Simultaneous Utilization of Two Different Pathways in Degradation of 2,4,6-Trinitrotoluene by White Rot Fungus Irpex lacteus

  • This study confirmed that white rot fungus Irpex lacteus was able to metabolize 2,4,6-trinitrotoluene (TNT) with two different initial transformations. In one metabolic pathway of TNT a nitro group was removed from the aromatic ring of TNT. Hydride-Meisenheimer complexes of TNT (H/sup -/-TNT), colored dark redo were confirmed as the intermediate in this transformation by comparison with the synthetic compounds. 2,4-Dinitrotoluene as a following metabolic product was detected, and nitrite produced by denitration of $H^-$-TNT supported this transformation. In the other TNT pathway, nitro groups in TNT were successively reduced to amino groups via hydroxylamines. Hydroxylamino-dinitrotoluenes and amino-dinitrotoluenes were identified as the intermediates. The activity of a membrane-associated aromatic nitroreductase was detected in the cell-free extract of I. lacteus. This enzyme catalyzed the nitro group reduction of TNT with NADPH as a cofactor, Enzyme activity was not observed in the presence of molecular oxygen.

Treatment of surface water using cold plasma for domestic water supply

  • This paper presents the results of using cold plasma to treat surface water for domestic use purpose. Experimental results showed that cold plasma was an effective method for destroying bacteria in water. After treatment with cold plasma, concentration of coliform and Escherichia coli dramatically reduced. Besides, cold plasma significantly removed water odor, increased dissolved oxygen and decreased the concentration of chemical oxygen demand. However, cold plasma significantly raised the concentration of nitrite and nitrate. Other disadvantages of treating with cold plasma were conductivity increase and pH reduction. Pretreatment steps of coagulation, flocculation, sedimentation and sand filtration followed by disinfection with cold plasma exhibited a high efficiency in surface water treatment. All parameters of surface water after treatment by using the prototype satisfied with the allowance standard of domestic water quality.

Antimutagenic and Antioxidative Effects of Seafood Bun with Kimchi (김치 해물만두의 항돌연변이 및 항산화 효과)

  • Antimutagenic, antioxidative and nitrite-scavenging effects of seafood bun were investigated. Each samples were extracted with methanol. Aflatoxin Bl(AFB1) was used as mutagen. Seafood bun added 20% kimchi(5mg/plate) reduced mutagenicity of AFB1 for Salmonella typhimurium TA and YG 1024 to 47% and 61%, rrespectivery. The reduction rate of seafood bun added 20% kimchi was higher relative to that of other samples. Mydrogen donating activity in all buns showed over 50% and seafood bun added 20% kimchi was higher than others, but the activity was lower than that of 0.1% BHT. The peroxide value for linoleic acid increased during the storage, the values in seafood bun added 20% kimchi was higher than others, but the activity was lower than that of 0.1% BHT. The peroxide value for linoleic acid increased during the storage, the values in seafood bun added 20% kimchi was significantly lower and the values in other buns were a lower than that of control, the values in all of the samples were higher than that in 0.1% BHT. Among the samples tested, the TBA value in the seafood bun added 20% kimchi for liver homogenate of rat was the lowest. Nitrite-scavenging effect in all the samples tested was higher than 50%.

The Influence of Some Soil-treated Herbicides on the Mineralization of Nitrogen Fertilizers II. In an upland soil (토양처리형 제초제가 질소비료의 무기화작용에 미치는 영향 II 밭 토양 조건)

  • Effect of Simazine(2-chloro-4,6-bis (ethylamino)-s-triazine), Nitrofen (2, 4-dichloro-4' -nitrodi­phenylether), Propanil (:3, 4-dichloropropionanilide), and Butachlor (2-chloro-2, 6-diethyl N-(buthoxy­menthyl) acetanilide on urea hydrolysis and subsequent nitrification was investigated in an upland soil incubated at $20\pm1^{\circ}C$. 1. All the herbicides tested had no effect on the hydrolysis of urea to ammonia at the recommended rates. Butachlor, at ten and fifty times the recommended rate, and Nitrofen, at fifty times the recommeded rate, depressed urea hydrolysis, resulting in reduction of ammonia. But the depressive effects were temporary, disappearing soon. Simazine and Propanil had no detrimental effect on urea decomposition at all the treated rates. 2. Also, all the chemicals tested had no effect on the nitrification process at the recommended rates. At higher concentrations of ten and fifty times the recommended rate Butachlor and Nitrofen inhibited the oxidation of nitrite, and propanil long inhibited the oxidation of ammonium to nitrite, but was inactive against nitrite oxidizer. These inhibitive effects of the chemicals, however, disappeared in the later period of incubation. Simazine had no effect on the nitrification process at all the treated rates. 3. The trend of change in soil pH of both the treated and untreated plots well reflected the change of soil nitrogen forms during incubation. No direct effect of the chemicals on soil pH was obserbed.

Effect of Influent C/N Ratio and DO on Denitrification of Nitrate Polluted Groundwater in a Biofilter Process (Biofilter 공정에서 유입 C/N비와 DO가 지하수의 질산성 질소제거에 미치는 영향)

  • In this study, effects of influent C/N(COD/Nitrate) ratio and dissolved oxygen(DO) concentration on biological nitrate removal from groundwater were investigated in the fixed-type biofilter. Influent nitrate of 30 mg/L was removed completely by biological denitrification at the C/N ratio of 10 and 4.0, while residual nitrate of 5 mg/L occurred at the C/N ratio of 2.0, which resulted from deficiency of organic electron donor. Furthermore, nitrite was accumulated up to about 5 mg/L as the C/N ratio decreased to 2.0. Increase in DO concentration also inhibited denitrification activity at the relatively high C/N ratio of 5.0, which decreased the nitrate removal efficiency. Although the influent DO concentration was reduced as low as 0.3 mg/L using sodium sulfite($Na_2SO_3$), effluent nitrite was up to 3.6 mg/L. On the other hand, nitrate was completely removed without detection of nitrite at the DO concentration of 0.3 mg/L using nitrogen gas($N_2$) sparging. The organic matter for denitrification in biofilter were in the range from 3.0 to $3.5gSCOD/g{NO_3}^--N$, while utilized these values increased at the high DO concentration of 5.5 mg/L. In addition to the high DO concentration and the low influent C/N ratio, DO control by chemical such as sodium sulfite affected on biological denitrification, which resulted in the reduction of nitrate removal efficiency and nitrite build-up in a biofilter.

  • The SBR(Sequencing Batch Reactor) process is ideally suited to treat high loading wastewater due to its high dilution rate. SBR operates by a cycle of periods consisting of filling, reacting, settling, decanting and idling. The react phases such as aeration or non-aeration, organic oxidation, nitrification, denitrification and other bio-logical reactions can be achieved in a reactor. Although the whole reactions can be achieved in a SBR with time distributing, it is hard to manage the SBR as a normal condition without recognizing a present state. The present state can be observed with nutrient sensors such as ${NH_{4}}^{+}-N$, ${NO_{2}}^{-}-N$, ${NO_{3}}^{-}-N} and ${PO_{4}}^{ 3-}-P.$ However, there is still a disadvantage to use the nutrient sensors because of their high expense and inconvenience to manage. Therefore, it is very useful to use common on-line sensors such as DO, ORP and pH, which are less expensive and more convient. Moreover, the present states and unexpected changes of SBR might be predicted by using of them. This study was conducted to get basic materials for making an inference of SBR process from ORP(oxidation reduction potential) of synthetic wastewater. The profiles of ORP, DO, and pH were under normal nitrification and denitrification were obtained to compare abnormal condition. And also, nitrite and nitrate accumulation were investigated during reaction of SBR. The bending point on ORP profile was not entirely in the low COD/NOx ratio condition. In this case, NOx was not entirely removed, and minimum ORP value was presented over -300mV. Under suitable COD/NOx ratio which complete denitrification was achieved, ORP bending point was observed and minimum ORP value was under -300m V. Under high COD/NOx ratio, ORP bending point was not detected at the first subcycle because of the fast denitrification and minimum ORP value was under -300mV at the time.

Influence of freeze-dried acid whey addition on biogenic amines formation in a beef and deer dry fermented sausages without added nitrite

  • Objective: Aim of this study was to evaluate the influence of freeze-dried acid whey addition and the use a game meat (fallow deer) on a microbial content and the biogenic amines formation in dry fermented sausages. Methods: The experiment involved dry fermented sausages made in two variants from beef and from fallow deer. Each variant was divided into five groups: control (with a curing mixture), reference (with a sea salt), sample with a liquid acid whey and two samples with the addition of reconstituted freeze-dried acid whey in different concentrations. Changes in lactic acid bacteria (LAB), Enterobacteriaceae content and biogenic amines content were determined. Results: The microbial content changes suggest that addition of acid whey slightly affected LAB content in comparison with the control and reference sample, but the addition of freezedried acid whey resulted in a reduction of Enterobacteriaceae content in the sausages from fallow deer or a similar level in the beef sausages compared with the control and reference sample. Both changes in LAB and Enterobacteriaceae content were more evident in case of sausages made from fallow deer. Addition of acid whey (liquid and a higher amount of freezedried) and use of fallow deer meat to produce the sausages resulted in a significant reduction of total biogenic amines content. Conclusion: The addition of acid whey (liquid and higher amount of freeze-dried) resulted in a significant reduction of total biogenic amines content in dry fermented sausages made from fallow deer meat.

Relationship between plasma asymmetric dimethylarginine and nitric oxide levels afects aerobic exercise training-induced reduction of arterial stifness in middle-aged and older adults

  • [Purpose] Aerobic exercise training (AT) reverses aging-induced deterioration of arterial stiffness via increased arterial nitric oxide (NO) production. Asymmetric dimethylarginine (ADMA), an endogenous inhibitor of NO synthase, was decreased by AT. However, whether AT-induced changes in ADMA levels are related to changes in nitrite/nitrate (NOx) levels remains unclear. Accordingly, we aimed to clarify whether the relationship between plasma ADMA and NOx levels afected the AT-induced reduction of arterial stifness in middle-aged and older adults. [Methods] Thirty-one healthy middle-aged and older male and female subjects (66.4 ± 1.3 years) were randomly divided into two groups: exercise intervention and sedentary controls. Subjects in the training group completed an 8-week AT (60%-70% peak oxygen uptake [${\dot{V}}O_{2peak}$] for 45 min, 3 days/week). [Results] AT signifcantly increased ${\dot{V}}O_{2peak}$ (P < 0.05) and decreased carotid β-stifness (P < 0.01). Moreover, plasma ADMA levels were significantly decreased while plasma NOx levels and NOx/ADMA ratio were significantly increased by AT (P < 0.01). Additionally, no sex diferences in AT-induced changes of circulating ADMA and NOx levels, NOx/ADMA ratio, and carotid β-stifness were observed. Furthermore, the AT-induced increase in circulating ADMA levels was negatively correlated with an increase in circulating NOx levels (r = -0.414, P < 0.05), and the AT-induced increase in NOx/ADMA ratio was negatively correlated with a decrease in carotid β-stifness (r = -0.514, P < 0.01). [Conclusion] These results suggest that the increase in circulating NOx with reduction of ADMA elicited by AT is associated with a decrease in arterial stiffness regardless of sex in middle-aged and older adults.

A Proposal of Sequencing the Combined Processes for Resources Recovery and Nitrogen Removal from Piggery Waste (슬러리형 돈사폐수에서 자원회수와 질소제거를 위한 순차적 결합공정 제안)

  • The combined ADEPT(Anaerobic Digestion Elutriated Phased Treatment)-SHARON(Single reactor system for High Ammonium Removal Over nitrite)-ANAMMOX(Anaerobic ammonium oxidation) processes were operated for resources recovery and nitrogen removal from slurry-type piggery waste. The ADEPT process operated at an acidogenic loading rates of 3.95 gSCOD/L-day, the SCOD elutriation rate and acid production rate were 5.3 gSCOD/L-day and 3.3 gVFAs(as COD)/L-day, respectively. VS reduction and SCOD reduction by the hydrolysis were 13% and 0.19 $gSCOD_{prod.}/gVS_{feeding}$, respcetively. Also, the acid production rate was 0.80 $gVFAs/gSCOD_{prod}$. In methanogenic reactor, the gas production rate and methane content were 2.8 L/day($0.3m^3CH_4/kgCOD_{removal}@STP$) and 77%, respectively. With these operating condition, the removals of nitrogen and phosphorus were 94.1% as $NH_4-N$(86.5% as TKN) and 87.3% as T-P respectively.
