The aim of this study was to provide basic information to allow improved nutritional management for velvet production by investigating the effects of dietary protein levels on dry matter intake and production and chemical composition of velvet antler in spotted deer (Cervus nippon). Twenty-four spotted deer stags were assigned to 4 unreplicated groups, Control (15% CP in diet, higher dry matter), CP10 (10% CP), CP15 (15% CP) and CP20 (20% CP). The velvet antlers were harvested from each stag on the 55th day after casting of the buttons from the previous set, measured for their size and weight, and the chemical composition of each antler was determined in three sections (top, middle, and base). Dry matter (DMI) and crude protein (CPI) intake were highest (p<0.05) for the Control and increased progressively (p<0.05) with increasing dietary protein level. Although not significant, mean length and girth of the main antler beam tended to be larger in either left or right beam with increasing protein level in the diet, longest in CP20 and shortest in CP10. Velvet antler production was lowest in CP10 and highest in CP20, which differed significantly (p<0.05). Only negligible differences were found between groups in chemical composition. It is concluded that dietary protein clearly influenced dry matter intake and velvet antler production, whereas there was comparatively little effect of dietary protein on chemical composition of antler in spotted deer.
Two different NokJoongTangs were prepared by boiling antler and various oriental medicinal materials. The phenolic contents and free radical scavenging effects against DPPH(1,1-diphenyl-2-picrylhydrazyl) radical and hydroxyl radical of the NokJoongTangs which are used as the health supplement foods, were investigated. NokJoongTang-II prepared dried antler with various oriental medicinal materials, had higher phenolic content than that of NokJoongTang-I prepared 1mm raw antler with the same materials. In addition, NokJoongTang-II exhibited higher scavenging activities than these of NokJoongTang-I against DPPH radical and hydroxyl radical. These result indicate that free radical scavenging activity of NokJoongTang showed a difference by using raw and dry antler for NokJoongTang preparation, and that the findings also show a positive correlation between radical scavenging activity and total phenolic contents.
본 연구는 꽃사슴에 있어서 사료내 단백질 수준이 녹용 생산성과 조성분 변화에 미치는 영향을 검토하여 녹용 생산성 향상을 위한 기초자료로 제시하고자 꽃사슴 18두를 3개의 처리구로 나누어 각각 사료내 단백질 수준을 10%(T1), 15%(T2) 및 20%(T3)로 하여 녹용 성장기에 사양실험을 실시했다. 건물섭취량 및 조단백질 섭취량 그리고 일당 증체량은 사료의 단백질 수준이 증가함에 따라 높아져 처리간 유의성(P<0.05)이 인정되었다. 생산된 녹용의 주간 길이 및 둘레의 경우도 사료의 단백질 수준이 높아질수록 증가하였으나 차이에 대한 유의성은 인정되지 않았다. 녹용 생산성은 역시 사료의 단백질 수준이 증가함에 따라 유의적으로 증가하여(P<0.05) T1구에서 가장 낮은 생산량을 나타냈고 T3에서 가장 높은 생산량을 기록했다. 녹용의 조단백질은 T1구에 비해 T3구에서 다소 낮았으며 조회분은 반대의 경향을 나타냈으나 유의적 차이는 인정되지 않았다. 결론적으로 녹용의 생산성을 증가시키기 위해서는 사료내 단백질 수준을 적정 수준으로 유지해 줄 필요가 있으나 녹용 품질에는 별 영향을 미치지 않는 것으로 판단된다.
본 실험은 기존 사슴사료에 있어서 육계분 발효사료의 대체 가능성을 검토하기 위해 숫사슴에 있어서 육계분 발효사료의 첨가수준을 0, 15, 및 30%로 달리하여 소화율, 증체량, 건물채식량 및 질소출납에 미치는 영향을 검토하였다. 모든 처리구에서 실험사슴들은 비슷한 수준의 채식량을 나타내어 건물기준으로 체중의 약 3% 정도의 채식량을 기록하고 있어 육계분 발효사료를 30%까지 첨가한 경우 숫사슴의 채식 기호도에는 부정적인 영향이 나타나지 않는 것으로 판단되었다. 건물소화율은 Control이 76.5%로 가장 높았고, T2가 70.3%로 가장 낮아 유의성(P<0.05)이 인정되었다. 조단백질의 경우도 Control구가 75.8%로 가장 높은 소화율을 나타냈으며, T2가 70.2%로 가장 낮은 소화율을 나타내어 처리간에 유의성(P<0.05)이 인정되었다. 조섬유 소화율은 Control구 70.8.%, T1구 62.7%, T2구 56.0%로 육계분 발효사료의 첨가수준이 높아짐에 따라 조섬유 소화율이 저하되어 Control구와의 사이에 유의성(P<0.05)이 인정되었다. 건물 및 가소화 건물섭취량 모두 육계분 발효사료의 첨가수준이 높아질수록 약간씩 저하되는 경향은 있었으나 그 차이에 대한 유의성은 인정되지 않았다. 실험사슴들의 일당 증체량은 T1구에서 가장 높았으며 Con- trol구에서 가장 낮았으나 처리간의 차이는 유의성이 인정되지 않았다. 질소섭취량은 T1구가 40.2g으로 가장 많았으며 T2구가 38.0g으로 가장 낮았으나 처리간의 유의성은 인정되지 않았다. 분 중 질소의 양은 T2구에서 가장 높았으며 뇨 중 질소의 양은 Control구가 가장 높았다(P<0.05). 체내 질소 축적량은 T1구에서 가장 높았으나 나머지 처리구들과 비슷한 수준을 유지했다.
Numerous plants have been documented to contain phenolic compounds. Thymol is one among these phenolic compounds that possess a repertoire of pharmacological activities, including anti-inflammatory, anticancer, antioxidant, antibacterial, and antimicrobial effects. Despite of the plethora of affects elicited by thymol, its activity profile on gastric cancer cells is not explored. In this study, we discovered that thymol exerts anticancer effects by suppressing cell growth, inducing apoptosis, producing intracellular reactive oxygen species, depolarizing mitochondrial membrane potential, and activating the proapoptotic mitochondrial proteins Bax, cysteine aspartases (caspases), and poly ADP ribose polymerase in human gastric AGS cells. The outcomes of this study displayed that thymol, via an intrinsic mitochondrial pathway, was responsible for inducing apoptosis in gastric AGS cells. Hence, thymol might serve as a tentative agent in the future to treat cancer.
Angiotensin I-converting enzyme (ACE) inhibitory activities of elk antler hydrolysates prepared with three kinds of proteases, pepsin, trypsin and $\alpha-chymotrypsin$, were investigated. The ACE inhibitory activity of the pepsinolytic hydrolysate was the highest with an $IC_{50}$ value of $9.3\mu g/mL.$ In addition, three kinds of pepsinolytic hydrolysates with relatively high molecular weights (over 10,000 Da), medium molecular weights (5,000 to 10,000 Da), and low molecular weights (below 5,000 Da) were fractionated using an ultrafiltration membrane system. The below 5,000 Da hydrolysate exhibited the highest ACE inhibitory activity. These results indicate that the pepsinolytic hydrolysates of elk velvet antler could be a good source of peptides with ACE inhibitory activity.
We applied enzymatic hydrolysis and tangential flow filtration (TFF) to purify a novel anticancer peptide from Mytilus coruscus (M. coruscus) and investigated its anticancer properties. To prepare the peptide, eight proteases were employed for enzymatic hydrolysis. Pepsin hydrolysates, which showed clearly superior cytotoxic activity on prostate cancer cells, were further purified using a flow filtration system using a TFF and consecutive chromatographic methods. Finally, a novel anticancer peptide was obtained, and the sequence was identified as Ala-Phe-Asn-Ile-His-Asn-Arg-Asn-Leu-Leu. The peptide from M. coruscus effectively induced cell death on prostate, breast and lung cancer cells but not on normal liver cells. This is the first report of an anticancer peptide derived from the hydrolysates of M. coruscus.
본 실험은 육림부산물 발효사료의 사슴용 사료가치를 평가하기 위해 꽃사슴에게 육림부산물 사일리지(FBS), 칡 사일리지(ARS) 및 수입 갈잎건초(OLH)를 급여하여 건물채식량, 소화율 및 질소출납을 상호 비교하였다. 건물소화율은 ARS가 가장 높았고 OLH가 가장 낮아 처리구간에 유의성(P$<$0.05)이 인정되었다. 조단백질 소화율의 경우는 OLH구가 FBS와 ARS구에 비해 현저히 낮은(P$<$0.05) 소화율을 나타냈다. 조섬유 소화율의 경우는 FBS가 가장 높고 OLH가 가장 낮았다. 건물채식량은 OLH가 가장 높아 ARS와의 사이에는 유의성(P$<$0.05)이 인정되었으나 FBS와의 사이에는 인정되지 않았다. 그러나 체내에서 이용 가능한 가소화 건물채식량으로 환산했을 경우 차이가 거의 없었다. 체내에 축적된 질소와 섭취질소에 대한 축적질소의 비율은 처리구간에 유의성은 인정되지 않았으나 FBS가 가장 높아 체내 질소의 이용효율이 가장 좋았던 것으로 평가되었다. 결과적으로 육림부산물 사일리지는 다양한 사료자원으로 구성되어 사슴의 기호성이 양호하고 높은 소화율과 건물채식량 및 영양소의 체내 이용성을 나타내고 있어 사슴용 사료로서 충분한 가치를 갖고 있는 사료자원으로 평가되었다.
The antioxidative activity of various enzymatic extracts from leaves of Perilla frutescens var. japonica was evaluated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. For this study, the leaves were enzymatically hydrolyzed by 8 carbohydrases (Dextrozyme, AMG, Promozyme, Maltogenase, Termamyl, Viscozyme, Celluclast, and BAN) and 9 proteases [Flavourzyme, Neutrase, Protamex, Alcalase, PP-trypsin (trypsin from porcine pancreas), papain, pepsin, $\alpha$-chymotrypsin, and BP-trypsin (trypsin from bovine pancreas)]. The DPPH radical scavenging activities of Promozyme and Alcalase extracts were the highest, and the $IC_{50}$ values were 77.25 and $109.66\;{\mu}g/mL$, respectively. All enzymatic extracts of the leaves scavenged hydroxyl radical, and the $IC_{50}$ values of Celluclast and pepsin extracts which were the highest activity were 243.34 and $241.86\;{\mu}g/mL$, respectively. The BAN and $\alpha$-chymotrypsin extracts showed the highest scavenging activities, and the $IC_{50}$ values were 21.13 and $33.23\;{\mu}g/mL$, respectively. The pepsin extracts from the leaves showed protective effect on $H_2O_2$-induced DNA damage. In addition, the pepsin extracts decreased cell death in PC-12 cells against $H_2O_2$-induced oxidative damage. The findings of the present study suggest that enzymatic extracts of the leaves possess antioxidative activity.
