been shown that DCN is expressed in the uterus during pregnancy and modulates implantation and decidualization for the establishment and maintenance of pregnancy in mice and humans. Expression of DCN in the uterine endometrium during pregnancy has not been investigated in pigs. Thus, this study investigated expression of DCN in the uterine endometrium during the estrous cycle and pregnancy in pigs. Uterine endometrial tissues were from day (D) 12 and 15 of the estrous cycle and D12, D15, D30, D60, D90, and D114 of pregnancy. Northern blot and real-time RT-PCR analyses showed that expression of DCN mRNA was detected throughout the estrous cycle and pregnancy with the highest levels during mid pregnancy. In situ hybridization analysis showed that DCN mRNA was localized to both luminal and glandular epithelia during the estrous cycle and pregnancy and also to chorionic membrane during mid pregnancy in pigs. To determine whether endometrial expression of DCN was affected by the somatic cell nuclear transfer (SCNT) procedure, DCN mRNA levels in the uterine endometrium from gilts with SCNT embryos on D30 of pregnancy were compared with those from gilts with normal embryos using real-time RT-PCR analysis. The result showed that DCN mRNA levels in the uterine endometrium were not significantly different between gilts with normal embryos and SCNT embryos. These results suggest that DCN may play an important role for endometrial tissue remodeling during mid pregnancy, and DCN expression is not affected by the SCNT procedure at the early stage of pregnancy in pigs.
