The mature zygotic embryos of the Hevea brasiliensis were cryopreserved through the use of the vitrification and encapsulation/dehydration techniques. In all the experiments, the zygotic embryos were pre-cultured for three days in the MS medium supplemented with 0.3 M sucrose before they were used for the cryopreservation technique. In the vitrification procedure, the effect of the plant vitrification solutions (PVS2 and PVS3) and exposure time were studied. The highest survival rate (88.87%) and regrowth (66.33%) were achieved when the precultured zygotic embryos were incubated in a loading solution for 20 minutes at $0^{\circ}C$. They were subsequently exposed to PVS2 for 120 minutes at $0^{\circ}C$ and plunged directly into liquid nitrogen. Cryopreservation by the encapsulation-dehydration method was successfully done by leaving the encapsulated zygotic embryos in a laminar flow for 4 hours prior to plunging into a LN. The survival rate and regrowth of the encapsulated zygotic embryos were 37.50% and 27.98%, respectively. The cryopreserved zygotic embryos were able to develop into whole plants.
A limnological survey was conducted in a reservoir, Lake Hoengsung located in Kangwondo, Korea, from July 2000 to September 2001 on the monthly basis. Phosphorus loading from the watershed was estimated by measuring total phosphorus concentration in the main tributary. Secchi disc transparency, epilimnetic (0-5 m) turbidity, chlorophyll a (Chl-a), total phosphorus (TP), total nitrogen(TN) and silica concentration were in the range of 0.9-3.5 m, 0.1-8.5 NTU, 0.3-32.4 mgChl $m^{-3}$, 5-46 mgP $m^{-3}$, 0.83-3.55 mgN $L^{-1}$ and 0.5-9.6 mgSi $L^{-1}$, respectively. Green algae and cyanobacteria dominated phytoplankton community in warm seasons, from July through October, 2000. In July a green alga (Scenedesmus sp.) was dominant with a maximum cell density of 10,480 cells mL. Cyanobacteria (Microcystics sp.) dominated in August and September with cell density of 3,492 and 295 cells mL ,respectively. Species diversity of phytoplankton was highest (2.22) in July. The trophic state of the reservoir can be classified as eutrophic on the basis of TP, Chl-a, and Secchi disc transparency. Because TP concentration was high in flood period, most of phosphorus loading was concentrated in rainy season. TP loading was calculated by multiplying TP and flow rate. The dam managing company measured inflow rate of the reservoir daily, while TP was measured by weekly surveys. TP of unmeasured days was estimated from the empirical relationship of TP and the flow rate of the main tributary; $TP=5.59Q^{0.45}\;(R^2=0.47)$. Annual TP loading was calculated to be 4.45 tP $yr^{-1}$, and the areal P loading was 0.77 gP $m^{-2}\;yr^{-1}$ which is similar to the critical P loading for eutrophication by Vollenweider's phosphorus model, 0.72 gP $m^{-2}\;yr^{-1}$.
In this study, the major operating factors in SND(simultaneous nitrification and denitrification) using bioreactor packed with submerged cilia media and granular sulfur such as variation of nitrification rate, organic matter removal efficiency and denitrification efficiency in different DO concentration were mainly evaluated. Synthetic wastewater and actual sewage were used as influent wastewater. Experiment with synthetic wastewater as influent wastewater was divided into three phases with the adjustment of DO concentration. As the results, nitrification efficiency and T-N removal efficiency in the Phase 3(DO 1.0~2.0 mg/L) were 99% and 52.3%, which is significantly greater than those in other two phases. Also, loading rate and denitrification efficiency of SCPGS(Submerged Cilia media Packed with Granular Sulfur) were calculated as $0.44kg\;NO_3^--N/m^3-day$ and 50%, respectively. On the other hand, nitrification rate was decreased from 99% to 64% according to the DO concentration with the variation from 3.0~3.5 mg/L(phase1) to 0.4~0.6mg/L(phase2). Although the nitrification rate was decreased in 64% according to the variation of the DO concentration, T-N removal rate was rapidly increased to 49% by increasing of the denitrification efficiency. Experiment with actual sewage as influent wastewater was carried out to evaluate efficiency of SCPGS in real operation condition of full-scale sewage water treatment plant. At the time, T-N removal rate in this experiment and full-scale wastewater treatment plants were given by 43% and 20%, respectively. The above results indicate that SCPGS can be used as an advanced treatment process for economical efficiency considered.

In this study, nitrogen removal characteristics of SBR3 process, which is two-sludge system of sequencing batch reactor(SBR) type, were investigated, with comparison of those of SBR1 process, which is conventional SBR process, and SBR2 process, which was designed to enhance denitrification efficiency through step-feeding of wastewater, using domestic wastewater. SBR3 process of two-sludge type can perform external nitrification, on which nitrification occurs in separated reactor, and enhanced denitrification using biosorbed organics. In the results with domestic wastewater, T-N removal efficiency of SBR3 process was better than those of SBR1 and SBR2 processes. It was observed that bigger difference of T-N removal efficiency between SBR3 process and SBR1 & SBR2 processes was showed at low C/N ratio than that at high C/N ratio resulting from more efficient use of organics by biosorption mechanism in denitrification of SBR3 process than those of SBR1 and SBR2 processes. In addition, T-N removal efficiency of SBR3 process according to influent T-N loading rate was better than those of SBR1 and SBR2, even though influent T-N loading rate of SBR3 process was higher than that of SBR1 and SBR2 process resulting from operation of SBR3 process in short hydraulic retention time(HRT) by external nitrification.
In order to develop constructed wetlands for treating hydroponic wastewater in greenhouse, actual constructed wetlands were used the obtained optimum condition in previous study, and the removal rate of pollutant in the water according to 4 kinds connection method of piping such as system A (UP-UP stream), system B (UP-DOWN system), system C (DOWN-UP stream) and system D (DOWN-DOWN stream) were investigated. Removal rate of biochemical oxygen demand (BOD), chemical oxygen demand (COD), suspended solids (SS), total nitrogen (T-N) and total phosphorus (T-P) by system A (UP-UP stream) connection method in actual constructed wetlands were slightly higher than other systems. At the system A, the removal rate of BOD, COD, SS, T-N and T-P were 88, 77, 94, 54 and 94%, respectively. Under different hydroponic wastewater loading, the removal rates of pollutants were higher in the order of $75L\;m^{-2}day^{-1}{\fallingdotseq}150L\;m^{-2}day^{-1}$$$\geq_-$$$300L\;m^{-2}day^{-1}$. Therefore, optimum connection method was system A for treating hydroponic wastewater in greenhouse.
Two climate change scenarios, the RCP (Representative Concentration Pathways) 4.5 and the RCP 8.5 in the fifth Assessment Report (AR5) by Intergovernmental Panel on Climate Change (IPCC), were applied in the Yocheon basin area using the SWAT (Soil and Water Assessment Tool) model to estimate changes in flow rates and pollutant loadings in the future. Field stream flow rate data in Songdong station and water quality data in Yocheon-1 station between 2013~2015 were used for model calibration. While $R^2$ value of flow rate calibration was 0.85 and $R^2$ value of water qualities were in the 0.12~0.43 range. The total study period was divided into 4 sub periods as 2030s (2016~2040), 2050s (2041~2070) and 2080s (2071~2100). The predicted results of flow rates and water quality concentrations were compared with results in calibrated periods, 2015s (2013~2015). In both RCP scenarios, flow rate and TSS (Total Suspended Solid) loadings were estimated to be in increasing trend while TN (Total Nitrogen) and TP (Total Phosphorus) loadings showed decreasing patterns. Also, flow rates and pollutant loadings showed larger differences between the maximum and the minimum values in RCP 4.5 than RCP 8.5 scenarios indicating more severe effect of drought and flood, respectively. Dependent on simulation period and rainfall periods in a year, flow rate, TSS, TN and TP showed different trends in each scenario. This emphasizes importance of considerations on time and space when analyzing climate change impacts of each variable under various scenarios.

Treatability tests were conducted to study the feasibility of EMMC process as a recycling-water treatment system in high density seawater aquaculture farm. To study the effect of organic and ammonia nitrogen loading rate on system performance, hydraulic retention time was reduced gradually from 12hr to 10min. The conclusions are can be summarized as follows. When the system HRT was reduced from 12hr to 2hr gradually, there was little noticeable change(reduction) in ammonia nitrogen removal efficiencies. However, removal efficiencies were decreased dramatically when the system was operated under the HRT of less than 2hr. In case of organics(COD), there was no dramatic change in removal efficiencies depending on HRT reduction. COD removal efficiencies were maintained successfully higher than 9% when the system was operated at tile HRT of 10 min. System performances depending on media packing ratio in the reactors were also evaluated. There were little differences in each reactor performances depending on media packing ratio in reactor when the reactors were operated under the HRT of longer than 1hr. However, differences in reactor performances were considerably evident when the reactors were operated under the HRT of shorter than 1hr. When comparing reactor performance among 25%, 50%,7 5% packed reactor, it can be judged that media packing ratio more than 50% plays no significant role in increasing reactor performance. For this reason, packing the media less than 50% is more reasonable way in view of economic. Such a tendency shown in COD removal efficiencies well agreed with the variation of ammonia-nitrogen removal efficiencies according to the media packing ratio in reactors at each HRT. Difference in effluent ammonia-nitrogen concentration between 50% media packing reactor and 75% media packing reactor was negligible. When comparing with the results of 25% packing reactor, difference was not so great.

The purpose of this study was to validate the EFDC model for the weir pool of Gangjeong-Goryong Weir located in Nakdong River, and evaluate the effect of flow control and phosphate loading reduction on the water quality and algae biomass by group (Diatom, Green, Cyanobacteria). As a result of model validation using 2018 experimental data,the time series of water level and vertical distribution of water temperature, DO, organic matter, nitrogen, and phosphorus time series were properly simulated. Seasonal fluctuations of algae biomass by group were adequately reproduced, but the deviations between measured and simulated values were significant in some periods. As a result of scenario simulations to control the water level and flow rate, the thermal stratification was resolved as the water level was lowered and the flow rate increased. The flow velocity at which the water temperature stratification was resolved was about 0.1 m/s, which is consistent with the previous study results of Baekje Weir in Geum River. Simulations of the 2Q flow scenario showed that Chl-a decreased by 8.7% and the cell density of diatom and green algae declined. The cell density of cyanobacteria increased, however, because the high concentrations of cyanobacteria in the upstream boundary conditions directly affected downstream due to increased flow velocity. In the scenario simulation of reducing the influent phosphate load concentration (average 0.056 mg/L) to 50%, Chl-a decreased by 13.6%.The results suggest that the upstream algae concentration and phosphorus load reduction should be considered simultaneously with hydraulic control to prevent algal overgrowth of Gangjeong-Goryong Weir.
The Keum river is one of the important river in Korea and has a drainage area of 9,873$\textrm{km}^2$. The Keum river is deepening pollution state due to development of the lower city and construction of a industrial complex. The water quality of the Keum river come to eutrophication state and belong to III grade of water quality standard. The concentration BOD in river is affected by the organic loading from a tributary and the algae biomass that largely happen to under eutrophication state. In the eutrophic water mass such as the Keum river, the autochthonous BOD was very important part for making a decision of water quality management, because it was accounted for majority of the total BOD. The purpose of this study was to survey the chatacteristics of water quality in summer and to estimate reaction coefficient. Also, we studied to correlationship between chlorophyll a and BOD(COD) for estimation of the autochthonous BOD. The correlationship between chlorophyll a and BOD(COD) were obtained through the culture experiment of phytoplankton in the laboratory. The results of this study may be summarized as follows ; The characteristics of water quality in summer were belong to III~IV grade of water quality standard as BOD and nutritive condition is very high. The BOD, ammonia nitrogen and phosphate loadings in Miho stream which inflowing untreated sewage from Chungju city was occupied with 64.07%, 26.36%, 46.08%, respectively. Maximum nutrient uptake (Vmax) was 0.4400$\mu$M/hr as substrate of ammonia nitrogen, 0.1652$\mu$M/hr as substrate of phosphate. Maximum specific growth rate ($\mu$max) was 1.2525$hr^{-1}$ as substrate of ammonia nitrogen, 1.5177$hr^{-1}$ as substrate of phosphate. The correlation coefficient between chlorophyll a and BOD by the culture experiment were found to be 0.911~0.935 and 0.942~0.947 in the case adding nutrient and no adding nutrient, respectively. The correlation coefficient between chlorophyll a and COD through the culture experiment were found to be 0.918~0.977 and 0.880~0.931 in the case adding nutrient and no adding nutrient, respectively. The autochthonous BOD(COD) was estimated to the relationship between BOD(COD) and chlorophyll a. The regression equation were found to be autochthonous BOD=(0.045~0.073)${\times}chlorophyll$ a and autochthonous $COD=(0.137~0.182){\times}chlorophyll$ a.
To effectively treat the domestic sewage that was produced on a small-scale in farming and fishing village in order to encourage an ecologically friendly environment, a small-scale sewage treatment apparatus using natural purification methods that consisted of an aerobic and an anaerobic plots were constructed. The efficiency of sewage treatment according to the sewage loading was investigated to obtain the optimum sewage loading in small-scale sewage treatment apparatus. Removal rate of pollutants according to the sewage loading were in the order of $150\;Lm^{-2}day^{-1}{\fallingdotseq}300\;Lm^{-2}day^{-1}>600\;Lm^{-2}day^{-1}$. Therefore, the optimum sewage loading was 300 L m-2 day-1. Under the optimum sewage loading, removal rate of BOD, $COD_{Mn}$, turbidity, T-N and T-P were 99, 94, 99, 49 and 89%, respectively. However, to satisfy the water quality standard in effluent in small-sclae sewage treatment apparatus for domestic sewage treatment, the low removal efficiency of T-N and T-P must be improved. So to improve the removal rate of T-N and T-P, the efficiency of sewage treatment according to the improved sewage treatment process such as, re-treatment at aerobic plot, anaerobic condition of aerobic plot, changing the filter media sizes and the depths in anaerobic plot, and also addition of oyster shells to filter media at anaerobic plot were investigated. In case of 150 cm depth in anaerobic plot with filter medium A (effectivity particle size 1.50 mm) and addition of oyster shells to filter media at anaerobic plot, removal rate of T-N and T-P in both plots were increased by 10 and 3%, and 14 and 7% in comparison with 100 cm depth in anaerobic plot with filter medium B(effectivity particle size 0.95 mm), respectively. The optimum improved sewage treatment process in small-scale sewage treatment apparatus were 150 cm depth in anaerobic plot with filter medium A and addition of oyster shells to filter media at anaerobic plot.
