Serological Investigation of Virus Diseases of Tobacco Plant (Nicotiana tabaccum L.) In Korea (혈청학적 방법에 의한 잎담배 바이러스병의 감염상 조사)

  • A total of 40 virus infected tobacco plants (Nicotiana tabaccum L.) with various symptom types Were collected from Bucheon and Jeonju area by its symptoms were investigated on the incidence of tobacco mosaic virus (TMV), cucumber mosaic virus (CMV), alfalfa mosaic virus (AMV), potato virus X (PVX) and potato virus Y (PVY) by serological methods. van Slogteren's microprecipitin test was applied for the testing of PVX and PVY from infected plants and Ouchterlony agar double diffusion test was used for CMV, TMV and AMV. Results obtained are as follows: 1. TMV, CMV, AMV, PVX and PVY wcre found to occur on the tobacco plants growing in Korea. 2. The prevalence of each of these viruses among the 40 tobacco plants investigated was in the order of AMV: $(67.5\%)>CMV:(60.0\%)>TMY:(47.5\%)>PVY:(17.5\%)>PVX: (10.0\%).$ 3. In Burley variety, the percentage of infection by TMV was $15\%$, whereas it was as high as $80\%$ in Hicks variety. 4. Among the 40 tobacco plants investigated, $37.5\%$ showed infection with one kind of virus whereas the remaining $62.5\%$, revealed mixed infection with more than two different viruses.

Functional Expression of Nicotiana tabacum Acetolactate Synthase Gene in Escherichia coli

  • Acetolactate synthase (ALS, EC 4.1.3.18) is the first common enzyme in the biosynthesis of leucine, isoleucine, and valine. It is the target enzyme for several classes of herbicides, including the sulfonylureas, the imidazolinones, the triazolopyrimidines, the pyrimidyl-oxy-benzoates and the pyrimidyl-thio-benzens. The sulfonylurea-resistant ALS gene (SurB) from Nicotiana tabaccum [Lee et al. (1988) The EMBO J. 7, 1241-1248] was cloned into the bacterial expression plasmid pT7-7. The resulting recombinant plasmid pT7-ALS was used to transform an ALS-deficient Escherichia coli strain MF2000. MF2000 cells transformed with pT7-ALS grew in the absence of valine and isoleucine. ALS activities of 0.042 and 0.0002 ${\mu}mol/min/mg$ protein were observed in the crude extracts prepared from MF2000 cells transformed with plasmids pT7-ALS and pT7-7, respectively. In addition, the former crude extract containing mutant ALS was insensitive to inhibition by K11570, a new chemical class of herbicides. $IC_{50}$ values for K11570 were $0.13{\pm}0.01$ mM. For comparison, a plasmid pTATX containing the wild-type Arabidopsis thaliana ALS coding sequences was also expressed in MF2000. ALS activities of 0.037 ${\mu}mol/min/mg$ protein were observed, and the wild type ALS was sensitive to two different classes of herbicides, K11570 and ALLY, a sulfonylurea. $IC_{50}$ values for K11570 and ALLY were $0.63{\pm}0.07$ and $80{\pm}5.6$ nM, respectively. Thus, the results suggest that the sulfonylurea-resistant tobacco ALS was functionally expressed in the bacteria, and that K11570 herbicides bind to the regulatoty site of ALS enzymes.

  • The effect of plant populations - 16,667, 14,285, 12,499 or 10,7l4/10a on the agronomic characteristics of an aromatic tobacco, Sohyang was investigated for two years. The growth and number of suckers produced decreased as the population increased. The leaf area index (L.A.I) was higher in thinner planting, but there was no significant difference in weight per unit leaf area between treatments. Yield was highest, 1 24 kg/ l0a, in 16,667 plantings per l0a, but quality was not different among densitites tested. Similar trends were observed in monetary value per l0a. Concentrations of total alkaloids and nitrogen were low in denser planting, but no significant difference was observed in total sugar. The results suggest that about 16,000/10a more would be optimum number of plants for Sohyang.

  • Dihaploid lines derived from the another of the $F_1$ of single cross, SC72 $\times$ Hicks (Nicotiana tabacum L.) were estimated the variation, heritablities, genetic correlations and path-co-efficients on agronomic characters and chemical compositions. Dinaploids exhibited reduced vigor and growth when comparecd with the mid-parent value but had increased nicotine contents, and the range of dihaploids characters were deviated more than that of their parents. Heritabilities of days to flowering, leaves per plant, nicotine. contents, leaf size and plant height appeared high value, and the yield, value per kg. and reducing sugar contents were low. There were positive genetic correlations between yield, and value per kg., leaf width, leaves per plant and reducing sugar contents and negative genetic correlations appeared between nicotine contents and the other characters except for index of leaf shape. Path-coefficient analysis of genotypic correlation coefficients showed that leaves per plant and leaf width lave a high direct effect, and plant height and days to flowering have a high indirect effect on yield, respectively.

Agroinfiltration-based Potato Virus X Replicons to Dissect the Requirements of Viral Infection

  • Extensive research of the Potato virus X(PVX) has been performed in in vitro transcription system using the bacteriophage T7 promoter. We constructed an efficient T-DNA based binary vector, pSNU1, and modified vectors carrying PVX replicons. The suitability of the construct to transiently express PVX RNA using Agrobacterium tumefaciens was tested by analysis of infectivity in plants. The expressed PVX RNA was infectous and systemically spread in three plant species including Nicotiana benthamiana, N. tabacum cv. Xanthi-nc, and Capsicum annuum cv. Chilsungcho. The PVX full length construct, pSPVXp31, was caused severe mosaic symptoms on N. benthamiana, severe necrotic lesions on C. annuum while milder symptoms and delayed mosaic symptoms were appeared on the systemic leaves on N. tabaccum. RT-PCR analysis confirmed the presence of PVX RNAs on both inoculated and systemic leaves in all three plant species tested. Our results indicated that PVX replicons were efficiently expressed PVX RNA in at least three tested species. Further investigation win be needed to elucidate the mechanism of PVX replication, translation, movement and assembly/disassembly processes.

Mathematical Analysis of Growth of Tobacco (Nicotiana tabaccum L.) II. A New Model for Growth Curve (담배의 생장반응에 관한 수리해석적 연구 제2보 담배생장곡선의 신모형에 관하여)

  • The experiment was conducted with three varieties (Hicks, Burley 21, and Sohyang) and cultivation type (Improved mulching, general mulching, and non mulching) of NC 2326 to model to curve of tabacco growth against time. The basic growth data were obtained by harvest method at intervals of ten days from transplanting at 7-8 times and analyzed by polynomial regression, orthogonal polynomial, and logarithmic transformation. It is shown that the C model of growth curve: T = A +$\sqrt{(1.4 AK + K)}$2K provides an excellent fit.

Characterization and Partial Nucleotide Sequence of Potato Virus X Isolated from Potato in Korea

  • Potato virus X (PVX-KO) showing mild mosaic and stunting symptoms on potato (Solanum tuberosum) in Kangwon area has been isolated and characterized. EM observation of the purified virus particles showed flexuous rod shape of about 520 nm in length. The coat protein (CP) of the virus had a molecular weight of 31 kDa in SDS-PAGE analysis, and the viral RNA was approximately 6.4 kb in size in denatured agarose gel electro-phoresis. In gel-immunodiffusion tests, it reacted strongly with an antiserum to common PVX from BIOREABAAG (USA). A rabbit antiserum was produced using purified virus and used for routine PVX detection by ELISA. Cultivated potatoes in Kangwon and other areas were frequently infected with PVX-KO. Both Datura stramonium and Nicotiana tabaccum cultivars developed necrotic local lesions 5 days after inoculation, and systemic mosaic symptoms with vein clearing 2 weeks after inoculation. All the features agree with the description of other PVX strains. To confirm and determine PVX strains, reverse transcription-polymerase chain reaction experiment was conducted using specific primers for viral CP. Amplified DNA fragments were cloned and sequenced. Results showed nucleotide sequence homologies of about 88 to 99% to other PVX strains. Based on CP amino acid sequence deduced from nucleotide sequences and host range studies PVX-KO is considered a member of the type X subgroup of PVX.

Expression and Inheritance Patterns of Gus Gene Driven by an Endosperm-Specific Promoter in Transgenic Tobacco (배유 특이 프로모터에 의해 유도된 GUS 유전자의 형질전환 담배 내에서의 발현 및 유전 양상)

  • This study was carried out to investigate the tissue-specific expression of ${\beta}$-glucuronidase (gus) gene driven by endosperm-specific promoter (Z4 promoter) in the transgenic tobacco and to find out inheritance pattern of transgene to the next generation. Tobacco (Nicotiana tabaccum cv. Havana SR1) was transformed with Agrobacterium tumerfaciens LBA4404 harboring BV3 construct containing gus gene driven by Z4 promoter and a kanamycin resistant gene. Seven hundred bp PCR products, indicating the presence of npt II gene, were found in the all eight transformants by PCR analysis using nptII primers. To study the expression pattern of the two different kind of promoters, leaf disks of the Z4pro-gus-transformed plants and 35Spro-gus-transformed plants were analyzed histochemically for gus activity. As a result, leaf disks of Z4pro-gus-transformed plants showed very weak and partial positive gus activity. In contrast, leaf disks of 35Spro-gus-transformed plants showed relatively strong positive gus activity. To investigate the expressed position of Z4 promoter, seeds from Z4pro-gus-transformed plants and 35Spro-gus-transformed plants were analyzed histochemically for gus activity. Z4pro-gus-transformed seeds showed positive gus activity restricted to the endosperm. However, the blue-colored product in 35Spro-gus-transformed seeds was observed in all the area including endosperm. Kanamycin resistance assay showed that transgenes were stably inherited to next generation in all lines.

Stress-induced biphasic ethylene and ROS biosynthesis are synergistically interacted in cell damage (스트레스에 의한 식물세포 손상에서 Biphasic Reactive Oxygen Species(ROS)와 Ethylene 생합성의 Synergism 효과)

  • Although reactive oxygen species (ROS) are inevitable by-products of many redox reactions in eukaryotic cells, they play a crucial role as signaling molecules in many cellular processes for development and defense response to abiotic stresses. The biphasic ROS production which was peaked twice in a first transient phase and a second massive phase was occurred after treatment of abiotic stress such as oxidative stress, high salinity. This biphasic generation of ROS was followed by the biphasic production of stress hormone, ethylene. The mechanism of interactions between ROS and ethylene biosynthesis is studied in tobacco (Nicotiana tabaccum L.) plants under the abiotic stresses. The stress-induced ethylene production was significantly inhibited in RbohD-AS and RbohF-AS, in which antisense expression of NADPH oxidase genes was performed. The accumulation of ROS, which was determined by DAB and DCFH-DA staining, was significantly decreased after abiotic stresses in transgenic plants. The suppression of signaling with ethylene and ROS induced more tolerance in response to abiotic stress. The transgenic plants were more tolerant in MS medium supplemented with salinity stress in contrast with wild-type. Stress-induced cell damage determined by DNA fragmentation was decreased at phase II in those transgenic plants. Therefore, the first burst of ROS is more responsible for making a role as a signaling molecule during stress-induced response. These results suggested that ethylene and ROS act in a positive feedback cycle that results in mutual enhancement of ethylene and ROS production during stress-induced cell death.
