Effects of Opuntia Ficus-indica on Ovalbumin-induced Asthma Model (Ovalbumin으로 유도된 천식 마우스 모델에서 백련초의 효과)

  • Objective : This study was undertaken to investigate effects of Opuntia ficus-indica (OP)extract on an asthma murine model. Methods : The total number of cells and eosinophils in BALF and the infiltration of inflammatory cells into lung tissues were determined by hematoxylin and eosin staining. Results : The number of OVA-induced total cells and eosinophils, a phenomenon of asthma, were decreased by treatment of the animals with OP extract (200 mg/kg) (respectively, p < 0.001 and p<0.01). Furthermore, we showed that OP extract reduced the increased immune cell infiltration induced by ovalbumin (p < 0.01). The levels of interleukin (IL)-4 in BAL fluid were measured by enzyme-linked immunosorbent assay, because the eosinophilia is associated with a T helper (Th) 2 response including IL-4. The level of OVA-induced IL-4 was decreased by OP extract in BAL fluid (p < 0.05). We investigated whether OP extract reduced nitrite (NO) production on lipopolysaccharide (LPS)-stimulated RAW 264.7 sells, because asthma is an inflammatory disease. The level of LPS-induced NO production was decreased by OP extract (50, 100 and 200 mg/ml) on RAW 264.7 cells (respectively, p < 0.05, p<0.01 and p<0.05). Conclusions : Our results indicate that OP extract has an inhibitory effect on lung eosinophilia of asthma and suggest that OP extract is a therapeutic candidate in the treatment of inflammatory disease, including asthma.

Modulation of Cytotoxicity by Nitric Oxide Donors during Treatment of Glioma with Anticancer Drugs

  • Objective : Nitric oxide[NO] is implicated in a wide range of biological processes in tumors and is produced in glioma. To investigate the role of NO and its interaction with the tumoricidal effects of anticancer drugs, we study the antitumor activities of NO donors, with or without anticancer drugs, in human glioma cell lines. Methods : U87MG and U373MG cells were treated with the NO donors sodium nitroprusside[SNP] and S-nitroso-N-acetylpenicillamine[SNAP], alone or in combination with the anticancer drugs 1,3-bis[2-chloroethyl]-1-nitrosourea[BCNU] and cisplatin. Cell viability, cell proliferation, DNA fragmentation, nitrite level, and the expression of Bcl-2 and Bax were determined. Results : NO was markedly increased after treatment with SNP or SNAP; however, the addition of the anticancer drugs did not significantly affect NO production NO donors or anticancer drugs reduced glioma cell viability and, in combination, acted synergistically to further decrease cell viability in a dose- and time-dependent manner. Cell proliferation was inhibited and apoptosis were enhanced by combined treatment. Bax expression was increased by combined treatment, whereas Bcl-2 expression was reduced. The antitumor cytotoxicity of NO donors and anticancer drugs differed according to cell type. Conclusion : BCNU or cisplatin can inhibit cell viability and proliferation of glioma cells and can induce apoptosis. These effects are further enhanced by the addition of a NO donor which modulates the antitumor cytotoxicity of chemotherapy depending on cell type. Further biological, chemical, and toxicological studies of NO are required to clarify its mechanism of action in glioma.

Anti-Arthritic Effect of Sogyunghwalhyel-tang-gamibang (소경활혈탕가미방(疎經活血湯加味方)의 관절염에 미치는 효과)

  • Objectives The aim of this study is to evaluate anti-inflammatory and anti-arthritic effects of Sogyunghwalhyel-tang-gamibang (SGHHTGB) in cell and animal models and also to suggest one of putative mechanisms underlying its anti-arthritic effects. Methods Enzyme-linked immunosorbent assay was applied to measure the concentrations of tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6 and prostaglandin E2 (PGE2) in culture medium and blood serum and nitric oxide (NO) was assayed by Griess reagent. The expressions of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) were analyzed by Western blot method. Results In a cell model using RAW264.7 macrophages stimulated with the endotoxin lipopolysaccharide (LPS), the drug, at its non-cytotoxic concentrations, inhibited the production of the pro-inflammatory cytokine TNF-α, IL-1β and IL-6. In addition, it suppressed the expression of the inflammatory enzyme iNOS and COX-2, and reduced the synthesis of the enzyme product NO (as stable nitrite) and PGE2 in activated macrophages. Meanwhile, in an animal model using rheumatic arthritis (RA) mice induced with injection of type II collagen antibody (CAb) and LPS, the drug improved clinical symptom of arthritis and reduced paw thickness and inflammatory cell infiltration. In blood of RA mice, the drug reduced serum levels of TNF-α, IL-1β, IL-6, nitrite, and PGE2, all inflammatory mediators produced by activated macrophages. Conclusions SGHHTGB may ameliorate CAb and LPS-induced RA in mice, presumably by inactivating macrophages that are capable of initiating joint inflammation by producing pro-inflammatory cytokines and expressing inflammatory enzymes.

Effect of Garlic and Onion Juice Addition on the Lipid Oxidation, Total Plate Counts and Residual Nitrite Contents of Emulsified Sausage during Cold Storage (마늘즙 또는 양파즙 첨가가 유화형 소시지의 저장 중 지방 산화, 총 미생물수 및 아질산염잔존량에 미치는 영향)

  • The objective of this study was to determine the antioxidant and antimicrobial effects of garlic juice and onion juice in emulsified sausage during cold storage. The sausages were into five groups: control, 1% garlic juice (T1), 3% garlic juice (T2), 1% onion juice (T3), and 3% onion juice (T4). Each sausage type was tested in triplicate and assigned to one of four storage periods: 0, 7, 14 and 21 days. As storage time increased, the presence of garlic juice and onion juice resulted in decreased pH, residual nitrite value, and increased peroxide value, TBARS (thiobarbituric acid reactive substance) values, and total plate counts. The pH value, peroxide value, TBARS, residual nitrite and total plate counts were significantly decreased by the addition of garlic and onion juice relative to the control (p<0.05). Especially, T2 was significantly (p<0.05) more effective in delaying lipid oxidation compared to the other treatment groups. However, no significant difference (p<0.05) was found in total plate counts among all the formulations on day 0. Also, the use of garlic juice resulted in much better antioxidant and antimicrobial effects than the use of onion juice and the control. In conclusion, this study demonstrates that the addition of 3% garlic juice (T2) to emulsified sausages tended to improve antioxidative and antimicrobial effects during storage relative to the other treatment groups.

Storage Stability of Cured Cooked Boneless Chicken Drumette under Vacuum and Its Acceptability as Raw Material for Chuncheon Dakgalbi (춘천닭갈비 원료로서 염지 육계 어깨살의 저장특성)

  • This study was conducted to evaluate the acceptability of cured cooked boneless chicken drumettes as a raw material for Chuncheon Dakgalbi and to observe its effect on pH, lipid oxidation, volatile basic nitrogen (VBN) content and total viable count (TVC) during refrigerated storage under vacuum. Boneless chicken drumettes were dry cured with a mixed curing agent (0.20% w/w) consisting of sodium chloride (93.1%), sodium nitrite (5.9%) and sodium bicarbonate (1.0%) for 24 h at $2{\pm}2^{\circ}C$. Cured samples were cooked using an oven at $150^{\circ}C$ for 20 min and stored at $5.5{\pm}0.5^{\circ}C$ for 60 days under vacuum. Non-cured cooked sample was used as a control. Sensory evaluation was performed on day 2 of storage using 22 untrained panelists and samples were mixed with Dakgalbi sauce and reheated using a microwave for 2 min. There were no significant differences between control and treatment group on appearance, flavor, texture, juiciness and overall acceptability. Cured cooked boneless chicken drumettes had significantly (p<0.05) higher pH, lower lipid oxidation, VBN and TVC consistently during storage in comparison with control. These results suggest that dry curing could be applied to boneless chicken drumettes as raw material for Chuncheon Dakgalbi and could extend its shelf life during refrigerated storage under vacuum.

Comparative Study of Anti-oxidant and Anti-inflammatory Activities between Curcumae longae Radix and Curcumae longae Rhizoma (울금과 강황의 항산화 및 항염증 활성 비교연구)

  • Objectives : In this study, we compared the anti-oxidant and anti-inflammatory activities of Curcumae longae Radix (CLRa) and Curcumae longae Rhizoma (CLRh). Methods : We performed 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and 2,2-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation scavenging assays, and determined total polyphenolic content to examine the anti-oxidant effects of CLRa and CLRh. We also evaluated the anti-oxidant effects of CLRa and CLRh against hydrogen peroxide ($H_2O_2$)-induced toxicity in PC12 cells using thiazolyl blue tetrazolium bromide (MTT) and reactive oxygen species (ROS) assays. Next, to compare the anti-inflammatory effects of CLRa and CLRh against lipopolysaccharide (LPS)-induced inflammation in microglia BV2 cells, we measured nitric oxide (NO) assay and inducible nitrite synthase (iNOS) using Western blotting analysis. Results : CLRa showed higher activity in DPPH and ABTS assays and lower total polyphenolic contents compared with CLRh. In PC12 cells, CLRa and CLRh showed no difference in H2O2-induced cell toxicity and ROS overproduction. In BV2 cells, CLRa showed higher effect than CLRh in NO and iNOS production induced by LPS. Conclusions : These results demonstrate that CLRa has higher radical scavenging activities and anti-inflammatory effect in BV2 cells comparing CLRh. However, CLRa and CLRh have no effect and no difference in $H_2O_2$-induced toxicity.

The Effects of Aerobic Exercise Training on Blood Lipid Profiles, Fibrinolytic Activities, and Nitric Oxide Levels in High-fat-diet induced Rats (유산소 운동이 고지방 식이 흰쥐의 지방량, 혈중지질, 혈전용해능 및 산화질소에 미치는 영향)

  • Although exercise training has been utilized to improve vascular function in animals and humans, the impact of moderate intensity exercise training on fibrinolytic activities and nitric oxide (NO) bioavailability has not been well documented. Therefore, the purpose of the current study was to examine the impact of moderate intensity aerobic exercise training on fat mass, blood lipid profiles, fibrinolytic activity, and NO levels in high-fat-diet induced rats. The body weight, fat mass, blood lipid profiles, fibrinolytic activity, and nitrite/nitrate were measured pre- and postexercise (10 weeks) training. The body weight and fat mass reduced significantly in the exercise (EX) group compared to the control (CON) group. Blood lipid profiles and low-density lipoprotein were unchanged in the EX group compared to the CON group. However, triglyceride and free fatty acid were significantly lower in the EX group compared to the CON group, and high-density lipoprotein was significantly greater in the EX group compared to the CON group. In addition, fibrinolytic activity and nitrite/nitrate were significantly greater in the EX compared to the CON group. These results suggest that 10 weeks of the moderated intensity aerobic exercise training improves blood lipid profiles, fibrinolytic activity, and the nitrite/nitrate ratio, which may improve vascular health and reduce obesity-related cardiovascular disease risks in high-fat- diet induced rats.

  • Rhodospirillum rubrum N-1 was inoculated to manipulated swine wastewater of 20,000 mg/L as Biochemical Oxygen Demand (BOD) to study the effect of aeration on swine wastewater deodorization. Biological and physico-chemical parameters were determined at 1 day interval for 9 days. Removals of BOD, volatile fatty acids (VFAs), and phosphate were 54.6%, 87.0%, and 54.5%, respectively. No significant changes were observed in the concentrations of total nitrogen, total phosphorus, nitrate, nitrite, hydrogen sulfide, and mercaptane.

  • For the destruction of toxic chlorinated organic compounds, this study proposes improved supercritical water oxidation method (multistep oxidation) using sodium nitrate as an oxidizer. This method solves the problems involved in the existing supercritical water oxidation method. Multistep oxidation means that $NaNO_3$ is oxidized to $N_2\;via\;NaNO_2$ and NO. Toxic and hard to destroy organic substances like para-dichlorobenzen(4-DCBz), polychlorinate biphenyl(PCB) ware oxidized to non toxic substances in a condition, in which rapid pressure and temperature rise is restrained as much as possible. 4-dichlorobenzene(4-DCBz) and Polychlorinate biphenyl(PCB) in condition$(450^{\circ}C,\;p_w=0.25g/cm^3,\;30min)$ Was discomposed perfectly.

Physicochemical Properties of Functional Herb Mixtures (기능성 천연물 복합제의 이화학적 특성 확인)

  • The herb mixtures that are known to lower blood pressure were selected through oriental medical books and prescriptions and the physicochemical properties of their water extracts were analyzed to examine the possibility as functional food materials. The total yield of 28 water extracts was in the range of $5.33{\sim}36.71%$. Total phenolics and flavonoid content were $204.89{\sim}2543.29mg%\;and\;59.79{\sim}1430.55mg%$, respectively, and especially No. 2, 5, 9, 17, 18, 20, 22, and 26 showed high rates of above 800 mg%. Electron donating ability (EDA) was $7.81{\sim}98.18%$, and the samples that showed high values in EDA were similar to the samples with high values in total phenolics and flavonoid content. Phenolics and flavonoid compounds of the herbs are considered to perform a major role in antioxidation. Nitrite scavenging ability reached the highest at pH 1.2 ($11.70{\sim}96.47%$) and the lowest at pH 6.0 (below 12.77%), which indicates that nitrite scavenging ability decreases when pH increases.
